Three strains of Cucumber mosaic virus (CMV) have been found to cause a lethal disease, referred to as fern leaf syndromes and mild mosaic symptoms in tomato (Lycopersicon esculentum Mill.) crops grown in Kuwait. CMV strains were detected and identified based on host range, symptomatology, serology, electron microscopy, and ribonucleic acid (RNA) electrophoresis in polyacrylamide gels. A high degree of viral genomic heterogeneity was detected among CMV strains isolated in Kuwait, with no apparent correlation to symptomatology in tomato host plants. Two different virus satellites of 'CMV associated RNA 5', designated CARNA 5, were detected in two virus strains that caused both lethal disease and mild symptoms, designated CMV-D1 and CMV-S1 respectively. CARNA5 was not detected in the third CMV strain that caused fern leaf syndromes designated CMV-F. All the three isolated strains were serologically indistinguishable from each other and may belong to one serotype according to Ouchterlony gel diffusion tests. These strains transmitted via aphids (Myzus persicae Sulz) in a non-persistent manner. Physical properties of the virus strains were very similar where thermal inactivation test showed that virus withstood heating for 10 min at $70^{/circ}$, dilution end point was $10^{-4}$, and the longevity in vitro at room temperature was less than 5 days for all virus strains. CMV-D1 and CMV-F were the most devastating diseases spreading in both greenhouse and field-grown tomato where aborted flower buds failed on fruit setting due to the viral infection. This is the first report to isolate three different strains of CMV in Kuwait.

Seeds of Tomato (Lycopersicon esculentum Mill.) have demanded high quality because of their high cost of seed. The optimization of the seed priming techniques that have positive effect on fast and uniform germination becomes important at the commercial level. Several factors such as solution composition, osmotic potential, and treatment duration affect seed priming response. In this study, osmotic potentials of priming solution and germination characteristics of primed seed were investigated to clarify the effects different inorganic salt types and the duration. Tomato seeds were primed in osmotic solutions that were osmotic potential ranged -1.54 to -0.45 MPa in an aerated solution of PEG 8000 (17%, 22%, 27%), and inorganic salt solution of $KNO_3$, $Na_2SO_4$, and $K_2SO_4$ (100, 200, 300mM). The seeds were treated at $20^{\circ}C$ for 2, 4, and 6 days. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$. Four replications of 25 seeds per each treatments were placed in 10-cm petri dishes containing two filter papers and 3 ml of $dH_2O$ and incubated at $20^{\circ}C/30^{\circ}C$ and $15^{\circ}C$ and seedlings evaluated for abnormality after 14 days of incubation. Seed water potential (${\psi}$) was correlated with water potential of priming solution ($r^2=0.86$). Seeds primed in 100mM $KNO_3$ resulted the highest germination rate (GR, $63.9 %{\cdot}day^{-1}$) and lowest mean germination time (MGT, 2.0 days) comparing to untreated control ($23.9%{\cdot}day^{-1}$ of germination rate and 4.1 days of MGT) at $20/30^{\circ}C$, even though 96% of germination percentage were not different. Seeds primed in 100mM $KNO_3$ (${\psi}=-0.45MPa$) for 4 days showed ${\psi}=-0.38MPa$. Priming in $Na_2SO_4$, $K_2SO_4$, and PEG solution for 6 days improved MGT and GR, but not significantly than 4 days of treatment. Additionally, stepwise osmotic solution treatment with 100mM and 300mM concentration for 6 day did not showed differences with single treatment. In relation to osmotic potentials, identical osmotic potential in different inorganic salt solution showed different effect on germination characteristics.

Effects of Jasmonic Acid and Wounding on Polyphenol Oxidase Activity in Senescing Tomato Leaves The effects of jasmonic acid(JA) and wounding on polyphenol oxidase(PPO) during leaf senescence was investigated by measuring the PPO activity in detached tomato(Lycopersicon esculentum Mill.) leaves of two-week-old seedlings. The PPO activity in the detached senescing leaves increased significantly in the dark. The leaf segments responded to the application of JA with accelerated senescence, as indicated by the loss of chlorophyll and rapid increase in the PPO activity. The senescence-promoting action of JA differed in the light and dark. Wounding the detached senescing leaves by scraping surface segments or making punctures with needles considerably delayed the loss of chlorophyll and had a significant effect on the PPO activity, the amounts of which were roughly proportional to the intensity of the wounding. In the dark, the combination of wounding plus JA resulted in stable levels of chlorophyll and PPO. JA and ABA acted similarly in both unwounded and wounded leaves, however, the amount of chlorophyll and PPO in the wounded segments was always higher than in the respective controls. JA was found to eliminate the senescence-retarding action of benzyladenine. In a histochemical localization test, the PPO activity was found to be localized in the cell walls of the parenchyma tissue, thereby indicating moderate cytoplasmic reactions. In the JA-treated plants, the PPO activity was intense in the cells of the cortex and phloem parenchyma. Accordingly, based on these observations it would appear that PPO is a component of a defense response maker, whereas JA plays an integral role in the intracellular signal transduction involved in inducible defense mechanisms.
The effects of wounding and jasmonic acid(JA) on polyphenol oxidase(PPO) in tomato(Lycopersicon esculentum Mill.) seedlings were investigated. PPO was strongly induced by wounding or JA, and the response was also shown to be systemically induced by wounding. Mechanical wounding in cotyledon or leaf produced a signal that caused the concentration of PPO to increase in the unwounded cotyledon, in the first leaves but not in the second leaves. Severity of wounding and light intensity also affected wound induced change in PPO activity, JA showed a stimulatory effect on the loss of chlorophyll and the rapid increase in PPO activity. The PPO was clearly more active in the wounded leaves than in controls. The potency and specificity of the JA indicate a close relationship between JA and wound-induced changes in PPO in tomato species. JA and abscisic acid(ABA) acted similarly on both unwounded and wounded leaves, but the amount of PPO in the wounded leaves was always more than the respective controls. The highest increase in PPO activity occurred in woundand JA-induced leaves of seedlings kept under bright lighting. Benzyladenine(BA) completely abolished JA- and ABA-induced PPO activity. The results suggest that JA-induced PPO activity is due to de novo PPO synthesis. Histochemical tests for PPO in stems of wound- and JA -treated tomato plants indicate that PPO was localized primarily, in the. outer .cortex . and xylem parenchyma. It is concluded that exogenously applied JA acts as stress agents and PPO may be a component of the inducible anti-hervivore defense response.
The purpose of this research was to develop ideal substrates for the production of good quality tomatoes in bag culture system and also to improve media with low or no environmental pollution by blending and mixing artificial substrate including peatmoss, perlite, vermiculite, granular rockwool, polyphenol resin foam, bark, and smoked rice hull. The highly efficient media proved by experiments were vermiculite, smoked rice hull, polyphenol resin foam, granular rockwool, and perlite, which showed good results in the early growth as well as the marketable yield of tomato in the bag culture. Tomato plants grown in the media mixed with peatmoss, vermiculite and granular rockwool at the ratio of 3 : 1 : 1(by volume) showed the highest marketable yield, and the next at the ratio of 2 : 1 : 1. The perlite-granular rockwool mixtures at the ratio of 2 : 3 and 1 : 4, and the peatmoss vermiculite mixtures at the ratio of 2 : 3 and 3 : 2, seemed to be promising media for bag culture.
The tomato ripening associated membrane protein (TRAMP) (Fray et al., 1994) is a member of the major intrinsic protein (MIP) family, defined as channels facilitating the passage of water and small solutes through membranes. During normal fruit ripening the TRAMP mRNA levels were increased whereas the expression levels of TRAMP in low ethylene ACO1-sense suppressed lines, Nr and rin fruits, were lower than at the breaker stage of wild type fruit. TRAMP mRNA is inhibited by $LaCl_3$, which is an inhibitor of $Ca^{2+}$-stimulated responses, treatment but drought condition did not affect TRAMP expression. The levels of TRAMP mRNA transcripts were substantially higher in the dark treated seedlings and fruits. These suggest that TRAMP function as a water channel may be doubted because of several reasons; no water content was changed during ripening in wild type, antisense and overexpression lines, TRAMP expression under light condition was lower than dark condition and TRAMP expression was not changed in drought condition. Co-suppression plant, 3588 was one of sense suppression lines, which contain CaMV 35S promoter and sense pNY507 cDNA, produced small antisense RNA, approximately 21-25 nucleotides in length, mediated post-transcriptional gene silencing. Therefore, TRAMP expression was inhibited by small antisense and multiple copies might induce gene silencing without any production of double strand RNA. Total seven selected volatile productions, isobutylthiazole, 6-methyl-5-hepten-2-one, hexanal, hexenal methylbutanal, hexenol, and methylbutanol, were highly reduced in sense line whereas total volatile production was increased in TRAMP antisense line. These results suggested TRAMP might change volatile related compounds.
To test a potential salt tolerance in phosphorus (P) efficient plants (T9 and T8), tomato seedlings were hydroponically grown in saline media. The tolerance was evaluated by comparing growth and metabolism against T5, non-P-efficient variety, at different salt concentrations: 0, 1, 5, 10 g/L. Fresh weights (FW) were measured weekly. Dry weight (DW), mineral contents, and stomatal resistance (Rs) were measured at the termination of experiment. At the lower two salt concentrations (0, 1 g/L), no significant difference was observed in terms of FW, DW, and Rs. At 5, 10 g/L of salt concentration, however, significant variation is evident: T9 and T8 outperformed T5. On the other hand, no difference was also in N, P, K, and Na contents at the corresponding salt concentration. These observations together indicate that P-efficient strain can better tolerate to salinity.
An anthocyanin-deficient tomato (Lycopersicon esculentum Mill.) strain, H957, shows an unusual tolerance to low phosphorus (P). To investigate whether the tolerance originates from a tissue/cellular strength, plant tissue culture procedure was employed which facilitate to characterize the tolerance independent of morphological features. The tolerance was analyzed by comparing H957 against H883, its maternal wild type, while each explant was co-cultured on minimal P media. Comparisons were made in fresh weight, dry weight, callus and shoot formation, mineral contents, and P utilization ratios at $0-400{\cdot}\bar{I}MP$, . Growth of the two strains was severely impaired at 0 and $12.5{\cdot}\bar{I}MP.\;At\;25-200{\cdot}\bar{I}MP$, however, H957 consistently showed a greater fresh and dry weight than H883. Shoot onset of H957 was less delayed than H883 compared to optimal P conditions. H957 tissue contains an overall lower P concentration than H883. These observations indicate that H957 may tolerate to low P by its tissue or cellular strength in P utilization side from its morphology.

This study was conducted to examine the potential of inducing salinity stress on cylindrical paper pot tomato seedlings to inhibit overgrowth. Potassium fertilizers, sulfate of potash (K2SO4), muriate of potash (KCl), and monopotassium phosphate (KH2PO4), were prepared as two solutions of (5 and 10) dS·m-1 salinity level, respectively, to investigate the influence on tomato (Lycopersicon esculentum Mill.) seedling growth. We also investigated the adaptability and survivability of treated tomato seedlings with high-salinity potassium (10 dS·m-1 KCl) to harsh environmental conditions (water deficit, low temperature, and storage conditions). Repeated addition of high-salinity level KCl, K2SO4, or KH2PO4 markedly decreased the dry matter of shoot and root, leaf area, and net assimilate rates (NAR) but increased the stem diameter of seedlings. Among the three sources, the relative growth rate of plant height (RGRH) was most sensitive to KCl addition; increasing salinity levels of KCl solution decreased the RGRH of seedlings. The compactness, which directly reflects the stocky growth index, increased in KCl or KH2PO4 treatments. After a week's water deficit, severely wilted seedlings were observed in control seedlings (untreated with KCl), but no wilted seedlings were observed in the KCl treated seedlings, and the relative water content (RWC) of the untreated seedlings significantly decreased by 23 %, while that of the pretreated seedlings only decreased by 8 %. The increase in ion leakage of KCl treated seedlings at low temperatures was less than that of untreated seedlings. Furthermore, there was far lower damage proportion on pretreated seedlings at (9, 12, and 15)℃ storage temperatures after 20 days, compared with on unpretreated seedlings. Our results suggest that high-salinity potassium fertilizer, especially KCl, is effective in preventing tomato seedling overgrowth, while it also improves tolerance.
The objective of this study is to investigate the effect of the level of $CO_2$ (370 and $650{\mu}mol\;mol^{-1}$) and temperature (ambient and ambient+$5^{\circ}C$) on tomato growth and fruit characteristics as affected by the application rate of N-fertilizer (68 and $204\;N\;kg\;ha^{-1}$), for the purpose of evaluating the influence of elevated $CO_2$ and temperature on tomato crop. The elevated atmospheric $CO_2$ and temperature increased the plant height and stem diameter for tomato crop, while the differences among the nitrogen(N) application rates were not significantly different. Under the elevated $CO_2$, temperature, and a higher N application rate, the biomass of aerial part increased. The fruit yield showed the same result as the biomass except for the elevated temperature. The elevated temperature made the size of fruit move toward the small, but the elevated $CO_2$ and the application of N-fertilizer were vice versa. The sugar content and pH of fruit juice were affected by nitrogen application rate, but not by the elevated $CO_2$ and temperature. These results showed that both the elevated $CO_2$ and temperature stimulated the vegetative growth of aerial parts for tomato, but each effects on the yield of fruit showed an opposite result between the elevated temperature and $CO_2$. In conclusion, the elevated $CO_2$ increased tomato yield and the ratio of large size of fruit, but the elevated temperature did not. Therefore, to secure the productivity of tomato as nowadays in future environment, it will need to develop new breeder as high temperature-tolerable tomato species or new type of cropping systems.
