A laboratory experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and cytotoxicity from methanol extracts of different plant parts of $T.$$officinale$ F. H. Wigg. Total phenolics [mg chlorogenic acid equivalents (FAE) $kg^{-1}$ DW] was highest in flower extracts (72.0 mg $kg^{-1}$), followed by leaf, root, and stalk extracts of $T.$$officinale$ ($p$ < 0.05). The result of total flavonoid level [mg naringin equivalents $kg^{-1}$ DW] had same tendency to differential total phenolics contents among plant parts, but showed lower ranges of amount. The antioxidant activity of the methanol extracts from all the plant parts dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was highest in flower extracts ($IC_{50}$ value = 624.3 mg $kg^{-1}$ ), and followed by leaf, root, and stalk extracts of $T.$$officinale$ ($p$ < 0.05). By means of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, cell viability of Calu-6 for human pulmonary carcinoma and SNU-601 for human gastric carcinoma showed the lowest $IC_{50}$ value in the flower extracts ($IC_{50}$ value = 85.7 and 311.4 mg $kg^{-1}$, respectively), indicating the highest cytotoxicity. The results suggested that total phenolics content and total flavonoids level in different plant parts of $T.$$officinale$ were highly correlated with antioxidative ($r^2$=0.7280 to 0.9971) or with cytotoxic activities ($r^2$=0.5795 to 0.9515).

Flavonoids are known to be effective scavengers of free radicals. In particular, proanthocyanidins are flavonoids that possess cardiovascular protection, antioxidative activities, and immunomodulatory activities. Here, we evaluated proanthocyanidin contents in the total polyphenolic compounds of pine needle extracts prepared by hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE). Analysis of each extract indicated that the ethanol extract contained the highest proanthocyanidin concentration. The HWH and hexane extracts also contained relatively high concentrations of proanthocyanidin. On the other hand, proanthocyanidin content analyses out of the total polyphenolic compounds indicated that the HWH extract contained the highest content. These results suggest that HWH extraction is a suitable method to obtain an extract with a high level of pure proanthocyanidins and a relatively high yield. The HWH extract possessed superior activity in diverse antioxidative analyses such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferrous ion chelating (FIC), and ferric-ion reducing power (FRAP) assays. In addition, upon assessing the effects of the pine needle extracts on macrophages (Raw 264.7 cell), the HWH extract exhibited the highest activity. In this study, we discerned an efficient extraction method to achieve relatively pure proanthocyanidins from pine needles and evaluated the biological functions of the resulting extract, which could potentially be used for its efficacious components in functional food products.
In this studies, contents and activities of antioxidative substances in the fruits of a pear cultivar and five Korean wild pear lines were investigated. The content of total phenolic compounds was highest with Pyrus pyrifolia var. culta "Shingo" (7.68 $mg{\cdot}g^{-1}$, D.W.) while total flavonoids was highest with P. ussuriensis var. acidula (5.66 $mg{\cdot}g^{-1}$, D.W.). The contents of total polyphenols and flavonoids varied according to plant species, but in general pyrifolia lines had higher total polyphenols than total flavonoids whereas in ussuriensis lines total flavonoid contents were higher than total polyphenols. The scavenging effects of DDPH radicals was highest with highest with P. ussuriensis var. seoulensis ($RC_{50}$=1.45 $mg{\cdot}mL^{-1}$) and that of ABTS radicals was highest with P. ussuriensis var. acidula ($RC_{50}$=0.69 $mg{\cdot}mL^{-1}$). A pear cultivar "Shingo" showed highest total polyphenol content, but scavenging activity of DDPH and ABTS radicals was lowest. The fruits of P. ussuriensis var. seoulensis contained low level of phenolic compounds, but their scavenging effects on two radicals was superior. Ferrous ion chelating effect was high in P. pyrifolia var. culta (98.93%), and in P. ussuriensis var. sinensis (68.71%), but two species had low scavenging activity. The content of phenolic compounds in wild pears was not always directly proportional to the radical scavenging activity. Neither radical scavenging activity was to ferrous ion chelating activity. In conclusion the fruits of P. ussuriensis var. acidula, P. ussuriensis var. seoulensis, and P. ussuriensis var. sinensis had great potential for the development of new antioxidative functional food because their antioxidative activity is higher or at least similar to P. pyrifolia var. culta. However, due to their different levels of antioxidative effects, the development of methods of using them jointly, not singly, to obtain maximum level of antioxidants is recommended.
In order to obtain basic data for utilization of the aerial parts of Cnidium officinale Makino (APCO), the antioxidant properties of the aerial parts and rhizomes of C. officinale were measured using DPPH and ABTS radicals, and nitrite scavenging assays. The ethyl acetate (EA) fraction prepared from the aerial parts of APCO showed the strongest antioxidant activities, and contained high level of total phenolic compounds (325.81 mgTE/g) and flavonoids (259.16 mgRE/g). The concentrations for 50% reductions ($RC_{50}$) values of the DPPH and ABTS radicals, and nitrite by the EA fraction of APCO were $11.27\;{\mu}g/m{\ell}$, $14.34\;{\mu}g/m{\ell}$, and $10.26\;{\mu}g/m{\ell}$, respectively. APCO exhibited approximately 3-9 times higher antioxidant activity than rhizomes of C. officinale. The antioxidant capacities of APCO were positively correlated with its total phenolic contents. Therefore, it was concluded that the aerial parts of C. officinale can be a useful and cost-effective source of natural antioxidant for food or cosmetics.
Monacolin K-enriched ganghwayakssuk (Artemisia princeps Pamp.) was developed by fermentation with Monascus sp. Among the 15 Monascus spp. isolated previously from Monascus fermentation products, Monascus pilosus KMU108 produced 2,219 mg/kg of monacolin K during ganghwayakssuk fermentation with no detectable citrinin. The optimum concentrations of ganghwayakssuk and glucose determined from the response surface methodology (RSM) design were 2.2% and 3.8%, respectively. By applying these conditions, the monacolin K productivity was increased to 3,007 mg/kg after 15 days of fermentation. On the other hand, other characteristics such as the total content of flavonoids and phenolic compounds, and the antioxidant activity were relatively unchanged. Therefore, Monascus-fermented ganghwayakssuk is an excellent biomaterial for the development of functional foods because of its high level of monacolin K, known to lower cholesterol levels.
The present study deals with the amelioration by Lagerstroemia speciosa Linn. leaf extract against hepatotoxicity induced by carbon tetrachloride ($CCl_4$), which was evaluated in terms of serum marker enzymes like serum glutamate pyruvate transaminase, serum glutamate oxaloacetate transaminase, alkaline phosphatase, serum total bilirubin, total protein levels along with concomitant hepatic and antioxidants like superoxide dismutase, catalase, glutathione, glutathione peroxidase and lipid peroxidation enzymes were monitored. These biochemical parameters altered by the single dose level of $CCl_4$ (0.75 ml/kg body weight, i.p). Pre treatment with L. speciosa prior to the administration of $CCl_4$, at the doses of 50 and 250 mg/kg. body weight/day, p.o. for 7 days, significantly restored all the serum and liver tissue parameters near to the normal levels, respectively. Silymarin was used as a reference standard, prior to the administration of $CCl_4$ to rats. These findings indicate the protective potential of L. speciosa against hepato toxicity which possibly involve mechanism related to its ability of selective inhibitors of (reactive oxygen species like antioxidants brought about significant inhibition of TBARS suggesting possible involvement of $O_2{\cdot}-$, $HO_2{\cdot}$, and ${\cdot}OH$. In conclusion, the amelioration may be attributed to the synergistic effects of its constituents rather than to any single factor as the leaves are rich in tannins, sterols, flavonoids, saponins etc.
Plastic house and laboratory experiments were conducted to determine the effects of substrate components and shade degrees on plant growth and physiologically-active substance content and antioxidant activity of Panax ginseng. No significant difference in growth characteristics among substrates was observed. However, shade treatment also significantly reduced leaf area, chlorophyll content, root length, and shoot and root fresh weight (P<0.05) with increasing of the degree. Total phenolics level [mg ferulic acid equivalents (FAE) $kg^{-1}$ DW] was higher in root parts (25.8 to 28.4 mg $kg^{-1}$) than shoot parts (20.7 to 23.8 mg $kg^{-1}$) of Panax ginseng, and with shade (23.7 to 28.4 mg $kg^{-1}$) than without shade (20.7 to 25.8 mg $kg^{-1}$). However, total flavonoids [mg naringin equivalents $kg^{-1}$ DW] was more detected in the shoot part (17.4 to 26.8 mg $kg^{-1}$) than in the root part (6.7 to 7.0 mg $kg^{-1}$) of Panax ginseng. Methanol extracts from the Panax ginseng plants dose-dependently increased DPPH free radical scavenging activity as a antioxidant potential, and the activity was higher in shoot parts (31.1 to 36.5%) than in root parts (24.9 to 28.7%), and without shade than with shade. In conclusion, shade affects plant growth as well as physiological activity of Panax ginseng under plastic house condition.
Rubus coreanus Miquel (RCM), a type of red raspberry, grows wild in Korea and China and its unripe fruit is used as a folk medicine for the treatment of impotence and as a diuretic. RCM was extracted with methanol and then further fractionated it into for different types. In this study, we investigated the antioxidant activity of a RCM extract (ext.) and its fraction (fr.). DPPH free radical scavenging activity assay, total polyphenols contents, total flavonoids contents assay were used to analyze antioxidant activity. The DPPH free radical scavenging activity $(RC_{50}:1.67{\mu}g/ml)$ and total polyphenols contents $(546.25{\mu}g/mg)$ were higher in butanol fraction than in other fr. And total flavonoids contents was higher in ethylacetate fr. $(141.78{\mu}g/mg)$. We applied comet assay to measure the DNA damage in the individual cells and exposed time course at $IC_{50}$. Comet assay is a rapid and sensitive fluorescent microscopic method to examine DNA damage and repair at individual cell level. Sprout ginseng paste were prepared with pine nut, dry oyster and dry shrimp to examine the antioxidant properties(total polyphenols, total flavonoids, and electron donating ability) and sensory test(attribute difference and acceptance). Sprout ginseng paste were measured based on color value, pH, viscosity, total bacteria cell numbers for 0 and 20 days at $4^{\circ}C$. The higher total polyphenol and total flavonoid content of sprout ginseng paste added with pine nuts, dry oyster, and dry shrimp were higher antioxidant capacity. DPPH radical scavenging activity was increased from 52.2% (SGP0) to 79.5.0 % with SGP5. The attribute test results reveal that the color intensity, bitter taste, and oily taste were decreased in SGP3 to SGP6. Taste, flavor, and coarseness did not show significant difference in SGP0 to SGP6. Thickness and After taste were increased in SGP4 to SGP6. The preference test results reveal that the appearance, flavor, and texture level did not show significant difference in SGP0 to SGP6. Taste and overall preference were increased in SGP4 to SGP6. L value, pH, decrease while a value and b value show no change in sprout ginseng paste with pine nut, dry oyster and dry shrimp. Total cell number was not detected during storage.
This study was carried out to compare the major nutrient components and antioxidant activities of Stachys sieboldii Miq leaf and root powders. For proximate compositions, crude protein and crude fat contents of leaf powder were higher than those of root powder, whereas contents of crude ash and carbohydrates were lower in leaf powder. The content of glutamic acid was greater in amino acids of leaf and root powders, and contents of total amino acids and essential amino acids were higher in leaf powder compared with root powder. Root powder contained a higher level of total unsaturated fatty acids than leaf powder. Total contents of organic acids were higher in leaf powder; tartaric acid was the major organic acid in leaf powder, and malic acid was the major organic acid in root powder. The content of vitamin A was higher in leaf powder than in root powder. However, vitamin E content was higher in root powder than in leaf powder. Total mineral contents of leaf powder were higher than those of root powder, and mineral contents of leaf and root powders were in the order of K>Ca>Mg. Extract yields of leaf and root powders were 27.21% and 58.51%, respectively. Total polyphenols and total flavonoids of leaf extract were 236.35 mg/g and 1.90 mg/g, respectively, which were higher than those of root extract. The $IC_{50}$ values of leaf and root ethanol extracts based on DPPH hydroxyl scavenging were 0.69 mg/mL and 5934.31 mg/mL, respectively, and antioxidative activities of ethanol extracts from all Stachys sieboldii Miq parts dose-dependently increased. These results suggest that Stachys sieboldii Miq can be recommended as an edible functional food material.
