고지방식이를 섭취한 흰쥐에서 유청단백질 가수분해물의 섭취가 지질 농도 및 식욕 관련 호르몬에 미치는 영향 (Effects of Whey Protein Hydrolysates on Lipid Profiles and Appetite-Related Hormones in Rats Fed High Fat Diet)

  • 본 연구에서는 고지방식으로 유도한 식이성 비만 모델 수컷 흰쥐에게 고지방식이와 함께 유청단백질 및 유청단백질 가수분해물을 급여하였을 때, 유청단백질 가수분해물의 섭취에 따른 체중, 장기 및 체지방, 지질 농도 및 식욕 조절호르몬에 대한 영향을 살펴보았다. 즉, 실험동물 흰쥐에게 18% 우지를 첨가한 고지방식을 기본으로 하여 질소원으로 유청단백질(whey protein concentrate 80: WPC80, 매일유업(주)) 10%를 4주간 공급한 후(control period), 4군으로 나누어서 고지방식을 기본으로 유청단백질, 유청단백질 가수분해물(whey protein hydrolysate), 유청 macropeptide fraction(whey macropeptide fraction; WH중 MW${\geq}$10,000)의 3종을 함유한 실험식이를 각각 6주 동안 급여하였다. 그 결과는 다음과 같다. 체중증가량 및 식이섭취량은 유청단백질 수준이나 가수분해물 섭취에 따른 실험군간 유의적 차이가 없었다. 간의 무게는 가수분해물 섭취에 의해 감소하였으나, 신장, 고환 및 비장의 무게는 실험군간 유의적 차이가 없었다. 한편, 신장의 무게는 고단백 식이에 의해 유의적으로 증가하였다(p<0.05). 지방조직의 무게는 신장 주변 지방과 부고환 주변 지방 모두 유청단백질 수준이나 가수분해물 섭취에 따른 차이를 보이지 않았다. 혈청의 총 지질과 중성지방은 가수분해물 섭취에 의해 유의적으로 낮게 나타났다(p<0.05). 반면, 혈청의 총 콜레스테롤과 HDL 콜레스테롤은 유청단백질 수준과 가수분해물 섭취에 따른 차이를 보이지 않았다. 간 조직의 총 지질, 중성지방과 총 콜레스테롤은 가수분해물 섭취에 따라 차이가 없었으며, 고단백식이에 의해 유의적으로 감소하였다(p<0.05). 일일 분 배설량은 고단백식이에 의해 유의적으로 증가하였다(p<0.05). 분 중 총 지질함량과 중성지방 함량은 가수분해물 섭취 시 그 함량이 유의적으로 증가하였다(p<0.05). 반면, 분 중 총 콜레스테롤 함량은 유청단백질 수준이나 가수분해물 섭취에 따른 영향을 받지 않았다. 혈청 중 glucose와 insulin 농도는 가수분해물 섭취 시 유의적으로 감소하였다(p<0.05). 혈청 중 leptin 농도는 유청단백질 수준이나 가수분해물 섭취에 따른 유의적인 차이를 보이지 않았다. 혈청 중 ghrelin 농도는 고단백 식이에 의해 유의적으로 감소하였으며(p<0.05), 특히, 가수분해물 섭취 시 감소하는 경향을 보였다. 이상의 결과로부터 유청단백질 가수분해물의 섭취는 intact 유청단백질보다 흰쥐의 혈청과 간의 지질 농도를 저하시키고, 분 중으로의 지질배설량을 증가시키는 것으로 나타났다. 또한 혈중 glucose 농도와 insulin 농도를 감소시킴으로서 인슐린 저항성을 개선시켰으며, ghrelin 감소를 통한 식욕조절 효과를 보였다. 한편, 본 연구에서 유청단백질 가수분해물이 체중감소나 조직의 지방량에는 영향을 미치지 않았는데, 이것은 본 연구에서 사용된 25% 단백질 수준이 체중감소나 조직 지방량에 영향을 미치기에는 부족한 수준이라 사료되며, 단백질 수준에서의 증가 또는 보다 장기간 동안의 섭취가 수행되어진다면 식욕 촉진 호르몬 분비 감소를 통해 체중 감소에 있어서 더 유의적인 효과가 나타날 것으로 사료된다.

철에 노출된 넙치, Pacalichthys olivaceus의 혈액화학적 변동 (Hemochemical Changes in Olive Flounder, Paralichthys olivaceus Exposed to Various Iron Concentrations)

  • 주요 양식어류인 넙치를 대상으로 $0.1\sim10mg/\ell$ 범위의 철 농도에서 순환식 방법에 의해 50일 동안 사육하면서 혈액화학변동을 조사하였다. 넙치의 혈청 iron과 magnesium 농도는 1.0 mg/$\ell$의 철 농도에서 20일째부터 유의한 증가를 나타냈다. 혈청 calcium농도는 철 농도 1.0 mg/$\ell$에서 는 30일째부터 유의한 감소를 나타냈으나, inorganic phosphate의 뚜렷한 농도변화는 관찰되지 않았다. 혈청 total protein, albumin, glucose 농도는 철 농도 1.0 mg/$\ell$에서 노출 30일째부터 유의하게 감소하였고, total cholesterol 농도는 1.0 mg/$\ell$ 철 농도에서 노출 40일째부터 유의한 증가가 관찰 되었다. 혈청 GOT, GPT, LDH 및 amylase 활성은 철 농도 1.0 mg/$\ell$에서 40일째부터 유의하게 증가하였으나, Al-P의 뚜렷한 변동은 관찰되지 않았다. 이상의 결과는 철에 노출된 넙치 일부의 혈청 무기성분, 유기성분 및 효소활성은 철 농도 1.0mg/$\ell$ 이상에서 적어도 30일째부터는 변동한다는 것을 의미한다.

Free Radical Scavenging Activity and Protective Ability of Methanolic Extract from Duchesnea indica Against Protein Oxidation and DNA Damage

  • The antioxidant potency of methanolic extract of Duchesnea indica (MDI; Indian strawberry) was investigated by employing various established in vitro systems, such as total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, reducing power assay, metal chelating assay, superoxide radical scavenging activity and protective ability of DNA damage and protein oxidation. MDI inhibited metal chelating by 75.57% at 2 mg/mL, scavenged 50% DPPH free radical at 29.13 ${\mu}$g/mL, and eliminated approximately 46.21% superoxide radical at the concentration of 1 mg/mL. In addition, MDI showed strong ability on reducing power, DNA damage protection and protein oxidation protection. Overall, results suggested that MDI might be beneficial as a potent antioxidant and effectively employed as an ingredient in food applications.

Insulin-like growth factor-I 이 치주인대세포의 생물학적 활성도에 미치는 영향에 대한 연구 (THE STUDY ON THE EFFECTS OF THE INSULIN-LIKE GROWTH FACTOR-I ON THE BIOLOGICAL ACTIVITY OF THE HUMAN PERIODONTAL LIGAMENT CELLS)

  • 치주조직재생에 중요하게 생각되는 요건으로는 치근면의 상태, 전구세포의 증식, 치유 부의 상피조직배제, 치유부의 안정화를 들 수 있으며 이중 가장 중요한 요건중의 하나가 치유부에 치주조직재생을 도모할 수 있는 전구 세포가 실수부로 이주하여 부착과 증식, 분화를 통하여 교원질섬유를 포함한 결체조직의 부착과 백악질, 골조직을 재형성하는 것이다. 최근에 이러한 전구세포들을 자극하고 원치 하는 세포들을 저지하기 위한 방법으로 성장 인자에 대한 연구가 활발히 진행되고 있다 골조직을 조절하는 인자로 알려진 인슐린유사성장인자- I (Insulin-like growth factor-I)는 폴리펩타이드계 성장인자로서 골세포의 증식, 기질합성 등을 촉진시킨다고 보고되고 있으나, 치주조직 재생에 대한 IGF- I 의 영향을 잘 규명되어 있지 않으므로 배양된 치주인대세포에 IGF- I 을 농도별로 주입하여 세포의 증식능, 교원질 및 단백질 합성능, 알카린인산효소활성도를 측정해 보므로써 IGF- I 이 치주인대세포의 활성에 미치는 영향을 알아보고자 하였다. 교정치료를 위해 내원한 환자로부터 건강한 제일소구치를 발거하여 치주인대세포를 분리, 배양하여 IGF- I 을 주입시키지 않은 군을 대조군으로 하고, IGF- I 을 각각 0.1, 1, 10, 100 ng//ml로 주입시킨 군을 실험군으로 하여 DNA합성능, 총단백질과 교원질 합성능 및 알카린인산효소활성도를 측정하여 다음과 같은 결과를 얻었다. DNA 합성능에 미치는 IGF- I 의 효과는 농도가 증가함에 따라 0.1ng/ml를 제외하고는 DNA 합성능이 증가하는 경향을 보였고, 대조군에 비해 10, 100ng/ml투여군에서 통계적으로 유의한 차이(P<0/05)를 나타내었다. 치주인대세포의 총단백질 합성양에 미치는 IGF- I 의 효과는 농도가 증가함에 따라 총단백질 합성양이 증가하는 경향을 보였으며, 대조군에 비해 1, 10, 100ng/ml 투여군에서 통계학적으로 유의한 차이(P<0.001)를 나타내었다. 총단백질을 교원질(collagenase digestible protein : CDP)과 비교원성 단백질(non-collagenous protein : NCP)로 분류하여 비교하였을때 IGF- I 의 농도가 증가함에 따라 비교원성 단백질 합성양과 교원질 합성양이 증가하는 경향을 보였으며, 비교원성 단백질 합성양이 교원질 합성양보다 약간 높게 나타났고, 대조군에 비해 1, 10, 100ng/ml 투여군에서 통계적으로 유의한 차이(P<0.05, P<0.001)를 나타내었다. 총단백질에 대한 교원질합성의 상대적 비율은 농도가 증가함에 따라 각 군당 별차이를 보이지 않았으며, 대조군에 비해 통계적으로 유의한 차이 (P>0.05)를 나타내지 않았다. 알카린인산효소활성도에 미치는 IGF- I 의 효과는 모든군에서 7일째보다 14일째에서 약간 높은 알카린인산효소활성도롤 나타내었으며, 7, 14일 모두 농도가 증가함에 따라 효소활성도가 증가하였으며, 7일째 대조군에 비해 100ng/ml 투여군에서 통계적으로 유의한 차이(p<0.05)를 나타내었다.

식이섬유 종류가 고콜레스테롤 식이를 급여한 흰쥐의 체내 지질대사 및 신장기능에 미치는 영향 (Effects of Dietary Fiber Sourecs on Lipid Metabolism and Kidney Function in Rats Fed High Cholesterol Diet)

  • This study was conducted to investigate the effects of dietary fibers (cellulose ; polydextrose ; pectin ; ricebran) supplementation on the kidney fucntion of hyperlipidemic-induced rats on a high cholesterol diet (5% cholesterol). Serum lipid concentrations were not influenced by dietary fiber sources, but the pectin group was effective in reducing serum lipid levels. Total serum protein and albumin concentrations tended to higher in the polydextrose group, and serum creatinine and urea-N concentrations were higher in the pectin group than those in the other groups. Even though total urinary protein was high in the pectin group, GFR was reduced (18~30% decrease) as compared to other groups. The urea-N level was elevated in the polydextrose group as compared to that of cellulose group(25% increase). Total lipid, triglyceride and cholesterol concentrations in the liver of pectin group were lower than those of other groups. Total lipid and cholesterol concentrations in the kidneys were reduced in pectin and ricebran groups, respectively. The total cholesterol concentration infecal was significantly high in the polydextrose and pectin groups(p<0.05), and the triglyceride was highest in the pectin group. These results indicate no significant effects of dietary fiber supplementation on the kidney function of hyperlipidemic-induced rats on a high cholesterol diet, but pectin was very effective to improve lipid metabolism and to reduce GFR.

곡아지실소시호탕(穀芽枳實小柴胡湯)의 간보호작용(肝保護作用)에 관(關)한 실험적(實驗的) 연구(硏究) (An Experimental Study on the Hepatoprotective Effect of Gokajisilsosiho-Tang)

  • This study was performed to elucidate the effects of Gokajisilsosiho-Tang(GJST) on the lactic dehydrogenase(LDH) release, cell viability and activity, lipid peroxidation, DNA synthesis and the changes of total protein synthesis and GSH changes in vivo and in vitro in rat cultured hepatocytes from hydrogen peroxide$(H_2O_2)$-induced injury. The GJST extract had not an effect on cytotoxicity in all experimental results. The treatment of GJST extract of $160{\mu}g/ml$, $320{\mu}g/ml$ showed the significant effect to decrease LDH leakage induced by t-BHP in cultured rat hepatocytes. The higher concentration of GJST extract than $160{\mu}g/ml$, showed the inhibitory effect on decreasing cell viability induced by t-BHP. The treatment of t-BHP to rat cultured hepatocytes resulted in a concentration dependent increase in TBARS, in the presence of GJST extract the production of TBARS induced by hydrogen peroxide was inhibited concentration dependently, significantly inhibited at $80{\mu}g/ml$ of GJST extract and above. The GJST extract simutaneously present with t-BHP prevented the loss of total protein and GSH in a concentration dependent manner. These results suggested that GJST extract may play a hepatoprotective role in oxidative damage induced by hydrogen peroxide and a therapeutic potential of inhibiting liver injury.

발효홍차가 당뇨쥐의 혈청 단백질농도 및 효소활성에 미치는 영향 (Effects of Tea Fungus/Kombucha Beverage on Serum Protein Levels and Enzyme Activity in Streptozotocin-Induced Diabetic Rats)

  • This study investigated the effect of tea fungus/kombucha beverage(TF) protein concentrations and enzyme activities in serum of both normal and diabetic male rats. Sprague Dawley growing rats were randomly assigned to one control and five diabetic groups. In five diabetic groups, D control group was fed drinking water and the other groups were fed drinking water supplemented with 20 or 40% TF (20 or 40% TFD group, respectively) and 20 or 40% disinfected TF(20 or 40% TFSD group, respectively) for 7 weeks. Diabetes was experimentally induced in all five diabetic groups by streptozotocin injection after 3 week feeding. The diabetic groups were significantly decreased the body weight( 29.4~ 48.6g) compared with those in control group(72.4g). The total liver and kidney weights in all diabetic groups were similar to those in control group, but those relative to body weights in all diabetic groups were heavier than those in control group. The total spleen weight in all diabetic groups was significantly decreased compared with those in control group, but those relative to body weights in all diabetic groups were similar to those in control group. The blood glucose levels were heigher in all diabetic groups than those in control group. The alkaline phosphatase activity in serum was higher in all experimental groups than those in control group, but it was lower in 40% TFD, 20% and 40% TFSD groups than those in D control group. The GPT activity was significantly increased in D control, 20% and 40% TFD groups than in control group. The GOT activity was significantly increased in D control goup than in control group, but those in all TFD and TFSD groups were similar to control group. The total protein concentration in all diabetic groups was significantly decreased compared with that in control group, but the albumin concentration showed almost the same levels in all the experimental groups. The ratio of albumin/globulin, and hem atocrit value were significantly increased in all diabetic groups than in control group. These results show that tea fungus/kombucha beverage with which diabetic rats were fed has not recovered the decreased body weight, lowered serum total protein level, hypertrophy of liver and kidney, hyperglycemia to the normal state.

소아의 다발성 치아우식증과 연관된 타액의 생화학적 특성 (BIOCHEMICAL CHARACTERISTICS OF SALIVA TO BE LINKED TO THE MULTIPLE CARIES IN CHILDREN)

  • Saliva is obviously potential medium to protect the dental caries by not only physical clearing effect, but aggregating action of protein with bacteria. Nevertheless, we still do not understand how the dental caries occur and what brings the individual difference in caries prevalence. In the regards of dental caries prevalence, we hypothesized that the composition of salivary protein might be different from caries susceptible group to caries resistant group. The purposes of this experiment were focused on the molecular analysis of salivary proteins from the subjects who were involved in multiple caries. Electrophoretic analysis was done on the whole saliva collected from the children with and without multiple caries. We found 86.2% of subjects with multiple caries has approximately 120 KDa protein band while 30.4% in the healthy subjects. And the concentration of the total protein on the subjects with multiple caries is significantly higher than that of the healthy group. However, it turned out that the difference of the salivary composition does not affect the bacterial adhesion to hydroxyapatite bead. With regards of enzymes in saliva, the activity of ${\alpha}-amylase$ and lactate dehydrogenase does not have any significant difference between both groups. However, the concentrations of $Na^+\;and\;Cl^-$ in saliva from multiple caries group is higher than that of the control group. Taken all together, it may be concluded that 120 KDa protein in saliva may be associated with the process of dental caries, also the high concentration of protein and $Na^+,\;Cl^-$ in saliva may be linked to dental caries development as a cofactors.

Black soldier fly larvae meal supplementation in a low protein diet reduced performance, but improved nitrogen efficiency and intestinal morphology of duck

  • Objective: Reduced crude protein (CP) diets offer potential benefits such as optimized feed efficiency, reduced expenses, and lower environmental impact. The objective of this study was to evaluate black soldier fly larvae (BSFL) meal on a low-protein diet for duck performance, blood biochemical, intestinal morphology, gastrointestinal development, and litter. Methods: The experiment was conducted for 42 days. A total of 210-day-old male hybrid ducklings (5 replicate pens, 7 ducks per pen) were randomly assigned to 6 dietary treatments (3×2 factorial arrangements) in randomized design. The factors were CP level (18%, 16%, 14%) and protein source feed soybean meals (SBM), black soldier fly larvae meals (BSFLM). Results: Reduced dietary CP levels significantly decreased growth performance, feed intake, the percentage of nitrogen, pH (p<0.05), and tended to suppress ammonia in litter (p = 0.088); increased lipid concentration; and enhanced relative weight of gastrointestinal tracts (p<0.05). In addition, dietary BSFL as a source of protein feed significantly increased lipid concentration and impacted lowering villus height and crypt depth on jejunum (p<0.05). Conclusion: In conclusion, the use of BSFLM in a low-protein diet was found to have a detrimental effect on growth performance. However, the reduction of 2% CP levels in SBM did not have a significant impact on growth performance but decreased nitrogen and ammonia concentrations.

정상치은열구액과 치주질환시 치은열구액의 단백질 조성비교에 관한 연구 (THE PROTEIN COMPOSITION OF GINGIVAL CREVICULAR FLUID SAMPLED FROM NORMAL SUBJECTS AND PATIENTS WITH PERIODONTAL DISEASE)

  • Gingival crevicular fluid (GCF) is a promising source for markers of destructive periodontal disease activity. This study was undertaken to evaluate the protein composition of GCF in varying stages of the gingival inflammatory response. GCF sampled from 26 people with clinically healthy gingiva and 18 people with periodontitis were examined via sodium dodecyl sulphate polyacrylamide gel electrophoresis(SDS/PAGE). The result were as follows. 1. Total amount of GCF protein of diseased group significantly different from that of normal group. But difference in protein concentration was not that significant. 2. In analyzing GCF with SDS/PAGE, it was suggested that albumin is used as indicator plasma protein leakage because of heavily staining bond of albumin in patients with periodontal disease. 3. In diseased group, overall bonds of protein and bands of high molecular weight protein were heavily stained. It was proved useful information on high molecular plasma protein leakage with increasing vascular permeability due to inflammation.
