This study was conducted to investigate the safety and efficacy of an oriental herbal composition, Kamihonghwatang(KH-19), for the reduction of the side effects of chemotherapeutic drug. KH-19 prevented the reduction of white blood cells including lymphocytes, monocytes and eosinophiles in C57BL/6 mice injected with fluorouracil, a commonly used anticancer drug. KH-19 also prevented the reduction of cell densities in bone marrow and spleen of fluorouracil-injected mice. To evaluate the safety of KH-19, single-dose toxicity test was conducted using SD rats. No dead animal was found and the minimum lethal dose of KH-19 was more than 5000 mg/kg.
In this research, we tried to develop the application method to water management and treatment using toxicity test method. When we measure the toxicity of environmental samples, we have to decide whether we take some countermeasures to reduce the toxicity or not. The first issue is how to set these action levels in each bioassays. A new idea was attempted to authorize indirect approach of each bioassays through the response characteristics against mixture of chemicals in water quality standard. The significant response in the cell-growth-inhibition bioassay was detected for standards-mixture(STDs). For acute toxicity assay, STDs-based implicit correlation between risks to humans and bioassay data showed a rational approach to set action levels in practical management. A simple model was proposed to describe and predict the changes in the total toxicity based on the concentrations of toxic-controlling chemicals during the ozonation of landfill leachates. On the basis of this simple model, toxicity reduction was predicted for pre-aggregation treatment before ozonation and ozone concentration during the ozonation. The method proposed in this study would be useful in optimizing water treatment processes and their running conditions in terms of the toxicity reduction efficacy.
Botulinum toxins are neurotoxic modular proteins composed of a heavy chain and a light chain connected by a disulfide bond and are produced by Clostridium botulinum. Although lethally toxic, botulinum toxin in low doses is clinically effective in numerous medical conditions, including muscle spasticity, strabismus, hyperactive urinary bladder, excessive sweating, and migraine. Globally, several companies are now producing products containing botulinum toxin for medical and cosmetic purposes, including the reduction of facial wrinkles. To test the efficacy and toxicity of botulinum toxin, animal tests have been solely and widely used, resulting in the inevitable sacrifice of hundreds of animals. Hence, alternative methods are urgently required to replace animals in botulinum toxin testing. Here, the various alternative methods developed to test the toxicity and efficacy of botulinum toxins have been briefly reviewed and future perspectives have been detailed.

A recent development in cosmetics has been the pursuit of wrinkling in the skin. The cosmetics composed of anti-wrinkle agent stand out from the point of view of environmental contamination and pollution. Among them, Indole-3-acetic acid (IAA), studied with wrinkling pigmentation and swelling conditions in the area of the eye, showed clinically significant reduction in depth of lines during one month trial using skin treatment. But, IAA has shown some problems when used in cosmetic formulations, such as stability, permeability and toxicity. The results of the clinical examination were shown that its permeability and toxicity didn't matter. To increase the stability of IAA, antioxidants such as Licorice, ubiquinone, tocopherol, Baicalin, ferulic acid, BHT, ascorbic acid, sodium metabisulfite, and so on were employed in cosmetic formulations. Our main purpose is the study for the stability efficiency and effect of each other of cream formulations containing optimal dosage antioxidants (o/w type emulsion), This study evaluated wrinkle reduction effect of IAA, which is used in cosmetics.
Colon cancer is the third most common malignant neoplasm in the world and it remains an important cause of death, especially in western countries. The toxic environmental pollutant, 1, 2-dimethylhydrazine (DMH), is also a colon-specific carcinogen. Tannic acid (TA) is reported to be effective against various types of chemically induced toxicity and also carcinogenesis. In the present study, we evaluated the chemopreventive efficacy of TA against DMH induced colon toxicity in a rat model. Efficacy of TA against the colon toxicity was evaluated in terms of biochemical estimation of antioxidant enzyme activities, lipid peroxidation, histopathological changes and expression of early molecular markers of inflammation and tumor promotion. DMH treatment induced oxidative stress enzymes (p<0.001) and an early inflammatory and tumor promotion response in the colons of Wistar rats. TA treatment prevented deteriorative effects induced by DMH through a protective mechanism that involved reduction of oxidative stress as well as COX-2, i-NOS, PCNA protein expression levels and TNF-${\alpha}$ (p<0.001) release. It could be concluded from our results that TA markedly protects against chemically induced colon toxicity and acts plausibly by virtue of its antioxidant, anti-inflammatory and antiproliferative activities.
In this study, Kami-honghwa-tang (KH-19) was designed and animal study was conducted to evaluate its efficacy on the reduction of the side effect of radiotherapy. Bone marrow toxicity is one of the major side effect of radiotherapy which cause the reduction of blood cells, and KH-19 was designed to protect and enforce blood. C57BL/6 mice were irradiated with 4 Gy of gamma ray, and divided into control group which was treated with water and KH-19 group which was treated with 1.5g/Kg of KH-19 up to 4 weeks. KH-19 group showed significantly increased white blood cells, lymphocytes and platelet count compared with control group (p<0.05). When bone marrows were examined, KH-19 group showed higher cell densities than control group (p=0.06). KH-19 may increase blood cell count after radiation by its protective effects on bone marrow.

The novel synthesis of 3-aminopropyl dihydrogen phosphate(3-APPA; 3-Aminopropane phosphoric acid), and its applicability to the skin as a cosmetic raw material in terms of its efficacy and toxicology were presented. The phosphorylation of 3-amino-1-propanol was carried out via cyclization into 6-membered 2, 6-oxaza-phosphoryl ring in the presence of phosphorous oxychloried and an organic base. The subsequent ring-opening hydrolysis and crystallization afforded the highly purified product in 90% isoloated yield. The method is much superior to the previous literature phosphorylation methodsm, as the procedure is simple and high-yielding. To confirm the efficacy of 3-APPA, several activities related to anti-aging capacity were measured. In-vitro human fibroblast, linear and 3-dimensional collagen matrix culture revealed that 3-APPA stimulated the proliferation of fibroblasts, and enhanced the synthesis of collagen, which showed 3-APPA's potency for skin wrinkle reduction. The toxicolgical aspect of 3-APPA was also extensively examined. In vivo toxicity tests such as acute oral toxicity, eye irritation, human patch, and the repeat insult human patch test proved 3-APPA to be a safe material. Thus 3-APPA can be used as an effective anti-aging agent for various cosmetic formulations.
Licorice has been wed in clinical medicine for thousands of years. However it is only in recent times that we have been able to employ scientific methods to prove its efficacy and to give us a better understanding of its mechanism of action. One of important mechanisms for its efficacy is related to mineralocorticoid activity increased by glycyrrhizic acid, the active ingredient in licorice. Also the main undesirable side-effects of Licorice relate to is mineralocorticoid activity resulting in a state of apparent mineralocorticoid excess (AME). These therapeutic and undesirable effects are explained by the inhibition of 11-$\beta$-hydroxysteroid dehydrogenase (11-$\beta$-HSD) activity. Recently, the reduction of serum testosterone in men by licorice was reported which would have important health implications in the context of fertility and sexual dysfunction. Here, health implication of licorice were reviewed In term of its pharmacodynamic and toxicodynamic mechanism.

Effect of nematicidal compounds on hatchability of sugar beet cyst nematode, Heterodera schachtii and its infective juveniles was investigated. The sugar beet cyst nematode was isolated from Chinese cabbage field in Samcheok in Korea. Acute toxicity of nematicidal compounds against infective juveniles was also tested to find the $LC_{50}$ by exposing juveniles to given dilutions of each compound. Hatchability and mortality of infective juveniles of H. schachtii were influenced by nematicidal compounds (Fluopyram 40% SC, imicyafos 30% SC, fosthiazate 30% SC, abamectine 1.68% SC, terthiophene, and Eclipta prostrata extract). Fluopyram and imicyafos yielded the lowest rates of hatching. Total hatched infective juveniles were significantly different among nematicidal compounds. Positive correlation in percentage reduction of hatching was observed in fluopyram. Furthermore, the highest mortality was also observed in the treatments of fluopyram and imicyafos ($LC_{50}$ of 0.0543 and 0.0178 ppm respectively). The study, therefore, demonstrated available alternative nematicidal compounds which could be used in the control of H. schachtii.

Stability, efficacy, absorption and toxicity of a new nasal spray formulation including salmon calcitonin were studied in the laboratory animals. After the effects of many excipients on the stability of salmon calcitonin were evaluated using HPLC system, we selected taurine. Our experimental composition of salmon calcitonin contains taurine as a stabilizer and HPMC (hydroxypropylmethyl cellulose) as an adhesive polymer. After intranasal administration of salmon calcitonin formulations, Mia$\^$(R)/, Men$\^$(R)/ and experimental composition, 22 IU to rats, the reduction percentages of calcium concentration in plasma (ΔD%) were 16.3%, 12.9% and 20.8%, respectively. After intranasal administration of Mia$\^$(R)/, Men$\^$(R)/ and experimental composition to rats, C$\sub$MAX/ (205${\pm}$161, 244${\pm}$117, and 330${\pm}$202 pg/$m\ell$, respectively) and AUC (41585${\pm}$22070, 41191${\pm}$19125, and 63357${\pm}$43126 pg. min/$m\ell$, respectively) were calculated. The permeation coefficients 10$\^$-7/,cm/sec) of salmon calcitonin in Mia$\^$(R)/, Men$\^$(R)/ and experimental composition using Ussing chamber with rabbit nasal mucosa were 4.7${\pm}$1.5, 0.75${\pm}$0.4 and 5.3${\pm}$1.1, respectively. The experimental composition with taurine and HPMC was proved to be excellent because it improved the stability of salmon calcitonin and inhanced the absorption of salmon calcitonin and was not irritative to the nasal mucosa.
