The improvement effect of anti-inflammation of Aronia extract that fermented by Lactic acid bacteria isolated from the fermented seafoods

  • Aronia (black chokeberry), a species of berries is source to a very large number of bioactive compounds like polyphenols, flavonoids, anthocyanins, and tannins in comparison to any other species. Owing to its antioxidant, anti-carcinogenic, anti-aging and anti-inflammatory properties. Fermentation- a bioconversion process exploiting the biological metabolic reaction of micro-organisms, has several benefits like improving the efficacy and safety of physiologically active substances, generation of new functional material, improving the adsorption rate and many others. Antigens like pathogens, food, pollen etc., generate a protective immune response in body tissues, and the process be referred to as inflammation. However, this when excessive results in a condition referred to as refractory inflammatory disease, whose incidence is increasing in the recent times, especially amongst children. The current study intended to assess the anti-oxidant activity, presence of polyphenols and anti-inflammatory efficacy of Aronia extract fermented by Lactic acid bacteria isolated from fermented sea foods. Aronia fruits collected from Sunchang, Chonbuk were lyophilized for fermentation. So as to maximise the efficacy of the fermented Aronia extract, the quantitative effects of lactic acid bacteria species, composition of extraction solution, influence of temperature and time on antioxidant activity and total polyphenol contents were investigated using an experimental design. Anti-inflammatory activity was evaluated on NO and cytokine ($TNF-{\alpha}$, IL-6) production in LPS activated Raw 264.7 cells. Results indicated that antioxidant effect and total polyphenol contents were best improved in extract of Aronia fermented by P. pentosaceus. In addition, NO and cytokine ($TNF-{\alpha}$, IL-6) levels were decreased significantly after fermentation. Thus, it was found that the anti-inflammatory activity of Aronia greatly increased after fermentation process using P. pentosaceus.

Preliminary pharmacognostical and phytochemical evaluation of Stachys tibetica Vatke

  • Stachys tibetica Vatke (Lamiaceae) is an important medicinal plant in the folk medicine of Ladakh, India and Tibet for the treatment of various mental disorders. Infusion and decoction of the whole plant is used as a cup of tea for a severe fever, headaches and to relieve tension. The recent study is aimed to evaluate the preliminary pharmacognostical and phytochemical nature of Stachys tibetica Vatke. The whole plant material was subjected to successive soxhlet extraction with petroleum ether (40 - $60^{\circ}C$), chloroform, ethyl acetate, methanol and finally decocted with water to get the respective extracts. The fluorescence characteristics of the powdered materials were analysed under ultraviolet light and ordinary light. Different physicochemical parameters such as ash value, extractive value, foaming index, pH values, loss on drying and determination of foreign matter were carried out as per WHO guidelines. The total fat, flavonoid, saponin and volatile contents were also determined. Macroscopical studies revealed the authentication of the plant drug. Physicochemical parameters helped to standardize the plant material while preliminary qualitative chemical tests of different extracts showed the presence of Glycosides, Carbohydrates, Phytosterols/triterpenoids, Saponins, Fixed oils, Fats and phenols/tannins. Quantification of the total flavonoids and saponins and contents were determined as $54.66{\pm}0.58mg/g$ and $75.42{\pm}0.48mg/kg$ respectively, while the volatile and fat contents were 6.5% and 0.7% respectively. Results may lay the foundation for the standardization of the drug and discovery of new molecules from S. tibetica for the treatment of various diseases.

Phytochemical Screening and Biological Studies of Boerhavia Diffusa Linn

  • Hexane, ethyl acetate and methanol extracts of whole plant of Boerhavia diffusa were screened for phytochemical and biological activities. Qualitative phytochemical screening via colorimetric method and the quantitative estimation of phenolic and flavonoid content were performed. Antioxidant assay using DPPH scavenging method was studied. Antimicrobial screening of plant extracts was done by cup diffusion technique. Cytotoxic activity of B. diffusa was studied by brine shrimp bioassay and anthelminthic activity was evaluated in vitro in Pheretima posthuma. This study revealed B. diffusa as a source of various phyto-constituents such as alkaloids, glycosides, saponins, tannins, carbohydrates, cardiac glycosides, flavonoids and terpenoids. Quantitative estimation of total phenol was found to be maximum in BEE i.e. $29.73{\pm}0.88$, BME $19.8{\pm}2.02$ and in BHE $9.15{\pm}0.304mgGAE/g$. Similarly, the total flavonoid content was found to be $17.44{\pm}0.75$ in BEE, $14.43{\pm}0.23$ in BHE and 3.678 mg QE/g in BME. Ethyl acetate extract showed its antibacterial activity against all tested pathogens except Escherichia coli whereas Staphylococcus aureus and Salmonella Typhi were resistant to methanol and hexane extract. The zone of inhibition (ZOI) of ethyl acetate extract against S. Typhi and B. cereus was found to be 18 mm and 14 mm respectively. The MIC value of BEE in S. Typhi was $3.125{\mu}g/ml$ and in B. cereus was $12.5{\mu}g/ml$. The preliminary screening of anticancer property of B. diffusa i.e. BSLT in methanol was found to be $165.19{\mu}g/ml$. B. diffusa was also found to contain anthelmintic property. The study helped in further exploration of medicinal properties of B. diffusa by phytochemical screening and biological activities paving the path for study and investigation in this plant.

Enhanced oxidative stability of meat by including tannin-rich leaves of woody plants in goat diet

  • Objective: The aim of this study was to evaluate the effect of dietary incorporation of tanninrich woody species on meat oxidative stability, carcass traits and meat quality in goats. Methods: Two tannin-rich species were tested using a three-treatments feeding trial, where treatments consisted of: Larrea divaricata and Acacia aroma both at 12.5% in dry matter basis of the diet and a control diet (alfalfa hay). All feeding diets were iso-protein and iso-energy. Carcass conformation, carcass compactness, carcass fatness and subcutaneous fat deposition were evaluated. Intake, liveweigh, Longissimus thoracis et lumborum muscles of goats were analyzed in order to evaluate quality parameters such as pH value, instrumental color evaluation, water holding capacity, total phenolic content, antioxidant activity, meat oxidative stability and fatty acid profiles in meat. Results: Feed intake, liveweight gain, carcass, and meat traits did not differ among treatments. Changes in meat lipid profile among treatments were observed for oleic and elaidic acid contents. Meat total phenolic content and antioxidant activity did not differ among treatments; although, meat oxidative status after storage at room temperature, as well as under refrigerated and frozen conditions were different between control and both supplemented groups. Conclusion: The inclusion of Acacia aroma and Larrea divaricata leaves in goat diet enhanced meat oxidative stability. Modulation of the ruminal biohydrogenation of fatty acids produced by condensed tannins of these plant species need to be further investigated.

고욤나무 열매의 영양성분 및 항산화 활성 (Nutrients and antioxidant activity of Diospyros lotus L.)

  • 본 연구는 고욤나무 열매 분말의 활용도를 높여 천연 기능성 식품의 재료로써 사용이 가능한지를 영양학적으로 판단하기 위해 실시하였다. 고욤나무 열매 분말의 IDL 단계에서는 산도가 높았으며, MDL 단계에서는 당도가 유의하게 높았다(p<0.05). 조지방, 셀레늄 및 타닌 함량은 고욤나무 열매 분말의 IDL 단계에서 MDL 단계로 갈수록 유의적으로 감소하였으나, 마그네슘과 칼슘 함량의 경우 IDL 단계보다 MDL 단계에서 유의적으로 증가하였다(p<0.05). IDL 단계에서 총 페놀 함량이 유의적으로 높았으나, DPPH 라디칼 소거활성은 MDL 단계에서 유의하게 증가하였다(p<0.05). 아질산염 소거능과 타닌 함량은 IDL 단계에서 높았다. 본 연구를 종합해 볼 때, 고욤나무 열매는 각 성장 시기별로 영양학적 특성을 가지므로, 이를 고려하여 고욤나무 열매를 분말화하여 다양한 기능성 식품의 원료로서 활용한다면, 좋은 먹거리 제공 식품으로서 활용도를 높일 것으로 판단된다.

Ensiled Green Tea Waste as Partial Replacement for Soybean Meal and Alfalfa Hay in Lactating Cows

  • The purpose of this study was to evaluate the effects of protein supplementation of green tea waste (GTW) on the performance of lactating cows. Another aim was to increase resource utilization and to eliminate any environmental negative impact from the tea waste. GTW from a beverage company was ensiled at a low pH (<4.0) and high acetic acid and lactic acid concentration, and it contained high crude protein (CP, 34.8%), total extractable tannins (TET, 9.2%) and condensed tannin (CT, 1.7%). Two experiments were conducted to investigate the palatability and performance in lactating cows fed GTW. In the palatability trial, three lactating cows were allocated to three dietary treatments in a 3$\times$3 Latin square design. The animals were offered a total mixed ration (TMR) including GTW at rates of 0, 2.5 and 5.0% on a dry matter (DM) basis. Total DM intake was not different among the treatments. In the performance trial, four lactating cows were used in a 2$\times$2 Latin square design with a 3 week sampling period. GTW was incorporated into TMR at a rate of 5.0% on a DM and 10.0% on a CP basis. Thus GTW replaced alfalfa hay and soybean meal at a level of 25.0% on a DM. DM and CP intake were not affected by the inclusion of GTW, whereas TET and CT intake were significantly increased (p<0.001). Milk production, milk composition and the efficiency of milk production were not altered by the GTW inclusion. Although ruminal pH and VFA, and blood urea nitrogen were not changed, ruminal $NH_{3}-N$ and plasma total cholesterol were relatively low in the GTW group, but not significantly different. The excretion of urinary purine derivatives and estimated MN supply were also not significantly affected by GTW treatment. It is therefore concluded that GTW can be used as a protein source without any detrimental effects on the performance of lactating cows.

Anti-ulcer and antioxidant activity of leaves of Madhuca indica in rats

  • The leaves of Madhuca (M.) indica J.f.Gmel. (Sapotaceae) have been used traditionally in folk medicine due to its astringent properties and are effective in treatment of eczema and snake bites. Methanolic extract of M. indica is rich in tannins and has been proven experimentally to possess antibacterial activity. The present study was undertaken to evaluate the anti-ulcer and antioxidant activity of M. indica in rats. The methanolic extract of leaves of M. indica was tested at various doses (75, 150 and 300 mg/kg, p.o.) for its effect on gastric secretion and gastric ulcers in pylorus-ligation and on ethanol- induced gastric mucosal injury in rats. The significant reduction in ulcer index in both the models along with an increase in the pH of the gastric fluid and mucin content of stomach, and the acid secretory parameters such as total acidity and volume of gastric fluid were also significantly reduced along with reduction in the pepsin activity in pylorusligated rats proved the anti-ulcer activity of M. indica. The increase in the levels of superoxide dismutase, catalase and reduced glutathione and decrease in lipid peroxidation in both the models proved the antioxidant activity of M. indica. Thus it can be concluded that M. indica possesses anti-ulcer activity, which can be attributed to its antioxidant mechanism of action.

The Effect of Yerba Mate (Ilex paraguarensis) Supplementation on the Productive Performance of Dorper Ewes and Their Progeny

  • Yerba Mate (Ilex paraguariensis), a tea known for its high antioxidant content, was supplemented to 30 of 60 ewes for 13 wks to assess its effect on their productive performance. A 2.5% inclusion rate of Yerba Mate (YM) in a pelleted concentrate diet decreased feed intake and live weight (LW) during the first few weeks post partum (p<0.001). Overall, the YM group ate less ($2,092{\pm}78g/d$) pellet than the control (CTRL) one ($2,434{\pm}83g/d$); similarly, LW was lower in the YM group compared to the CTRL one, $64.9{\pm}1.6kg$ and $67.3{\pm}1.4kg$, respectively. Lambs' birth weight and growth rates were not affected. At birth, lambs' LW were similar between the Yerba Mate and control groups ($4.2{\pm}0.5kg$ and $4.1{\pm}0.4kg$, respectively. At the end of the trial, Yerba Mate lambs weighed $15.7{\pm}0.4kg$ while CTRL lambs weighed $16.1{\pm}0.4kg$. Average daily growth rate was similar between the two groups and ranged from $176{\pm}19$ to $234{\pm}24g/d$. The inclusion of Yerba Mate in a pelleted diet increased milk fat, protein and total solids content while it decreased milk lactose content. Further work is required to investigate the mechanisms by which Yerba Mate supplementation affects feed intake and milk composition.

포도종실 에탄올 추출물의 항균 활성 (Antimicrobial Activity of Grape Seed Ethanol Extract)

  • 포도종실 에탄올 추출물의 각 검정 균주에 대한 생육 억제효과를 페이퍼디스크법 및 액체배양법에 의해 조사한 결과, B. subtilis ATCC 6538 혹은 S. aureus. ATCC 6538에 대하여는 농도 의존적인 증식억제 효과를 나타냈으며 P. aeruginosa IFO 3080 및 Sal. enteritidis. IFO3313에 대해서는 제한적인 증식억제 효과를 나타내기도 하였으나 E. coli ATCC 25922 에 대해서는 뚜렷한 항균 효과를 확인하기 어려웠다. 또한 포도종실 에탄을 추출물의 순차 용매 분획물 중에서 에틸아세테이트 분획과 부탄올 분획물만이 각각 35.7 mg/g과 20.2 mg/g의 카테킨을 함유하고 있었는데 에틸아세테이트 분획에 비하여 부탄을 분획물이 각 검정 균주에 대해 뚜렷한 항균활성 을 나타내었다. 부탄올 분획물은 에틸아세테이트 분획물에 비하여 카테킨의 절대 함량은 적었지만 Cl8 카트리지로 소분획하였을 때 고분자체 중심의 폴리페놀 화합물로 구성되어 있음을 알 수 있었다. 따라서 포도종실 에탄올 추출물 의 항균 활성은 카테킨과 같은 플라반-3-올 화합물의 절대 함량과 더불어 플라반-3-올 화합물의 중합도가 관계하는 것으로 예측되었다.

Antifungal Activity of Lagenaria breviflora Fruit Extracts Against Wood Rotting Fungi on Vitex doniana Wood

  • As a result of contemporary environmental concerns, a number of studies from plants' tissues as one of the alternatives to conventional chemicals are increasingly investigated. In tandem with these trends, Lagenaria breviflora (LB) fruit, reputed as antiviral and depilatory agents in the Yoruba folkloric medicine was examined on Vitex doniana wood to ascertain its antifungal activity. Fungicides of 25%, 50%, 75%, and 100% LB fruits formulations (concentrations) were developed through simple one-step mechanical-forming process, including control. In this study, the yield, the chemical compositions, the absorption capacity of the fungicides and wood weight losses (WWL) analysis were evaluated to investigate the antifungal activity of LB fruit on wood. The fruit extract yielded 35.4% of fresh juice weight. LB fruits contained total: alkaloids ($8.78{\pm}0.21mg/mL$), flavonoids ($2.01{\pm}0.02mg/mL$), phenol ($7.42{\pm}0.09mg/mL$), saponins ($11.00{\pm}0.10mg/mL$) and tannins ($5.47{\pm}0.05mg/mL$) contents. All the formulations provided effective protection against the tested wood fungi compared to control. Interestingly, the antifungal activity of 50% and 25% formulations of 6.8% WWL and 9.9% WWL satisfied the excellent fungal resistance class description against white rot fungus (Ganoderma lucidum) and brown rot fungus (Fibroporia vaillantii), respectively according to ASTM D 2017. These results thus, support LB fruit as a strong potential source of natural antifungals for industrial wood production.
