금은화 추출물을 이용한 미생물 발효 생성물의 항산화 특성 (Antioxidant Properties in Microbial Fermentation Products of Lonicera japonica Thunb. Extract)

  • 최근에 합성 항산화제를 대체할 천연물에서 새로운 항산화 물질을 찾는 연구가 활발하게 진행되고 있다. 천연물 중에 Lonicera japonica Thumb은 항균, 항염증, 항바이러스, 해독작용, 혈중 지질 감소, 해열작용 등 약리적인 효능뿐 아니라 다양한 항산화 물질을 포함하고 있는 것으로 보고되어 있다. 본 연구는 금은화 열수, 알코올 추출물을 기질로 전통발효식품에서 분리한 Lactobacillus plantarum NHP1 균주로 발효과정을 진행한 후 항산화 물질 및 래디컬 소거능이 변화하는지를 관찰하기 위해 진행되었다. 전통 발효식품으로부터 발효를 위한 균주를 분리한 후 16S rDNA 염기서열을 비교하여 동정한 결과 Lactobacillus plantarum NHP1로 명명하였다. 발효를 위한 기질인 Lonicera japonica Thumb는 열수 및 70% ethanol을 이용하여 추출하였다. 열수 및 ethanol 추출물의 발효는 2 L 발효기에 1.2 L 추출물을 함유한 배지를 공급하고, $30^{\circ}C$ 및 100 rpm의 조건으로 48 시간 수행하였다. 발효 전 후의 총 페놀, 총 플라보노이드, DPPH 소거능 및 ABTS 변화를 측정한 결과, 발효 전 총 페놀양은 열수 추출물과 ethanol 추출물에서 $56.5{\pm}4.9$ GAE mg/g, $72.7{\pm}5.3$ GAE mg/g이었고, 발효 후 각각 30.2%, 12.9% 증가되었다. 총 플라보노이드 함량은 모든 용매에서 발효 후에 유의적으로 약 75% 이상 증가되었다. 발효 후 총 페놀 함량과 플라보노이드 함량이 증가한 것에 비해 DPPH 소거능은 비례적으로 증가되지 않았으나, ABTS 항산화능은 발효에 의해 50% 증가되었다. 그동안 선행 연구에서 금은화의용매 추출물에 대한 기능성연구를 진행한 것이 대부분인 것에 비해, 본 연구 결과를 바탕으로 금은화 추출물에 발효기술을 접목한다면 항산화 기능 및 다른 기능성이 대폭 증가되어 기능성 건강식품 및 미용식품의 소재로의 사용이 활성화 될 수 있을 것이라 사료된다.

수산폐기물 재활용을 위한 미더덕껍질 용매 추출물의 생리활성 탐색 (Screening of Biological Activity of Solvent Extract from Styela clava Tunic for Fishery Waste Recycling)

  • Styela clava tunic is generated in large amounts as a waste from S. clava processing plants and causes environmental problem. Although biological activities of S. clava were reported by many investigators, study on S. clava tunic was little. In this study, therefore, tyrosinase inhibition and antioxidative activities of extracts from S. clava tunic using different solvent were investigated for recycling of the fishery waste. Among extraction methods tested, autoclaved extraction (25.7%) and hot water extraction (18.2%) appeared to be effective for extraction. The highest total phenolic content was 46.6 mg/g in autoclaved extract while the highest flavonoid content was 23.0 mg/g in chloroform extract. All extracts possessed tyrosinase inhibition activity and the inhibition activity was concentration-dependent. Inhibition concentration ($IC_{50}$) against tyrosinase activity was $0.36{\times}10^4$ mg/ml in ethanol extract, $0.11{\times}10^3$ mg/ml in acetone extract and 0.27 mg/ml in n-butanol extract. Among extracts tested, hot water and autoclaved extracts displayed higher antioxidative activity than organic solvent extracts. Therefore, our data suggest that extract from S. clava tunic may potential candidate for cosmetic product with whitening effect and medicine for diseases caused by various oxidative stresses.

클러스터 분석을 이용한 황련 추출물 항산화 활성 최적화 추출공정 (Optimization of the Extraction Process and Antioxidant Capacity of Coptis chinensis Franch Extract through Cluster Analysis)

  • 황련은 여러 가지 질병 치료에 사용되는 효과적인 전통약용식물이다. 하지만 황련 추출물의 생리활성성분 함량에 영향을 미치는 주요 인자 및 생리활성성분과 항산화 효능의 상관관계는 아직 알려지지 않았다. 따라서 본 논문에서는 용매, 온도 및 추출 시간이 황련 추출물의 생리 활성성분의 함량과 항산화 활성에 미치는 영향을 조사하였다. 클라스터를 이용하여 분석한 결과, $35^{\circ}C$에서 추출시간 30 min인 에탄올(50% 에탄올 : 50% 물) 조건으로 추출 시, 생리활성성분 함량 및 항산화 활성이 제일 높으므로 공장에서 사용하기에 가장 적합한 추출조건임을 확인하였다. 다중선형회귀분석 결과, 총 페놀화합물 함량은 DPPH소거 활성에 기여하였으며 총 알칼로이드 함량과 총 플라보노이드 함량은 FRAP활성에 기여하였다. 이상의 결과들로부터 황련의 추출 조건이 생리 활성 화합물의 수율 및 항산화 활성을 조절가능하며, 이는 공업적인 응용에 중요한 정보를 제공할 수 있을 것으로 사료된다.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.

건조방법에 따른 고구마 식물체 부위별 항산화특성 (Antioxidant Characteristics of Sweet Potato (Ipomoea batatas (L.) Lam.) according to Different Plant Parts and Drying Methods)

  • This study investigated the antioxidant characteristics of sweet potato according to different plant parts and drying methods. The sweet potato plant parts were divided into root tubers, stems, stalks, leaves, and tips, and the drying methods were freeze-drying and hot air drying. Total polyphenol and flavonoid contents and radical scavenging activity of the sweet potato plant parts were significantly different depending on the plant parts and drying methods. The total polyphenol content of freeze-dried sweet potato leaves and tips were 52.76 and 46.19 mg chlorogenic acid equivalents/g sample, and the total flavonoid contents were 222.47 and 214.12 mg quercetin equivalents/g sample, respectively, and decreased with hot air drying. DPPH radical scavenging activity was higher in freeze-drying than hot air drying and was significantly different depending on the plant parts. The ABTS radical scavenging activity of freeze-dried sweet potato leaves and tips were 43.48 and 44.68 mg Trolox equivalents/g sample, respectively, and decreased with hot air drying. Therefore, additional studies on the functionality of using by-products from sweet potato cultivation are needed.

녹차 추출물 및 프로폴리스의 항산화 및 갈변억제 효과 (Antioxidative Activities and Antibrowning Effects of Green Tea Extracts and Propolis)

  • This study was designed to investigate the properties related to browning of hot water and 80% ethanol extracts of green tea (Camellia sinensis) and propolis. The extracts were assessed for polyphenol oxidase (PPO) inhibitory activity and total phenolic and flavonoid contents. In addition, apples were cut into 15 mm thick slices and dipped for 1 min in 0.5% of the green tea extracts and the propolis solutions and stored at room temperature for 24 h. The PPO inhibitory activity of the green tea extracts was better than that of the propolis. The highest DPPH free radical scavenging activity (90.81%), total phenol contents (170.42 mg/mL) and the metal chelating effects (45.93%) were measured in the green tea hot water extracts. The mineral content of the green tea water extracts was 69,328.44 ppm potassium and 2,409.42 ppm magnesium. After 24 h, the ${\Delta}E$ value of the apple slices treated with the green tea water extracts was the lowest (1.35). The antibrowning effects of the green tea extracts were higher than those of propolis in the apple slices.

Anti Tumoral Properties of Punica Granatum (Pomegranate) Peel Extract on Different Human Cancer Cells

  • Background: Medicinal plants, especially examples rich in polyphenolic compounds, have been suggested to be chemopreventive on account of antioxidative properties. Punica granatum (PG) (pomegranate) is a well known fruit in this context, but its cytotoxicity in cancer cells has not been extensively studied. Here, we investigated the antiproliferative properties of a peel extract of PG from Iran in different human cancer cells. Materials and Methods: A methanolic extract of pomegranate peel (PPE) was prepared. Total phenolic content(TPC) and total flavonoid conetnt (TFC) were determined by colorimetric assays. Antioxidant activity was determined by DPPH radical scavenging activity. The cytotoxicity of different doses of PPE (0, 5, 20, 100, 250, 500, $1000{\mu}g/ml$) was evaluated by MTT assays with A549 (lung non small cell cancer), MCF-7 (breast adenocarcinoma), SKOV3 (ovarian cancer), and PC-3 (prostate adenocarcinoma) cells. Results: Significant (P<0.01) or very significant (P<0.0001) differences were observed in comparison with negative controls at all tested doses (5-$1000{\mu}g/ml$). In all studied cancer cells, PPE reduced the cell viability to values below 40%, even at the lowest doses. In all cases, IC50 was determined at doses below $5{\mu}g/ml$. In this regard, MCF-7 breast adenocarcinoma cells were the most responsive cells to antiprolifreative effects of PPE with a maximum mean growth inhibition of 81.0% vs. 69.4%, 79.3% and 77.5% in SKOV3, PC-3 and A549 cells, respectively. Conclusions: Low doses of PPE exert potent anti-proliferative effects in different human cancer cells and it seems that MCF-7 breast adenocarcinoma cells are the most cells and SKOV3 ovarian cancer cells the least responsive in this regard. However, the mechanisms of action need to be addressed.

벌꿀과 벌집채꿀의 페놀성분 및 항산화 효과 비교 (Comparative Phenolic Composition and Antioxidant Properties of Honey and Honeycomb Extracts)

  • 본 연구에서는 야생화로부터 채밀한 제주 지역 벌꿀 및 벌집채꿀의 페놀성분 분석과 항산화 활성 비교평가를 시행하였다. 총 폴리페놀 및 플라보노이드 함량 측정 결과 물을 용매로 사용한 벌집채꿀에서 가장 높은 함량을 나타냈다. 또한 벌꿀 및 벌집채꿀 모두 농도 의존적으로 항산화 활성이 증가하는 양상을 보였으며, 이를 EC50 및 EC0.5 값으로 환산한 결과 벌집채꿀이 벌꿀에 비해 통계적으로 유의하게 높은 항산화 활성을 나타내었다. 상기의 결과를 종합하여 봤을 때 총 폴리페놀 및 플라보노이드 함량과 항산화 활성간의 높은 연관성이 있다는 것을 추측 해볼 수 있었고 이를 상관관계를 통해 분석한 결과, 총 페놀 및 플라보노이드 성분의 함량과 항산화 활성이 통계적으로 유의한 상관관계를 나타냄을 확인할 수 있었다. 이러한 결과들은 잠재적 천연식품소재로의 벌집채꿀의 가능성을 시사하며 향후 이에 대한 체계적인 연구가 필요하다 사료된다.

찔레꽃 추출물의 용매별 항산화 활성 (Antioxidant Activities of Solvent Extracts from Rosa multiflora)

  • 본 연구는 찔레의 열수추출물과 에탄올추출물간의 항산화 활성 비교를 위해 in vitro에서 총 페놀 및 플라보노이드 함량을 측정하였으며, DPPH, ABTS, NO radical 소거능 및 환원력을 $10{\sim}500{\mu}g/ml$의 농도에서 실험하였다. DPPH radical 소거능은 열수추출물(4.95~92.15%)과 에탄올추출물(2.76~90.55%)이 모두 농도 의존적으로 유의성 있게 높아졌으며 열수추출물이 더 높은 소거능을 보였다. ABTS radical 소거능을 비교한 결과 열수추출물(4.44~92.99%)이 에탄올추출물(2.63~87.12%)에 비해 유의적으로 높은 소거활성을 나타내었다. 환원력은 열수추출물(0.256~0.775)이 에탄올추출물(0.234~0.568)에 비해 유의적으로 높은 활성을, 그리고 농도 의존적으로 높아지는 것을 보였으며, NO radical 소거능을 비교한 결과 열수추출물이 에탄올추출물보다 조금 높은 활성을 보였고 농도 의존적으로 높아지는 것을 확인하였다. 찔레추출물의 항산화력의 $IC_{50}$값을 비교한 결과 DPPH radical 소거능, ABTS radical 소거능에서 각각 열수추출물에서 79.73, $79.82{\mu}g/ml$으로 나타났고, 에탄올추출물의 경우 145.85, $159.03{\mu}g/ml$로 나타났다. 총 페놀함량은 열수추출물에서 140.74 mg/g, 에탄올추출물에서 37.83 mg/g 플라보노이드 함량은 열수추출물에서 45.31 mg/g, 에탄올추출물에서 42.68 mg/g로 에탄올 추출물에 비해 열수추출물이 높게 나타났다. 결론적으로 찔레꽃 추출물은 항산화 효능을 나타내는 실험들에서 모두 높은 수치를 나타내어 천연 항산화제로 이용가치가 높다고 사료된다.

해동조건에 따른 냉동마늘의 품질 특성 (Effects of Various Thawing Conditions on Quality Characteristics of Frozen Garlic)

  • This study investigated the effects of thawing conditions on physiological activities and quality of peeled garlic. Peeled frozen garlic was analyzed after thawing at low temperature ($4^{\circ}C$), room temperature ($20^{\circ}C$), tap water ($20^{\circ}C$), radio frequency of 27.12 MHz, and 2.45 GHz in a microwave. As a result, the time required to thaw garlic to $0^{\circ}C$ by various thawing methods was shortest at2.45 GHz in a microwave, followed by $20^{\circ}C$ tap water, radio frequency of 27.12 MHz, $20^{\circ}C$, and $4^{\circ}C$. Microwave thawing was faster than other methods, but it resulted in significant non-uniformity of heating. The hardness of peeled garlic significantly decreased upon freeze-thawing, whereas it showed improved hardness upon radio-frequency thawing. Total color difference in garlic increased upon freeze-thawing, and it was not improved by various thawing methods. Antioxidant activities were determined for DPPH radical scavenging ability, SOD-like activity, and reducing power. Total phenolic compounds and flavonoids in garlic extract were measured as $3.222{\pm}0.214{\mu}g$ GAE/g and $0.149{\pm}0.03{\mu}g$ QE/g, respectively. The content of total phenolic compounds was significantly reduced by 2.45 GHz microwave thawing ($1.90{\pm}0.02{\mu}g$ GAE/g); however, flavonoid contents were slightly reduced under freezing and thawing conditions. The DPPH radical scavenging ability of garlic extracts was not affected by thawing methods; however, SOD-like activity and reducing power were slightly reduced by freeze-thawing. These results indicate that physiological activities were not improved by radio-frequency thawing; however, thawing time and maintain hardness were reduced compared with conventional thawing methods.
