Transfer of Potato Virus Y (Necrotic strain) Resistance from Nicotiana africana to Nicotiana tabacum (Nicotiana Tabacum과 Nicotiana africana의 종간교배에 의한 감자바이러스 Y 저항성 유전자원 개발)

  • This study was conducted to transfer the potato virus Y-necrosis strain resistance from Nicotiana africana to Nicotiana tabacum (cv. NC82) N. africana was crossed with NC82, GErmination of the cotyledon stage, at which time most of the seedling died. However, surviving seedlings continued to grow normally. Chromosomes of the these interspecific self-sterile F1 hybrids were doubled by tissue culture. Amphidiploid of F1 hybrid was self-fertile. Starting with amphidipliod, a wystematic backcross (BC) program was set up with NC82 as recurrent parent. In the BC5C2 generation, the resistant plant was selected. This resistant line, KF8833-1, had 48 chromosome and secreting glandular trichomes. It flowered 2days later than NC82, and stalk height, leaves per plant, leaf length and leaf width were similar to those of NC82.

Effect of Yellowing Times During Yellowing Stage on Physico-chemical Properties of Immature Tobacco Leaves in Bulk Curing (Bulk건조시 미숙엽의 황변시간 조절이 잎담배 이화학성에 미치는 영향)

  • Leaves harvested were separated with visual characters into 2 classes such as immature and mature leaves. In the curing process, the prolonged yellowing treatment during yellowing stage was automatically controlled at the different stalk position, and condition of curing process after this period was all the same with conventional ones. In case of prolonged yellowing in immature leaves, increase of price per kg reached to 8 % compared with those of conventional ones. In physical properties, filling capacity and shatter index was decreased with the degree of maturity, and it was equal level in filling capacity of immature leaves between curing method, while shatter index was decreased in prolonged yellowing treatment than that of conventional ones. There was no difference in chemical components between immature leaves of prolonged yellowing and conventional ones. As to the prolonged yellowing of immature leaves, there was decreased in citric and malic acid contents of the nonvolatile organic acids, and it was equal level in all higher fatty acids content of leaves cured by prolonged yellowing treatment compared with in that of conventional curing method. The contents of key compounds such as solanone, damascenone, damascone in the essential oil were lower in prolonged yellowing of immature leaves than those of mature leaves cured by conventional ones.

Effect of Air Flow During Curing Process on Physical Properties of Bulk Cured Leaves (황색종 Bulk건조시 송풍량 조절에 따른 건조엽의 물리성 변화)

  • In recent years, the line of increase in policy on tobacco production triggered a rise in the loading volume per bulk curing barn, and manufacturers boosted the output of their blowers in order to prevent dirty leaves in the process of curing. for this reason, we studied the effect of the reducing air flow in bulk curing chamber from the color fixing stage to the stem drying stage of flue curing process on physical properties of cured leaves. The control of air flow was composed of the reducing air velocity of blower by means of a voltage regulator (slidac), and condition before color fixing stage was all the same with the conventional curing method. As a result, an effectiveness of improvement in the physical properties of cured leaves were observed. The leaves cured by this method were somewhat orange in color of upper stalk position, better bodied, and less brittle compared with the leaves produced by conventional curing. However, the leaves cured by this method had a little sharpness and harshness. As to the physical properties, there was decreased in occurrence of flat leaves than that of conventional ones. On the other hand, in case of reducing air flow during the curing process, increase of price per kg reached to about 5 % compared with those of conventional curing method.

  • Nitrate nitrogen was absorbed dominantly among the inorganic nitrogen nutrients by tobacco plant. Transport and reduction of $NO_3-N$ in plant tissue were the important metabolism for supplying synthetic N compounds to developing tissues during growth period. Under field and environment-controlled condition tobacco plants were grown and seperated to leaf tissues at stalk positions for investigation of nitrogen transport and assimilation ability during period of rapid vegative growth. The results of studies were summarized as follows: 1. $NO_3-N$ absorbed from roots was transported as inorganic nitrogen through the vascular tissue of leaf veins as resulting from the high $NO_3-N$ ratio of the nitrogen content in leaf veins, but these ratios in mesophyll tissue of the same leaf laminae decreased remarkably in disregard of higher accumulation of nitrogen being compared to midvien. 2. Mesophyll tissue of mature leaves appeared higher value of nitrate reductase activity (NRA) comparing with other tissues, stem, leaf vien, and meristmatic tissue at emergence point with young leaves. 3. Matured leaves at lower position being reducing nitrate nitrogen vigorously observed thick laminae and kept high amount of water in them. 4. Mature leaves of young plant reduced $NO_3-N$ vigorously for supply synthetic N compounds to meristmatic tissues at growing point by the reason of narrow and few leaves at young stage, but in advancing growth period NRA of mature leaves along upper position reached to lower value. This appearence attributed to distribution of organic-N compound demanding for growth to increasing numbers of wide leaves.

Colorimetric and Chemical Characteristics of Grey Leaf Tobacco (Grey 증상 잎담배의 색채 및 화학성분 특성)

  • This experiment was conducted to get the fundamental information about the difference in color and some chemical components of grey flue-cured tobacco cultivated in paddy field to compare with normal tobacco. Cured leaves of NC82 were separated with visual characters into three classes of slight, fair and severe symptoms of grey leaf by percentage of grey parts to whole leaf area, and compared to normal ones. Occurrence of grey leaves tended to be higher in lower stalk position than in upper ones. Cl, Mn and Fe content of grey leaves was much higher when it compared with normal leaves, and within the same leaf, grey parts were increased in Fe, Mn and Cl content than in those of normal parts but there was no appearent difference in total sugar, nicotine and Ca contents. Chemical analysis of grey leaf samples indicated that they contained more Fe, Mn and Cl contents than those of normal leaves. These tendencies in chemical traits were accordance with the degree of grey symptoms. Grey leaves showed remarkably lower b*value and slightly lower L*value than those of normal leaves, while a*value was mostly higher in grey leaves. In general, grey appearence of grey flue-curd tobacco seemed to be from less yellowish color, poorer brightness with more brownish or reddish color.

Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars.   • This experiment was carried out to investigate control efficacy by cultural practice and fumigation of tacterial wilt caused by Pseudomonas solancearum in resistant tobacco line NC82 at the Korea Ginseng and Tobacco Research Institute, Eumseong Experiment Station in 1982 and 1983. The bacterial wilt of tobacco occurred severely from mid-July to last August in applicable temperature and soil humidity for increasing bacteria. Disease severity appeared low and slowly at fumigation and resistant variety treatment. Incidence of bacterial wilt in tobacco line NC82 was $44.7\~55.8\%$ being compared with susceptible variety, NC2326 and $95\~99\%$ when resistant variety, NC32 was cultivated with soil fumigation treatment. Control efficacy of cultural practices appeared low with $0.8\~20\%$ and was not different from resistant variety and fumigation treatment. Control system against bacterial wilt in flue-cured tobacco was accomplished by control efficacy over $95\%$ when resistant variety(NC82) was cultured after treatment of cultural practices (Tillering after harvest and before transplanting, stalk and root destruction, early transplanting early removal of the mulching film) and soil fumigation(Cylon).

Detection of Virus in Fruit and Seed of Vegetables Using RT-PCR (RT-PCR에 의한 과채류 열매 및 종자의 바이러스 검정)

  • Tobacco mosaic tobamovirus (TMV), cucumber mosaic cucumovirus (CMV), cucumber green mottle mosaic tobamovirus (CGMMV) and zucchini yellow mosaic potyvirus (ZYMV) from individual fruits and seeds of hot pepper and cucumber were detected by the reverse transcription-polymerase chain reaction (RT-PCR). The dilution end-points for RT-PCR in curde sap from TMV. and CMV - infected hot pepper leaves and CMV - and CGMMV-infected cucumber leaves were 10-5. However, the amount of PCR product obtained from preparation of ZYMV-infected cucumber leaf was 10-fold lower than those of CMV or CGMMV-infected cucumber leaves. In hot pepper, both TMV and CMV were detected in all parts of the fruit wall tissue, but the yields of PCR products in the fruit stalk and its surrounding tissues were higher than those of the end parts of the fruit. On the other hand, in cucumber fruit infected with CMV, CGMMV or ZYMV, the fruit wall tissue and seed located in both stalk and end parts showed higher yields of PCR products than those of intermediate parts. Of five viruses that were analysed, only TMV in hot pepper seed, and CGMMV and CMV in cucumber seed were detected in testa parts.

  • Using a flue-cured tobacco variety, BU 109, effect of growth regulators(Fatty Alcohol and C-MH) on the change of invertase activity and sugar metabolites were investigated. Invertase activity in untreated leaf tissue was decreased along with maturation of leaf, However, a momentary increase of the activity was observed in leaves by the dual treatment of fatty alcohol and C-MH regardless of leaf position while sole C-MH treatment resulted in activity increase by 14 days after the treatment. Similar tendency was observed in stalk. Sugar content in leaf was increased immediately after the treatment but no significant increase at large resulted until 14 days after treatment. After harvest, reducing sugar was increased by the growth regulators. Nevertheless, in case of dual treatment, the total sugar content was not different with that of untreated control. R/T ratio was gradually increased after topping stage and reached maximum at 14 days after treatment of growth regulators. It decreased a little after harvest but the RR ratio was relatively higher due to increase of reducing sugar resulted by the treatment of the chemicals. Upon treatment of growth regulators, reducing sugar content was increased in lugs and leaves compared to untreated control and the content of sugar metabolites was increased by the use of the chemicals either at lower or higher than recommended dose. Effect of Total Sugar Content on Chemical, Smoke and Sensory properties in Flue-cured Leaf Tobacco (황색종 잎담배의 전당 함량이 화학성분, 연기 및 관능의 특성에 미치는 영향)

  • This study was conducted to investigate the effect of the total sugar contents on chemical, smoke and sensory properties and to estimate the desirable chemical composition contents from sensory properties in flue-cured leaf tobacco. The leaves used in the study were in 2005 and in $1997{\sim}2006$ crop year. Nicotine and total nitrogen contents, filling value, impact, irritation and bitterness were reduced with increasing the total sugar contents of B2O(leaf) and C2L(cutters), whereas tar and CO contents and puff number were increased. The desirable sugar contents estimated from the best overall tobacco taste of smoke were 25.9% in B2O and 26.9% in C2L. All of regression equations among total sugar/nicotine ratio, total sugar, nicotine and total nitrogen contents were significant($p{\leqq}0.05$). There were no significant difference between the 4 grade(A2O, B2O, C2L, D2L) average and the whole plant(12 grade) average contents in total sugar, nicotine and total nitrogen contents. All of regression equations between the ratio of each grade content to the average content of twelve grades and stalk position[number ; from bottom(1) to top(12)] in total sugar, nicotine and total nitrogen contents were significant($p{\leqq}0.05$). The estimated desirable chemical properties which based on the best overall tobacco taste were 23.2% of total sugar, 2.39% of nicotine, 9.7 of total sugar/nicotine ratio, 2.57% of total nitrogen, 1.08 of total nitrogen/nicotine ratio in average of four grades. With decrease the total sugar content from 32.0% to 24.0%, filling value was enhanced by 14.1% in average of B2O and C2L, also tar and CO contents of smoke were decreased by 13.2% and 10.5%, respectively. These results suggest that the estimated desirable chemical composition contents may be useful to produce good leaf tobacco, and to enhance filling value and to reduce tar and CO contents of smoke in flue-cured leaf tobacco.
