열처리(熱處理)에 의한 홍삼(紅蔘)엑기스의 성분변화(成分變化) (Quality Changes in Red Ginseng Extract during High Temperature Storage)

  • 열처리(熱處理)가 홍삼(紅蔘)엑기스의 성분변화(成分變化)에 미치는 영향(影響)을 조사(調査)하기 위하여 저온(低溫)에서 추출(抽出), 조제(調劑)된 홍삼(紅蔘)엑기스를 각온도별(各溫度別)로 $40^{\circ}Bx$까지 농축(濃縮)하고 다시 온도별(溫度別), 시간별(時間別)로 숙성(熟成)시켜 숙성과정중(熟成過程中)에 일어나는 pH, 당(糖), saponin 및 색상(色相) 등을 조사(調査)하여 다음과 같은 유의성(有意性)있는 결과(結果)를 얻었다. (1) 홍삼(紅蔘)엑기스의 pH는 농축(濃縮) 및 숙성(熟成)중에 차차 감소(減少)하였으며 $100^{\circ}C$에서 96시간(96時間)까지 숙성(熟成)하면 pH가 4.15까지 감소(減少)하였다. (2) HPLC에 의한 방법(方法)으로 rhamnose, glucose, fructose, sucrose, maltose 둥의 당(糖)을 동정(同定) 및 정량(定量)하였으며 $100^{\circ}C$에서의 당(糖)의 변화(變化)는 sucrose가 30시간(30時間)에서 95%이상 감소(減少)하였고 glucose는 50시간(50時間)까지는 증가(增加)하다가 감소(減少)하였고 fructose는 10시간(10時間)까지 약간 증가(增加)되다가 감소(減少)하였으며 rhamnose는 큰 변화(變化)가 없었다. (3) 수삼(水蔘)이나 백삼(白蔘)에는 없는 rhamnose가 홍삼(紅蔘)엑기스중에서는 동정(同定)되었으며 이는 인삼중(人蔘中)의 일부 배당체(配糖體)의 분해(分解)에 의한 것으로 생각되며 온도(溫度)의 변화(變化)에 따라 큰 변화(變化)가 없는 것으로 보아 다른 당(糖)에 비(比)해 amino-carbonyl 반응(反應)에 적게 관여(關與)하는 것으로 생각된다. (4) 열처리중(熱處理中)의 홍삼(紅蔘)엑기스중의 saponin은 ginsenoside-Re와 $-Rg_1$이 감소(減少)하는 반면(反面) $ginsenoside-Rg_2$와 -Rh group은 증가(增加)되어 saponin group간(間)의 상호변환(相互變換)으로 추정(推定)할 수 있다. (5) 열처리(熱處理)에 의한 홍삼(紅蔘)엑기스의 갈변색소(褐變色素)는 벤젠층에 비해 부탄올층(層)이 약(約) 10배(10倍), 수층(水層)이 약(約) 100배(100倍)로 나타나 홍삼(紅蔘)엑기스의 갈변색소형성(褐變色素形成)은 비효소적(非酵素的) 갈변반응(褐變反應)인 amino-carbonyl 반응(反應)이 주도적(主導的) 역할(役割)을 하고 있음을 알 수 있다. (6) 총당(總糖)과 갈변반응속도(褐變反應速度)는 유의성(有意性)이 있었으며 $100^{\circ}C$의 경우 20시간(20時間)에 가장 색도(色度)가 높아 갈변반응속도(褐變反應速度)가 0.2로 나타났다.

분쇄방법에 따른 고려홍삼분말의 품질특성 (Quality Characteristics of Korean Red Ginsneg Powder on Pulverizing Methods)

  • 홍삼 분쇄의 신 가공 기술로서 비충격(非衝擊)분쇄방식인 cell cracker의 공장 적용 가능성을 제시하고 홍삼분말의 품질고급화에 기초자료로 활용하고자 기존의 hammer mill(충격분쇄방식)과 cell cracker에 의한 분쇄방식으로 홍삼분말을 제조한 후 색도, 밀도 및 관능적 특성을 비교 조사한 결과를 요약하면 홍삼분말의 색도는 cell cracker로 분쇄한 홍삼분말의 경우 색의 밝기를 나타내는 L값은 68.15로 hammer mill로 분쇄한 홍삼분말 72.08 보다 낮았으나 a(적색도) 와 b(황색도)값은 각각 4.21과 5.73, 26.56과 28.36으로 높게 나타났다. 물 추출물의 색도는 시간이 경과함에 따라 두 가지 시료 모두 흡광도가 증가하였으며 전반적으로 hammer mill로 분쇄한 홍삼분말이 cell cracker로 분쇄한 홍삼분말 보다 흡광도가 높게 나타났다. 밀도는 분말의 유동성 특성을 나타내는 이완겉보기밀도, 충격겉보기밀도, 체적감소율은 hammer mill로 분쇄한 홍삼분말보다 cell cracker로 분쇄한 홍삼분말이 모두 높게 나타났다. 미Rm럼각은 쌓임각 보다 낮았으며 이 두 가지 모두 hammer mill로 분쇄한 홍삼분말 보다 cell cracker로 분쇄한 홍삼분말이 낮게 나타났다. 관능검사결과 촉감에 있어서는 두 시료간에 유의 차가 없는 것으로 나타났지만, 홍삼분말 제품의 품질 특성에 큰 영향을 미치는 색도는 cell cracker가 좋게 평가되었으며, 전체적인 선호도에 있어서도 좋게 나타났다. 이상의 연구결과를 종합해볼 때 홍삼분말의 품질 특성에 미치는 요소로서 분쇄방법이 중요한 인자로 작용하고 있음을 알 수 있었으며 홍삼분말의 제조에는 분쇄기 선정이 무엇보다 중요하며 특히 색깔 등 관능적 품질이 강조되는 홍삼분말 제품을 제조할 경우는 기존 충격식 분쇄보다 비충격식 분쇄시스템을 적용하는 것이 품질향상 효과가 클 것으로 생각된다.

The effects of Internet addiction on the lifestyle and dietary behavior of Korean adolescents

  • We performed this study to examine lifestyle patterns and dietary behavior based on the level of Internet addiction of Korean adolescents. Data were collected from 853 Korean junior high school students. The level of Internet addiction was determined based on the Korean Internet addiction self-scale short form for youth, and students were classified as high-risk Internet users, potential-risk Internet users, and no risk Internet users. The associations between the students' levels of Internet addiction and lifestyle patterns and dietary behavior were analyzed using a chi-square test. Irregular bedtimes and the use of alcohol and tobacco were higher in high-risk Internet users than no risk Internet users. Moreover, in high-risk Internet users, irregular dietary behavior due to the loss of appetite, a high frequency of skipping meals, and snacking might cause imbalances in nutritional intake. Diet quality in high-risk Internet users was also worse than in potential-risk Internet users and no risk Internet users. We demonstrated in this study that high-risk Internet users have inappropriate dietary behavior and poor diet quality, which could result in stunted growth and development. Therefore, nutrition education targeting high-risk Internet users should be conducted to ensure proper growth and development.

연포장재 필름으로 개별 포장된 수삼의 저장 중 품질 변화 (Changes in Quality of Fresh Ginseng Packed Individually with a Soft Film during Stnrage)

  • 수삼을 물로 세척하고 기능성 연포장재 필름주머니(ONY/LDPE/L-LDPE; 20$\times$30 cm, 93${\mu}m$)로 개별 포장하여 $25^{\circ}C$에서 20일간 저장하면서 수삼의 외관품질 변화 및 관능적 특성을 조사하였다. 수삼의 외관품질은 저장 후 10일째부터 열화되기 시작하였고 15일이 경과되면서 열화빈도가 급격히 증가하여 20일째에는 전체 수삼의 75%가 열화되었다. 저장 중 수삼의 외관품질 열화는 주로 경도 저하에 의하여 일어났으며 곰팡이 발생 빈도는 낮았다. 20일이 경과된 수삼 중 정상 수삼은 조직이 단판하고 인삼냄새와 쓴맛이 강했던 반면, 경도가 저하된 수삼은 조직이 물렁거리고 끈적이는 정도가 심하였으며 자극적인 냄새, 신냄새, 단맛 및 신맛이 강한 경향을 나타냈다.

제1, 2차 한국근로환경조사: 한국과 EU회원국의 비교 (First and Second Korean Working Conditions Survey: A Comparison between South Korea and EU Countries)

  • Purpose: In this study, I am going to figure out Korean workers' actual exposure to risk factors and percentage of the workers who complain of health problems by performing a comparative analysis of the characteristics of the workers and working conditions in Korea and EU. Methods: The data used in this study includes the $1^{st}$ working conditions survey conducted in 2006 in Korea, the $2^{nd}$ working conditions survey in 2010, the $4^{th}$ EU working conditions survey, and the $5^{th}$ EU working conditions survey. I have compared the changes to Korean working conditions and those to EU working conditions in the categories of demographic characteristics, quality of labor, exposure to risk factors, and health problems included in the data. Results: The analysis of the characteristics of the demographic characters of Korean workers shows that aged and female workers register increased labor force participation. The analysis of labor quality shows that Korea has a long working time but with a lower work intensity compared to the countries included in EU working conditions survey. As for risk factors, Korea registers a low level of exposure as compared to countries included in EU working conditions survey and characteristically shows a decreasing exposure to tobacco smoke. The survey shows sharply increasing complaints of muscle pain in the upper and lower limbs. Conclusion: In this study, I have identified vulnerable social groups by using quantified values in a comparison of the working conditions of Korea and those of EU.

인삼제품(人蔘製品)의 품질안정성(品質安定性)에 관한 연구(硏究) -2. 인삼정분(人蓼精粉)과 시판다류제품(市販茶類製品)의 품질안정성(品質安定性) 비교(比較)- (Studies on Stability for the Quality of Ginseng Products -2. Comparative Study on Keeping Quality of Red Ginseng Extract Powders(RGEPs) and Commercial Dehydrated Tea Products-)

  • 유통과정(流通過程)중의 품질안정성(品質安定性) 유지(維持)가 항상 문제시되고 있는 홍삼정분(紅蓼精粉)(동결(凍結) 및 분무(噴霧))의 안정성(安定性)을 시판차류정품(市販茶類精品)과 비교(比較)하기 위하여 이들 제품(製品)의 흡습특성(吸濕特性)을 비교(比較)하였고 또 초기수분함량(初期水分含臺), 조지방함량(粗脂肪含量) 및 밀도(密度)와의 관계(關係)를 조사(調査)하여 홍삼정립(紅蓼精粒)의 안정성(安定性) 미치는 영향(影響)을 비교(比較)하였다. (1) 홍삼정분(紅蔘精粉) 및 시판차류제품(市販茶類製品)의 입도(粒度)는 100 mesh 이하였으며 동결건조제품(凍結乾燥製品)이 분무건조제품(噴霧乾燥製品)보다 입도(粒度)가 컸다. (2) 홍삼정분(紅蓼精粉)(동결(凍結) 및 분무(噴霧))이 3.85% 및 4.56% 로 초기수분함량(初期水分舍量)이 높은데 비해 커피(동결(凍結) 및 분무(噴霧))가 2.17% 및 3.52%로 낮았으며 홍삼정분(紅蔘精粉)이 커피 보다 1.3$\sim$l.8배의 높은 초기수분함량(初期水分含量)을 나타냈다. (3) 탱이나 쌍화차가 흡습율(吸濕率)이 가장 낮아 높은 안정성(安定性)을 나타냈는데 이는 탱과 쌍화차의 지방함량(脂肪含量) (4.12% 및 0.62%)이 높을 뿐만 아니라 밀도(密度)(0.89g/ml 및 0.59g/ml)도 다른 제품(製品)에 비해 훨씬 높기 때문으로 판단된다. (4) 커피의 흡습율(吸濕率)이 홍삼정분(紅參精粒)보다 컸으나 커피의 안정성(安定性)이 홍삼정분(紅蔘精紛)보다 더 높은 것은 커피의 초기수분함량(初期水分含量) (2.17${\sim}$3.52%)이 홍삼정분(紅蓼精粉) (3.85${\sim}$4.56%)보다 낮을 뿐만 아니라 커피의 지방함량(脂肪含量)(0.31${\sim}$0.38%)도 홍삼정분(紅蔘精粉)(0.12${\sim}$0.16%) 보다 훨신 높기 때문으로 판단된다. (5) 따라서 홍삼정분(紅蓼精粉)의 안정성(安定性)을 높이기 위해서는 현행 물추출방벙(抽出方法)을 용매추축(溶媒抽出)로의 전환(轉換)이나 지방피막(脂肪皮膜) 처리(處理)에 의한 흡습물성(吸濕物性)을 개선(改善)함이 바람직할 것으로 판단된다.

  • Human and monogastric animals can not synthesize 10 out of the 20 amino asids and therefor need to obtain these from their diet. The plant seed is a major source of dietary protein. It is particular important in their study to increase nutritional quality of the seed storage proteins. The low contents of lysine, asparagine and threonenein various cereal seeds and of cystein and methionine. In legume seeds is due to the low proportions of these amino acids in the major storage proteins, we have tried to apply the three strategies; (1) mutagenesis and selection of specific amino acid analogue resistance, (2) cloning and expression study of lysine biosynthesis related gene, (3) transfomation of lysine rich soybean glycinin gene. The 5-methyltryptophan (5MT) resistant cell lines, SAR1, SAR2 and SAR3 were selected from anther derived callus of rice (Oryza sativa L. "Sasanishiki"). Among these selected cell lines, two (SAR1 and SAR3) were able to grow stably at 200 mg/L of 5MT. Analysis of the freed amino acids in callus shows that 5MT resistant cells (SAR3) accumulated free tryptophan at least up to 50 times higher than those that of the higher than of SAS. These results indicated that the 5MT resistant cell lines are useful in studies of amino acid biosynthesis. Tr75, a rice (Oryza sativa L., var. Sasanishiki) mutant resistant to 5MT was segregated from the progenies of its initial mutant line, TR1. The 5MT resistant of TR75 was inherited in the M8 generations as a single dominant nuclear gene. The content of free amino acids in the TR75 homozygous seeds increased approximately 1.5 to 2.0 fold compared to wild-type seeds. Especially, the contents of tryptophan, phenylalanine and aspartic acid were 5.0, 5.3 and 2.7 times higher than those of wild-type seeds, respectively. The content of lysine is significantly low in rice. The lysine is synthesized by a complex pathway that is predominantly regulated by feedback inhibition of several enzymes including asparginase, aspatate kinase, dihydrodipicolinat synthase, etc. For understanding the regulation mechanism of lysine synthesis in rice, we try to clone the lysine biosynthetic metabolism related gene, DHPS and asparaginase, from rice. We have isolated a rice DHPS genomic clone which contains an ORF of 1044 nucleotides (347 amino acids, Mr. 38, 381 daltons), an intron of 587 nucleotides and 5'and 3'-flanking regions by screening of rice genomic DNA library. Deduced amino acid sequence of mature peptide domain of GDHPS clone is highly conserved in monocot and dicot plants whereas that of transit peptide domain is extremely different depending on plant specie. Southern blot analysis indicated that GDHPS is located two copy gene in rice genome. The transcripts of a rice GDHPS were expressed in leaves and roots but not detected in callus tissues. The transcription level of GDHPS is much higher in leaves indicating enormous chloroplast development than roots. Genomic DNA clones for asparaginase genes were screened from the rice genomic library by using plaque hybridization technique. Twelve different genomic clones were isolated from first and second screening, and 8 of 12 clones were analyzed by restriction patterns and identified by Southern Blotting, Restriction enzyme digestion patterns and Southern blot analysis of 8 clones show the different pattern for asparaginase gene. Genomic Southern blot analysis from rice were done. It is estimated that rice has at least 2-3 copy of asparaginase gene. One of 8 positive clones was subcloned into the pBluescript SK(+) vector, and was constructed the physical map. For transformation of lysine rich storage protein into tobacco, soybean glycinin genes are transformed into tobacco. To examine whether glycinin could be stably accumulated in endosperm tissue, the glycinin cDNA was transcriptionally fused to an endosperm-specific promotor of the rice storage protein glutelin gene and then introduced into tobacco genomic via Agrobacterium-mediated transformation. Consequently the glycinin gene was expressed in a seed-and developmentally-specific manner in transgenic tobacco seeds. Glycinin were targeted to vacuole-derived protein bodies in the endosperm tissue and highly accumulated in the matrix region of many transgenic plant (1-4% of total seed proteins). Synthesized glycinin was processed into mature form, and assembled into a hexamer in a similar manner as the glycinin in soybean seed. Modified glycinin, in which 4 contiguous methionine residues were inserted at the variable regions corresponding to the C - teminal regions of the acidic and basic polypeptides, were also found to be accumulated similarly as in the normal glycinin. There was no apparent difference in the expression level, processing and targeting to protein bodies, or accumulation level between normal and modified glycinin. glycinin.

Electron Beam 조사가 인삼분말의 색도 및 관능적 품질에 미치는 영향 (Effects of Electron-Beam Irradiation on Color and Organoleptic Qualities of Ginseng Powders)

  • Electron beam, electrically produced from an electron accelerator, was compared with gamma energy in terms of its influence on color and organoleptic qualities of ginseng powders when exposed to the energy used for their microbial decontamination. Hunter color L and b values were suitable for measuring color characteristics of ginseng powders, which were not significantly changed by the exposure to 5 to 7.5 kGy electron beam and gamma energy. Fifty percent ethanol extracts of irradiated ginseng powders at 10 key showed negligible differences from the non-irradiated control in the pattern of absorption spectra at 280∼800 am, but showed increased values in overall color difference (AE) as compared with powdered samples. Irradiation more than 10 kGy and storage at ambient temperature for 4 months caused browning of powdered samples. Irradiation at more than 10 kGy of electron beam was found a critical level to bring about appreciable changes (p<0.05) in or-ganoleptic qualities such as color and odor of sterilized samples, and red ginseng powder was more susceptible than white one to organoleptic changes by irradiation.

PC방에서의 간접흡연에 따른 요중 코티닌의 농도

  • Many adolescents stay long time in the PC game room and are exposed to much of tobacco smoke. To evaluate the effect of passive smoking in the PC game room, airborne nicotine concentrations and urinary cotinine concentrations were measured for 20 adolescents at 2 PC game rooms in Sung-nam city. And the subjects were interviewed for duration and time in PC game room and smoking pattern. Subjects are composed of each of 10 smokers(5 males and 5 females) and 10 non-smokers(5 males and 5 females). They stayed for three hours in the PC game room without smoking. Concentrations of nicotine in smokers and non-smokers were 129.72 $\mu\textrm{g}$/㎥ and 99.99 $\mu\textrm{g}$/㎥, respectively. Urinary cotinine concentrations were increased as time goes on after exposure to nicotine and showed maximum value at 9.45 hours after nicotine exposure and were 32.21 and 110.66 $\mu\textrm{g}$/L for non-smoker and smokers. Urinary cotinine has a tendency to be increased by passive smoking. Therefore, it is recommended that the effective control for indoor air quality and extensive research be needed to reduce nicotine concentration by passive smoking in the PC game room.

살균처리가 인삼엽록차의 화학성분에 미치는 영향 (Effects of Decontamination Treatments on Chemical Components of Panax Ginseng-Leaf Tea)

  • 수출용으로 제조되고 있는 인삼엽록차에 대하여 위생적 품질개선을 목적으로 처리된 ethylene oxide(EO) 훈증제와 적정선량의 감마선 조사가 시료의 몇 가지 화학성분에 미치는 영향을 검토하였다. 인삼엽록차에 함유된 사포닌과 지방산은 상업적 조건의 EO 처리나 적정선량의 감마선 조사(5kGy 범위)에 의해 변화되지 않았다. 그러나 유리당과 아미노산에서 glucose, lysine 및 histidine은 EO 처리시 유의적으로 감소되어(p<0.05) 살균방법 중 EO gas는 감마선조사에 비해 엽록차 성분에 미치는 영향이 큰 것으로 화인되었다.
