Dietary supplementation of Eucommia leaf extract to growing-finishing pigs alters muscle metabolism and improves meat quality

  • Objective: The objective of this study was to investigate the influence of dietary supplementation of Eucommia ulmoides leaf extract (ELE) on muscle metabolism and meat quality of pigs with and without pre-slaughter transportation. Methods: In a 43-day feeding experiment, a total of 160 pigs with an initial body weight 60.00±2.00 kg were randomly assigned into four groups in a completely randomized design with 10 replicates. Pigs in groups A and C were fed a basal diet and pigs in groups B and D were fed a basal diet supplemented with 0.5% ELE. Pigs were slaughtered with (group B and D) or without (group A and C) pre-slaughter transport. Muscle chemical composition, postmortem glycolysis, meat quality and muscle metabolome were analyzed. Results: Dietary ELE supplementation had no effect on the proximate composition of porcine muscle, but increased free phenylalanine, proline, citruline, norvaline, and the total free amino acids in muscle. In addition, dietary ELE increased decanoic acid and eicosapentaenoic acid, but decreased heptadecanoic acid, oleic acid, trans-oleic acid, and monounsaturated fatty acids in muscle. Meat quality measurement demonstrated that ELE improved meat water holding capacity and eliminated the negative effects of pre-slaughter transport on meat cooking yield and tenderness. Dietary ELE reduced muscle glycolytic potential, inhibited glycolysis and muscle pH decline in the postmortem conversion of muscle to meat and increased the activity of citrate synthase in muscle. Metabolomics analysis by liquid chromatographic tandem mass spectrometric showed that ELE enhanced muscle energy level, regulated AMP-activated protein kinase (AMPK) signaling, modulated glycogenolysis/glycolysis, and altered the metabolism of carbohydrate, fatty acids, ketone bodies, amino acids, purine, and pyrimidine. Conclusion: Dietary ELE improved meat quality and alleviated the negative effect of pre-slaughter transport on meat quality by enhancing muscle oxidative metabolism capacity and inhibiting glycolysis in postmortem muscle, which is probably involved its regulation of AMPK.

야생 망초(Conyza canadensis L) 추출물의 영양성분 분석 (Nutritional Analysis of Wild Conyza canadensis L. Extract)

  • 배경: 야생 망초에는 다양한 물질들이 존재하는 것으로 일부가 보고되었으나 전체적인 성분분석은 아직 미흡하다고 생각한다. 목적: 본 연구에서는 야생 망초에서 항산화 물질 및 다양한 영양성분에 대하여 추출조건을 설정하고 검색하는 것이 목적이었다. 방법: 망초 추출물을 다양하게 추출하여 다양한 분석기기를 활용하여 성분분석의 조건을 제시하고 분석하였다. 결과: 망초 추출물의 일반영양성분에서는 탄수화물이 46.4%, 조단백질 26.5%, 수분 16.3%, 조회분 9.5%, 조지방 1.2% 순이었다. 식용섬유 함량은 30.76%이었다. 유리당은 fructose, glucose, sucrose, maltose가 분리되었고, fructose의 함량이 4,144.2 mg/100 g으로 높았다. 무기성분 분석에서는 K가 64.87 mg/g, P가 4.09 mg/g, Mg이 1.77 mg/g, Na이 나트륨이 0.16 mg/g이 확인되었다. 지방산은 모두 20종이 확인되었으며, 불포화지방산은 octadecenoic acid, octadecadienoic acid, linolenic acid 가 함유하였고, linolenic acid가 54.1%로 높았다. 포화지방산 함량은 0.39 g/100g 이었고, 트랜스지방산 함량은 0.01 g/100g이었다. 망초에서는 콜레스테롤은 확인하지 못하였다. riboflavin이 0.4 mg/100g, 필수 아미노산 6종과 24종의 아미노산을 확인하였다. 결론: 망초 추출물에는 다양한 영양성분과 기능성 성분이 존재하여 이를 이용한 자연치유적 활용성이 기대 된다.

개불의 지질에 관한 연구 -지질, 지방산 및 Sterol 조성에 관하여- (Studies on Lipids of Urechis unicintus -on the Composition of Lipids, Fatty acid and Sterol-)

  • 개불지질의 성상을 밝힐 목적으로 그 지방산조성 및 sterols 조성에 대하여 분석 검토 하였는데, 그 결과는 다음과 같다. (1) 조지질과 부검화물함량은 각각 $1.9\%,\;14.3\%$였으며, 요오드값은 111이었다. (2) 조지질의 조성은 인지질이 $39.8\%$, 유리 sterols이 $27.7\%$, triglyceride가 $21.4\%$였다. (3) 지방산조성은 $C_{20:4}$$19.4\%$로 가장 많았고, 다음으로 $C_{16:0},\;C_{20:1},\;C_{18:1}$이 각기 $13.7\%,\;11.3\%,\;10.4\%$였으며 또 polyene산 monene 산 및 포화산으로 계산하면 각각 $39.7\%,\;31.9\%$$26.6\%$였다. (4) 5종의 sterols이 검출되었는데 cholesterol이 $57.6\%$로 가장 많았고 다음이 brassicasterol이 $20.3\%$, 24-methylene cholesterol of $17.7\%$, 22-dehydro cholesterol of $3.0\%$, 22-trans-24-norcholesta-5,22-dien-3-${\beta}$-ol 이 $1.4\%$였다.

Crystal Structure of Acyl-CoA Oxidase 3 from Yarrowia lipolytica with Specificity for Short-Chain Acyl-CoA

  • Acyl-CoA oxidases (ACOXs) play important roles in lipid metabolism, including peroxisomal fatty acid ${\beta}$-oxidation by the conversion of acyl-CoAs to 2-trans-enoyl-CoAs. The yeast Yarrowia lipolytica can utilize fatty acids as a carbon source and thus has extensive biotechnological applications. The crystal structure of ACOX3 from Y. lipolytica (YlACOX3) was determined at a resolution of $2.5{\AA}$. It contained two molecules per asymmetric unit, and the monomeric structure was folded into four domains; $N{\alpha}$, $N{\beta}$, $C{\alpha}1$, and $C{\alpha}2$ domains. The cofactor flavin adenine dinucleotide was bound in the dimer interface. The substrate-binding pocket was located near the cofactor, and formed at the interface between the $N{\alpha}$, $N{\beta}$, and $C{\alpha}1$ domains. Comparisons with other ACOX structures provided structural insights into how YlACOX has a substrate preference for short-chain acyl-CoA. In addition, the structure of YlACOX3 was compared with those of medium- and long-chain ACOXs, and the structural basis for their differences in substrate specificity was discussed.

Supplementation Effects of $C_{18:2}$ or $C_{18:3}$ Rich-oils on Formations of CLA and TVA, and Lipogenesis in Adipose Tissues of Sheep

  • The present study was conducted to investigate the supplementation effects of $C_{18:2}$ rich-soybean oil or $C_{18:3}$ rich-perilla oil (7% of total diet, DM basis) for 12 weeks on plasma metabolites, fatty acid profile, in vitro lipogenesis, and activities of LPL and FAS in adipose tissue of sheep. The treatments were basal diet (Control), $C_{18:2}$ rich-soybean oil supplemented diet (SO-D) and $C_{18:3}$ rich-perilla oil supplemented diet (PO-D). All the sheep were fed the diets consisting of roughage to concentrate in the ratio of 40:60 (DM basis). Oil supplemented diets (SO-D and PO-D) slightly increased contents of triglyceride (TG) and total cholesterol (TC), proportions of both cis-9 trans-11 and trans-10 cis-12 CLA and TVA, but lowered (p<0.01) those of $C_{18:0}$ compared to the control diet. No differences were observed in the contents of TG and TC and proportions of fatty acids in plasma between supplemented oils. Oil supplemented diets slightly increased the proportions of cis-9 trans-11 and trans-10 cis-12 types of CLA in subcutaneous adipose tissue of sheep compared to the control diet. The rate of lipogenesis with acetate was higher (p<0.01) for intermuscular- and subcutaneous adipose tissues than that for intramuscular adipose tissue, while that with glucose did not differ among fat locations in sheep fed SO-D. No differences were observed in the rate of lipogenesis between substrates in all fat locations. The rates of lipogenesis with glucose increased only in the intermuscular- (p<0.01) and subcutaneous adipose tissue (p<0.005) compared to those with acetate. The rates of lipogenesis with acetate were the highest in the intermuscular and intramuscular adipose tissue of the sheep fed PO-D. Oil supplemented diets slightly increased the rate of lipogenesis with glucose for all fat locations. Supplementation of oils to the diet numerically increased the fatty acid synthase activity but did not affect the lipoprotein lipase activity in subcutaneous adipose tissue.

대전, 충천 지역 제빵류의 조지방 및 트랜스 지방산 함량 조사 (Analysis of Trans Fatty Acid and Crude Fat Contents of Bakery Foods in Chung-cheong Province)

  • 대전, 충청(청주, 천안)지역의 제과점에서 수거한 제빵류 42개의 시료에서 조지방을 추출하고 이것을 Gas Chromatography(GC)를 이용하여 트랜스 지방의 함량을 분석하였다. 수거한 시료는 생크림 케이크 9개, 페이스츄리 9개, 슈크림 7개, 크로켓 9개, 찹쌀 도넛 8개였다. 추출한 시료의 조지방 함량은 페이스츄리 10.67$\sim$34.26%로 가장 높은 조지방 함량을 보였으며, 찹쌀 도넛은 1.44$\sim$4.34%로 가장 낮은 조지방 함량을 보였다. 트랜스 지방의 함량은 생크림 케이크 0.03$\sim$1.31 g/100 g of food, 페이스츄리가 0.21$\sim$2.69 g/100 g of food, 슈크림 0.09$\sim$0.43 g/100 g of food, 크로켓 0.22$\sim$1.99g/100 g of food, 찹쌀 도넛 0.02$\sim$0.38 g/100 g of food로 페이스츄리가 시료들 중에서 가장 높은 함량을 나타내었으며, 분석한 시료들 중에서 트랜스지방의 함량이 2 g/100g of food를 초과하는 시료가 모두 페이스츄리에서 검출되었다.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

  • A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

바이오디젤 생산을 위한 어유의 에스테르화 및 전이에스테르화 반응 (Esterification and Trans-esterification Reaction of Fish Oil for Bio-diesel Production)

  • 본 연구에서는 유리지방산을 4% 포함한 어유로부터 바이오디젤을 제조하기 위해 산촉매를 이용한 에스테르화 반응과 염기촉매를 이용한 전이에스테르화 반응을 수행하였다. 실험에 사용된 어유는 GS바이오사(社)로부터 공급받은 베트남산 메기(catfish)에서 추출된 오일을 사용하였다. 에스테르화 반응에 대하여 불균질계 고체 산촉매로 Amberlyst-15와 Amberlyst BD-20을 이용하였으며 균질계 산촉매로 황산을 사용하였다. 에스테르화 반응에 의한 유리지방산 제거율이 가장 높은 촉매는 황산으로 나타났으며 반응시간도 가장 짧게 나타났다. 3종의 염기촉매 KOH, $NaOCH_3$, NaOH를 이용하여 어유의 전이에스테르화 반응 특성을 조사한 결과 KOH 촉매가 가장 적합한 것으로 나타났다. $NaOCH_3$와 NaOH 촉매의 경우 전이에스테르화 반응시 글리세롤과 바이오디젤이 일정한 조건에서 고형화 현상이 관찰되었으며 비누화 반응이 진행된 것으로 판단된다. KOH 촉매를 이용하여 초기 원료 산가와 메탄올 투입량이 전이에스테르화 반응에 미치는 영향을 조사한 결과 초기 원료오일의 산가는 낮을수록 좋았으며 메탄올과 오일의 몰비는 9:1이 적합한 것으로 도출되었다.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.

식용(食用) 대두유(大豆油) 경화시(硬化時) 반응조건(反應條件)이 이성체(異性體) 생성(生成)에 미치는 영향(影響) (The Effects of Various Reaction Conditions on Trans Isomer Formation in Hydrogenating Edible Soybean Oil)

  • 국내에서 생산된 정제 대두유의 열, 산화 안정성을 향상시키고 풍미, 물성의 개량과 이용도를 높이기 위하여 수소첨가에 의한 식용경화 대두유 제조시 반응 조건을 달리하여 주요 지방산 조성, 옥소가 및 trans 이성체의 생성에 미치는 영향을 조사하였다. $180^{\circ}C$의 반응조건에서 trans 산의 생성은 압력 $3.0{\;}kg/cm^{2}H_{2}$에서 $0.5{\;}kg/cm^{2}H_{2}$에서 보다 6.2배 증가하였고 linolenic acid는 감소하였다. 그러나 경화온도 $200^{\circ}C$의 경우 압력 $0.5{\;}kg/cm^{2}H_{2}$에서 $3.0{\;}kg/cm^{2}H_{2}$보다 trans 이성체 생성이 4.6%가 많았으며, linolenic acid, linoleic acid함량은 각각 0.51%, 2.5%가 감소하였다. 경화온도 $180^{\circ}C$, 압력 $3.0{\;}kg/cm^{2}H_{2}$와 sulphur 4 ppm을 첨가한 조건에서 옥소가와 linolenic acid가 감소하여 유리하였으나, trans 이성체의 생성이 가장 많아 불량하였다. 그러나 경화온도 $200^{\circ}C$, 압력 $0.5kg/cm^{2}H_{2}$$3.0kg/cm^{2}H_{2}$의 조건하에서 trans 이성체의 생성이 비교적 적게 생성되었고, 옥소가와 linolenic acid도 감소되어 최적 조건이라 생각된다.
