  • This study was conducted to determine the changes of nitrogen compounds in lamina and midrib during the curing process with the different curing methods, such as priming and stalk-cut curing. After KB 108 burley tobacco was cultivated by the different fetilization levels such as standard and 20% higher, only the tips and leaf were harvested. Though the contents of total alkaloid and total nitrogen were similar in lamina, midrib showed a very low contents of those components by the different curing method and fertilization levels. The content of nitrate-nitrogen in lamina increased during a middle of curing process, and then these compound was decreased during an end of curing process by stalk-cut curing method. Nitrate-nitrogen contents in the lamina by the priming curing showed a high level caused by rapid drying process during an end of curing process. That component in midrib was 5-6 times higher than that of lamina. The contents of ammonia-nitrogen in the lamina and midrib were increased during a curing process. Though those amount in tips showed a similar by a different fetilization and curing method, midrib of stalk-cut curing showed a slow increasing during a curing process.

  • Objective: This experiment aimed to explore the protective action of dietary supplementation with isoquinoline alkaloids (IA) from Macleaya cordata on lipopolysaccharide (LPS)-induced liver injury in broilers. Methods: Total 216 healthy broilers were selected in a 21-d trial and assigned randomly to the following 3 treatments: control (CON) group, LPS group, and LPS+IA group. The CON and LPS groups were provided with a basal diet, whereas the LPS+IA group received the basal diet supplemented with 0.6 mg/kg Macleaya cordata IA. Broilers in LPS and LPS+IA groups were intraperitoneally injected with LPS (1 mg/kg body weight) at 17, 19, and 21 days of age, while those in CON group were injected with equivalent amount of saline solution. Results: Results showed LPS injection caused systemic and liver inflammation in broilers, inhibited immune function, and ultimately lead to liver injury. By contrast, supplementation of IA ameliorated LPS-induced adverse change in serum parameters, boosted immunity in LPS+IA group. Furthermore, IA suppressed the elevation of hepatic inflammatory cytokines and caspases levels induced by LPS, as well as the expressions of genes related to the toll-like receptor 4 (TLR4)/myeloid differentiation primary response 88 (MyD88)/nuclear factor-kappa B (NF-κB) pathway. Conclusion: Dietary inclusion of 0.6 mg/kg Macleaya cordata IA could enhance immune function of body and inhibit liver damage via inactivating TLR4/MyD88/NF-κB signaling pathway in broilers.

  • Effects of Panax ginseng were tested on morphine-and steroids-induced immunosuppression, focusing on mechanism and identification of active components. To investigate overall effects of morphine and ginseng total saponin (GTS) on immune system, body weight and lymphoid organ weight were measured. Morphine significantly reduced body weight, spleen/body weight, and thymus/body weight ratio. GTS, at 100mg/kg (oral), restored spleen/body weight ratio. Because morphine is known to increase corticosterone level, serum corticosterone level was measured by radioimmunoassay. Serum corticosterone was increased by morphine and it was restored to the control level by GTS 100mg/kg (oral). In vitro proliferation studies were also conducted to study the effects of ginseng on steroids-induced immunosuppression. While ginsenoside Rg$_1$ and ginseng alkaloids were effective on proliferation and dexamethasone-induced death of thymocytes, 50% ginseng ethanol extract and polysaccharides were effective on splenocytes. In vivo mprohine-induced apoptosis of thymus was partly protected by GTS.

  • Pharmacological investigations were carried out with aqueous methanolic extract (AME) of Swietenia mahagoni (L.) Jacq. (Meliaceae) leaves. Acute toxicity studies revealed that the $LD_{50}$ dose of AME was 600 mg/kg, i.p. AME was found to possess significant anti-inflammatory activity in acute, sub-chronic and chronic models of inflammation. AME selectively inhibited cyclooxygenase (COX)-2 activity, which is involved in arachidonic acid metabolism and biosynthesis of prostaglandins under inflammatory conditions. Treatment with AME significantly enhanced total peritoneal cell count and the number of macrophages in normal mice, which revealed that AME may also alter the immune response along with its anti-inflammatory effect. The saponins or the alkaloids present in AME may be responsible for the anti-inflammatory activity.

  • Six flue-cured cultivars of Nicotiana tabacum L., 15 possible $F_1$ hybrids and 15 $F_2$ populations among them, and 15 haploid populations of $F_1$ hybrids and 6 haploid from parents, were evaluated. The variances of general combining ability were predominant for all characters studied in $F_1$ hybrid and $F_2$ population, and in haploid populations of $F_1$ hybrids except leaf width, value and reducing sugar. There were significant correlations between generations in variances and ranks of parental GCA effects estimated from general combining ability analysis of $F_1,\;F_2$ and haploid generation of $F_1$ hybrids for yield, stem height, days to flower. leaves per plant, value and total alkaloids.

  • Basic substances were obtained from five Korean Aconitum, A. koreanum, A. chiisanense, A. ciliae, A. pseudoproliferum and unknown Aconitum species and compared TLC patterns of the substances. Soluble substances in ammonia alkali showed 8 spots on TLC, while insoluble substances in ammonia alkali 7 spots. These patterns are all similar in all species used in the investigation.

  • An efficient Pictet-Spengler type reaction of N,O-acetal TMS ethers for the practical synthesis of 1-substituted tetrahydroquinolines, medicinally important alkaloids, has been accomplished. To demonstrate the versatility of this novel procedure, the total synthesis of calycotomine, a representative 1-hydroxymethyl substituted tetrahydroisoquinoline, is also described.

  • The alkaloid components in the root of Codonopsis lanceolata were studied. From the ether soluble alkaloid fraction, four $\beta$-carboline alkaloids were isolated in crystalline state by chromatographic purification process; comp. I mp $178^{\circ}$, isolated yield 4.5$\times$$10^{-5}%$, comp. II mp $166^{\circ}$, 6.0$\times$$10^{-5}%$, comp. III mp $164^{\circ}$, 1.8$\times$$10^{-4}%$ and comp. IV mp $197^{\circ}$, 3.5$\times$$10^{-5}%$. They were identified by spectral analysis and by total synthesis as being a new component $N_9$-formylharman, and already known components in other plant 1-carbomethoxy-$\beta$-carboline, perlolyrine and norharman.

  • Azasugars, which have been called the "sugar-shaped" alkaloids from plants. are reversible. competitive inhibitors of glycosidases. The purpose of these natural products is possibly to inhibit the carbohydrate metabolism and consequently the growth of plant consuming pests. Since selective glycosidase inhibitors have a large number of interesting potential applications including treatment of AIDS, diabetes. and tumor metastasis. they have received a considerable attentions. • Cytoplasmic male- sterile Hyangchio, derived from male- sterile Burley 21 containing N. megalosiphon cycloplasm, and male- fertile Hyangchio (Korean local aromatic variety) were evaluated in replicated pot trials in green-house. Male- sterile Hyangchio was comparable to the male- fertile Hyangchio for six agronomic traits and total alkaloids content. The only significant difference was that the male- sterile Hyangchio flowered 2.3 days later than the male- fertile Hyangchio. M ale- sterile Hyangchio has increased the yield than the male- fertile Hyangchio significantly in case of non- topped.
