남해무인도서 백도와 서귀포 인근 섶섬에서 맹독성 저서와편모조류 Gambierdiscus spp.의 출현 (Occurrence of the Toxic Benthic Dinoflagellate Gambierdiscus spp. in the Uninhabited Baekdo Islands off Southern Coast and Seopsom Island in the Vicinity of Seogwipo, Jeju Province, Korea)

  • Gambierdiscus toxicus, Adachi et. Fukuyo, is a benthic ciguatoxin-producing armored dinoflagellate, often attached to macroalgae. This organism is the primary causative agent of ciguatera fish poisoning which occurs in tropical and subtropical regions. However, regardless of the fact that the population of Gambierdiscus spp. has expanded to such temperate areas from sub-trophic and trophic areas, monitoring of G. toxicus has been lacking in the Korean coastal waters of temperate areas. This study was performed at the uninhabited Baekdo Islands off the southern coast of Korea and at Seopsom Island in the vicinity of Seogwipo, Jeju Province during April and May, 2011. Cell densities of Gambierdiscus spp. on macroalgae at Baekdo and Jeju Island ranged from zero to 56.4 cells $g^{-1}$. Maximum density was recorded on the brown alga Cladophora japonica at St. 3 of Jeju Island. In particular, the cell densities of Gambierdiscus spp. were influenced by the substrate characteristics of macroalgae. In the future, the continuous monitoring of toxic benthic dinoflagellate is necessary to predict and prevent ciguatera poisoning in Korean coastal waters.

Presence of benthic dinoflagellates around coastal waters of Jeju Island including newly recorded species

  • A study on the presence of benthic dinoflagellates in the intertidal zone along the coasts of Jeju Island, Korea was conducted during 2011 and 2012. Identification and taxonomic observations were made of the benthic dinoflagellate samples using light and epifluorescence microscopy. Thirty-seven dinoflagellate taxa belong to five orders, nine families, 18 genera and 30 species, which are new records for Korean waters, were detected in this study. The detailed nomenclature, references, distribution, and illustrations are presented here. The commonly occurring genera were Amphidinium, Coolia, Ostreopsis, Prorocentrum, and Thecadinium. Among the recorded species, 26 were found only in sand sediment, seven in macroalgal samples, and four were found in both sand and macroalgal samples. Of the 37 species, nine were potentially toxic. These results suggest that diversified benthic dinoflagellates including several potentially toxic species occur in sand sediment and macroalgae in the intertidal zone along the coasts of Jeju Island. The morphological features of the identified species were more or less similar to observations made by previous studies in Korea and elsewhere. The presence of known toxic species may indicate a potential risk of toxicity in the marine ecosystem of Jeju Island. The present study can be helpful for further detailed taxonomic, toxicological, molecular phylogenetic studies and may help in the management and conservation of Jeju Island's marine ecosystem.

Benthic dinoflagellates in Korean waters

  • The occurrence of benthic dinoflagellates, many of which are known to be toxic, is a critical concern for scientists, government officers, and people in the aquaculture, dining, and tourism industries. The interest in these dinoflagellates in countries with temperate climate is increasing because tropical or subtropical species introduced into temperate waters by currents are able to survive the winter season in the new environment owing to global warming. Recently, several species from the benthic dinoflagellate genera Amphidinium, Coolia, Ostreopsis, Gambierdiscus, and Prorocentrum have been reported in the waters of the South and East Sea of Korea. The advent of the benthic dinoflagellates in Korean waters is especially important because raw or slightly cooked seaweeds, which may harbor these benthic dinoflagellates, as well as raw fish, which can be potentially intoxicated by phytotoxins produced by some of these benthic dinoflagellates, are part of the daily Korean diet. The recent increase in temperature of Korean coastal waters has allowed for the expansion of benthic dinoflagellate species into these regions. In the present study, we reviewed the species, distribution, and toxicity of the benthic dinoflagellates that have been reported in Korean waters. We also provided an insight into the ecological and socio-economic importance of the occurrence of benthic dinoflagellates in Korean waters.

The occurrence of the ciguatera fish poisoning producing dinoflagellate genus Gambierdiscus in Pakistan waters

  • Five benthic species of the genus Gambierdiscus (Dinophyceae) were observed for the first time in the coastal waters of Pakistan, Northern Indian Ocean. The morphology of the epiphytic, ciguatera-related toxic species G. toxicus, G. belizeanus, G. polynesiensis, G. australes and G. cf. yasumotoi are presented here, described by the Kofoid system of thecal plates Po, 3', 7", 6c, 8s, 5"', 1p, 2"" with differences in cell shape, cell size, plates, pores around the apical pore plate by using light and scanning electron microscopy. The occurrence of these potentially toxic dinoflagellate species in Pakistani coastal areas of Manora Channel and Balochistan during high temperatures of 28-$32^{\circ}C$ is cause of concern for human health impacts from ciguatera fish poisoning.

Methods for sampling and analysis of marine microalgae in ship ballast tanks: a case study from Tampa Bay, Florida, USA

  • Ballasting and deballasting of shipping vessels in foreign ports have been reported worldwide as a vector of introduction of non-native aquatic plants and animals. Recently, attention has turned to ballast water as a factor in the global increase of harmful algal blooms (HABs). Many species of microalgae, including harmful dinoflagellate species, can remain viable for months in dormant benthic stages (cysts) in ballast sediments. Over a period of four years, we surveyed ballast water and sediment of ships docked in two ports of Tampa Bay, Florida, USA. Sampling conditions encountered while sampling ballast water and sediments were vastly different between vessels. Since no single sample collection protocol could be applied, existing methods for sampling ballast were modified and new methods created to reduce time and labor necessary for the collection of high-quality, qualitative samples. Five methods were refined or developed, including one that allowed for a directed intake of water and sediments. From 63 samples, 1,633 dinoflagellate cysts and cyst-like cells were recovered. A native, cyst-forming, harmful dinoflagellate, Alexandrium balechii (Steidinger) F. J. R. Taylor, was collected, isolated, and cultured from the same vessel six months apart, indicating that ships exchanging ballast water in Tampa Bay have the potential to transport HAB species to other ports with similar ecologies, exposing them to non-native, potentially toxic blooms.

춘계 제주 연안에서 유독 저서성 와편모류 Ostreopsis sp.의 분포와 분자계통학적 위치 (Distribution and Molecular Phylogeny of the Toxic Benthic Dinoflagellate Ostreopsis sp. in the Coastal Waters off Jeju Island, Korea)

  • 본 연구는 2017년 4월 춘계 제주 연안에서 총 7개의 정점(협재, 이호테우, 함덕, 성산, 표선, 남원, 사계)을 선정하여 해조류에 부착하여 서식하는 유독 착생 와편모류 Ostreopsis의 출현양상을 조사하고 분자계통학적 분석을 실시하였다. 본 연구 해역의 표층 수온은 $15.7^{\circ}C-18.3^{\circ}C$의 범위를 보였으며, 염분은 33.4-34.9의 범위로 나타났다. 각 연구 정점에서 채집된 전체 13종의 해조류 가운데 8종에서 Ostreopsis가 출현하였으며, 정점 6에서 출현한 홍조식물 참지누아리(Grateloupia filicina)에서 해조류 단위 무게당 Ostreopsis의 출현밀도($cells\;g^{-1}$)가 $157.5cells\;g^{-1}$로 가장 높은 농도로 출현하였다. Ostreopsis가 출현한 4개의 연구정점에서 분리한 종주들의 LSU rDNA D8/D10 영역의 염기서열은 모두 100% 동일한 것으로 나타났다. LSU rDNA 염기서열 정보를 이용한 분자계통수에서 이들은 모두 Ostreopsis cf. ovata의 잠재종(cryptic species)으로 알려진 Ostreopsis sp. 1의 분기군에 속하는 것으로 나타났다. Ostreopsis sp.1 종주를 이용하여 수온과 염분에 따른 생장 반응을 측정한 결과, $10-30^{\circ}C$의 광범위한 수온과 20-35의 염분 범위에서 뚜렷한 생장을 나타내었고, 수온 $25^{\circ}C$와 염분 30에서 $0.49d^{-1}$의 최고 생장률을 나타내었다. 또한, 수온 $10^{\circ}C$의 저온에서도 염분 35에서 뚜렷한 생장을 보여, 이들은 온대 해역에서 적응하여 정착한 종으로 판단된다.

유독 와편모조류 Alexandrium속의 출현에 미치는 휴면포자의 발아율 (Germinability of Resting Cysts Associated with Occurrence of Toxic Dinoflagellate Alexandrium Species)

  • 패독발생의 원인규명을 위한 모니터링의 일환으로 원인생물의 출현과 휴면포자의 발아에 의한 현장 개체군의 유입가능성을 조사하기 위하여 1993년 9월부터 진해만을 중심으로 Alexandrium속의 발생현황, 원인생물의 잠재력을 제공하는 저질중의 휴면포자의 분포 및 실내 발아율을 조사하였다. 만내의 조사된 모든 정점에서 1월부터 Alexandrium속이 출현하여 대곡리 정점(칠천도)에서 3월에 533 개체/l의 최고치를 보였으며, 다른 지점에서는 100개체 이하를 나타냈으나 수온상승기인 4월에는 출현량이 감소하였다. 저질중의 Alexandrium속 휴면포자의 분포밀도는 채집한 정점간의 차이가 많아 100-800 개체/$cm^3$의 밀도를 보였으나, 채집지역의 저질상부층$(0\~4cm)$에서 $700\~800$ 개체/$cm^3$의 분포 지점이 많았다 휴면포자의 발아율은 수정리 정점에서 1월$(72.5\%)$, 4월$(68.0\%)$, 7월$(44\%)$ 및 10월$(9.0\%)$ 순으로 채집 계절에 따른 차이를 나타냈으며, 1월 채집한 만내의 다른 2 정점의 휴면포자도 $71\~97|%$의 높은 발아율을 보였다. 한편, 수심이 얕은 다대포 지점의 휴면포자는 6월$(86.5\%)$과 10월$(70.0\%)$에도 높은 발아율을 보여 진해만의 각 지점과는 대조적인 결과를 보였다. 이상의 결과로부터 Alexandrium속 휴면포자의 발아는 지역환경에 따라 차이를 보일 수 있으며 비교적 수심이 깊은 지역에서는 온도 등의 환경요인의 변동 외에 내재적 발아리듬의 영향이 클 것으로 추정되었다. 따라서 진해만에서는 저수온기일지라도 휴면포자의 발아에 의한 Alexandrium속의 출현이 놀게 나타나며, 이 지역의 패독발생의 원인생물로 기여할 가능성이 높은 것으로 시사되었다.
