Characteristics of proteolytic microorganisms and their effects on proteolysis in total mixed ration silages of soybean curd residue

  • Objective: The objective of this study was to isolate proteolytic microorganisms and evaluate their effects on proteolysis in total mixed ration (TMR) silages of soybean curd residue. Methods: TMRs were formulated with soybean curd residue, alfalfa or Leymus chinensis hay, corn meal, soybean meal, a vitamin-mineral supplement, and salt in a ratio of 25.0: 40.0:30.0:4.0:0.5:0.5, respectively, on a basis of dry matter. The microbial proteinases during ensiling were characterized, the dominate strains associated with proteolysis were identified, and their enzymatic characterization were evaluated in alfalfa (A-TMR) and Leymus chinensis (L-TMR) TMR silages containing soybean curd residue. Results: Both A-TMR and L-TMR silages were well preserved, with low pH and high lactic acid concentrations. The aerobic bacteria and yeast counts in both TMR silages decreased to about 105 cfu/g fresh matter (FM) and below the detection limit, respectively. The lactic acid bacteria count increased to 109 cfu/g FM. The total microbial proteinases activities reached their maximums during the early ensiling stage and then reduced in both TMR silages with fermentation prolonged. Metalloproteinase was the main proteinase when the total proteinases activities reached their maximums, and when ensiling terminated, metallo and serine proteinases played equally important parts in proteolysis in both TMR silages. Strains in the genera Curtobacterium and Paenibacillus were identified as the most dominant proteolytic bacteria in A-TMR and L-TMR, respectively, and both their proteinases were mainly with metalloproteinase characteristics. In the latter ensiling phase, Enterococcus faecium strains became the major sources of proteolytic enzymes in both TMR silages. Their proteinases were mainly of metallo and serine proteinases classes in this experiment. Conclusion: Proteolytic aerobic bacteria were substituted by proteolytic lactic acid bacteria during ensiling, and the microbial serine and metallo proteinases in these strains played leading roles in proteolysis in TMR silages.

Estimation of Shelf-life of Frankfurter Using Predictive Models of Spoilage Bacterial Growth

  • The aim of this research was to develop predictive models for the growth of spoilage bacteria (total viable cells, Pseudomonas spp., and lactic acid bacteria) on frankfurters and to estimate the shelf-life of frankfurters under aerobic conditions at various storage temperatures (5, 15, and $25^{\circ}C$). The primary models were determined using the Baranyi model equation. The secondary models for maximum specific growth rate and lag time as functions of temperature were developed by the polynomial model equation. During 21 d of storage under various temperature conditions, lactic acid bacteria showed the longest lag time and the slowest growth rate among spoilage bacteria. The growth patterns of total viable cells and Pseudomonas spp. were similar each other. These data suggest that Pseudomonas spp. might be the dominant spoilage bacteria on frankfurters. As storage temperature increased, the growth rate of spoilage bacteria also increased and the lag time decreased. Furthermore, the shelf-life of frankfurters decreased from 7.0 to 4.3 and 1.9 (d) under increased temperature conditions. These results indicate that the most significant factor for spoilage bacteria growth is storage temperature. The values of $B_f$, $A_f$, RMSE, and $R^2$ indicate that these models were reliable for identifying the point of microbiological hazard for spoilage bacteria in frankfurters.

Indoor distribution characteristics of airborne bacteria in pig buildings as influenced by season and housing type

  • Objective: A concentration of airborne bacteria generated from swine houses is recognized to be relatively higher than other work places and it is essential to optimally manage it to prevent farmers' respiratory diseases. This study was conducted to assess the distribution characteristics of airborne bacteria in swine houses located at South Korea. Methods: A total 27 pig buildings of the enclosed type operated with mechanical ventilation system by a side wall fan and deep-pit manure system with slats were surveyed. Air samples were collected at 1.0 m above the middle floor in pig housing room. A six-stage viable particulate cascade impactor was used to identify the distribution of the sizes of particles in diameter. Results: Seasonal mean levels of airborne bacteria in the housing rooms of gestation/farrowing pigs, nursery pigs and growing/fattening pigs were 3,428(${\pm}1,244$) colony forming unit $(cfu)/m^3$, $8,325({\pm}3,209)cfu/m$, and $13,254({\pm}6,108)cfu/m^3$ for spring; $9,824({\pm}2,157)cfu/m^3$, $18,254({\pm}5,166)cfu/m^3$, and $24,088({\pm}9,274)cfu/m^3$ for summer; $1,707({\pm}957)cfu/m^3$, $4,258({\pm}1,438)cfu/m^3$, and $8,254({\pm}2,416)cfu/m^3$ for autumn; and $2,322({\pm}1,352)cfu/m^3$, $6,124({\pm}1,527)cfu/m^3$ and $12,470({\pm}4,869)cfu/m^3$ for winter, respectively. Conclusion: Concentrations of airborne bacteria according to pig housing type were highest in growing/fattening housing room followed by nursery housing room and gestation/farrowing housing room. In terms of seasonal aspect, the pig building showed the highest levels of airborne bacteria in summer followed by spring, winter and autumn. The respirable airborne bacteria which are ranged between 0.6 and $4.7{\mu}m$ accounted for approximately 60% compared to total airborne bacteria regardless of pig housing type.

  • Around and in the area of Wolgot-Muon, Gimpo-Gun, Kyunggi province, I examined total bacteria, general Pseudomonas spp., fluorescent Pseudomonas spp., in soil layers and also in different kinds of soil of respective diseased, uncultivated, and healthy areas, and found the followings. 1. In the diseased and uncultivated areas, the content of moisture and silt was greater than in the healthy area. 2. Contrary to the above, the healthy area contained a greater amount of inorganic elements such as $P_2O_5$, K, Ca and of soil particle such as Cs and Fs. The degree of pH and content of Mg were even in three types of soils. 3. Total bacteria were found in abundance in the healthy soil. It was observed that in all types of areas, bacteria reside in abundance in the rhizosphere, i.e., 10-15 cm layers and that the closer the surface, the greater the numbers of the bacteria. 4. General Pseudomonas spp. were also found to the greater in number on the surface of the soil, especially so in the rhizosphere, with the numbers decreasing as the soil layers increase. Numbers of this bacteria in all types of area were nearly uniform. 5. A great number of fluorescent Pseudomonas spp. were found in the diseased area, especially so in the rhizosphere.

Microbial Inactivation of Chicken Cage Litter by Aqueous Chloride Dioxide (이산화염소수를 이용한 계사 내 깔짚의 미생물 수 저감화)

  • We evaluated microbial inactivation in chicken cage litter, to ensure microbial safety, using aqueous chloride dioxide. Contamination by coliforms, Escherichia coli, Listeria spp., yeasts and molds, total aerobic bacteria, and Salmonella spp. was detected in fresh cage litter, and microbial populations increased if litters were repeatedly used. Aqueous $ClO_2$ treatment (500 ppm) significantly decreased the populations of coliforms, E. coli, Listeria spp., yeasts and molds, total aerobic bacteria, and Salmonella spp. in all litter samples tested. In particular, aqueous $ClO_2$ treatment on fresh litter reduced the initial populations of coliform, E. coli, Listeria spp., yeasts and molds, and total aerobic bacteria by 4.47, 1.29, 1.23, 3.24, and 5.2 log CFU/g, respectively. In addition, when litters used for 1 and 5 weeks were tested, treatment significantly reduced microbial populations. The results suggest that aqueous $ClO_2$ treatment is useful to reduce microbial hazards in chicken cage litter and to improve the microbial safety of slaughtered chickens.

Assessment of Microbiological Quality of Cooked Dried-Seafoods in School Foodservice Operations (학교급식에서 제공되는 건포류 조리식품의 미생물적 품질평가)

  • This study was aimed to provide safety management guidance by evaluating the microbial quality of cooked dried-seafoods in school foodservice operations. Nineteen seafood items were collected from six elementary schools, those were dried-anchovy, dried-seaweed and dried-fish, which were classified as cooking process. The temperatures at receiving and after cooking were measured and the analyses of cooking processes and microbial quality were performed. The temperatures of all foods after cooking were higher than the temperature limit of $74\^circC$. The number of total aerobic bacteria and S. aureus in dried-anchovy over the limit of $10^5$ and even the level of S. aureus was found to be unsatisfactory. The count of total aerobic bacteria was 2.1x$10^8$ CFU/g and the number of total aerobic bacteria after cooking was over the limit in one school. The level of E. coli (3.1x$10^3$ CFU/g) was over the limit at one school and the number of S. aureus (1.2×$10^4$ CFU/g) was considered as unacceptable. Dried- tangle and green laver were contaminated with total aerobic bacteria showing the over the limit. The numbers of total aerobic bacteria in dried- filefish, pollack and squid were 4.3x$10^6$, 3.4x$10^6$-3.9x$10^7$ and 4.6x$10^5$-4.1x$10^7$ CFU/g, respectively, which were in acceptable or unsatisfactory level. The E. coli in dried- filefish and pollack were over the limit. The total aerobic bacteria levels, 4.6x$10^5$-1.5x$10^6$ CFU/g in dried-pollack and 8.0x$10^5$-2.2x$10^7$ CFU/g in dried-squid, were over the limit after cooking except dried-filefish. The E. coli levels, 4.3x$10^3$ CFU/g in dried-filefish and 2.5x$10^2$ CFU/g in dried-pollack, were over the limit of $10^2$ CFU/g. The numbers of Enterobacteriaceae were either acceptable (3.3x$10^3$ CFU/g) or unsatisfactory (1.6x$10^4$ CFU/g) level in dried-pollack. S. aureus was unsatisfactory level (6.5x$10^4$ CFU/g) in dried-filefish while unacceptable in dried-pollack both before and after cooking. Unacceptable levels of S. aureus, 2.4x$10^4$ and 1.3x$10^5$ CFU/g were found from two schools, respectively. These results suggest that the contamination of raw materials and the seasonings added after cooking should be controlled to manage the microbial safety of cooked dried-seafoods.

Exposure Assessment of Dust and Airborne Microorganisms among Workers in a Clothing Shopping Center (의류쇼핑센터 근로자의 분진 및 부유미생물에 대한 노출평가)

  • Objectives: The purpose of this study is to assess the exposure level to dust and airborne microorganisms among employed workers in a clothing shopping center. Materials and Methods: On-site investigation of a clothing shopping center was performed between October and November 2012. The hazardous substances measured in this study are particulate matter(Total dust, respirable dust) and airborne microorganisms (Total airborne bacteria, total airborne fungi). Results: The highest geometric mean levels of particulate matter(total dust, respirable dust) for personal sampling were $1.735(SD:0.883)mg/m^3$ for total dust and $0.0711(SD:0.008)mg/m^3$ for respirable dust, respectively. Those for area sampling were $0.625(SD:0.091)mg/m^3$ for total dust and $0.0718(SD:0.012)mg/m^3$ for respirable dust, respectively. The highest geometric averaged concentrations of airborne microorganisms(Total airborne bacteria, total airborne fungi) were detected at $1,181(SD:105)cfu/m^3$ for total airborne bacteria and $683(SD:114)cfu/m^3$ for total airborne fungi, respectively. Concentrations of particulate matters and airborne microorganism in clothes shopping center did not correlate significantly with environmental factors such as temperature or relative humidity. Conclusions: Exposure levelshave not been established for service workers. Thus, health risk assessment for this group is very difficult. Health guidelines for service workers should be established as soon as possible.

Gender and Age Differences in Hand Hygiene Practices among the General Population (성별 및 연령별 손 위생관리의 비교)

  • Objective: This study was performed to evaluate gender and age differences in hygienic behavior among the general population, focusing on hand-washing habits and the microbial load of hands. Methods: A self-administered questionnaire survey and a separate microbial examination were performed. The Pearson's correlation between hand-washing habits and microbial load was analyzed. Results: In the questionnaire survey on hand-washing habits, gender differences were found in hand-washing frequency, use of hand-washing agents, and hand drying methods (p<0.05). Age differences were found in numbers of washing parts of the hands and also in hand drying methods (p<0.05). Females showed better habits washing hands than did males, as did older people compared to younger. In the microbial examination of indicator bacteria on their hands, younger people tended to show a higher load of total aerobic bacteria than did the older, and females showed a higher load of total coliforms than did males (p<0.05). There were significant relationships between the load of total aerobic bacteria and hand-washing frequency, duration, and method of turning-off water (p<0.05). Conclusions: Although females were expected to show a better practice of hand-washing than were males based on the survey results, they showed a higher level of total coliforms in the hand examination. The older age group showed better hand-washing habits than did the younger age group and had less total aerobic bacteria on their hands. These inter-gender and age differences highlight the need for development and implementation of gender-and age-specific educational programs or campaigns.

Quality Characteristics of Apple Vinegar using Various Acetic Acid Bacteria (초산균을 달리하여 제조한 사과식초의 품질특성)

  • To develop farm-made high effective vinegar, this study prepared apple vinegar using four kinds of acetic acid bacteria isolated from a natural fermentation liquid of 'Cheongsoo' grapes and analyzed vinegar samples fermented 93% apple juice and 7% alcohols at 30℃ for 20 days. To accomplish this, quality characteristics such as pH, total acidity, reducing sugar, organic acid, color, total polyphenol contents, and antioxidant activity contents were determined. The pH decreased while total acidity of all samples gradually increased during fermentation period. The vinegar with AP 21 strain tended to increase the total acidity quicker than other stains. Reduced sugar content remained high until the last fermentation day. Furthermore, reduced sugar contents of all vinegars increased as fermentation progressed by decomposing sucrose present in apples. When physiological activities were compared, apple vinegars fermented with AP 21 and 30 strains had higher total polyphenol and flavonoid contents than other samples. However, there was no significant difference in antioxidant activity between samples. These results indicate that strain 21 is the most suitable starter as acetic acid bacteria for producing farm-made vinegar.

Changes of Microorganism during the Storage of Spices as Affected by Irradiation (방사선 조사한 향신료의 저장중 미생물 변화)

  • Spices(Tumeric, Celery-seed, Cumin, Black pepper, Coriander) were irradiated at dose levels of 0, 5, 7 and 9kGy using Co60 source and stored at room temperature. After 12 months of storage following 9kGy irradiated spice, total aerobic bacteria were significantly decreased. When the irradiation dose was 7kGy, microbial levels showed l02-103 levels. However, total lactic acid bacteria and E-coli were shown negative at below 5kGy.
