This study was conducted to determine the changes of nitrogen compounds in lamina and midrib during the curing process with the different curing methods, such as priming and stalk-cut curing. After KB 108 burley tobacco was cultivated by the different fetilization levels such as standard and 20% higher, only the tips and leaf were harvested. Though the contents of total alkaloid and total nitrogen were similar in lamina, midrib showed a very low contents of those components by the different curing method and fertilization levels. The content of nitrate-nitrogen in lamina increased during a middle of curing process, and then these compound was decreased during an end of curing process by stalk-cut curing method. Nitrate-nitrogen contents in the lamina by the priming curing showed a high level caused by rapid drying process during an end of curing process. That component in midrib was 5-6 times higher than that of lamina. The contents of ammonia-nitrogen in the lamina and midrib were increased during a curing process. Though those amount in tips showed a similar by a different fetilization and curing method, midrib of stalk-cut curing showed a slow increasing during a curing process.
In order to obtain the basic information for the successful breeding of aromatic tobacco (Nicotiana tabacum L.), the heterosis, combining ability and gene distributions in 28 Fl hybrids by diallel crosses between Korea local and Oriental varieties were investigated. The yield were higher, stalk height, leaf length and leaf width were larger and days to flower was shorter for the Fl hybrids when compared to mid-parent. Estimates of general combining ability were significant for all characters investigated, and specific combining ability were significant for yield, stalk height, leaves Per Plant, leaf width and days to flower. In vr-wr graph, partial dominane was exhibited by all characters investigated.
A tobacco ripeness grading meter was designed and constructed using the color sensor, its performance was evaluated. A degree of ripeness grading of a leaf is very closely related to the measured tobacco leaf color. Measuring the small amount of the reflectance precisely depends on the apparatus including color sensor, light source, detector sensitivity, and geometric characteristics of appratus. To analyze and minimize the variational effects, experiments to select the proper condition were performed. Because of the combined effect mentioned above, the system has some variation on its response. Basis on the results of the experiments, prototype was developed and interfaced to a computer system. The main components of prototype included a tungsten lamp as a light source, Amorphous full color sensor with three filters, regulated D.C. power supply, OP - AMP(741 TC) for amplification, AR - B3001 board for interfacing to a computer with analog to digital conversion, and a compatible IBM PC XT computer. The experimental results of the developed ripeness tobacco leaf measurement system are summarized as following: [1] The output readings of ripeness grade meter for tobacco leaf, which is based on harvesting time, showed the apparent difference in variety of different quality. It was considered suitable that three filters(red, green, blue) in Amorphous full color sensor could be used in four different ripeness degree measurement of tobacco leaf. [2] The output readings of ripeness grade meter for tobacco leaf, which is based on government procurement, showed apparent difference in variety of different quality. Tobacco leaf varieties to stalk position are divided into tips, leaf, cutters, and primings, It is considered suitable that only red filter in the sensor could be used to classify the grade of tobacco leaf within the same kind tobacco stalk. However, the ripeness grade meter was not adequate to classify all the tobacco grades in the four different tobacco leaves.
It would be clear that the constituents of the leaf surface lipid is ye비 important as an evaluation index of tobacco leaf quality since the quality of tobacco specific aroma with leaf species depends on the contents of the lipid and the strength of the aroma is determined by the amounts of the lipid secreted. For the reason, a rapid and peproducible method to quantify DVT, which is a kind of lipid, has been studied. The biosynthesis procedure of DVT in leaf growing processes, and the volatile or decompositional characters of DVT in leaf drying processes were also discussed. In consequence, it might be possible to get the data available to the cultivation of better tobacco leaf and the manufacture of cigarettes with better aroma and taste. The results obtained from this study are as follows. 1. Chloroform/dichloromethane solvent was better than chloroform alone for DVT extraction. The extraction yields of the leaf surface lipid were about 5% 2. The extractives with dichloromethane were treated by silylation with BSTPa and the quantitative analysis of DVT was carried out using SE -54 fused silica capillary column. It was found that rapid and reproducible data could be obtained from these methods. 3. In flue - cured tobacco species, DVT contents were $30.3\mu\textrm{g}/cm^2$ in the beginning stage of leaf drying processes and $12.1\mu\textrm{g}/cm^2$ corresponded to 30% levels of the beginning stage, in the end stage. 4. DVT contents in Burley mere 2 times as large as those in fluecured tobacco. DVT in the upper stalk position of leaf was 3 times larger than that in the lower stalk position. 5. DVT of tobacco leaves was decomposed by $SO_2$ gas or the sun light. The decomposition rate was largest in the sample used methanol as a extraction solvent.
This experiment was conducted to study the nature of gene effects for the leaf characters in flue-cured tobacco. The genetic populations were derived from crosses between NC 2326 and Mc Nair 373, and NC 628 and DG-72. The generation means experiment Included the Pl, P2. Fl, F2, Bl and B2, which were frown at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1984. Seedlings were transplanted to the field in a randomized block design with 3 replications. In each block, parental and Fl Plots contained 15 plants in a single row, F2, Bl and B2 plots being composed of 75 plant, in 5 rows. Leaf characters were measured of largest (middle leaf) and 5th leaf(top leaf) from the top after topping. Measurements of the length and width of leaf were obtained from the fresh the middle and top leaves, and weight of leaf, weight and width of midrib were from the satrap leaf after curing. Estimates of additive and dominance genetic variance were analyzed according to Gamble's biometrical model. The results obtained are as follows: 1. The additive gene effects were significant and larger than the dominance gene effects for all leaf and midrib characters in both stalk positions. 2. The dominance gene effects were significant for the length and width of leaf, and weight of midrib in the middle leaves. 3. The digenic epistatic effects were significant for the length and width of leaf in both stalk positions. 4. The additive gene effects were larger in the top than in the middle leaves and midrib characters.
Effects of application time And rate of chlorine on the chlorine absorption and distribution at different stalk positions of the plants were Investigated in burley tobacco grown under the pot condition. Treatment consisted of 6 application times (from 2 weeks before transplanting to 8 weeks after transplanting with 2 weeks' interval) and 2 application rates (2g and 4g-KCl/pot) by 6X2 factorial experimental design. The tobacco plants took up the chlorine rapidly just after chlorine application, and the chlorine content of leaf was increased proportionally to the rate of chlorine application. The chlorine was accumulated when much in lower leaves when chlorine was applied early, while the chlorine accumulated much in upper leaves when chlorine was applied late. The chlorine content of leaf was negatively correlated to the degree of brightness, red color and yellow color of cured leaf. This experiment was conducted to examine characteristics of agronomic characters and estimate of gene effect for several mutant characters. The genetic populations were derived from cross between 83H-5 and Hicks. There were significant difference for plant height, stlk height, leaf shape and bacterial wilt disease index except leaf number, leaf length, and what is more, F3 variance is more than Bl and B2 generation from cross 83H-5 X Hicks. Gene actions for stalk height and bacterial wilt disease were estimated by 3-parameter, and by 6- parameter model for all characters except above two characters but stalk height and bacterial wilt disease index are not significant in the additive and dominance effects. Dominant$\times$dominant epitasis for plant height, dominant and dominant$\times$dominant epistasis for leaf length, additive and additive$\times$additive and dominant$\times$dominant epistasis for leaf width, and additive and additive$\times$dominant epistasis for days to flower were appeared significant in gene action.
This study was conducted to investigate the interaction between chemical compositions and agronomic characteristics of perique tobacco leaf. At the topping stage, the thickness of leaf was positively correlated with total sugar($\textrm{0.975}^{**}$), reducing sugar($\textrm{0.975}^{**}$), alkaloid($\textrm{0.518}^{*}$) and petroleum ether extract($\textrm{0.801}^{**}$) contents. The length of upper leaf was negatively correlated with its chemical contents. Total sugar contents of the harvested leaf showed a sigmoid pattern of distribution along the leaf stalk positions.
The changes of contents of organic acids in flue-cured tobacco leaves depending on stalk position and maturity viz immature, mature and overmature were studied in 1982-83.
