In the study, we endeavored to assess the convergence effect of Silybum marianum-derived silymarin and epidemiologically-correlated alcohol intake on vascular contractility and to determine the mechanism involved. There were few reports addressing the question whether thin or thick filament modulation is included in ethanol and silymarin-induced regulation. We hypothesized that ethanol at a low concentration and silymarin play a role in agonist-dependent regulation of vascular contractility. Denuded arterial muscles of Sprague-Dawley male rats were suspended in organ baths and isometric tensions were transduced and recorded using isometric transducers and an automatic data acquisition system. Interestingly, both silymarin and ethanol didn't encourage silymarin alone-induced inhibition in agonists-induced contraction suggesting that endothelial nitric oxide synthesis might be involved in ethanol or silymarin-induced modulation of vascular contractility and additional pathways besides endothelial nitric oxide synthesis such as ROCK inactivation might be involved in the silymarin-induced modulation of vascular contractility.

In this study, the optimal driving frequency was derived through finite element analysis (FEA) to optimize the developed piezoelectric ultrasonic medical devices(PUMD) for bone surgery. The core of the PUMD is the piezoelectric ceramic (PZT), which is a vibrator that generates vibration energy. The piezoelectric ceramic shows the maximum current value with respect to the input voltage at the resonance frequency, which generates the maximum mechanical vibration. In the past, various studies have been conducted related to the analysis of PUMD, but most of the research so far has been limited to free vibration analysis. However, in order to derive the accurate resonant frequency, the initial stress generated by bolt tightening in the bolt-clamped Langevin type transducer (BLT) must be considered. In this study, after designing a PUMD, the driving performance according to the bolt tightening value was analyzed through FEA, and this was experimentally verified. First, the resonance mode and frequency response were confirmed through modal and harmonic analysis at 20-40 kHz, which is known as the optimal driving frequency band of PUMD for bone surgery. In addition, the design of the PUMD was confirmed by checking the mechanical behavior of the tip and the piezoelectric ceramic at the resonant frequency. Consequentially, the characteristic evaluation was performed, and it was confirmed that the resonant frequency result derived through the FEA was reasonable. Through this study, we presented a more rational FEA method than before for BLT transducers. We expect that this will shorten the time and cost of developing a PUMD, and will enable the development of more stable and high-quality products.

Clinical ultrasound (US) is a widely used imaging modality with various clinical applications. However, capturing a large field of view often requires specialized transducers which have limitations for specific clinical scenarios. Panoramic imaging offers an alternative approach by sequentially aligning image sections acquired from freehand sweeps using a standard transducer. To reconstruct a 3D volume from these 2D sections, an external device can be employed to track the transducer's motion accurately. However, the presence of optical or electrical interferences in a clinical setting often leads to incorrect measurements from such sensors. In this paper, we propose a deep learning (DL) framework that enables the prediction of scan trajectories using only US data, eliminating the need for an external tracking device. Our approach incorporates diverse data types, including correlation volume, optical flow, B-mode images, and rawer data (IQ data). We develop a DL network capable of effectively handling these data types and introduce an attention technique to emphasize crucial local areas for precise trajectory prediction. Through extensive experimentation, we demonstrate the superiority of our proposed method over other DL-based approaches in terms of long trajectory prediction performance. Our findings highlight the potential of employing DL techniques for trajectory estimation in clinical ultrasound, offering a promising alternative for panoramic imaging.
An automated ultrasonic flaw detection system was developed for thin-walled and short tubes such as Zircaloy-4 tubes used for cladding heavy-water reactor fuel. The system was based on the two channels immersion pulse-echo technique using 14 MHz shear wave and the specially developed helical scanning technique, in which the tube to be tested is only rotated and the small water tank with spherical focus ultrasonic transducers is translated along the tube length. The optimum angle of incidence of ultrasonic beam was 26 degrees, at which the inside and outside surface defects with the same size and direction could be detected with the same sensitivity. The maximum permissible defects in the Zircaloy-4 tubes, i.e., the longitudinal and circumferential v notches with the length of 0.76mm and 0.38mm, respectively and the depth of 0.04 mm on the inside and outside surface, could be easily detected by the system with the inspection speed of about 1 m/min and the very excellent reproducibility. The ratio of signal to noise was greater than 20 dB for the longitudinal defects and 12 dB for the circumferential defects.
The shear wave velocity is related with the stiffness of granular skeleton and mass density. The shear stiffness of the granular skeleton remains unaffected by the presence of the fluid. Bender elements are convenient shear wave transducers for instrumenting soil cells due to optimal soil-transducer coupling. This study addresses the principles of the shear wave, the design and implementation of bender elements including electromagnetic coupling prevention, directivity, resonant frequency, detection of first arrival, and near field effects. It is shown that electromagnetic coupling effects can be minimized using parallel-type bender elements. Thus, the in-plane S-wave directivity is quasi-circular. The resonant frequency of bender element installations depends on the geometry of the bender element, the anchor efficiency and the soil stiffness. One of the most cumbersome parts in the bender element test is near field effects, which affect the selection of arrival time. The selection of the first arrival within the near field Is effectively solved by the multiple reflection technique and signal matching technique. Bender elements, which requires several considerations, may be effective tools for the subsurface characterization by using S-wave.
Rock bream iridovirus (RBIV) is a megalocytivirus widely infected in various fish species in Korea, causing symptoms of acute inflammation and enlargement of spleen. In our previous study, RBIV induced the initial upregulation but later down-regulation of proinflammatory cytokines and IFN1 gene expression. Signal transducers and activators of transcriptions (STAT) are transcription factors involved in the regulation of immune genes including IFNs. This study was conducted to analyse the expression of STAT2. The expressional study of STAT2 gene was performed in head kidney and spleen upon RBIV infection and immune stimulants like LPS or poly I:C in vitro. Consequently, STAT2 gene expression pattern was different in head kidney and spleen as it was significantly up-regulated by LPS from 4 h to 8 h but down-regulated at 24 h while up-regulated by poly I:C at 8 h in head kidney while, in spleen, STAT2 gene expression was down regulated by LPS but significantly up-regulated by poly I:C. Upon RBIV stimulation, STAT2 gene expression was significantly down-regulated by high dose RBIV at 4 h but up-regulated at 8 h and 24 h in head kidney. In spleen cells, it was up-regulated by medium dose RBIV at 4 h and by high dose RBIV at 4 h and 8 h but down regulated later then. In vivo, STAT2 gene expression was not significantly affected by RBIV infection while significant up-regulated by vaccination at day 7 post-vaccination, indicating STAT2 gene can be involved in adaptive immune response in rock bream.

Obesity-induced inflammation is crucial in the pathogenesis of insulin resistance and type 2 diabetes. In this study, we investigated the effects of the Gracilaria chorda (GC) on lipid accumulation and obesity-induced inflammatory changes or glucose homeostasis in cell models (3T3-L1 adipocytes and RAW 264.7 macrophages). Samples of GC were extracted using solvents (water, methanol, and ethanol) and subcritical water (SW) at different temperatures (90, 150, and 210℃). The total phenolic content of GCSW extract at 210℃ (GCSW210) showed the highest content compared to others, and GCSW210 highly inhibited lipid accumulation and significantly reduced gene expressions of peroxisome proliferator-activated receptor-γ, CCAAT/enhancer-binding protein-α, sterol regulatory element-binding protein-1c, and fatty acid synthase in 3T3-L1 adipocytes. In addition, GCSW210 effectively downregulated the pro-inflammatory cytokine regulator pathways in RAW 264.7 macrophages, including mitogen-activated protein kinase, signal transducers and activators of transcription and nuclear factor-κB. In co-culture of 3T3-L1 adipocytes and RAW 264.7 macrophages, GCSW210 significantly reduced nitric oxide production and interleukin-6 levels, and improved glucose uptake with dose-dependent manner. These findings suggest that GCSW210 improves glucose metabolism by attenuating obesity-induced inflammation in adipocytes, which may be used as a possible treatment option for managing obesity and associated metabolic disorders.

If the velocities of longitudinal, transverse and leaky surface acoustic waves in an isotropic material are given, the elastic constants and density of the material can be deduced analytically. Those velocities have been measured using three ultrasonic transducers with different vibrational modes so far. In this paper a line-focusing PVDF transducer with divided electrodes was newly proposed and designed for measuring approximate velocities of the three waves. The measurement method established in this study for each waves using the transducer was applied to several isotropic materials including fused quartz. The elastic stiffness constants and densities of the materials were calculated using the measured velocities, and the accuracies were discussed. It was shown that the obtained results are in good accord with the reference values.
In the present study, polyacetal bone mimics with circular cylindrical pores were used to investigate variations of speed of sound and attenuation coefficient with porosity and microarchitecture in bone. The speed of sound and attenuation coefficient of the 6 bone mimics with porosities from 0 % to 65.9 % were measured by a through-transmission method in water, using a pair of broadband, unfocused transducers with a diameter of 12.7 mm and a center frequency of 1.0 MHz. Independently of the structural properties of the bone mimics, the speed of sound decreased almost linearly with the increasing porosity. The attenuation coefficient measured at 1.0 MHz exhibited linear or nonlinear correlations with the porosity, depending on the structural properties of the bone mimics. These results are consistent with those previously published by other researchers using bone samples and mimics, and advances our understanding of the relationships of the ultrasonic parameters for the diagnosis of osteoporosis with the bone density and microarchitecture in human bones.
Biased signaling or functional selectivity refers to the ability of an agonist or receptor to selectively activate a subset of transducers such as G protein and arrestin in the case of G protein-coupled receptors (GPCRs). Although signaling through arrestin has been reported from various GPCRs, only a few studies have examined side-by-side how it differs from signaling via G protein. In this study, two signaling pathways were compared using dopamine D2 receptor (D2R) mutants engineered via the evolutionary tracer method to selectively transduce signals through G protein or arrestin (D2G and D2Arr, respectively). D2G mediated the inhibition of cAMP production and ERK activation in the cytoplasm. D2Arr, in contrast, mediated receptor endocytosis accompanied by arrestin ubiquitination and ERK activation in the nucleus as well as in the cytoplasm. D2Arr-mediated ERK activation occurred in a manner dependent on arrestin3 but not arrestin2, accompanied by the nuclear translocation of arrestin3 via importin1. D2R-mediated ERK activation, which occurred in both the cytosol and nucleus, was limited to the cytosol when cellular arrestin3 was depleted. This finding supports the results obtained with D2Arr and D2G. Taken together, these observations indicate that biased signal transduction pathways activate distinct downstream mechanisms and that the subcellular regions in which they occur could be different when the same effectors are involved. These findings broaden our understanding on the relation between biased receptors and the corresponding downstream signaling, which is critical for elucidating the functional roles of biased pathways.
