Heat/Mass Transfer Augmentation in a Square Duct . Roughened with Angled Discrete Ribs Having Narrow Gaps (정사각 덕트 내에서 열/물질전달 촉진을 위한 경사진 단락 요철의 좁은 틈새 효과)

  • Local heat/mass transfer and friction loss in a square duct roughened with various types of continuous and discrete rib turbulators are investigated. The combined effects of the gap flows of the discrete ribs and the secondary flows are examined for the purpose of the reduction of thermally weak regions and the promotion of the uniformity of heat/mass transfer distributions as well as the ;augmentation of average heat/mass transfer. The rib-to-rib pitch to the rib height ratio (p/e) of 8 and the rib angles of 90° and 60° are selected with e/D$\_$h/=0.08. The vortical structure of the secondary flows induced by the parallel angled arrays are quite distinct from that induced by the cross angled arrays. This distinction influences on heat/mass transfer and friction loss in all the tested cases. The gap flows of the discrete ribs reduce the strength of the secondary flows but promote local turbulence and flow mixing. Consequently, the angled discrete ribs with the small gaps provide a more uniform heat/mass transfer distribution sustaining high average heat/mass transfer.

  • The purpose of this study was to development of transgenic cow using the nuclear transfer. To secrete hFSH in urea, the vector was constructed with UPII promoter. The fetal fibroblast cells (KbFF) were constructed from pregnant day 45 male fetus. The hFSH genes were cotransfected with pcDNA3 (neo) vector to KbFF cells by electroporation. (omitted)

Improved Transformation of the Filamentous Fungus Aspergillus niger Using Agrobacterium tumefaciens

  • Since it is known that Agrobacterium tumefaciens, which has long been used to transform plants, can transfer the T-DNA to yeast Saccharomyces cerevisiae during tumourigenesis, a variety of fungi were subjected to transformation to improve their transformation frequency. In this study, I report the A. tumefaciens-mediated transformation of filamentous fungus Aspergillus niger. Transfer of the binary vector pBIN9-Hg, containing the bacterial hygromycin B phosphotransferase gene under the control of the Aspergillus nidulans trpC promoter and terminator as a selectable marker, led to the selection of $50{\sim}100$ hygromycin B-resistant transformants per $1{\times}10^7$ conidia of A. niger. This efficiency is improved $10{\sim}20$ fold more than reported elsewhere. In order to avoid the difficulties in selection transformant from the over-growing non-transformant, I used top agar containing 900 ${\mu}g/ml$ of hygromycin. Genomic PCR and Southern analysis showed that all transformants contained single T-DNA insert per fungal genome. This technique offers an easier and more efficient method than that of using protoplast.

  • Dane particle was prepared from the plasma of chronic HBsAg carrier with high levels of HBsAg activity. DNA extracted front Dane particle core after a DNA polymerase reaction with $\alpha$-($^{32}$P) dNTP, was identified as HBV DNA by liquid scintillation counter and agarose gel electrophoresis-G.M. counting. To produce Hepatitis B surface antigen for use as a vaccine against Hepatitis B virus infection, yeast strains harboring recombinant plasmid with Apase promoter was used. Recombinant plasmid was construced from pHBV 130 and pAN 82, transformed into E coli, and then transferred into yeast strains. HBsAg was produced by derepression in Burkholder minimal medium with controlled inorganic phosphate concentration. The kinetics of HBsAg production was also investigated. Total HBsAg activity increased rapidly between 3 and 6 hours after transfer to phosphate-free medium and reached a maximum at around 9th hour. The transfer into phosphate-free medium after 6 hours in high phosphate cell growth medium gave maximum activity.

  • Baculovirus was originally isolated from the alfalfa looper and contains a 134-kbp genome with 154 open reading frames (ORF). The major capsid protein VP39 together with some minor proteins forms the nucleocapsid ($21nm{\times}260nm$) that encloses the DNA with p6.9 protein. They are double-stranded, circular, supercoiled DNA molecules in a rod-shaped capsid. Wild-type baculoviruses exhibit both lytic and occluded life cycles that develop independently throughout the three phases of virus replication. Recombinant baculoviruses can transfer their vectors and express their recombinant proteins in a wide range of mammalian cell types. Especially, inclusion of a dominant selectable marker in these baculoviral vectors can express diverse recombinant genes in many cells. Baculoviral vectors were reconstructed with cytomegalovirus (CMV) promoter,uroplakin II promoter, polyhedron promoter, vesicular stomatitis virus G (VSVG), enhanced green fluorescent protein (EGFP), protein transduction domain (PTD) gene and so on. These reconstructed vectors were infected into various cell and cell lines. We performed transfection and expression of these recombinant vectors comparison with other control vectors. From this study, we knew that transfection and expression of these recombinant vectors have higher efficacy than any control vector.   • Total RNAs were isolated from cultured roots of Scopolia parviflora, $poly(A)^+$ RNA was obtained through the mRNA purification, cDNA library of Hnh6h was constructed. Recombinant baculoviruses in Spodoptera frugiperda (Sf) cells were constructed by use of the transfer vector pBacPAK, which has the AcNPV sequence under the polyhedrin promoter. The expression vector carrying Hnh6h gene was transferred to S. parviflora and obtained transgenic hairy root lines. Our results confirmed the over expression of the H6H protein was used by anti-pBacPAK about cDNAs of S. parviflora. This study will served for production of tropane alkaloids by metabolic engineering.

Expression of the FLP recombinase of the 2 $\mu$m plasmid of yeast in the cultured cells of Bombyx mori using a transient expression vector (Yeast 2 $\mu$m 플라스미드 유래 FLP recombinase 유전자의 곤충 배양세포내 발현)

  • In order to express the FLP recombinase in B. mori cultured cell line, BmN-4, transient expression system using a heat shock protein gene (hsp70) promoter of Dorosophilla melnogaster was constructed. This vector was designated as pHsSV. Activity strength of the hsp70 promoter was compared with that of immediate early gene (IE-1) and polyhedrin gene of BmNPV employing the E. coli $\beta$-galactosidase gene as a reporter gene. The result showed that the pHs $\beta$-gal plasmid vector expressed the $\beta$-galactosidase at 2nd and 3rd day after the transfer of plasmid DNA into BmN-4 cells, which was similar to that of pIE1 $\beta$-gal vector, but different from that of a recombinant virus, vBm $\beta$-gal. For the construction of FLP recombinase transient expression vector, the FLP recombinase gene was cloned by polymerase chain reaction technique. To express the FLP recombinase, this gene was inserted into pHsSV plasmid vector, under the control of the hsp70 promotor, and tranfected in BmN-4 cells. The expressed FLP recombinase was estimated at 44kDa on a 12.5% SDS-PAGE.

A Numerical Study on Performance of Air-to-Air Plastic Plate Heat Exchanger

  • The purpose of this research is to develop high efficiency plastic plate heat exchangers which can be substituted for conventional aluminum plate heat exchangers. Four simulation models of plastic plate heat exchangers are designed and simulated: that is, flat plate type, turbulent promoter type, corrugate type and dimple type heat exchanger. The flat plate type is designed as the reference model in order to evaluate how much thermal performance increases. The turbulent promoter type is fabricated with cylindrical-type vortex generators and rib-type turbulent promoters. The corrugate type is obtained from the conventional stainless steel compact heat exchangers, which are called the herringbone-type compact heat exchangers. The dimple type has a number of dimples on its surface. In this study, the flow and heat transfer characteristics of the plastic plate heat exchanger are investigated using numerical simulation and compared with experimental results. Numerical simulation is carried out using the FLUENT code. The flows are assumed as a three-dimensional, incompressible and turbulent model. The computational analysis and experimental results both show that the friction coefficient and Nu number is highest in the corrugate type. The tendency of numerical simulation results is in good agreement with that of the experimental results.
