Insert Transmission of Paulownia Witches'-Broom Mycoplmsma-Like Organism to Periwinkle Plant by Tobacco Leaf Bug, Cytropheltis tenuis Reuter (담배장님노린재에 의한 오동나무 빗자루병 MLO의 일일초로의 전반)

  • Paulowina witches'-broom mycoplasma-like organism (PWB-MLO) was transmitted experimentally to periwinkle (Catharanthus roseus L.) plant by tobacco leaf bug (Cyrtopeltis tenuis Reuter). Adults of the leaf bugs were allowed to feed on the witch's-broom infected paulownia (Paulownia tomentoas Steud.) trees for three weeks to insure the acquisition of PWB-MLO and then transferred to healthy seedlings of periwinkle and paulownia plants. In 25∼35 days after transfer of the viruliferous leaf bugs, six out of the ten periwinkle plants showed‘little-leaf’symptoms, while the paulowina seedlings remained symptomless. Presence of MLO in the infected periwinkle tissue was diagnosed by fluorescence microscopy and MLO particles were observed under electron microscope, confirming the transmission of PWB-MLO to periwinkle.

Salicylic Acid as a Safe Plant Protector and Growth Regulator

  • Since salicylic acid (SA) was discovered as an elicitor of tobacco plants inducing the resistance against Tobacco mosaic virus (TMV) in 1979, increasing reports suggest that SA indeed is a key plant hormone regulating plant immunity. In addition, recent studies indicate that SA can regulate many different responses, such as tolerance to abiotic stress, plant growth and development, and soil microbiome. In this review, we focused on the recent findings on SA's effects on resistance to biotic stresses in different plant-pathogen systems, tolerance to different abiotic stresses in different plants, plant growth and development, and soil microbiome. This allows us to discuss about the safe and practical use of SA as a plant defense activator and growth regulator. Crosstalk of SA with different plant hormones, such as abscisic acid, ethylene, jasmonic acid, and auxin in different stress and developmental conditions were also discussed.

Influence of Growth Environment of Anther - Donor Plant and Chilling treatments to Flower Bud on Haploid Plantlets Production in Anther culture of Nicotiana tabacum L. (연초 약배양시 Anther-donor 식물체의 생육조건 및 약의 저온처리가 반수체 출현빈도에 미치는 영향)

  • The present experiments were conducted to investigate some of the factors affecting the number of haploids derived from anther culture of Nicotiana tabacum. Anther - donor plants grown under controlled environment room at 3$0^{\circ}C$ yielded more haploid than room at 18, 25 and 26-22-18$^{\circ}C$ in anther culture. Donor plants starved of fertilizer yielded more haploids as compared to those of the well fed with fertilizer in anther culture. Pretreatment of exercised flower bud at 5$^{\circ}C$ was shown to be more effective in anther culture than pretreatment at 7 and 1$0^{\circ}C$, and the optimum temperature and period of pretreatment were 4 or 6 days at 5$^{\circ}C$.

Transgenic tobacco plants overexpressing the Nicta; CycD3; 4 gene demonstrate accelerated growth rates

  • D-type cyclins control the onset of cell division and the response to extracellular signals during the G1 phase. In this study, we transformed a D-type cyclin gene, Nicta;CycD3;4, from Nicotiana tabacum using an Agrobacterium-mediated method. A predicted 1.1 kb cyclin gene was present in all of the transgenic plants, but not in wild-type. Northern analyses showed that the expression level of the Nicta;CycD3;4 gene in all of the transgenic plants was strong when compared to the wild-type plants, suggesting that Nicta;CycD3;4 gene driven by the CaMV 35S promoter was being overexpressed. Our results revealed that transgenic plants overexpressing Nicta;CycD3;4 had an accelerated growth rate when compared to wild-type plants, and that the transgenic plants exhibited a smaller cell size and a decreased cell population in young leaves when compared to wild-type plants.

  • The natural ingredient has been utilized variety of the food and medicine. The aromatherapy well known prevent disease and healthy promotion using the essential oil derived extracted plants. In this work, natural ingredient include herb granulated for application of tobacco filter. That used by granulation using fluidized bed granulator(top-spray, bottom-spray and tangential-spray) and wet granule method. According to ingredient can used granulating method selectivity. So, we used fluid-bed granulator and wet granule method. Grapefruit extract coated sugar particle using the bottom-spray method and red ginseng granule granulated red ginseng powder using the tangential-spray method in a fluidized bed. Then, these granules applied the tobacco filter after due consideration add amount and operation efficiency. As a result, wet granule was loaded dual filter because that similarity carbon granule. And it was fitted in added 3mg/mm, per tip in the tobacco end part. Another type, fuidized bed granules was filled cavity filter because it has high hardness, sphere shape.
