Evaluation of Streptomyces saraciticas as Soil Amendments for Controlling Soil-Borne Plant Pathogens

  • Soil-borne diseases are the major problems in mono cropping. A mixture (designated LTM-m) composed of agricultural wastes and a beneficial microorganism Streptomyces saraceticus SS31 was used as soil amendments to evaluate its efficacy for managing Rhizoctonia solani and root knot nematode (Meloidogyne incognita). In vitro antagonistic assays revealed that SS31 spore suspensions and culture broths effectively suppressed the growth of R. solani, reduced nematode egg hatching, and increased juvenile mortality. Assays using two Petri dishes revealed that LTM-m produced volatile compounds to inhibit the growth of R. solani and cause mortality to the root knot nematode eggs and juveniles. Pot and greenhouse tests showed that application of 0.08% LTM-m could achieve a great reduction of both diseases and significantly increase plant fresh weight. Greenhouse trials revealed that application of LTM-m could change soil properties, including soil pH value, electric conductivity, and soil organic matter. Our results indicate that application of LTM-m bio-organic amendments could effectively manage soil-borne pathogens.

Effect of Cu Species Distribution in Soil Pore Water on Prediction of Acute Cu Toxicity to Hordeum vulgare using Terrestrial Biotic Ligand Model (토양 공극수 내 Cu의 존재형태가 terrestrial biotic ligand model을 이용한 보리의 급성독성 예측에 미치는 영향)

  • In this study, the predictive toxicity of barley Hordeum vulgare was estimated using a modified terrestrial biotic ligand model (TBLM) to account for the toxic effects of $CuOH^+$ and $CuCO_3(aq)$ generated at pH 7 or higher, and this was compared to that from the original TBLM. At pH values higher than 7, the difference in $EA_{50}\{Cu^{2+}\}$ (half maximal effective activity of $Cu^{2+}$) between the two models increased with increasing pH. As Mg concentration increased from 8.24 to 148 mg/L in the pH range of 5.5 to 8.5, the difference in $EA_{50}\{Cu^{2+}\}$ increased, and it reached its maximum at pH 8. The difference in $EC_{50}[Cu]_T$ (half maximal effective concentration of Cu) between the two models increased as dissolved organic carbon (DOC) concentration increased when pH was above 7. Thus, for soils with alkaline pH, the toxic effect of $CuOH^+$ and $CuCO_3(aq)$ are greater at higher salt and DOC concentrations. The acceptable Cu concentration in soil porewater can be estimated by the modified TBLM through deterministic method at pH levels higher than 7, while combination of TBLM and species sensitivity distribution through the probabilistic method could be utilized at pH levels lower than 7.

Single and mixed chelants-assisted phytoextraction of heavy metals in municipal waste dump soil by castor

  • The phytoextraction of some toxic heavy metals from municipal waste dump soil by castor plant (Ricinus communis) was tested under natural and single or mixed chelant-assisted scenarios in pot microcosms. A sandy loam with total metal contents (mg/kg): Cd (84.5), Cu (114.5), Ni (70.3), Pb (57.8), and Zn (117.5), was sampled from an active dumpsite in Calabar, Nigeria and used for the study. Castor (small seed variety) was grown under natural phytoextraction or single/binary chelant (citric acid, oxalic acid, and EDTA) applications (5-20 mmol/kg soil) for 63 days. Castor exhibited no visual phytotoxic symptoms with typically sigmoid growth profiles at the applied chelant doses. Growth rates, however, decelerated with increase in chelant dose. Post-harvest biomass yields were higher under chelant application than for natural phytoextraction. Both root and shoot metal concentrations (mg/kg) increased quasilinearly and significantly ($p{\leq}0.05$) with increase in chelant dose, furnishing maximum levels as: Cd (55.6 and 20.9), Cu (89.5 and 58.4), Ni (49.8 and 19.6), Pb (32.1 and 12.1), and Zn (99.5 and 46.6). Ranges of translocation factors, root and shoot bioaccumulation factors were 0.21-3.49, 0.01-0.89 and 0.01-0.51, respectively. Overall, the binary chelant treatments were less toxic for R. communis growth and enhanced metal accumulation in shoots to a greater extent than the single chelant scenarios, but more so when EDTA was present in the binary combination. This suggests that the mixed chelants could be considered as alternative treatments for enhanced phytoextraction and revegetation of degraded waste dump soils.

A Study on Application Test of Cut-slope Revegetation Measures with Organic Soil Amendment Materials (유기질계 토양개량재를 이용한 절토비탈면 녹화공법 적용시험 연구)

  • This study was conducted to suggest the ecological restoration methods of the decomposed granite cut-slope by organic soil amendment materials. Field test carried out for the cut-slope with organic soil amendment materials method and other three revegetation methods in Cheongwon. Test revegetation plants were Festuca arundinacea, Lolium perenne, Dactylis glomerata, Lespedeza cyrtobotrya, and Arundinella hirta. The result of this study can be summarized as follows; 1. The soil hardness, the soil acidity, and the soil humidity of organic soil amendment materials method were at a suitable value for plants growth. And it was better as compared with other three revegetation methods of cutting-rock slopes. 2. The result of toxic substance investigation, all items were at a suitable for standard law. 3. During one year after seeding, most plants germinated and especially Festuca arundinacea and Dactylis glomerata grows well. Seedling numbers were 336.7 per $m^2$(after 6 months), 183.3 per $m^2$(after 10 months), and 353.3 per $m^2$(after 6 months). Ten months later after seeding, plants showed 80% ground coverage. Visual rate, plant height, and growth rate were excellence, Also, high plant growth in spring better than autumn.

A Study of PAH (polyaromatic hydrocarbon) Biodegradation in Soil by Bacillus subtilis mixed with Wooden Media (목재재질의 담체를 혼합한 토양에서 Bacillus subtilis에 의한 PAH분해에 대한 연구)

  • PAHs are aromatic hydrocarbon compunds with two or more benzene rings. Because they are mostly toxic to human life, they need to be converted to non-toxic compunds or to be degraded completely. This work aims to degrade PAHs (phenanthrene and pyrene) using Bacillus bacteria covered on cork or sawdust. The results show that media effect on phenanthrene was negligible whereas biodegradation ability of sawdust carrying the bacteria was better than that of biofilm-covered cork when pyrene was tested. PAH removal was also affected by soil moisture content with $45\~55\%$ of the optimal content.

Toxic Effect of Cadmium Assay in Contaminated Soil Earthworm Cell Using Modified Sensor

  • A voltammetric toxic metal of cadmium detection was studied using a fluorine doped graphite pencil electrode (FPE) in a seawater electrolyte. In this study, square wave (SW) stripping and chronoamerometry were used for determination of Cd(II) in seawater. Affordable pencils and an auxiliary electrode were used as reference. All experiments in this study could be performed at reasonable cost by using graphite pencil. The application was performed on the tissue of contaminated soil earthworm. The results show that the method can be applicable for vegetables and in vivo fluid or medicinal diagnosis.

Effects of Pb, Cu, and Cr on Anaerobic Biodegradation of Diesel Compounds by Indigenous Bacteria (혐기성 토착미생물의 디젤 생분해에 대한 Pb, Cu, Cr의 영향)

  • Anaerobic biodegradation of diesel with coexisting heavy metals (Pb) was monitored in batch mode. Two different groups of the indigenous bacteria from a site contaminated with diesel and lead were used in this research: the first group was composed of a single species and the second group was composed of several species. The effect of heavy metals on the microbial population was monitored and confirmed the biodegradation mechanism in each combined contaminant. Growth of the microorganisms in 21 days was observed Diesel > Diesel + Pb > Diesel + Cu > Diesel + Pb + Cu > Diesel + Cr > Diesel + Pb + Cr. Indigenous microorganisms showed the adaptation in the Pb contaminate. Interactive toxic effect using AMES test observed larger synergistic effect than antagonistic in Diesel + Cr and Diesel + Pb + Cr. Therefore, the main effects of diesel biodegradation in the present of heavy metals are likely to exist other factors as well as toxic of heavy metals. This is a necessary part of the future studies.

  • Corn (Zea may, L.) was grown alternatively in nutrient solution and hydroxy Al or Al-citrate solution to identify the form of Al which induces Al toxicity on Corn seedlings. Corn seedlings exposed to hydroxy Al solution was very toxic but Al-citrate solution did not show any toxic symptoms. At pH 7 with Al-citrate solution, severe Fe, deficiency was induced probably by the decrease of stability constant of Al-and Fe-oganic complexes and subsequent precipitation of Al-and Fe-as a hydroxide form. Addition of humic acid ameliorated the Al toxicity somewhat at pH 4.7 with hydroxy-Al solution but at pH 7 it induced more severe Fe deficiency.

  • The global environment is deteriorating at an alarming rate despite of enhanced international environmental regulation. Many studies have been performed to reduce pollutants. Recently, phytoremediation, plant-based technology for the removal of toxic contaminants from soil, water, and air, has been receiving large attention. Arsenic-contaminated soil is one of the major pollutant sources fur drinking water. The fern brake (Pteris erotica) has been reported as a hyper-accumulate arsenic from soils. In this study, we investigated the arsenic absorption effect on sap flow inside xylem vessels of a fern brake. The synchrotron X-ray micro-imaging technique was employed to monitor flow inside the plant non-invasively. The captured phase-contrast X-ray images show both anatomy and transport of water inside the fern brake. The refilling process of water containing arsenic inside the xylem vessels of fern brake's leaves and stems was clearly observed. These results would provide important information needed fur understanding the mechanisms of accumulation, translocation, and transformation of toxic materials in plants.
