High Cell Density Culture of Alcaligenes eutrophus and Poly-$\beta$-hydroxybutyrate Production by Optimization of Medium Compositions (배지조성 최적화를 통한 Alcaligenes eutrophus의 고농동 세포배양 및 Poly$\beta$-hydroxybutyrate 생산)

  • The medium compositions of Alcaligenes eutrophus were optimized for increasing PHB productivity. It is very important to optimize the concentrations of inorganic salts and trace eleme- nts as well as carbon and nitrogen sources to maximize cell growth rate and productivity. The fed-batch culture of Alcaligenes eutrophus by dual feeding of ammonia water and glucose under optimized initial medium concentrations was carried out. Glucose was fed manually according to glucose consumption rate and ammonia water by pH-stat. The final cell concentrations and PHB content in 30 hours were 122 g/l and 65% of dry cell weight(yielding 79 g of PHB/l), respectively and 2.64 g/l/hr of PHB production rate was obtained.

Selection of Medium Components by Plackett-Burman Design for Cell Growth of a Newly Isolated Methylobacterium sp. WJ4

  • We isolated a novel methylotrophic bacterium from soil collected in Dongducheon Environment Affairs Agency. The isolate was identified as Methylobacterium sp. WJ4 based on phylogenetic analysis. Plackett-Burman design was employed for screening eight parameters of nitrate mineral salts (NMS) medium for cell growth of a newly isolated Methylobacterium sp. WJ4 with experimental validation. Trace element solution and vitamin stock were found to affect cell growth, which can be further optimized for increased cell growth. This is the first report of screening parameters of NMS medium which affect cell growth of strain belonging to the genus Methylobacterium using Plackett-Burman design.

  • The quantitative analysis of various trace metals including fission products in lithium molten salts has been performed using a inductively coupled plasma atomic emission spectrometer (ICP-AES). The spectral interferences of lithium content, 500, 1,000 and 2,000 mg/L, in the sample solution were investigated using an optimum wavelength for the respective metal species. As a result, the line intensities for Y, Nd, Sr, and La had no influences from the lithium content up to 2,000 mg/L, while Mo, Ba, Ru, Pd, Rh, Zr and Ce showed spectral interferences of 10% to 50%. The group separation of metals from lithium in the molten salts solution was carried out by adding ammonia water into the solution. The recovery of Ru, Y, Rh, Zr, Nd, Ce, La and Eu was found to be over 90%, while Mo, Ba, Pd, and Sr provided low recovery percentages.

  • These investigations were conducted to standardize several chemical and physical environments affecting in vitro propagation of gametophytes of Climacium japonicum. Propagation of this moss species was established on basal medium containing Knop macro salts and Nitsch and Nitsch trace elements. Primary cultures were initiated from apical shoots of gametophytes. Gametophyte production was accessed using chopped gametophytes, apical shoots and basal shoots. Seven ty pes of culture media and four concentrations of total nitrogen and five strengths of sucrose were tested for in vitro gametophyte production. Light and temperature factors were also evaluated. Apical shoots were the greatest among three types explants used for gametophyte propagation. Medium containing Knop macro salts and Nitsch and Nitsch trace elements was more effective than other types of media. Higher sucrose concentrations showed a positive effect on the elongation and multiplication of gametophytes. Both nitrogen deficiency and excessiveness inhibited gametophyte growth. Light intensity variation showed highly significant changes in numbers, length and fresh weight of gametophytes. Optimum light intensity for gametophyte growth seemed to be around 3000-4000 lx. Both lower and higher temperature had a negative effect on gametophyte propagation and production. This study will provide large scale and high quality propagules, and effective moss propagation system.

Heavy Metal Accumulation in Halophyte Salicornia europaea and Salt Marsh in West-coast of Korea (서해안 염생식물 퉁퉁마디와 염습지의 중금속 축적)

  • Harvesting of marshfire glasswort (Salicornia europaea) by local people has been increased recently since this plant was known for a well-being food. Even though some harvesting sites are facing high risk of environmental pollution, Salicornia europaea is still harvested on a large scale. Therefore, to investigate safety of Salicornia europaea as a food resource, salt marsh environment, potential harvestable biomass, element and heavy metal accumulations in Salicornia europaea has been studied in three salt marshes, west-coast of Korea. Salicornia europaea showed 150 - 230$g/m^2/yr$ harvestable biomass. Biomass and nitrogen contents of Salicornia europaea were closely related to soil nitrogen and carbon concentrations. Average Na, Zn, Fe, Cr accumulations in Salicornia europaea were 41479, 18, 297, 1.5 (mg/kg), indicating valuable trace element contents. However, average heavy metal accumulations such as As, Cd, Cu, Pb, Hg accumulations were 1.5, 7.1, 4.2, 1.5, 0.1 (mg/kg), which were even or higher than national standards (0.5, 0.5, 2, 2, 0.1) of salts. These results imply that harvesting and eating of Salicornia europaea in west-coast research sites would be harmful because the contents would be much higher if it is calculated as only considering salts and minerals. Therefore, harvesting of Salicornia europaea from some salt marsh in west-coast of Korea should be done cautiously.

Optimization of Endoglucanase Production from Fomitopsis pinicola Mycelia (Fomitopsis pinicola 균사체로부터 Endoglucanase의 최적생산)

  • The culture conditions to maximize the production of endoglucanase (EC 3.2.1.4) from the brown rot fungus Fomitopsis pinicola MKACC 54347 mycelia were investigated. Among the tested media for endoglucanase production, Mandel's mineral salts medium (MSM; 1% cellulose, 0.1% peptone, 0.14% $(NH_4)_2SO_4$, 0.03% urea, 0.2% $KH_2PO_4$, 0.03% $MgSO_4{\cdot}7H_2O$, 0.03% $CaCl_2$, and 0.1% trace metal solution (19.8 mM $FeSO_4$, 13.0 mM $MnSO_4$, 12.2 mM $ZnSO_4$, and 15.4 mM $CoCl_2$)) produced the highest activity of the enzyme. To optimize the medium composition for enzyme activity, the effects of various carbon, nitrogen, phosphorus, and inorganic sources were investigated in MSM. Maximal enzyme production was accomplished using a medium containing 2% carboxymethyl cellulose (CMC), 2% yeast extract, 0.2% $KH_2PO_4$, 0.03% $MnSO_4$, and 0.3% trace metal solution. Different physiological conditions, like incubation period and temperature, were also examined to assess their influence on enzyme production. Enzyme production from F. pinicola reached its highest level after cultivation for 8 days at $25^{\circ}C$. Nondenaturing polyacrylamide gel electrophoresis (PAGE), followed by the endoglucanase activity staining using CMC as the substrate, was performed to identify the endoglucanase under the culture conditions studied. Zymogram analysis of the culture supernatant revealed an endoglucanase band with a molecular mass of 52 kDa. The optimum pH and temperature for enzyme activity were $55^{\circ}C$ and pH 5.0, respectively.

  • Carboxymethylated polyamine-polyurea resin loaded with rubeanic acid (RCCPPI resin) was obtained by 1 step chemical reaction between chlorocarboxymethylated polyamine-polyurea(CCPPI) resin as matrix polymer and rebeanic acid. This resin was confirmed with infrared spectrometry, elemental analysis, and thermal analysis(DSC). The adsorption characteristics of the heavy metal's on the resin were studied by measuring distribution coefficient($K_d$) with changing pH of the solutions and frontal chromatography. The enrichment, recovery, and analysis of trace heavy metals, such as cadmium, cerium, copper, nikel, lead, and zinc, in the presence of high concentrations of sodium, calcium, and acetate salts was possible quantitatively by a column packed with the resin at each optimum pH. Preconcentration factors were more than 25. To elute the adsorbed heavy metals on the resin, 0.025M EDTA solution(pH 9.0) was used.

  • The measurement of trace amounts of uranium(VI) and thorium(VI) in the solutions containing high concentration of dissolved salts was carried out. The procedure using reversed phase liquid chromatography with trace enrichment techniques has been developed to cope with the high salt content of the samples. 2 ml of sample were passed through a small C_{18}$ reversed phase enrichment column with ${\alpha}$-HIBA eluent (0.11 M, pH 5.5) where the uranium and thorium were separated from other constituents and concentrated. The uranium and thorium were then backflushed from the column onto a deactivated C_{18}$ reversed phase analytical column where furthe separation was achieved with a mixed eluent (pH3.0, 0.17M ${\alpha}$-HIBA/0.0038 M 1-pentanesulfonate). The separated species were determined spectrophotometrically by postcolumn reaction with Arsenazo III, the chromogenic reagent. Detection limits were found within 1 ppb range for both species.

  • In this study, solid-phase microextraction (SPME) technique using GC/FID was studied as a possible alternative to liquid-liquid extraction for the analysis of EGBE, DGBE, DBM and TGME in water, and an optimization condition of trace analysis of EGBE, DGBE, DBM and TGME using the factorial design was described. Experiments used a fractional factorial design method followed by central composite design allowing optimization of a number of factors as well as statical analysis of results. The response surface analysis showed that the extraction efficiency can be described by a second-order polynomial equation in which the salts concentration, extraction temperature, extraction time and sonication time are the major influences. Using DOE, a new data-dependent method was developed that improved the quantity of confidently analyzed EGBE, DGBE, DBM and TGME in water samples.

  • An effecient gas chromatographic method is described to be used routinely for the rapid profiling and identification of biochemically important organic acids. It involves the solid-phase extraction of organic acids from aqueous samples using Chromosorb P as the solid sorbent and diethyl ether as the eluting solvent, with subsequent triethylamine treatment. The resulting triethylammonium salts of acids were directly converted to volatile tert-butyldimethylsiyl derivatives, which were simultaneously analyzed by two capillary columns of different polarity, DB-5 and DB-1701, under the identical temperature programming condition. The retention index (RI) and area ratio (AR) values of each peak measured on DB-5 and DB-1701 enabled rapid identification of acids by computer RI library search and AR comparison. The application of the present method to the organic acid profiling of various complex samples is demonstrated.
