Effects of whole-body gamma-irradiation on the peripheral blood of ICR mouse (ICR 마우스에서 방사선 조사 용량에 따른 혈액변화의 관찰)

  • The effects of ionizing radiation on the peripheral blood elements of ICR mouse were examined after varying doses of whole-body gamma-irradiation. ICR mice (n=50) were exposed to 0, 2, 4, 6 and 8 Gy gamma-ray ($^{60}Co$) at 10 Gy/min. The animals were studied for their hematological response on days, 3, 7, 14, 21, 42 and 56 post irradiation. No significant change was noted in erythrocyte, hemoglobin and hematocrit values after irradiation with dose of 2 Gy. Decreasing erythrocyte, hemglobin and hematocrit values occured after irradiation with doses of more than 4 Gy on day 7 after irradiation followed by a sharp fall on day 14. A recovery in these values was noted after 3 weeks of irradiation. Thrombocyte counts decreased on day 3, reaching minimal values on day 7. The total number of leukocytes was reduced on day 3, mainly because of a decrease in the lymphocyte population. An evident lymphopenia and neutropenia occur almost on the day 3 and last up to the day 28 after irradiation. All of the hematological values decreased in the blood in a dose-dependent manner at the same time.

Effects of same TDF Factors on Body Weight of Mice and Peripheral Blood Picture (상이한 분할조사가 마우스의 체중과 말초혈액에 미치는 효과)

  • Using same TDE factors, the authors studied the effects of whole abdominal irradiation on body weight and peripheral blood picture in $30{\pm}3$ day old mice. Fractions of 100 and 200 cGy were given five times a week to the final TDF factors l7, 33, and 49. Total 80 mice were irradiated with orthovoltage x-ray machine. Our results can be summarized as follows : There were no significant differences by sex. Body weight was progressively decreased by the duration for irradiation, but no remarkable difference by fractional dose. Hemoglobin level showed no remarkble change by fractional dose or TDF factors. Leukopenic changes showed that severity of decrease seems to be more related to TDF factors. There were no significant changes of differential count by TDF factors of total doses.

  • In the event a population of animals is exposed to ionizing radiation, proper disposition of animals will minimize loss to the farmer and protect the public from unwholesome meat and dairy products. Clinical response is an important factor in considering salvage of animals for food following exposure to ionizing radiation whether the dose is lethal or not. It is the purpose of this report to present the discussions of blood picture and growth of y ung growing rabbits exposed to cobalt-60 gamma-irradiation. The experimental animals were Hyrnalayan rabbits of 65 days old. The body weight for all animals of the time of irradiation was approximately 450g. The 96 rabbits used in the experiment were allotted t groups of eight. Rabbits in one group served as controls and the others were exposed to single doses of 100, 200, 300, 400, 500, 600 and 700 Roentgens (R). The exposure does rate averaged 15 R per minute. Central blood was obtained by heart puncture. Erythrocyte and leukocyte enumerations and, determinations of hemoglobin in blood and hematocrit value were made in the usual manner. Erythrocyte counts, concentrations of hemoglobin in blood, and hematocrit readings dropped to the lowest value between 1 and 4 weeks postirradiation, returning towards the control values. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. There was a progressive decrease with increasing radiation does in the above values. Mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were not affected by the irradiation. After irradiation of each does the numbers of total leukocyte and the absolute numbers of various types of white cell began to decrease rapidly, and reached a minimum after 2 weeks, returning towards normal levels. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. The reductions in total leukocyte and differential counts as the radiation does increases were apparent. Relative neutrophilia was observed in each irradiated group because of more expense of lymphocytes after irradiation. Growth rate of the rabbits was not affected by 100 R of whloe-body irradiation, while rabbits irradiated with 200, 300, 400 and 500 R showed marked depression of growth rate. As the radiation does increases, a depression of growth rate was apparent. The rabbits exposed to 600 and 700 R ceased from growing and recovery was not recognized during the experimental period.

Comparison of the growth characteristics of Aurcularia auricula-judae according to gamma ray irradiation dose (감마선 조사량에 따른 Aurcularia auricula-judae의 생육특성 비교)

  • In this study, we examined the effects of gamma irradiation dosage on the mycelial growth of Auricularia auricula-judae and performed analyses of fruiting body yield, growth characteristics, taste, fragrance, and mineral composition. Assessments of mycelial growth in response to gamma irradiation at different intensities revealed an enhancement in the growth of fungi exposed to irradiation at 200 Gy. Fruiting body yield was also highest at 200 Gy, followed by 800 Gy and the control group. On the basis of these observations, we subsequently applied gamma ray doses of 200 and 800 Gy to examine the effects of irradiation on fungal quality characteristics. In terms of the taste of fruiting bodies, we detected no significant differences among the control, 200 Gy, and 800 Gy groups. Contrastingly, with respect to fragrance, we found that fungi treated with 200 Gy were characterized by a pattern that differed from those of the control and other treatment groups. Furthermore, whereas we detected no significant difference among treatments with respect total dietary fiber content, calcium content was found to be higher in the treatment groups compared with the control group, with the highest content being measured in fungi exposed to 800 Gy irradiation. Copper content was confirmed to be higher in the control group, whereas there were no significant differences between the fungi irradiated with 200 and 800 Gy. Contrastingly, the highest levels of zinc were detected in response to 200 Gy irradiation, followed by 800 Gy. Collectively, our findings thus indicate that gamma irradiation can contribute to promoting increases in the fruiting body yield and mineral contents of mushrooms.

  • Male rats of the Albino strain received methylene blue in the dose of 40mg/kg by subcutaneous injection and were subjected to total body X-irradiation, 300 roentgen, at 30 minutes after the injection. The protective effect of methylene blue against the single total body X-irradiation was studied for 24 days after X-irradiation with regard to the levels of liver glycogen, blood glucose, and electrolytes in serum. 1. Total body X-irradiation generally casued an increase in the levels of the liver glycogen and blood glucose in both methylene blue treated and control group during the entire experiment. 2. Methylene blue has been shown to delay slightly the increase of the levels of the liver glycogen and blood glucose when comparing with both groups which were given methylene blue and control saline injection before irradiation in the rats. 3. The delay in the increase in the levels of liver glycogen, in experimental group injected with methylene blue, significantly came in two phases. The first phase appeared at there days after the exposure, the second followed at eighth day. It appeared that the recovery phase was at nineteenth day. 4. During the experimental days the levels of the blood glucose increased generally, methylene blue, however, caused delay in two phases; the first at fifth day, the second at eighteenth day after the exposure to X-rays. 5. In electrolytes, there was not a significant difference. The levels of chloride were, however, slightly decreased in both groups, levels of potassium appeared different in two phases at first day and twelfth day, and the levels of sodium appeared to show irregular changes at the same levels, but there was no significant difference. 6. It may be considered that methylene blue greatly reduces the sensitivity of rats to X-rays, provided that methylene blue is given before the exposure.

Effects of Single Total Body Irradiation (TBI) on the Peripheral Blood of Piglets (돼지에서 단일 전신 방사선 조사 용량에 따른 혈액변화 관찰)

  • We examined total body irradiation (TBI)-induced effects by complete blood count (CBC) and fluorescence-activated cell sorter analysis (FACS) in the piglet following radiation irritation. A CBC included red blood cell count, white blood cell count, and platelet cell count. Four piglets were examined in this study and each piglet was divided by irradiation dose, two piglets with 4 Gy, two with 6 Gy, one with 8 Gy. All piglets showed leukopenia, thrombocytopenia after irradiation. In 6 and 8 Gy group, three piglets showed severe hemostatic disorder and gastrointestinal disorder suchas diarrhea and anorexia, and they died between 10 and 15 days after radiation irritation. In 4 Gy, two piglets showed no clinical sign after radiation injury, but persistent leukopenia was shown in blood examination. We suggest that a single TBI dose less than 6 Gy is adequate for conditioning piglet for bone marrow transplantation.

A study on the dose distribution for total-body & hemibody irradiation using clinical photon beams (광자선을 이용한 전신 및 반신조사의 선량분포에 관한 고찰)

  • We have discussed that the total body irradiation(TBI) dose distribution of 6 and 10 MV photon beams, also differences between calculation dose use of compensator sheet and measurements in humanoid phantom. Total body irradiation and hemi-body irradiation(HBI) can be effectively performed when uniformity of dose distribution is estabilished. The method of TBI and HBI dosimatry requires special considerations related to technique, long distance and very large field, machine parameter, patient positioning. TBI and HBI with megavoltage photon beams requires basic dosimatric data which have to be measured directly or derived from the standard beam data. The semiconductor detector and ion chamber were positioned at a dmax depth, mid depth, and its specific ratio was determined using a scanning data by RFA-7 3-dimensional water phantom and solid phantom. The effective source axis distance 380 cm, the field size from 120 cm to 152 cm, isodose distributions were analyzed as a function of the thickness in phantom. Also, have discussed that the measurement of basic data for clinical photon beams for dosage calculations, data calculation sheet and the use of tissue compensation to improve dose uniformity. We have improved a dose uniformity in the TBI and HBI method.

  • Total body irradiation in the treatment of childhood leukemia, which is one of the pre-treatment with stem cell transplantation is being used, the current organization using compensators are treated. However, under the terms of the compensator organization long-term impact on the human body, it is difficult to assess directly. In this study, we use the mathematical simulation of radiation exposures body energy and the distance to the crew and the patient (source surface distance, SSD), and patients with tissue compensators change of the distance along the body of the organ doses were evaluated. As a result, the surface dose of energy 4 MV, SSD 280 cm, tissue compensators and the patient when the distance 30 cm 5.84 G / min showed the highest levels. In addition, patients with tissue compensators and the distance apart when 30 cm TBI represents the ideal dose distribution was found.

Prediction of Midline Dose from Entrance and Exit Dose Using OSLD Measurements for Total Body Irradiation

  • Background: This study aims to predict the midline dose based on the entrance and exit doses from optically stimulated luminescence detector (OSLD) measurements for total body irradiation (TBI). Materials and Methods: For TBI treatment, beam data sets were measured for 6 MV and 15 MV beams. To evaluate the tissue lateral effect of various thicknesses, the midline dose and peak dose were measured using a solid water phantom (SWP) and ion chamber. The entrance and exit doses were measured using OSLDs. OSLDs were attached onto the central beam axis at the entrance and exit surfaces of the phantom. The predicted midline dose was evaluated as the sum of the entrance and exit doses by OSLD measurement. The ratio of the entrance dose to the exit dose was evaluated at various thicknesses. Results and Discussion: The ratio of the peak dose to the midline dose was 1.12 for a 30 cm thick SWP at both energies. When the patient thickness is greater than 30 cm, the 15 MV should be used to ensure dose homogeneity. The ratio of the entrance dose to the exit dose was less than 1.0 for thicknesses of less than 30 cm and 40 cm at 6 MV and 15 MV, respectively. Therefore, the predicted midline dose can be underestimated for thinner body. At 15 MV, the ratios were approximately 1.06 for a thickness of 50 cm. In cases where adult patients are treated with the 15 MV photon beam, it is possible for the predicted midline dose to be overestimated for parts of the body with a thickness of 50 cm or greater. Conclusion: The predicted midline dose and OSLD-measured midline dose depend on the phantom thickness. For in-vivo dosimetry of TBI, the measurement dose should be corrected in order to accurately predict the midline dose.

Radioprotective Effect of Ginseng Components on Antioxidant Enzymes, Glutathione and Lipid Peroxidation of Liver in ${\gamma}$-Irradiated Mice (홍삼 분획물이 감마선을 비사한 생쥐 간에서 항산화물질과 지질과산화에 미치는 방사선 보호효과)

  • In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.
