Effect of Rudbeckia laciniata Extract on Physiological Activity of HaCaT Cells (삼잎국화 추출물의 피부세포 생리활성 효과)

  • The objective of the present investigation was to obtain vitamin, mineral, flavonoid, and polyphenol profiles of Rudbeckia laciniata (RL), and to examine the effects of extract of RL (RLE) on various physiological activities of HaCaT keratinocyte for the utilization of RL as natural raw materials to develop functional food. To accomplish this purpose, we checked the contents of the general nutrients of RL. The contents of vitamin A, vitamin $B_1$ and vitamin $B_2$ were $7.49{\mu}g/g$, $51.96{\mu}g/g$, and $132{\mu}g/g$ respectively, while vitamin C and vitamin $D_3$ were not detected. The contents of mineral such as Ca, K and Fe were 2.01 mg/g, 6.06 mg/g and 0.03 mg/g respectively. Total flavonoid contents of RLE were 0.25 mg/g, and total polyphenol were estimated as 1.43 mg/g. Because RL contains high levels of vitamin A which is associated with skin aging, we investigated the effect of RLE on physiological function of keratinocytes with respect to skin aging. We found that RLE significantly increased the growth rate of HaCaT cells and reduced ultraviolet radiation B (UVB)-induced cellular toxicity. Also, the extract of Rudbeckia laciniata attenuated the UVB-induced reactive oxygen species (ROS) generation in a dose-dependent manner in HaCaT cells. In addition, treatment with the extract dose-dependently increased migration activity of HaCaT cells. Thus, these findings indicated that RLE could regulate the physiological activity of keratinocytes, and may be used to develop functional foods.

Effects of Dietary Attitudes on the Nutritional Status of Nurses in Kyungnam Province

  • This study has investigated the effect of dietary attitudes on nutritional status of nurses living in Kyungnam province. A total of 249 muses working at hospitals in the Kyungnam area participated in this study. The general characteristics and dietary attitudes of the subjects were surveyed using a self-administered questionnaire, and nutrient intakes were examined using one day 24-hour recall method from November 14 to December 20, in 2001. The data of nutrient intakes were analyzed by the Computer Aided Nutritional Analysis Program and then the diet quality was estimated using the Index of Nutritional Quality (INQ). The subjects were classified as the high score group (17.3 %), medium score group (54.2%), and low score group (28.5%) based on dietary attitude score. The results were as follows: Average age of subjects was 27.1 years old, average nursingexperience was 5.7 years, and 70.7% of the subjects graduated from a junior college. The rate of shift work (45.4%) and non-shift work (54.6%) were similar. The average score on dietary attitudes in thehigh score group was 78.8, and those in he medium score group and in the low score group were 58.6 and 40.8 out of 100 points respectively. The average intake of energy (p < 0.001), carbohydrate (p < 0.001), protein (p < 0.05), animal protein (p < 0.01), animal fat (p < 0.05), vitamin $B_1$ (p < 0.01), vitamin $B_2$ (p < 0.0l), niacin (p < 0.001), Ca (p < 0.001), Fe (p < 0.001), animal Fe (p < 0.01), P (p < 0.001), and crude fiber (p < 0.00l), were significantly higher in the high score group than those in the medium score group and low score group. In the high score group, the average intake of protein, vitamin $B_1$, niacin, vitamin C and P were above 125% of the Korean RDA, and in medium score group, those of vitamin C and P, and in low score group, that of vitamin C were above 125% of the Korean RDA. In the high score group, nutrient of intake below 75% of the Korean RDA was Ca, and those in medium score group were vitamin $B_2$, Ca, and Fe, and those in low score group were energy, vitamin $B_2$, Ca and Fe. The intake ratio of Ca and P showed an unbalanceof 1:2, and that of animal protein/plant protein, and that of animal fat/plant fat were high (1.45-1.64) in all group;. The dietary attitude had a positive correlation (p < 0.00l) with nutrition intake except vitamin A and vitamin C.

A Study on Nutritional Status of Iron in the Rural Elderly (일부 지역 농촌 노인의 철분 영양상태에 관한 연구)

  • The purpose of this study was to examine nutritional status of iron in 27 rural elderly subjects. The daily intake, urinary excretion and serum level of iron were determined by 24-hr food collection method, and 24-hr urine and serum collection for 3 days. The results were summarized as follows. 1. The mean age, height, weight, and blood pressure were 67.7 years, 163.2cm, 55.9kg, and 121.1/80. 0mmHg in men and 69.9 years, 146.5cm, 47.6kg, and 108.9/71.1mmHg in women, respectively 2. The daily intake of energy, protein, vitamin A, vitamin B$_1$, vitamin B$_2$, niacin, vitamin C, and iron were lower than Korean recommended dietary allowances. 3. The daily iron intake was 7.82mg. The proportion of heme iron and nonheme iron to the total iron intake were 8.05% and 91.94%, respectively Major iron sources of the rural elderly were cooked rice, Ko Chu Jang, beef blood, anchovy, soybean paste, and leafy radish. 4. The hemoglobin, hematocrit, serum iron, and total iron binding capacity were 14.3g/㎗, 42.9%, 79.4 $\mu\textrm{g}$/㎗, and 268.4 $\mu\textrm{g}$/㎗ in men and 12.9g/㎗, 38.8%,92.4 $\mu\textrm{g}$/㎗, and 277.9$\mu\textrm{g}$/㎗ in women, respectively. 5. There was a significant positive correlation between dietary intake and urinary excretion of iron.

Use of Response Surface Methodology for Optimization of Clarified Mixed Apple and Carrot Juice Production (반응표면 분석을 이용한 사과.당근 혼합주스의 청징공정 최적화)

  • Response surface methodology was used to investigate the quality of clarified mixed apple and carrot juices using ultrafitration. Apple and carrot juices were blended at the ratio of 1:3, 1:1, and 3:1. A three-variable, three-level central composite design was employed where the independent variables were the blend ratio, temperature and average transmembrane pressure (ATP). With increasing temperature and pressure, flux linearly increased regardless of blending ratio. Blend juice with 75% apple showed the highest soluble sugar and total sugar content in apple and carrot blend juices. Soluble solid contents were more affected by blending ratio than temperature and ATP. Total sugar contents were greatly affected by temperature; increasing temperature led to higher total sugar content up to $25^{\circ}C$. Higher carrot ratio led to higher vitamin C content. In general, higher acidity was achieved by higher apple content and acidity was increased with increasing temperature. Turbidity increased for all samples as APT increased, with the blending ratio of 1:1 (apple:carrot) showing the highest turbidity. Viscosity was greatly changed in the blending ratio of 3:1 (apple:carrot) juice. The polynomial models developed by RSM were satisfactory to describe the relationships between the studied factors and the responses. Analytical optimization gave $flux=0.216\;L/m^2.h$, soluble $solids=10.39^{\circ}Brix$, total sugar=71.32 mg/mL, vitamin C=315.18 mg%, acidity=7.78 mL, turbidity=0.017, and viscosity=1.44 cp, when using a $temperature=44.97^{\circ}C$, ATP=113.57 kPa, and blend ratio=28.50%.

Effect of Preparation Methods on Yulmoo Kimchi Fermentation (열무 물김치의 담금방법이 발효숙성에 미치는 영향)

  • Effect of preparation method on Yulmoo kimchi(watery kimchi prepared with Yulmoo, leaf radish) fermentation was investigated by measuring physicochemical, microbiological and sensory properties for 20 days. Yulmoo kimchi was fermented at 4$^{\circ}C$ after keeping at room temperature(27$\pm$0.5$^{\circ}C$) for 8 hours. Four types(A, B, C, D) of Yulmoo kimchi were prepared. Sample A was made without wheat flour paste and red pepper powder and sample B was made with wheat flour paste. Sample C was made with red pepper powder, and sample D was made with wheat flour paste and red pepper powder. pH was slowly lowered and stabled after 10 days in all samples. Total acid content increased in all samples. Reducing sugar content initially increased and decreased thereafter. Especially, reducing sugar content of sample B increased in the early stage of fermentation and maintained initial reducing sugar content after 20 days. Total vitamin C content reduced during fermentation in all samples. Especially initial total vitamin C content of sample D increased more than those of other samples relatiely. Lightness and yellowness showed no difference, but on the other hand redness increased gradually in all samples. The number of lactic acid bactgeria reached maximum value in 10~13 days with the total cell numbers and gradually decreased thereafter in all samples. The number of lactic acid bacteria and total cell number of sample D was much more than those of any other samples. As a result of the sensory evaluation, Yulmoo kimchi showed significant difference in all characteristics. Sample D showed the highest scores in all characteristics before 10 days of fermentation. However, after 10 days sample B showed the highest scores.

  • To compare varietal differences in Rubus coreanus Miq. (Rosaceae), we evaluated antioxidant activity and the contents of functional substances. In this study, five new cultivars selected as superior berries in yield and weight of fruit, R. coreanus (Jungkeuam 1, 2, 3, 4, and 5), and one foreign berry, R. fructicosus were used. The contents of the major anthocyanin, cyanidin-3-glucoside (C3G) in berries and antioxidant capacity including, free radical scavenging activity and reducing power were measured. The contents of total phenol and vitamin C were also investigated. Among 5 cultivars and one foreign berry, Jungkeuam 1 had the richest C3G and high levels in vitamin C. DPPH radical scavenging activity of berries was ranged from 46.58 up to 78.55%.

Effect of Onion on Dongchimi Fermentation (양파 첨가가 동치미의 발효숙성에 미치는 영향)

  • The purpose of this study was to investigate the effects of onion on the physicochemical, microbiological and sensory properties of Dongchimi. Dongchimi with levels(0, 5, 10, 20%) of onion was fermented at twodifferent temperatures : Group A was fermented at 4$^{\circ}C$ and group B was fermented at 4$^{\circ}C$ after fermentation at room temperature(27$\pm$0.5$^{\circ}C$) for 24 hours. During the fermentation up to 45 days, total acidity increased while pH decreased gradually. Total acidity of group B was relatively higher than that of group A. The content of reducing sugar was reached the maximum value in 6 days of fermentation in the group A and on 3 days in the group B. Especially, the highest content of reducing sugar was shown on the Dongchimi added with 5% onion in the group A and 20% onion in the group B. Vitamin C was reduced in the early stage of fermentation, and increased gradually after 10 days. However, vitamin C of Dongchimi without onion increased rapidly in the early stage of fermentation and remarkably decreased thereafter. After 20 days, vitamin C was maintained in Dongchimi added with 10% onion of both two groups. The number of lactic acid bacteria was reached the maximum value on 1 day in the Dongchimi without onion and on 3 days in the Dongchimi added 5, 10 and 20% onion of group A, while gropu B was reached on 1 days and then decreased thereafter. As a result of the sensory evaluation, Dongchimi added 5% onion in the group A showed the highest scores in texture and overall acceptability. Whereas, in the group B, Dongchimi added 20% onion was the most preferable one.

  • To investigate the nutritional contents of lunchbox of students of elementary school located in Sokcho city Kangwon province, the survey was conducted from April 23 to April 26, 1991. Main dishes and side dishes in lunchbox were weighed and nutrients were analyzed from food composition table. Total daily energy and nutrients intake were reach to RDA's, except for intake of calcium, iron, vitamin A, vitamin Be and vitamin C. Children's height was positively correlated with energy and protein Intake. Fat intake was positively correlated with monthly income and intake of vitamin A was positively correlated, with number of family, This survey suggests that it need nutritional education program and school feeding to improvement of nutritional status for children.

Re-evaluation of the Optimum Dietary Vitamin C Requirement in Juvenile Eel, Anguilla japonica by Using L-ascorbyl-2-monophosphate

  • This study was conducted to re-evaluate the dietary vitamin C requirement in juvenile eel, Anguilla japonica by using L-ascorbyl-2-monophosphate (AMP) as the vitamin C source. Five semi-purified experimental diets were formulated to contain 0 ($AMP_0$), 30 ($AMP_{24}$), 60 ($AMP_{52}$), 120 ($AMP_{108}$) and 1,200 ($AMP_{1137}$) mg AMP $kg^{-1}$ diet on a dry matter basis. Casein and defatted fish meal were used as the main protein sources in the semi-purified experimental diets. After a 4-week conditioning period, fish initially averaging $15{\pm}0.3$ g (mean${\pm}$SD) were randomly distributed to each aquarium as triplicate groups of 20 fish each. One of five experimental diets was fed on a DM basis to fish in three randomly selected aquaria, at a rate of 3% of total body weight, twice a day. At the end of the feeding trial, weight gain (WG) and specific growth rate (SGR) for fish fed $AMP_{52}$ and $AMP_{108}$ were significantly higher than those recorded for fish fed the control diet (p<0.05). Similarly, feed efficiency (FE) and protein efficiency ratio (PER) for fish fed $AMP_{52}$ were significantly higher than those for fish fed the control diet (p<0.05). Broken-line regression analysis on the basis of WG, SGR, FE and PER showed dietary vitamin C requirements of juvenile eel to be 41.1, 41.2, 43.9 and 43.1 (mg $kg^{-1}$ diet), respectively. These results indicated that the dietary vitamin C requirement could range from 41.1 to 43.9 mg $kg^{-1}$ diet in juvenile eel when L-ascorbyl-2-monophosphate was used as the dietary source of vitamin C.
