For the regeneration of periodontal tissues, the microenvironment for new attachment of connective tissue fibers should be provided, At this point of view, cementum formation in root surface plays a key role for this new attachment. This study was performed to figure out which factor promotes differentiation of cementoblast Considering anatomical structure of tooth, we selected the cells which may affect the differentiation of cementoblast - Ameloblast, OD11&MDPC23 for odontoblasts, NIH3T3 for fibroblsts and MG63 for osteoblasts. And OCCM30 was selected for cementoblast cell line. Then, the cell lines were cultured respectively and transferred the conditioned media to OCCM30. To evaluate the result, Alizarin red S stain was proceeded for evaluation of mineralization. The subjected mRNA genes are bone sialoprotein(BSP), alkaline phosphate(ALP) , osteocalcin(OC), type I collagen(Col I), osteonectin(SPARC ; secreted protein acidic and rich in cysteine). Expression of the gene were analysed by RT-PCR, The results were as follows: 1. For alizarin red S staining, control OCCM30 didn't show any mineralized red nodules until 14 days. But red nodules started to appear from about 4 days in MDPC-OCCM30 & OD11-OCCM30. 2. For results of RT-PCR, ESP mRNAs of control-OCCM30 and others were expressed from 14 days, but in MDPC23-OCCM30 & OD11-OCCM30 from 4 days. Like this, the gene expression of MDPC23-OCCM30 & OD11-OCCM30 were detected much earlier than others. 3. For confirmation of odontoblast effect on cementoblast, conditioned media of osteoblasts(MG63) which is mineralized by producing matrix vesicles didn't affect on the mineralized nodule formation of cementoblasts(OCCM30). This suggest the possibility that cementoblast mineralization is regulated by specific factor in dentin matrix protein rather than matrix vesicles. Therefore, we proved that the dentin/odontoblast promotes differentiation/mineralization of cementoblasts. This new approach might hole promise as diverse possibilities for the regeneration of tissues after periodontal disease.
Most of cervical abrasion and erosion lesions show gingival margin where the cavosurface angle is on cementum or dentin. Composite resin restoration of cervical lesion shrink toward enamel margin due to polymerization contraction. This shrinkage has clinical problem such as microleakage and secondary caries. Several methods to diminish contraction stress of composite resin restoration, such as modifying cavity form and building up restorations in several increments have been attempted. The purpose of this study was to compare polymerization contraction stress of composite resin in Class V cavity subjected to cavity forms and placement methods. In this study, finite element model of 5 types of Class V cavity was developed on computer tomogram of maxillary central incisor. The types are : 1) Box cavity 2) Box cavity with incisal bevel 3) V shape cavity 4) V shape cavity with incisal bevel 5) Saucer shape cavity. The placement methods are 1) Incisal first oblique incremental curing 2) Bulk curing. An FEM based program for light activated polymerization is not available. For simulation of curing dynamics, time dependent transient thermal conduction analysis was conducted on each cavity and each placement method. For simulation of polymerization shrinkage, thermal stress analysis was performed with each cavity and each placement method. The time-temperature dependent volume shrinkage rate, elastic modulus, and Poisson's ratio were determined in thermal conduction data. The results were as follows : 1. With all five Class V cavifies, the highest Von Mises stress at the composite-tooth interface occurred at gingival margin. 2. With box cavity, V shape cavity and saucer cavity, Von Mises stress at gingival margin of V shape cavity was lower than the others. And that of box cavity was lower than that of saucer cavity. 3. Preparing bevel at incisal cavosurface margin decreased the rate of stress development in early polymerization stage. 4. Preparing bevel at incisal cavosurface margin of V shape cavity increased the Von Mises stress at gingival margin, but decreased at incisal margin. 5. At incisal margin, stress development by bulk curing method was rapid at early stage. Stress development by first increment of incremental curing method was also rapid but lower than that by bulk curing method, however after second increment curing final stress was the same for two placement methods. 6. At gingival margin, stress development by incremental curing method was suddenly rapid at early stage of second increment curing, but final stress was the same for two placement methods.
The purpose of this in vitro study was to evaluate the microleakage of 5 curing methods in class V composite restorations which are composed of two-step light curing, pulse-delay cure, low curing-light intensity, moderate curing-light intensity and high curing-light intensity. In this study, class V cavities were prepared on buccal and lingual surfaces of 50 extracted human upper or lower molars on cementum margin. Single Bond adhesive and Z-100 shade A2 were applied for each group following the manufacture's instruction. The experimental teeth were randomly divided into 5 groups of 10 samples (20 surfaces) each. Group 1: two-step light curing; Group 2: pulse-delay cure; Group 3: low curing-light intensity; Group 4: moderate curing-light intensity; Group 5: high curing-light intensity. After 500 thermocycling between $5^{\circ}C$ and $55^{\circ}C$, the 60 teeth were placed in 2% methylene blue dye for 24 hours, then rinsed with tab water. The specimens were embedded in clear resin, then sectioned buccolingually through the center of restoration with a low speed diamond saw. The dye penetration on each of the specimen was then observed with a stereomicroscope at ${\times}20$. The composite resin/tooth interfaces were examined under Scanning Electron Microscopy. The results were statistically analyzed using the Kruskal-Wallis One Way ANOVA and Dunn's Method. The results of this study were as follows. 1. In all groups, the leakage values seen at the enamel margin were significantly lower than those seen at the dentin margin(P<0.05). 2. No group in this study showed significant differences in leakage values at both the enamel and the dentin margins(P<0.05). 3. In all groups, the gaps seen at the enamel margin were significantly lower than those seen at the dentin margin(P<0.05). 4. The gaps in this study showed significant differences and two-step light-curing and low curing-light intensity produced significant less gap than high curing-light intensity(P<0.05).
치아이동시 골세포간 활성 조차에서 세포간 교통의 중요성이 차츰 인식되고 있으며 세포간 교통을 위한 교통반의 존재가 확인되고 있다. 동물실험 모델에서 실험적 치아이동을 통해 조성된 압박 및 견인측 치주인대에서 교통반 단백의 일종인 connexin 43의 발현을 관찰함으로써 인접 연조직 및 골 조직 내에서 세포 신호 전달 양상의 한 부분을 파악하고자 하였다. Sprague-Dawley계 백서 27마리를 대조군(3마리)과 실험군(24마리)으로 나누었으며, 실험군은 견인력(75g)을 가한 후 12시간, 1일, 4일, 7일, 14일, 28일이 경과한 후 각각 4마리씩 희생시켜, connexin 43의 발현을 면역조직화학적으로 관찰한 바 다음과 같은 결과를 얻었다. 1. 대조군에서의 connexin 43 발현은 치은, 상아질, 치주인대, 치수, 골세포에서 경미하였다. 2. 교정적 치아이동에 반응하는 치수, 치주인대, 골모세포, 파골세포, 골세포부위에서 connexin 43의 발현이 많았으나 치은, 상아질, 상아모세포에서의 발현은 견인력 적용기간에 관계없이 대조군과 큰 차이가 없었다. 3. 치수에서 connexin 43의 발현은 교정력을 가한 4일에서부터 증가하여 7일째까지 크게 증가한 후 14일째부터 감소되었으며 28일째에는 대조군과 차이가 없었다. 4. 치주인대에서 connexin 43의 발현은 주로 혈관을 중심으로 치조골 면을 따라 압박측에서 주로 나타났는데 교정력을 가한 12시간에서부터 증가하여 4일에서 가장 많은 발현을 보인 후 7일부터 감소하기 시작하였다. 5. 골모세포 및 골세포에서 발현은 실험 1일째부터 출현해 4일째에 가장 많은 발현을 보였고 7일 이후에는 크게 감소하였으며 파골세포에서의 발현은 골모세포와 골세포보다 실험 4일째에 더 많이 관찰할 수 있었다.기계화와 자동화(26.2%), 정성부족으로 인한 맛의 감소(21.6%) 순으로 나타났고, 성별(p<0.01), 연령별(p<0.05)로 유의적 인 차이를 보였다. 7. 전통음식 패스트푸드점이 생기면 이용도에 관한 견해는 '가끔 이용할 것이다'가 56.5%, '많이 이용할 것이다'가 19.8%로 나타났고, 연령별(p<0.05), 주거상태별(p<0.05)로 유의적인 차이를 보였는데 연령이 많을수록 '이용치 않겠다'는 비율이 높았고, 특히 주거상태별로는 '전혀 이용치 않을 것이다'의 비율이 하숙생이 높게 나타났다. 본 연구는 유행에 민감하고 문화의 변화를 주도하며 패스트푸드를 가장 많이 이용하는 세대인 대학생들을 대상으로 우리의 전통음식을 패스트푸드화 하였을 때의 견해를 알아보고 그 전망을 살피기 위한 연구였다. 연구결과에서 나타난 바와 같이 유행의 첨단을 달리는 신세대라 하더라도 많은 비율의 대학생들이 전통음식의 패스트푸드화에 전망을 밝게 보고 있었으며 이를 위해서는 많은 문제점도 지적하였다. 이에 전통음식의 패스트푸드화와 상품화에 대한 인식이 고무적인 방향으로 변화하고 있어 앞으로 계속 그 추이를 연구할 가치가 있다고 사료된다.ematured infants during the early weeks of life. 모든 치근단 수준에서 비표준화 medium 크기 master cone 사용군이 ISO 표준화 규격의 master cone 사용군에 비해 유의하게 높은 gutta-percha 면적비를 나타내었다 (p < 0.01). 비표준화 medium크기 master cone 사용군에서는 표준화 규격의 master cone 사용군에 비해 유의하게 적은 수의 accessory cone이 사용되었다 (p < 0.01). 대학생간에는 유의한 차이(p<0.05)가 인정되었다. 응답자의 체형은
치아에 압하력을 elastic chain을 이용하여 지속적으로 교정력을 평균 9개월된 유성견의 소구치에 가한 직후 치아와 그 주위조직에서 일어나는 조직학적인 변화와 압하력을 제거한 후 일정기간이 지난 후 변화되었던 조직의 회복양상을 고찰하고, 이를 토대로 교정치료를 위한 생물학적인 근간을 마련하기 위하여 본 연구를 시행하였으며, Hematoxylin & Eosin 및 Masson's Trichrome 염색법을 이용하여 치아와 주위조직의 반응을 치경부, 치근 중앙부, 치근단부로 나누어 광학현미경하에서 관찰하여 다음과 같은 결론을 얻었다. 1. 대조군의 HE 소견에서 치주인대는 치근단부에서 치근경부까지 비교적 균일한 두께를 나타냈고, 치주인대의 주행방향은 치근경부에 수평 또는 약간 경사져 있었으며, 치근 중앙부와 치근단부에서는 사선으로 주행하였다. 2. 대조군의 MT 소견에서 치경부에서는 조골세포 및 파골세포가 나타났으며 치경부, 치근 중앙부 그리고 치근단부에 약한 골흡수와 신생골 침착이 보였다. 3. 실험 1 군에서는 치근단부에서 뚜렷한 파골세포의 증가와 혈관 충혈이 보였으며, 신생골 형성과 불규칙한 반전선들이 나타나 골흡수하는 양상을 보였다. 치주인대는 치경부에서 치근단에 이르기까지 뚜렷한 증가를 보였으며 치근단부에서 더 심하였다. 4. 실험 2군에서는 대조군에 비해 조골세포가 치근단부에서 혈관 충혈과 함께 여전히 관찰되었다. 치주인대 폭경은 치근단부에서 대조군에 비해서는 증가되었고 1 군에 비해서는 감소된 양상을 보였다. 치주인대 주행방향은 1군과 같았고 단지 치근단부에서 혼합된 양상으로 관찰되었다. 이상과 같은 소견으로 보아 유성견 하악 소구치 압하시 초기 조직변화 에서 압박을 받는 치근단부에서 파골세포의 뚜렷한 증가, 출혈상 및 간격 확장, 그리고 골흡수와 백악질 흡수가 현저히 나타났으며, 치주인대 주행 방향도 다양하였다. 또한 압하력을 제거한 후에 변형되었던 치아와 치아 주위조직들이 완전한 회복 양상을 보이지는 않았다.
Flowable composite resin has lower filler content, increased flow, and lower modules of elasticity. It is suggested that flowable composite resin can be bonded to the tooth structure intimately and absorb or dissipate the stress. Therefore, it may be advantageous to use flowable composite resin for the base material of class II restoration and for the class V restoraton. The purpose of this study was to evaluate the microleakage and shear bond strength of four flowable composite resins (Aeliteflo, Flow-It, Revolution, Ultraseal XT Plus) compared to Z100 using Scotchbond Multi Purpose dentin bonding system. To evaluate the microleakage, notch-shaped class V cavities were prepared on buccal and lingual surfaces of 80 extracted human premolars and molars on cementum margin. The teeth were randomly divided into non-thermocycling group (group 1) and thermocycling group (group 2) of 40 teeth each. The experimental teeth of each group were randomly divided onto five subgroups of eight samples (sixteen surfaces). The Scotchbond Multi-Purpose and composite resin were applied for each group following the manufacturer's instructions. the teeth of group 2 were thermocycled five hundred times between 5$^{\circ}C$ and 55$^{\circ}C$. The teeth of group 2 were placed in 2% methylene blue dye for 24 hours, then rinsed with tab water. The specimens were embedded in clear resin, and sectioned longitudinally with a diamond saw. The dye penetration on each of the specimen were observed with a stereomicioscope at $\times$20 magnification. To evaluate the shear bond strength, 60 teeth were divided into five groups of twelve teeth each. The experimental teeth were ground horizontally below the dentinoenamel junction, so that no enamel remained. After applying Scotchbond Multi-Purpose on the dentin surface, composite resin was applied in the shape of cylinder. The cylinder was 4mm in diameter and 2mm in thickness. Shear bond strength was measured using Instron with a cross-head speed of 0.5mm/min. After shear bond strength measurement, mode of failure was evaluated with a stereomicroscope at $\times$30 magnification. All data were statistically analyzed by One Way ANOVA and Student-Newman-Keuls method. The correlation between microleakage and shear bond strength was analyzed by linear regression. The results of this study were as follows ; 1. In non-thermocycling group, the leakage value of Z100 was significantly lower than those of flowable composite resins at the enamel and dentin margin, margin, except that Revolution showed the lower leakage value than that of Z100 at the dentin margin (p<0.05). 2. In thermocycling group, the leakage values of Z100 and Ultraseal XT Plus were lower than those of other subgroup at the enamel and dentin margin, except that Flow-It showed the lower leakage value than that of Ultraseal XT Plus at the dentin margin (p<0.05). 3. The leakage value of Z100 and Ultraseal XT Plus in thermocycling group were not higher than that in non-thermocycling group at the enamel margin. The leakage value of Z100 in thermocycling group was not higher than that in non-thermocycling group at the dentin margin (p<0.05). 4. As for the shear bond strength measurement, there were no statistically significant differences among groups (p<0.05). The shear bond strengths given in descending order were as follows: Z100(16.81$\pm$2.98 MPa), Flow-It(14.8$\pm$4.43 MPa), Aeliteflo(14.34$\pm$3.69 MPa), Revolution(13.46$\pm$4.23 MPa), Ultraseal XT Plus(12.83$\pm$3.16 MPa). 5. Failure modes of all specimens were adhesive failures. 6. There was no correlation between microleakage and shear bond strength.
