  • Dihaploid lines were derived from the anthers of the F, of single cross, "Va 115" x"SC 72" (Nicotiana tabacum L.). The path-analysis, correlation coefficient, and variation of characteristics among dihaploid families were evaluated. Dihaploid lines displayed much greater variation than their parents. The genetic correlation coefficient between yield and total alkaloids of dihaploids was significantly lower than that of conventional varieties and lines. In path-analysis which relates yield components to yield, number of leaves harvested and leaf width had a direct effect on yield. a direct effect on yield.

Identification of tobacco Burley species specific marker in several tobacco species by AFLP (AFLP 방법을 이용한 담배 버어리종 특이 프라이머의 개발)

  • AFLP(Amplified Fragment Length Polymorphism) analysis was conducted to cultivars of tobacco, Nicotiana tabacum in order to select the cultivar-specific markers. AFLP results using 12 primer sets revealed genetic diversity among 12 field grown tobacco cultivars. Polymorphic fragments amplified by PCR was purified and cloned to identify their nucleotide sequences. From the sequences of them, 40 primer sets were designed to select cultivar-specific markers. When genomic DNA isolated from tobacco were used as PCR template, a set of primers, BrSF/BrSR showed Burley-specific band patterns. The results indicate that AFLP technique used in this experiments is useful for identifying tobacco cultivars in a rapid manner.

  • The metabolic enzymes, nitrate reductase, amylase and peroxidase and the Photorespiratory enzyme, Slycolate oxidise in Nicotiana tabacum varieties were studied at various growth stages. The enzyme activities of young leaves with rapid growth were different from those of old ones with stationary growth. In young leaves, activity of nitrate reductase was higher than that in mature ones and amylase activity was fairly constant in all stages. Activities of glycolate oxidise and peroxidase were found to be significantly lower in young leaves than in mature ones. Activity of glycolate oxidase in mature middle leaves was 2.45 times higher than that of young ones and inhibited by 36% when the enzyme was treated with 0.16 mM isonicotinic hydrazide.

Effects of Curing Methods on the Quality of Cured Leaves in Burley Tobacco (N. tabacum L.) (버어리종 건조방법이 건엽의 품질에 미치는 영향)

  • This study was conducted to obtain the basic information on the burley tobacco (Nicotiana tabacum L.) curing methods. The effects of the three kinds of curing methods on the dry weight losses during curing, chemical contents and physical properties of cured leaves, and organoleptic and smoking qualities were observed. The dry weight losses during curing was the highest in the stalk curing, and the lowest in the conventional priming. The longer the curing periods was, the more the loss of dry weight. The percent of the excessive dried leaves was high and the price per kilogram was low in the conventional priming. The chemical contents and the physical properties of the cured leaves in the conventional priming were similar to those of yellowing or browning stage of curing. But, the chemical contents and the physical properties of the cured leaves in the stalk curing were comparable to those of the air curing. The smoking quality was the best in the stalk curing.

Degree of Heterosis in Diallel Crosses of Burley and Chinese Sun-cured Tobacco (Nicotiana tabacum L.) (버어리종 연초와 중국 양건종의 이면교잡에 의한 양적형질의 잡종강세정도)

  • This study was conducted to obtain the basic information for breeding of burley(N. tabacum cv. burley) and sun-cured tobacco(N. tabacum cv. sun-cured) variety. Two burley(Burley 21, KB 108), and T.I. 1068 and three sun-cured varieties(Yeonbyeon 3, Jaheungyeon and Jaraehong) were diallel crossed and 15 combinations of F$_1$generations were made in 1998, respectively. Six parental varieties, 15 F$_1$s were grown under the complete randomized block design with 3 replications at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 2000. The data of 12 quantitative characters were measured for degree of heterosis. The results obtained are as follows. Heterosis of F$_1$generation was positive in weight per leaf, plant height, stem diameter, yield, leaf length and total alkaloid content. Cross combinations between either Burley 21 or KB 108 and sun-cured varieties showed comparatively high heterosis in each characters.

  • The present study examined various environmental and cultural media conditions for in vitro "rescue" of cross-fertilized ovules formed through sexual crosses between Nicotiana rustica and N. tabacum cv. BY4. The response ovules to two cultural procedures was compared; ovules were cultured either separately or left attached to the placenta. Total yield of seedlings and percent of normal seedlings were increased by culturing individual ovules separately, rather than on excised placenta. Total yield of seedlings and number of normal seedlings were produced following in vitro culture of individual fertilized ovules of N. rustica X M tabacum cv. BY4 at four days post-pollination on NN medium containing 2% sucrose. In the in vitro culture of fertilized ovules, high sucrose concentration increased the frequency of seedlings of abnormal appearance. Therefore, sucrose should be supplied to developing ovules at gradually decreased concentrations. Culture of fertilized ovules from three to eight days after pollination gave increased number of seedlings, but with delayed cultral time the number of morphologically normal seedling were decreased. Hybrids were uniform in appearance and showed vegetative heterosis but flower characteristics were generally intermediate between those of the parents. All hvbrids evaluated were self-sterile.f-sterile.

  • Feeding experiment of [$^3$H] 5-epi-aristolochene (5-EAS) demonstrated in suspension cultures of tobacco (Nicotiana tabacum L.) that 5-EAS hydroxylase activity was absent from control cells, but induced to a maximum level within 18 h after the addition of cellulase, and was very similar to induction pattern of sesquiterpene cyclase. This result suggest that the conversion of 5-EAS to capsidiol is catalyzed by at least one elicitor-inducible hydroxylase. Cytochrome P450 inhibitors, ancymidol and ketoconazole, suppressed the elicitor-induced capsidiol accumulation by inhibiting hydroxylase activity, suggesting that the hydroxylase may be a Cyt P450 type enzyme.

Effects of Weather Conditions on Sunburn in Stalk Curing of Burley Tobacco

  • The effects of stalk cutting time and environmental factors such as air temperature, leaf temperature, solar radiation and leaf moisture content during harvesting and curing in burley tobacco(Nicotiana tabacum L.) on weight loss of fresh stalks and sunburning in leaves were investigated at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1996 and 1997. Twelve to fifteen percent of the fresh weight was lost in 3 to 4 hours after stalk-cutting, and sunburned leaves could be observed in case of stalk cutting between 11:00 and 15:00 O'clock on a clear sunny day, when the air temperature was 34 to 35$^{\circ}C$, leaf temperature 52 to 54$^{\circ}C$, and solar radiation 700 to 940 w/$m^2$. The leaves exposed to this weather condition were sunburned within 1 hour after stalk cutting. But low temperature (below $25^{\circ}C$) with high solar radiation(above 700w/m2) or high temperature(above 3$0^{\circ}C$) with low solar radiation (below 600w$m^2$) did not induce the sunburn damage in leaves. As the leaf temperature and leaf moisture content were higher, the sunburned leaves increased. The leaves at the higher stalk position were more easily sunburned than those at the lower. This result indicates that the immature leaves with higher chlorophyll content might be more susceptible to sunburning.

The Cause of Calcium Deficient leaf of Burley Tobacco (Nicotiana tabacum L.) (Burley종 연초의 칼슘결핍엽 발생원인)

  • Field experiment was conducted to find out the cause of calcium deficiency of burley tobacco (Nicotiana tabacum L.), Liming materials and application rate were CaSO4 : Ca 35kg/10a, CaCO3 : 35kg/10a and agricultural lime : Ca 142kg/10a. The breakdown in tissues at tips and margins of upper leaves was developed at maximum growth stage. If complete breakdown and death does not occur and growth takes place later, giving the leaf a scalloped appearance. Upper leaves contained lower calcium content than other leaves. The stem and midvein of calcium deficient plants contained lower calcium and calcium minus oxalic acid, but higher oxalic acid contents than those of normal plant. Fresh leaves of limed plot contained higher calcium and oxalic acid, but not significant increment of calcium minus oxalic acid than those of unlimed plot. Since calcium oxalate is insoluble, it could precipitate within the culls if the calcium and oxalic acid are accessible to each other. It suggest that high level of oxalic acid in stem and midvein could be interfering with translocation of calcium to upper leaves.

  • Field test were conducted to select the best method for soil phosphorus test and to obtain the critical levels of each method for P fertilizer recommendations based on tobacco production. Truog method was recognized to be the mort suitable one for soil P test. N. Carolina, Bray No.1 and Olsen methods were followed in order. Bray No. 2 and Lancaster methods showed the least satisfaction. The critical levels of available P by lancaster, Bray No. 1, N. Carolina,01sen, and Truog methods were 55ppm, 32ppm, 21ppm, 19ppm, and 22ppm, respectively. Below the critical level a large yield response can be expected with 10kg $P_2O_5$/10a, above critical level little or no response is expected. In soils the more the initial available p, the more available P was carried over, while the more the total amount of silt and clay, the less the residual P was found. The increase in residual P by 10kg of $P_2O_5$ was greater than that of 5kg/10a. The chemical composition and the price of the cured leaf were not affected by the $P_2O_5$ application, indicating that the increase in the rate of $P_2O_5$ application would not exert on the quality of tobacco.
