The author studied on the effects of X-ray irradiation to the development of periodontal ligament in gestation rats. They were irradiated in their abdomen with 100, 200 and 300 rads respectively in one shot irradiation with deep radiation therapy equipment (MAXIMAR 250-Ⅲ), In 7th, 14th, 21th and 28th day after delivery, those new born rats were respectively sacrificed with ether anesthesia and removed of their mandibles. After removal, those mandibles were fixed in 10% neutral buffer formalin, decalcified with 5% trichloroacetic acid for 5 days and embedded with paraffine. Staining was performed with H-E, Van Gieson, Mallory azan, Bielshowsky-Gomori silver stain and Halmi's oxytalan fiber stain. The results were as follows: 1. Before tooth eruption, all the fiber: components in dental sac were almost always oriented near the outer enamel epithelial \layer. But in irradiated new born rats, those collagen fiber orientation was more irregular than those of control groups, and this phenomenon was more severe in proportion to the amount of irradiation in the gestation period. 2. Before tooth eruption, the connective tissue fibers in periodontal ligament were stained with lighter in the irradiated groups than those of control groups. Oxytalan fibers of irradiated groups were thin and splitting pattern of their fiber morphology to compare with those of control groups. 3. After tooth eruption, the periodontal ligament fibers of irradiated groups were oriented functionally and their morphology was thick, fine and heavy staining. Oxytalan fibers were revealed with oblique parallel arrangement in the periodontal ligament of irradiated groups.
Background: The size of the tooth whitening market and toothpaste market is increasing worldwide. The purpose of this in vitro study is to confirm and compare the coffee stain removal effects of commercial whitening toothpaste in sound and demineralized teeth, respectively. Methods: A total of 112 flat permanent bovine teeth specimens were manufactured. Half of the surface of the specimen was coated with an acid-resistant varnish and deposited in an artificial demineralizing solution for 65 hours. The varnish applied to half of the specimen was removed and deposited in a coffee solution for 96 hours to induce coloring. Two control and five experimental group toothpastes for teeth whitening were selected and the main components were investigated. Toothbrushing was performed 50, 100, and 150 times for each toothpaste group. A total of four images were obtained: before the start and after 50, 100, and 150 times of brushing to obtain the lightness (L*) values of the sound and the demineralized tooth surfaces. The difference in the average value between toothpaste groups at each treatment period was analyzed by one-way ANOVA. The difference in the L* average value according to the number of the brushing was analyzed by repeated measure ANOVA. Results: All toothpastes in the seven groups contained abrasive agents and had different ingredients for each product. Compared to before brushing, the L* value changed significantly in all toothpaste groups after brushing 50 times (p<0.05). This was common in both the sound and demineralized teeth surfaces. Demineralized teeth had significantly lower L* values at all brushing times than that in sound teeth (p<0.05). Conclusion: The effect of whitening teeth was different for each toothpaste. Demineralized teeth were more likely to cause coloration than sound teeth, and the coloration was not removed well.
The purpose of this investigation was to study the efficacy and safety of 6% hydrogen peroxide gel as a daily home tooth bleaching gel. The subjects consisted of 20 male dental students representing a variety of acquired stain and each subject participated for a 4-week period. Tooth color analysis(Shade determination), sulcus bleeding index, probing depth and probing attachment level were done and recorded at baseline and at the end of each week of study. The results indicated that home bleaching gel containing 6% hydrogen peroxide was effective and caused no gingival inflammation. Sulcus bleeding index, probing depth and probing attachment level showed no change. In conclusion, 6% hydrogen peroxide gel is an effective and safe agent for daily home tooth bleaching.
To test flavonoids for antibacterial activity against oral micraorganisms, flavonoids, quercetrin and naringenin, were incorporated into two pharmaceutical preparations in the form of tooth paste. Samplees of dental plaque, the msot accused dental deposit which initiates the gingival and periodental diseases, were collected from the teeth surface of ten dental students at one week interval before and after using placebo, followed by two formulae of tooth paste containing 0.1% of quercetrin and naringenin (formulas I and II, respectively). The amount of dental plaque was assessed by the quigley and Hens index. Then plaque samples were subjected to bacteriological examination of Gram stain and plate counts of microorganisms. The amount of dental plaque was assessed by the Quigley and Hens index. Then plaque samples were subjected to bacteriological examination of Gram stain and plate counts of microorganisms. The results revealed that most of Gram negative cocci and bacilli were highly affected by the two formulae : the number of actinomycetes were decreased after using formula I and disappeared completely by the sue of formula II, while the number of Gram positive streptococci was highly decreased after the treatment with the two formulae. These results indicate a possible use of flavonoids to inhibit dental plaque formation.
Human teeth vary widely in color. Practitioner and patients are concerned with preventing and correcting discolored or dark teeth to achieve and maintain stain-free, white teeth. Tooth brushing cannot alter tooth color but it can remove adhering films and stains. The esthetics of natural dentition can be improved by bleaching and this process can be applied to intrinsically and extrinsically stained teeth. The need for a brighter, more attractive smile has made rapid growth in the market for tooth whiteners. There is no doubt these products work as whiteners, at least on mild to moderate stains, but the safety of these products are unclear. In this experiment, the effect of tooth whitener application on the color and microhardness of extracted human enamel was measured. RMS, RMT and NWT were used as tooth whiteners, and tooth paste(ETQ) and hydrogen peroxide solution(HPO) were used as controls. 35 caries-free extracted human molars were embedded and polished with the exposed enamel diameter of 4 mm. The tooth whiteners and control agents were applied according to the manufacturers' instructions or clinically simulated procedures for eight weeks, and measurements were repeated every two weeks. Value(L*) difference was measured using Differential Colorimeter(Model TC-6FX, Denshoku Co., Japan), and microhardness was measured using microhardness tester(Mitsuzawa Seiki Co., Japan). The results were as follows; 1. After application of agents for eight weeks, the Vickers hardness increased significantly in the ETQ, RMS and RMT application group(p〈0.01), and that decreased significantly in NWT application group(p〈0.01), but in HPO application group there was no significant change. The change in microhardness was greatest in NWT application group(p〈0.01). 2. After application of tooth whiteners and controls for eight weeks, the value change of toothpaste application group was significantly lower than those of other agents groups(p〈0.01), and there was no significant difference in value(L*) change among tooth whitener groups(p〉0.01). 3. The application of tooth paste and paste type tooth whitener made gradual value change, but hydrogen peroxide gel type tooth whitener and hydrogen peroxide solution made rapid value change during initial application period.
Background: Dental caries and periodontal disease are bacterial infectious disease, mainly caused by plaque, a bacterial colony deposited on the tooth surface and gum tissue. Dental plaque disclosants easily stain the dental plaque, making them effective for scaling and tooth brushing education. As the erythrosine typically contained in dental plaque disclosants is highly cytotoxic, a low toxicity additive is needed. In this study, we aimed to examine the natural pigments with negligible cytotoxicity but can effectively stain the dental plaques for use in dental plaque disclosants. Methods: The pigmentation of eight types of natural pigments was tested on bovine tongue and teeth, as well as on head and neck tissue sections of experimental ICR mice. The cytotoxicity of gingival epithelial cells was measured via MTT assay. Pigmentation was performed on the bovine tongue and tooth surface. Pigmentation in the oral environment was observed in four mandibular incisors. A 2 Tone was used as a control. Results: Of the eight types of natural pigments, purple and blue pigments were effective in coloring dental plaques on the enamel surface as well as in the head and neck tissue sections. Additionally, purple and blue pigments were visible on the surface of the bovine tongue. Red, pink, orange, green, purple, and yellow pigments showed strong cytotoxicity, whereas brown and blue pigments had relatively low cytotoxicity. Blue pigment was effective in staining the dental plaque of four mandibular incisors. Conclusion: We suggest that the blue pigment derived from Gardenia jasminoides Ellis (Rubiaceae), which is effective for coloring dental plaques and has low cytotoxicity, is useful as a naturally derived dental disclosant.
Statement of problem: The proper contact relation between adjacent teeth in each arch plays an important role in the stability and maintenance of the integrity of the dental arches. Proximal contact has been defined as the area of a tooth that is in close association, connection, or touch with an adjacent tooth in the same arch. Purpose: The aim of this study was to develop a digital device for measuring the proximal tooth contact tightness by pulling a thin stainless steel strip (2mm wide, 0.03mm thick) inserted between proximal tooth contact. Material and method: This device consists of measuring part, sensor part, motor part and body part. The stainless steel strip was connected to a stain gauge. The strain gauge was designed to convert the frictional force into a compressive force. This compressive force was detected as a electrical signal and the electrical signal was digitalized by a A/D converter. The digital signals were displayed by a micro-processor. The pulling speed was 8mm/s. Results: For testing reliability of the device in vivo, two healthy young adults (A, B) participated in this experiment. The tightness of proximal tooth contact between the second premolar and the first molar of mandible (subject A) and maxilla (subject B) was measured fifteen times for three days at rest. We double-checked the accuracy of the device with a Universal Testing Machine. Output signals from the Universal Testing Machine and the measuring device were compared. Regression analysis showed high linearity between these two signals. In vivo test, no significant differences were found between measurements. Conclusion: This device has shown to he capable of producing reliable and reproducible results in measuring proximal tooth contact. Therefore, it was considered that this device was appropriate to apply clinically.
The author observed the effects of the cobalt-60 irradiation on the amelogenesis and dentinogenesis of the albino rat fetuses by means of histological and histochemical methods. Females in oestrus were mated overnight and examined the next morning for evidence of copulation. The lower left abdomen of mothers were exposed to cobalt-60 irradiation on the 10th day of gestation, l00R 200R and 300R respectively. The fetuses were removed from the mothers on the 18th day of gestation. The employed histochemical methods were PAS reaction, colloidal iron reaction, aldehyde fuchsin stain, α-amino acid reaction, -SH radical reaction and methyl- green pyronin stain. The results were as follows; 1. The group irradiated by l00R made no histological differences in comparison with the control group. 2. Increasing the irradiation to 200R, abnormal dentin formation occured, and resulted in enamel hypoplasia and in atrophy and necrosis of odontoblasts. In dentinal papilla, the dilation and the degeneration of the blood vessels, excessive reticular atrophy and osteodentin were revealed. 3. With the more irradiation (200R-300R), the positive material of PAS, α-amino acid and aldehyde fuchsin tended to decrease in the ameloblast and the odontoblast. No significant changes appeared in DNA, the stainability of methylgreen pyronin.
It is known that radiation therapy is a kind of treatment choices of the maxillofacial tumors. This study is designed to investigate the irradiation effects on rat's periodontal tissues as functional tissues which relate to tooth-support, hard tissue formation and destruction. 20 rats (Sprague-Dowley branch, male) were devided into control group of 4 and experimental groupe of 16. Experimental group was singly exposed to Co-60 irradiation with 10 Gy in the head and neck region. Animals were sacrificed on 2 days, 1 week, 2 weeks and 3 weeks after the irradiation. The specimens were observed by histopathological examination employing H-E stain, van-Gieson stain and PA-ACH fluorescent stain. The results were as follows: 1. Cementoblasts and osteoblasts were gradually lost and rearranged along the external surfaces of the cementum and alveolar bone, but osteoclasts were almost not affected. 2. The cell numbers of the periodontal ligament were decreased due to the cellular atrophy and degeneration, but recovered almost normally on the 3rd week after irradiation. 3. The collagen fibers within the periodontal ligament were irregularly oriented, became finer and decreased in number. 4. The vessels of the periodontal ligament were decreased at the initial stage but increased again on the 2nd week after irradiation, and the hemorrhagic appearances, occurred within the tissues, due to the arterial destruction, were lasted until 3 weeks after irradiation. 5. The glycogen within the periodontal ligament was gradually increased and stored in the matrices of the cemental side on the 1st week after irradiation, but recovered almost normally on the 3rd week after irradiation.
