Identification of Carotenoids from Green Alga Haematococcus pluvialis by HPLC and LC-MS (APCI) and Their Antioxidant Properties

  • Haematococcus pluvial is, a green alga, accumulates astaxanthin (3,3'-dihydroxy-$\beta$,$\beta$'-carotene-4,4'-dione) upto 2-3% on a dry weight basis. In the present study, identification of carotenoids from Haematococcus cyst cell extract by HPLC and LC-MS (APCI) and their antioxidant properties were evaluated in in vitro model systems. The extract exhibited 89% and 78% antioxidant activities in the $\beta$-carotene linoleate model and the hydroxyl radical scavenging model, at 9 ppm of total carotenoid, respectively. The extract also showed 80%, 85%, and 79% antioxidant activities against lipid peroxidation in the kidney, brain, and liver of rats. Low-density lipoprotein oxidation induced by $Cu^{2+}$ ions was also protected (45%, 64%, and 75%) by the extract in a dose-dependent manner with different carotenoid levels. Thiobarbituric acid reactive substances concentration in the blood, liver, and kidney of rats were also significantly (p<0.005) decreased in H. pluvialis-treated rats. The potent antioxidant activity is attributable to various carotenoids present in the extract.

Carotenoid Pigments from Suspended and Sinking Particulate Matter in Prydz Bay, Antarctica

  • Suspended and sinking particles were collected during ODP Leg 119 to the Indian Ocean sector of the Antarctic Ocean. Field work was carried out at four sampling sites in Prydz Bay. Two of these sites were located in the Outer Bay, and two in the Inner Bay. At the four locations, a total of ten deployments of a sediment trap array were made. The concentrations of carotenoids both in suspended and sinking particulate matter in Prydz Bay were analyzed using HPLC. Fucoxanthin was the dominant carotenoid pigments both in suspended and sinking particles. The present study also indicates that 19'-hexanoyoxyfucoxanthin-containing prymesiophytes (Phaeocystis spp.) was abundant in the study area. The flux rates of carotenoids were generally highest at 50 m, and approximately double the flux rates at deeper horizons, however, at Inner Bay sites, the mean flux rates of carotenoids were greatest at 200 m, and 3 times greater than that of 50 m. Such anomalous high fluxes at 200 m imply that grazers were locally abundant between 100 m and 200 m at these sites close to land, and this hypothesis is supported by visual evidence of lots of fecal pellets in the 200 m trap. Integrates standing stocks versus sinking pigments data support that particulate material in Prydz Bay was not recycled rapidly.

Assessment of Phytochemicals, Quality Attributes, and Antioxidant Activities in Commercial Tomato Cultivars

  • To assess South Korean commercial tomato cultivars, regular and cherry tomato cultivars were grown in the greenhouse and evaluated for color attributes, titratable acidity, pH, total soluble solids, carotenoids (lycopene and ${\beta}$-carotene), total phenols, flavonoids, vitamin C, and antioxidant activity. Significant differences (p < 0.05 using Duncan's multiple range test (DMRT)) were observed in the levels of most phytochemicals, quality parameters, and antioxidant activity among the twenty South Korean tomato cultivars tested. Lycopene and ${\beta}$-carotene contents varied significantly (p < 0.05 using DMRT), from $0.95mg{\cdot}100g^{-1}$ to $5.12mg{\cdot}100g^{-1}$ and $0.65mg{\cdot}100g^{-1}$ to $3.56mg{\cdot}100g^{-1}$ of fresh weight, respectively. ${\beta}$-carotene contents exhibited the highest genetic variation (59.2%), followed by naringenin (52.8%) and other phytochemicals. Most of the cherry tomato cultivars had statistically higher levels (p < 0.05 using DMRT) of carotenoids, phenols, flavonoids, vitamin C, and antioxidant activity compared to the regular tomato varieties, suggesting their higher nutritional value. Lycopene content was highest in the cultivars YoYo, Jicored, Titi-Chal, TY-Endorphin, and Rubyking. Cultivars Rubyking, TY-Endorphin, and Titi-Chal also showed relatively higher antioxidant activities in three assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric ion reducing antioxidant power (FRAP) assays. All the antioxidants, except luteolin, were positively correlated with antioxidant activities; the highest correlation was observed between total phenol and antioxidant activities, followed by the correlation between rutin and vitamin C. Cultivars identified to have superior nutritional status would be useful in tomato breeding programs to further improve quality and health benefits of tomatoes for the fresh and processed markets.

저장중 호박분말의 카로테노이드 색소 변화 (Changes of Carotenoids of the Pumpkin Powder during Storage)

  • 열풍건조한 호박분말을 2$0^{\circ}C$에서 1개월 저장하였을 때 저장 초기 총 카로테노이드 함량이 14.46mg%이던 것이 저장 1개월후 빛의 유무에 관계없이 약 65~70% 감소하였으며, TLC 분리에서 빛의 영 향에 관계없이 5개 의 반점으로 분리되었으나 저장시일의 경과에 따른 카로테노이드 색소의 차이를 구별하기는 어려웠다. HPLC에서 lutein의 경우 빛을 조사한 분말은 저장 15후 부터 함량이 다소 증가하였고, 빛을 차단한 분말은 1개월간 함량 변화가 일어나지 않았으며, lycopene은 모두 저 장 말기까지 거의 변화가 일어나지 않았다. $\beta$-Carotene 의 경우 빛을 조사한 처리구가 15일경부터 그 함량이 감소한 반면 빛을 차단한 분말은 저장말기에 가서야 감소하였으며, $\alpha$-carotene은 저장 전 기간을 통해 두 처리구 모두 변화를 나타내지 않았다. 색도조사에서 황색 도인 b값은 두처리구 모두 저장 10일 이후부터 감소하 였으나 빛 차단구의 경우가 그 감소폭이 적은 것으로 나타났다.

우렁쉥이 껍질 카로테노이드의 가열분해로 생성되는 휘발성 화합물의 특성 (Volatile Compounds Formed by Thermal Degradation of Ascidian Tunic Carotenoids)

  • 우렁쉥이 껍질로부터 추출한 카로테노이드 색소의 식품첨가물로서의 이용 가능성을 알아보기 위하여 SDE(simultaneous distillation and extraction/concentration) 장치를 이용하여 휘발성 화합물을 추출하였다. 이를 GC 및 GC-MS로 분석하여 총 63성분의 휘발성 화합물을 동정하였고, 대부분의 화합물이 카로테노이드의 가열분해로 생성되어지는 것으로 나타났다. 동정된 화합물중 대표적인 화합물은 1,3,5-trimethylbenzene, 3,5,5-trimethyl-3-cyclohexen-1-ol, 3,5,5-trimethyl-3-cyclohexen-1-one, 1,1,2,3-tetramethyl-2-cyclohexen-5-ol, 1,1,2,3-tetramethyl-2-cyclohexen-5-one, 2,3,4,4-tetramethyl-6-hydroxy-2-cyclohexene-1-one, 1,2,3,8-tetrahydro-3,3,6-trimethyl-1-naphtol, dihydroacetinidolide, ${\beta}-ionone$, 2-(1,1,5-trimethyl-3-hydroxy-5-cyclohexen-6-yl)-1-to-lylethene, 2,6-dimethyl-8-(1,1,5-trimethyl-3-hydroxy-5-cyclohexen-6-yl)-1,3,5-octatriene-7-yne 등 이었다. 이중 몇가지 화합물의 생성기작도 추정해 보았다.

배합사료 내 색소 원료에 따른 비단잉어 홍백C(yprinus carpio) 치어의 성장과 체색 변화 (Effect of Dietary Carotenoids Sources on Growth and Skin Color of Red- and White-colored Fancy Carp Cyprinus carpio var. koi)

  • A feeding trial was conducted to investigate the effects of dietary carotenoids sources on growth and skin color of red- and white-colored fancy carp Cyprinus carpio var. koi. Nine experimental diets (designated as Con, CP, PA, SP, OP, MB, TO, BE and PO) were formulated to contain Carophyll Pink, red paprika, Spirulina, Opuntia, mandarin bark, tomato, beet and Porphyra, respectively. Each experimental diet was fed to two replicate groups of fish (22.9 g/fish) to visual satiation three times a day for 8 weeks. Weight gain and feed efficiency of fish fed the diets containing Spirulina or Opuntia were higher than those of fish fed the control diet (P<0.05). The values of $a^*$, $L^*$ and $b^*$ of fish skin were significantly changed by dietary carotenoids sources (P<0.05). The $a^*$ values of fish fed the diets containing Carophyll Pink, red paprika and Spirulina were higher than those of fish fed other diets (P<0.05). The skin total carotenoids of fish fed the diets containing Spirulina and Opuntia were higher than those of others (P>0.05). Based on the results of this study, it can be concluded that dietary inclusion of red paprika and Spirulina pacifica could increase the skin redness of red- and white-colored fancy carp.

  • 소나무좀에 감수성(感受性)인 소나무류(類) 잎에는 carotenoid의 함량(含量)이 많고 저항성(抵抗性)인 소나무류(類) 잎에는 그 함량(含量)이 적다는 흑목등(黑木等)의 보고(報告)에 따라 소나무 곰슬 리기다소나무 잣나무 리기테-다소나무 태-다소나무의 소나무좀에 대한 저항성정도(抵抗性程度)를 밝히기 위하여 이들 소나무류(類)의 일년생(一年生) 묘목(苗木)의 침엽(針葉)으로부터 Carotenoid를 추출분석(抽出分析)하여 얻어진 결과(結果)를 다음과 같이 요약(要約)할 수 있다. 이들 소나무류(類)에 함유(含有)되어 있는 carotenoid의 가시부(可視部) 흡수(吸收)spectrum은 매우 비슷하였다. 이들 일정침엽량(一定針葉量)에 함유(含有)된 carotenoid의 량(量)은 잣나무>리기다소나무>곰솔>리기테-다소나무>테-다소나무의 순(順)이였다. Carotenoid는 아가시아 밤나무 편백 희마라야시-다 잎에도 함유(含有)되어 있었고 이들 9수종(樹種)의 carotenoid의 총함량비교(總含量比較)는 아가시아>리기다소나무>곰솔>밤나무>리기테-다소나무>테-다소나무>편백>희마라야시-다 순(順)으로 나타나 이것은 carotenoid의 함량(含量)과 소나무좀에 대한 저항성정도(抵抗性程度)를 직결(直結)시켜서 설명(說明)하는데에는 난점(難點)이 있다고 생각되었다. carotenoid의 thin layer chromatography에 의하면 소나무 잣나무 리기다소나무에서는 13개(個)의 spot기외(其外)의 소나무류(類)에서는 12개(個)의 spot가 자외선형광등(紫外線螢光燈) $3,600{\AA}$$2,537{\AA}$하(下)에서 나타났다. 소나무 잣나무 리기다소나무의 8번째 spot는 다른소나무류(類)와 소나무류외(類外)의 수종(樹種)(밤나무 아가시아 편백 희마라야시-다)에서는 나타나지 않았으며 특(特)히 소나무의 8번째 spot는 자외선형광등(紫外線螢光燈) $2,537{\AA}$하(下)에서 cobalt blue의 강(强)한 형광(螢光)을 발(發)하였다.

Bioavailability of plant pigment phytochemicals in Angelica keiskei in older adults: A pilot absorption kinetic study

  • BACKGROUND/OBJECTIVES: Angelica keiskei is a green leafy vegetable rich in plant pigment phytochemicals such as flavonoids and carotenoids. This study examined bioavailability of flavonoids and carotenoids in Angelica keiskei and the alteration of the antioxidant performance in vivo. SUBJECTS AND MATERIALS: Absorption kinetics of phytochemicals in Angelica keiskei were determined in healthy older adults (> 60 y, n = 5) and subjects with metabolic syndrome (n = 5). Subjects consumed 5 g dry Angelica keiskei powder encapsulated in gelatin capsules with a low flavonoid and carotenoid liquid meal. Plasma samples were collected at baseline, 0.5, 1, 2, 3, 4, 5, 6, 7, and 8 h. Samples were analyzed for flavonoids and carotenoids using HPLC systems with electrochemical and UV detection, respectively, and for total antioxidant performance by fluorometry. RESULTS: After ingestion of Angelica keiskei increases in plasma quercetin concentrations were observed at 1-3 and 6-8 hr in the healthy group and at all time points in the metabolic syndrome group compared to baseline (P < 0.05). Plasma lutein concentrations were significantly elevated in both the healthy and metabolic syndrome groups at 8 hr (P < 0.05). Significant increases in total antioxidant performance were also observed in both the healthy and the metabolic syndrome groups compared to baseline (P < 0.05). CONCLUSIONS: Findings of this study clearly demonstrate the bioavailability of phytonutrients of Angelica keiskei and their ability to increase antioxidant status in humans.

Fatty Acid and Carotenoid Production by Sporobolomyces ruberrimus when Using Technical Glycerol and Ammonium Sulfate

  • The production of carotenoids, lipid content, and fatty acid composition were all studied in a strain of Sporobolomyces ruberrimus when using different concentrations of technical glycerol as the carbon source and ammonium sulfate as the nitrogen source. The total lipids represented an average of 13% of the dry weight, and the maximum lipids were obtained when using 65.5 g/l technical glycerol (133.63 mg/g). The optimal conditions for fatty acid production were at $27^{\circ}C$ using 20 g of ammonium sulfate and a pH range from 6 to 7, which produced a fatty acid yield of $32.5{\pm}1\;mg/g$, including $1.27{\pm}0.15\;mg$ of linolenic acid (LNA), $7.50{\pm}0.45\;mg$ of linoleic acid (LLA), $5.50{\pm}0.35\;mg$ of palmitic acid (PA), $0.60{\pm}0.03\;mg$ of palmitoleic acid (PAL), $1.28{\pm}0.11\;mg$ of stearic acid (SA), $9.09{\pm}0.22\;mg$ of oleic acid, $2.50{\pm}0.10\;mg$ of erucic acid (EA), and $4.25{\pm}0.20\;mg$ of lignoceric acid (LCA), where the palmitic, oleic, and linoleic acids combined formed about 37% of the total fatty acids. The concentration of total carotenoids was 2.80 mg/g when using 20 g of ammonium sulfate, and consisted of torularhodin (2.70 mg/g) and $\beta$-carotene (0.10 mg/g), at $23^{\circ}C$ and pH 6. However, the highest amount with the maximum specific growth rate was obtained (${\mu}_{max}=0.096\;h^{-1}$) with an ammonium sulfate concentration of 30 g/l.

고추건조기의 최적운전조건 (Optimum Drying Conditions of On-Farm Red Pepper Dryer)

  • 농가에서 현재 사용되고 있는 고추건조기의 운전조건을 고추의 건조 모델과 품질변화 모델을 수치해석적 최적화방법인 Box의 complex 방법과 연결하여 최적화를 시도하였다. 주어진 carotenoids. 보존을 만족시키는 제한조건 하에서 에너지소비를 최소화하기 위한 열풍온도, 열풍재순환율, 풍량이 결정되었다. 품질제한 조건이 엄격할수록 낮은 열풍재순환율과 높은 풍량이 사용되어 에너지소비는 증가하고 전체 건조시간은 짧아졌다. 건조 중 건조기내에서 고추의 혼합이 에너지 사용효율과 제품품질을 향상시킬 수 있었다. 현재 사용되고 있는 풍량은 최적운전을 위해서는 증가되어야 하는 것으로 판단되었다. 풍량을 일정하게 적정조건으로 고정하고 열풍의 온도와 재순환율을 2단계로 변화시키는 건조방식이 에너지소비를 더욱 감소시킬 수 있었다. 이러한 2단계 최적건조에서는 단계이동은 전체 건조시간의 약 1/3 부근에서 이루어지고 열풍온도는 첫 단계에서 낮다가 두 번째 단계에서 증가하고 열풍재순환율은 단계변화에 따라 약간 증가하였다. 얻어진 최적제어변수는 건조기내에서의 건조과정과 carotenoids 파괴 kinetics에 의하여 해석될 수 있었다. 얻어진 결과의 실험적 확인이 요청된다.
