배추 유래의 cytosolic glutathione reductase 유전자 (BcGR1)의 지속적 발현과 형질전환 식물체의 oxidative stress에 대한 내성과의 관계를 분석하기 위하여, BcGR1 유전자를 CaMV 35S promoter의 하류에 연결한 다음, 담배에 형질전환하였다. PCR 및 Southern blot 분석을 통하여 BcGR1 유전자가 정상적으로 삽입된 32 계통의 T$_{0}$ 식물체를 선발하였다. Northern blot 분석 결과, 도입된 유전자가 형질 전환 식물체 내에서 항상적으로 발현된다는 것을 확인하였으며, 도입 유전자의 copy number와 발현량 사이에는 정의 상관관계를 보이지 않았다.
This study was conducted to develop a resistant tobarro against Potato virus Y (PVY) by transformation of the plants with genetically engineered viral genes. The complimentary DNAs (cDNAS) of potato virus Y-necrosis strain (PVY-Vn) replicase mutant genes (3'-deleted, 5'-deleted and ADD-mutant Nlbs) were synthesized through RT-PCR by using purified PVY-VN RNA and synthesized primers, and cloned in the sense orientation into a plant expression vector (pMBPI), The cDNAS of the genes were transferred into Agrobacterium tumefaciens LBA 4404, and then transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Regenerated plants were tested for PVY resistance by inoculation test; 13 transgenic plants including 7 for 3'-deleted Nlb, 3 for 5'-deleted Nlb, and 3 for ADD-mutant Nlb appeared to be resistant at 4 weeks after inoculation with PVY-VN. Among the 13 transgenic tobacco plants, 8 plants had no symptom up to 14 weeks after inoculation. The progenies ($T_1$) from self-fertilization of the transgenic lines varied 0.0% to 81.2% in their resistance (% of resistant plants). The analysis of Nlb-31deleted, -5'deleted and -ADD mutant in the $T_1$ plants by polymerase chain reaction (PCR) showed that Nlb-3'deleted, -5'deleted and -ADD mutants were detected in all of the resistant plants. These results suggest that the PVY resistance was inherited in the $T_1$ generation.
This study was conducted to obtain the transgenic Brnssica napus plants with tobacco Apase gene using the binary vector system of Agrobacteriurn fumefociens. The results obtained were summarized as follows: A repressible acid phosphatase gene of Saccharon~yces cerevisiae, pho105 was used for screening of tobacco Apase cDNA. In order to identify Apase gene in tobacco genome, Southern blot analysis was pcrformed and the Apase gcnc may be present as a single copy, or at most two or three copies, in tobacco genome. To isolate the tobacco Apase gene, tobacco cDNA library was constructed using purifed mRNA from -Pi treated tobacco root and the plaque forming unit of the library was 2.8 x $10^5$ pfu/m${\ell}$, therefore the library might cover all expressed mRNAs. Using pho5 as a probe. tobacco Apase cDNA was cloned, and restriction mapping and Southern blot analysis of cDNA insert were revealed that the 3.6 kb cDNA contained tobacco acid phosphatase cDNA. Plasmid pGA695 -tcAPl was constructed by subcloning tobacco Apase cDNA into the Hind site of pGA695 with 35s promoter which can be expressed constitutively in plants. The Brassica napus cotyledonary petioles were cocultivated with the ,4 grobacteriunz and transferred to the selection medium. The transformed and regenerated plants were transplanted to soil medium. Southern blot analysis was done on the transformed plants, and it was confirmed that a foregin gene was stably integrated into the genonies of B. nnpus plants.
Background: Protopanaxatriol (PPT) is an aglycone of ginsenosides, which has high medicinal values. Production of PPT from natural ginseng plants requires artificial deglycosylation procedures of ginsenosides via enzymatic or physicochemical treatments. Metabolic engineering could be an efficient technology for production of ginsenoside sapogenin. For PPT biosynthesis in Panax ginseng, damarenediol-II synthase (PgDDS) and two cytochrome P450 enzymes (CYP716A47 and CYP716A53v2) are essentially required. Methods: Transgenic tobacco co-overexpressing P. ginseng PgDDS, CYP716A47, and CYP716A53v2 was constructed via Agrobacterium-mediated transformation. Results: Expression of the three introduced genes in transgenic tobacco lines was confirmed by Reverse transcription-polymerase chain reaction (RT-PCR). Analysis of liquid chromatography showed three new peaks, dammarenediol-II (DD), protopanaxadiol (PPD), and PPT, in leaves of transgenic tobacco. Transgenic tobacco (line 6) contained $2.8{\mu}g/g$ dry weight (DW), $7.3{\mu}g/g$ DW, and $11.6{\mu}g/g$ DW of PPT, PPD, and DD in leaves, respectively. Production of PPT was achieved via cell suspension culture and was highly affected by auxin treatment. The content of PPT in cell suspension was increased 37.25-fold compared with that of leaves of the transgenic tobacco. Transgenic tobacco was not able to set seeds because of microspore degeneration in anthers. Transmission electron microscopy analysis revealed that cells of phloem tissue situated in the center of the anther showed an abnormally condensed nuclei and degenerated mitochondria. Conclusion: We successfully achieved the production of PPT in transgenic tobacco. The possible factors deriving male sterility in transgenic tobacco are discussed.
식물의 미토콘드리아에서 eukaryotic 유전자 발현하는 양상을 관찰한다는 것은 매우 중요하게 알여져 왔다. 본 실험에서는 식물의 미토콘드리아에서 발현하는 유전자와 GFP가 미토콘드리아에 발현하는지를 구명하였다. 미토콘드리아(mt)에서 발현 하는 AtBI-1 유전자와 GFP 유전자를 35S promoter를 가진 pBin vector에 재조합한 후, 유전자총을 이용한 형질전환법으로 담배의 잎과 cotyledon에 형질전환하여 재분화 된 shoot를 얻었다. mt에서 그 유전자가 발현 되는 것을 현미경하에서 GFP가 발광하는 것으로 확인하였다. 또한 PCR분석과 Southern분석에서도 미토콘드리아에서 AtBI-1 유전자가 발현함을 확인하였다. 따라서 본 연구의 결과로 mt에 관련된 유전자를 식물의 조직에 형질전환 하여 1개 이상의 유전자가 식물의 mt에 삽입되어 그 유전자의 특성이 발현되는데 이용되어 질수 있을것이라 생각된다.
For genetic engineering to be commercially viable, an efficient transformation system is needed to produce transgenic plane from diverse genotypes ("generalized protocol"). Development of such a system requires optimization of a number of components such as gene transfer agent, plant tissues competent for both regeneration and transformation, and control of transgene expression. Although several novel gene transfer methods have been developed for plane, a majority of stably transformed plane express the introduced genes at low levels. Moreover, silencing of selectable marker genes shortly after their incorporation into plant chromosomes may result in low recovery of transgenic tissues from selection. Matrix attachment regions (MARs) are DNA sequences that bind to the cell's proteinaceous nuclear matrix to form DNA loop domains. MARs have been shown to increase transgene expression in tobacco cells, and reduce position in mature transgenic plants. Flanking an antibiotic resistance transgene with MARs should therefore lead to improved rates of transformation in a diversity of species, and may permit recalcitrant species and genotypes to be successfully transformed. Literature review and recent data from my laboratory suggest that MARs can serve as a transformation booster in recalcitrant plant species.
These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.
As one way to approach to cold defense mechanism in plants, we previously identified the gene for protein-tyrosine phosphatase (CaTPP1) from hot pepper (Capsicum annuum) using cDNA microarray analysis coupled with Northern blot analysis. We showed that the CaTPP1 gene was strongly induced by cold, drought, salt and ABA stresses. The CaTPP1 gene was engineered under control of CaMV 35S promoter for constitutive expression in transgenic tobacco plants by Agrobacterium-mediated transformation. The resulting CaTPP1 transgenic tobacco plants showed significantly increased cold stress resistance. It also appeared that some of the transgenic tobacco plants showed increased drought tolerance. The CaTPP1 transgenic plants showed no visible phenotypic alteration compared to wild type plants. These results showed the involvement of protein tyrosine phosphatase in tolerance of abiotic stresses including cold and drought stress.
Tobacco plants(Nicotiana tabacum cv. NC82) transformed with TMV CP cDNA were self-fertilized until 8th generation (R$_{8}$), and the transgenic plants from 6th to 8th generation were analized for their resistance to tobacco mosaic virus(TMV) and stability of the gene expression. The 6th generation of the plants(R$_{6}$) showed high resistance(81-91 %) to TMV at eight weeks after artificial inoculation with the virus. The transgenic cell line 601 was the most prominant in the expression of resistance. 98 % of the plants showed no symptom without any agronomic phynotepe variation when they were inoculated with the virus in a experimental field. However, 2% of the plants were revealed as delay type of symptom with mild mosaic on a few leaves. The viral resistance in greenhouse tests of the 7th generation (R$_{7}$) was 54-64%, and the number of delay type plants were increased than that of 6th generation plants. In the 8th generation, 81 % of the plants was complete resistant to the virus. The TMV CP cDNA of the transgenic plants of each generation was also confirmed by genomic PCR, and there was no systemic viral multiplication in the resistant plants. It suggests that the viral resistance and gene expression of the transgenic plants might be stable through the generations.ons.s.
Interleukin-18 (IL-18), otherwise known as interferon-gamma-inducing factor (IGIF), is one of several well characterized and important cytokines that contribute to host defenses. The complementary DNA (cDNA) of mature human interleukin-18 gene (hIL-18) was fused with the signal peptide of the rice amylase 1A gene (Ramy1A) and introduced into the plant expression vector under the control of a duplicated CaMV 35S promoter. The recombinant plasmid was transformed into tobacco (Nicotiana tabacum L. cv Havana) using the Agrobacterium-mediated transformation method. The integration of the hlL-18 gene into the genome of transgenic tobacco plants was confirmed by polymerase chain reaction (PCR) amplification and its expression was observed in the suspension cells that were derived from the transgenic plant callus by using Northern blot analysis. The hlL-18 protein was detected in the extracts of the transgenic callus and in the medium of the transgenic tobacco suspension culture by using immunoblot analysis. Based upon enzyme-linked immunosorbant assay (ELISA) results, the expression level of the hlL-18 protein approximated $166{\mu}g/L$ in the suspension culture medium. Bioassay results from the induction of $interferon-{\gamma}$ from a KG-1 cell line indicated that the hlL-18 secreted into the suspension culture medium was bioactive.
