고오지 종류에 따른 식혜 고추장의 숙성중 미생물 및 효소 역가의 변화 (Changes in the Microflora and Enzyme Activities of Kochujang Prepared with Different Koji during Fermentation)

  • 전통 고추장의 품질개선을 위하여 고추장에 전통메주와 전통 메주에서 분리된 우수 균주를 접종한 고오지를 일정비율로 첨가하고 일부 시료는 가열처리하여 저장하면서 고추장의 미생물 및 효소역가를 분석하였다. 세균수는 전체적으로 발효 40일째 약간 증가하였으며 처리구간의 뚜렷한 차이는 없었고 곰팡이는 계속 감소하여 처리구에 따라 발효 40일 또는 60일 이후에는 검출되지 않았고 효모는 P-1 고오지 함량이 많은 처리구일수록 적은 수가 검출되었고, 열처리에 의해서 효모의 증식이 억제되었다. 고추장 숙성 중 발생한 가스의 누적량은 효모의 증식경향과 일치하였는데 전통메주 첨가시 가장 많았고 P-1 고오지 첨가시 가장 적었으며 열처리구는 발효기간 내내 가스가 전혀 발생되지 않았다. 고추장 숙성 중 효소는 ${\alpha}-amylase$ 경우 전체적으로 활성이 매우 낮았으며, ${\beta}-amylase$는 처리구에 따라 불규칙한 경향을 보이면서 전반적으로 전통메주 첨가구(CON)와 전통메주와 P-2 고오지 첨가구(MKP2)가 다른 처리구에 비하여 낮은 활성을 보였다. 산성 protease 활성도는 증가와 감소를 반복하다가 발효 100일째 활성이 최대였고, 중성 protease는 발효 60일까지 증가하다 점점 감소하였고 두 효소 모두 처리구간의 뚜렷한 차이는 없었다. 결과적으로 고추장에 선발 균주를 접종한 고오지를 첨가하였을 때 세균 및 곰팡이수, 효소역가는 전통 메주를 사용할 때와 뚜렷한 차이를 보이지 않은 반면 효모 생성과 가스 발생 억제에는 매우 효과적이므로 고추장 발효 관리의 한 수단으로 미생물 관리방법이 제안될 수 있다.

명태를 첨가한 김치의 품질특성 (Effects of Alaska Pollack Addition on the Quality of Kimchi (Korean salted cabbage))

  • 명태를 $0.5{\times}0.5\;cm^2$ 로 잘라 절인배추의 무게 비로 6, 12, 18% 첨가한 김치를 제조한 후 $10^{\circ}C$에서 20일간 발효숙성시키면서 4일 간격으로 pH, 산도, 염도, 환원당, 젖산균의 변화, 색도, 아미노산, 관능 등 품질 특성의 변화를 관찰하였다. pH와 환원당은 발효가 진행될수록 전 실험군에서 낮아지는 경향을 보였으며 명태 첨가량이 높아질수록 pH와 환원당의 값이 높았다. 산도는 발효가 진행되어 갈수록 전체적으로 높아지는 변화를 보였는데 명태 첨가군이 높은 값을 나타내었다. 염도는 저장기간별로 차이가 없었지만 첨가량에 따라서는 유의적으로 차이를 보여 명태첨가량이 많을수록 염도가 낮아졌다. 젖산균의 수는 발효가 진행될수록 점차 높아져 명태를 가장 많이 첨가한 18% 첨가군은 8일째에, 나머지군은 12일에 가장 높은 수준을 보인 후 다시 감소하는 양상을 보였다. 색도에서는 명태 첨가량이 많아질수록 명태 자체의 연한 황백색이 영향을 주어 L값, a값 및 b값 모두 증가하는 경향을 보였고 발효가 진행되면서 L값은 유의적인 차이가 나지 않았지만 a값은 감소되는 경향을 보였으며 b값은 서서히 증가하여 숙성 8~9일에 최고에 달하였다가 그 이후 감소하였다. 유리 아미노산은 명태첨가량에 따라 유리 아미노산의 총량이 증가하였고 발효가 진행 될수록 감소되는 경향을 보였다. 관능검사 결과로는 외관, 조직감, 맛, 전반적인 선호도는 6% 첨가군이 가장 높은 점수를 나타내었으며, 비린향미는 명태첨가량이 많아질수록 강도가 높았으나 숙성 적숙기에는 유의적인 차이를 보이지 않았으며, 짠맛, 신맛, 숙성된 맛도 유의적인 차이를 보이지 않았다. 이상의 결과로 김치를 제조할 때 6%의 명태를 첨가하는 것이 외관과 조직감, 맛이 좋으며, 비린향미가 다소 있기는 하지만 중간 이하 수준으로 기호도에 큰 영향을 미치지 않으며 풍부한 유리 아미노산의 용출로 인해 발효에 긍정적인 영향을 주어 우수한 품질의 김치를 얻을 수 있음을 보여주었다.

글리신을 활용한 저염 오징어 및 명란 젓갈의 미생물 안전성 확보 (Effects of glycine on microbial safety of low-salted squid and myungran jeotgal)

  • 기존 대비 염함량을 50% 내외 감량한 염도 4-6%의 저염 오징어 및 명란 젓갈의 미생물 안전성을 확보하기 위해 상업적 항균제 7종의 적용 실험을 진행하여 다음과 같은 결과를 얻었다. 저염 젓갈 내 자생하고 있는 균총을 살펴본 결과 간균인 Lactobacillus sp.와 구균인 Streptococcus sp., Pediococcus sp., Luconostoc sp. 등의 젖산균이 80%를 차지하는 우점균으로 나타났으며, 오징어 젓갈에는 yeast가 명란젓에는 fungi가 추가로 관찰되었다. 젖산균 계통의 미생물의 살균 및 생육억제에 효과가 있다고 알려진 5종의 상업적 항균제를 저염 오징어 젓갈 중량 대비 0.5%(w/w)를 조미배합 과정에서 첨가, 혼합하여 총균수의 변화를 고찰한 결과 글리신이 94.20%, 폴리라이신이 92.78%의 살균률을 보여 $10^1CFU/g$ 내외의 감균 효과를 나타내었으며, 저염 명란 젓갈에서도 0.5% (w/w) 글리신이 90.87%의 살균률을 나타내었다. 저염 젓갈의 균 제어에 효과적인 글리신은 조미료로도 많이 사용되는 아미노산 계통의 항균성분으로 그람 음성균 및 양성균 모두에 대해 살균효과를 가지고 있으며, 특히 유기산과 병용 사용 시 감균 및 생육 억제에 있어 효과적인 것으로 알려져 있다. 저염 오징어 젓갈에 0.5% (w/w)의 글리신을 혼합 후 저장기간별로 총균수의 변화를 고찰한 결과 $10^{\circ}C$의 냉장조건에서 21일 경과 후에도 90%의 살균률이 유지되어 $10^1CFU/g$의 감균 효과가 지속되었으며, 저염 명란 젓갈의 경우에도 저장시 97%의 살균효과가 지속되어 무처리구 대비 $10^2CFU/g$ 내외 감소하였다. 따라서 글리신은 다양한 수산물을 원료로 사용한 저염 젓갈의 균 제어에 활용할 수 있는 효과적인 상업적 항균제로 판명되었다.

Kinetics of Chemical Properties and Microbial Quantity in Soil Amended with Raw and Processed Pig Slurry

  • Pig slurry is a good soil amendment not only because of its high organic matter content, but also because of its ability to provide various nutrients. The objective of this study was to estimate the influence of raw and processed pig slurry application on pot soil over chemical fertilizer and non-amended control soil. Change in the chemical parameters (pH, organic matter (OM), organic carbon (OC), macro and micronutrients) and microbial mass of the treated soils were monitored over 30 to 90 days. Pot soil was treated with the recommended dose of pig slurry and chemical fertilizer, and was sampled after 30, 60 and 90 days of incubation. The least significanct difference (p<0.05) was observed on Fe, Cu, Zn, available P and K between treatments. All treatments increased N, P and K content and microbial mass of soil over control soil. Interestingly, no significant effects were detected on OM, OC, total bacteria, actinomycetes and fungi mass in soil irrespective of treatments given. However fungal and bacterial counts, as well as available nutrients, were found to be higher in processed slurry (PS)-treated soil compared to other soils. In general a significant correlation existed between the fungal count and OM, OC, Zn, T Kjeldahl N (TKN), available P and K of soil. A strong negative correlation was observed between pH and Fe in soil. This study clearly demonstrated that the use of processed manure as a fertilizer could be a key for sustainable livestock agriculture.

개심술 환자에서의 면역기능의 변화;T lymphocyte subset의 변화에 대한 고찰 (Changes in Lymphocyte Subsets following Open-Heart Surgery ; A Study for Changes in Lymphocyte Subsets)

  • Cell mediated immunity is depressed following surgical procedure and the degree of immunosuppression is directly related to the magintude of the procedure, blood transfusion, and length of operation. So we would expect cardiac operations to be highly immunosuppressive, although little is konwn about their immunosuppressive effect. The nearly complete consumption of complement factors and decreased levels of IgM and IgG resulting in an impaired opsonizing capacity. Additionally, peripheral blood mononuclear cell counts including T-and B-lymphocytes and T-cell subsets are reduced. Depression of cell-mediated immunity following open-heart surgery is potentially detrimental because it could increase the susceptability of patients to viral and bacterial infection. We reviewed 20 patients after cardiac operation to search for changes in peripheral blood lymphocyte subsets. Lymphocyte subsets were measured by flow cytometer and the preoperative values of lymphocyte subsets were compared with those from the first, fourth, and seventh days after operation. After cardiac operation, total mumbers of T lymphocyte was severely depressed on the first postoperative day and returned to the preoperative level by the seventh day after operation. CD3, CD4, and CD8 lymphocytes were decreased on the first postoperative day and returned to the preoperative level by the seventh day also. There was four cases of wound infection and these patients had increased CD4 lympocyte and more decreased CD19 lymphocyte compared with the non-infected group. It is concluded from these data that cell-mediated immunity is significantly depressed for at least one week following open-heart surgery and this result was closely related to the postoperative infection.

감마선 조사에 의한 고로쇠 수액의 효율적인 저장방법 (Efficient Storage of Gorosoe(Acer mono Max.) Sap by Gamma Irradiation)

  • Effects of gamma irradiation on microbiological changes of Gorosoe sap were characterized during a post-irradiation storage at $4^{\circ}C$. The aseptically collected sap was irradiated and stored at $4^{\circ}C$ for 0 to 60 days and analysed for standard plate counts and 16S rDNA. There were significant differences in the total number of colony forming units(CFUs) of bacteria between irradiated and non-irradiated control sap. Bacteria of non-irradiated sap were present at levels of $1.5{\times}10^4{\sim}2.3{\times}10^8\;CFU/m{\ell}$, whereas no viable microbial cells were detected in sap after 10 kGy of irradiation during storage. According to the 16S rDNA sequence analysis, bacterial community structures decrease with time and the most abundant strain was Pseudomonas species. Our results suggested that gamma irradiation can be used to enhance the shelf-life of Gorosoe sap.

Effects of Various Levels of Cassava Hay on Rumen Ecology and Digestibility in Swamp Buffaloes

  • Four, mature rumen fistulated male swamp buffaloes with an average initial weight of $426{\pm}25kg$ were randomly allocated to receive dietary treatments according to a $4{\times}4$Latin square design. Four dietary treatments with varying proportions of ureatreated rice straw (UTRS) and cassava hay (CH) were offered (100:0, 75:25, 50:50, 0:100). Each feeding period lasted 21 days, the first 14 days for feed adjustment and intake measurement and the final 7 days for rumen fluid and faecal collections. The results demonstrated the potential use of both UTRS and CH as roughage sources. As levels of CH increased in the diets pH values were maintained (6.5-7.0). Ruminal $NH_3-N$ concentrations were significantly (p<0.05) raised as higher levels of CH were incorporated into the diets. Moreover, cellulolytic and proteolytic bacterial populations were enhanced while total protozoal counts were decreased (p<0.05). In addition, DM, OM and CP digestibilities and their digestible intakes as well as estimated energy increased with increasing levels of CH in the diets. The results suggest a favorable effect of CH substituting for UTRS.

Microbial Evaluation of Commercially Packed Kimchi Products

  • Commercially packed kimchi products from 6 different manufacturers, which are exported overseas as well as sold domestically, were analyzed to determine their microorganism distributions and presence of pathogenic bacteria. All samples showed decreasing pH levels (from 5.7-6.2 to 3.9-4.3) and increasing titratable acidities (from 0.3-0.4 to 0.8-1.2%) during 15 days of storage at $4^{\circ}C$. Total bacterial counts ranged from $2.1{\times}10^5-1.9{\times}10^6\;CFU/mL$ in the initial kimchi samples, and then increased to $1.1{\times}10^8-1.8{\times}10^9\;CFU/mL$. The coliform numbers decreased from approximately $2.5{\times}10^2-1.7{\times}10^4\;CFU/mL$ to zero. Major foodborne pathogens such as Salmonella spp., Staphylococcus aureus, Escherichia coli O157:H7, Listeria monocytogenes, Bacillus cereus, Yersinia enterocolitica, and Shigella spp. were not detected in any of the samples. However, 2 out of the 6 samples carried E. coli, emphasizing the need for improved hygiene practice. Interestingly, Hafnia alvei, belonging to the Enterobacteriaceae family, was isolated in all of the samples. Further study is needed on this newly reported bacterium in kimchi.

Effects of Edible Films Containing Procyanidin on the Preservation of Pork Meat during Chilled Storage

  • Procyanidins, which are natural antioxidants and antimicrobials found in grapes, enhance the quality and extend the shelf life of meat. We explored the effects of edible films incorporating procyanidins on pork loin stored for various times. Procyanidins (0, 0.1, and 0.3%, w/w) were incorporated into the edible films. We assessed meat color, pH, levels of volatile basic nitrogen (VBN) and 2-thiobarbituric acid-reactive substances (TBARS), and microbial populations for 14 d. The chromaticities and pH values of pork loin wrapped in film containing procyanidins (0.1% and 0.3%) generally increased (p<0.05) with storage time. VBN and TBARS levels, and total bacterial and Escherichia coli (E. coli) counts, significantly decreased (p<0.05) in the procyanidin groups. In particular, procyanidins strongly inhibited TBARS formation. Thus, our findings suggest that edible film impregnated with procyanidins inhibits lipid oxidation and microbial growth, thereby enhancing the quality and shelf life of pork meat.

연잎을 첨가한 탁주의 발효 특성 (Fermentation Characteristics of Takju Prepared with Lotus Leaf)

  • In this study, lotus leaf was added to a Takju (Korean milky rice wine) preparation to evaluate its effects on physicochemical and fermentation properties. For rapid saccharification Takju was stored at 25$^{\circ}C$ for 2 days and then temperature was switched to 18$^{\circ}C$ to mature the wine for 21 days. From the start of fermentation to 2-days, the pH of Takju decreased rapidly and acidity increased. Maltose, a byproduct of starch saccharification, increased for 2 days then rapidly decreased. Glucose fluctuated for 7 days and of the 21-days fermentation. The ethanol production rate was highest during the first 7 days, then slowed. Total viable yeast and lactic acid bacterial counts increased rapidly for 2 days and then decreased gradually thereafter. Leuconostoc spp. grew rapidly for 1 day and sharply disappeared with decreasing pH. DPPH radical scavenging activity was significantly higher for Takju prepared with lotus leaf than without. The overall acceptance of Takju tended to increase when it was prepared with lotus leaf.
