  • During titration, several chemical reactions result in changes not only in the potential of chemicals, but also in the colors of the indicator. In a potentiometric titration, a titration curve is obtained by measuring the abrupt change in the potential at the endpoint. Generally, acid-base titration is performed by observing the color change caused by an indicator to determine the endpoint. The method of determining the endpoint by measuring the potential difference has been well established and commercialized; however, the devices that can obtain the endpoint by observing the color change are limited. Consequently, in this study, a simple and precise spectral endpoint detector was manufactured using a drop-counter comprising an infrared emitter and a phototransistor, a white light LED as the light source and photodetector, and an analog-to-digital converter (Arduino). Spectrator, a new named, showed excellent results in terms of the reproducibility of acid-base titration using thymol blue as an indicator. Herein, we present the results of the Spectrator-manufacturing process as well as the experimental results.

Determination of Free Acid in U(VI)-Al(III) Solutions by Gran Plot Titration (Gran Plot 적정법을 이용한 U(VI)-AI(III) 용액의 자유산 농도 측정)

  • The determination method of free acid in spent U-Al nuclear fuel solutions by Gran plot titration was described. Effect of U(VI) and Al(III) on the alkalimetric titration of nitric acid was investigated in oxalate complexing media as well as in noncomplexing media. Positive biases were observed in both titration media when the end-point was estimated by the Gran plot method. It was found that the cause of the bias was U(VI) in the oxalate complexing media, but Al(III) in the noncomplexing media. The relative error was less than 1% in the titration of 0.1 M $HNO_3$ at a U(VI) : Al(III) : $H^+$ mole ratio of up to 2:12:1 as long as the pH of the oxalate titration media was sustained to be below 5.0 at the beginning of titration. The method was successfully applied to the determination of nitric acid in a solution of HANARO reactor fuel with U:Al mole ratio of 1:6.

Sleep Fragementation Decreases during the nCPAP Titration Night in Obstructive Sleep Apnea Syndrome (폐쇄성 수면무호흡증에서 지속적 상기도 양압술 압력 처방 검사 시 나타나는 수면분절의 감소)

  • Objectives: Obstructive sleep apnea syndrome (OSAS) not only causes respiratory disturbances during sleep but also decreases the quality of nocturnal sleep through sleep fragmentation and sleep structure change. We aimed at comparing the changes in sleep fragmentation and structure between baseline (diagnostic) nocturnal polysomnography (NPSG) and nCPAP (nasal continuous positive airway pressure) titration trial. Methods: One hundred and three patients with a baseline night of respiratory disturbance index (RDI) of 5 or greater and reduced RDI score during nCPAP titration night were retrospectively selected for the study. Sleep fragementation and sleep structure between baseline NPSG and the NPSG during nCPAP titration were compared. Sleep fragmentation index (SFI) was defined as the total number of awakenings and shifts to stage 1 sleep divided by the total sleep time in hour. SFI and other polysomnographic parameters were statistically compared between the two nights. Results: SFI during baseline NPSG and nCPAP titration nights were $29.0{\pm}13.8$ and $15.2{\pm}8.8$, respectively, indicating a significant SFI decrease during nCPAP titration (t=9.7, p<0.01). SFI showed significant negative correlations with sleep efficiency (r=-0.60, p<0.01) and total sleep time (r=-0.45, p<0.01) and a positive correlation with RDI (r=0.28, p<0.01). Conclusion: Use of nCPAP, even during the titration, significantly decreases sleep fragmentation and improves sleep structure in OSAS patients. We suggest that SFI may be utilized as a measure of assessing OSAS severity and nCPAP efficacy.

Development of alternative titration method without mercuric acetate for the assay of difemerine hydrochloride in Korean Pharmaceutical Codex (디페메린염산염 시험에 사용되는 유해시약인 수은 대체 시험법의 개발)

  • Currently nonaqueous titration method for the assay method using the hazardous reagent, mercury acetate for difemerine hydrochloride has been used in Korean Pharmaceutical Codex. We developed an alternative titration assay method by substituting the use of the hazardous reagent, mercury acetate to the use of less toxic ones like ethyl alcohol. The linearity of the calibration curves in the desired concentration range was good (r>0.999). Precision was less than 0.64%. Accuracy was obtained with recoveries in range of 99.10% and 99.71%. The developed assay could be expected to become valuable tools for revising the Korean Pharmaceutical Codex.

  • The varation of potential and completeness of reaction in an oxidation-reduction titration of Red$_1$ ($a\;Ox_1+N_1e{\leftrightarrow}b\;Red_1)\;by\;Ox_2\;(C\;Ox_2+n_2e{\leftrightarrow}d\;Red_2)$ is analyzed on the basis of general expressions. It is shown that the reaction deficiency as well as the rate of variation of the potential with titration fraction change in proportion to the $[n_2(a-b)-n_1(c-d)]$ th power of the concentration of the reagents. In particular, at the equivalence point, the expression of the potential contains a concentration dependent term that is proportional to ac-bd. Thus, the equivalence-point potential varies with the concentration of the reagents unless ac=bd is satisfied. It is also shown that the sharpness of the potentiometric titration curve is shown to occur either prior to or after the equivalence point depending upon whether $n_1cn_2b$, the deviation being the same order of magnitude as the relative deficiency at the equivalence point.

Intravenous Nefopam Reduces Postherpetic Neuralgia during the Titration of Oral Medications

  • Background: The recently known analgesic action mechanisms of nefopam (NFP) are similar to those of anticonvulsants and antidepressants in neuropathic pain treatment. It is difficult to prescribe high doses of oral neuropathic drugs without titration due to adverse effects. Unfortunately, there are few available intravenous analgesics for the immediate management of acute flare-ups of the chronic neuropathic pain. The aim of this study was to determine the additional analgesic effects for neuropathic pain of NFP and its adverse effects during the titration of oral medications for neuropathic pain among inpatients with postherpetic neuralgia (PHN). Methods: Eighty inpatients with PHN were randomly divided into either the NFP or normal saline (NS) groups. Each patient received a 3-day intravenous continuous infusion of either NFP with a consecutive dose reduction of 60, 40, and 20 mg/d, or NS simultaneously while dose titrations of oral medications for neuropathic pain gradually increased every 3 days. The efficacy of additional NFP was evaluated by using the neuropathic pain symptom inventory (NPSI) score for 12 days. Adverse effects were also recorded. Results: The median NPSI score was significantly lower in the NFP group from days 1 to 6 of hospitalization. The representative alleviating symptoms of pain after using NFP were both spontaneous and evoked neuropathic pain. Reported common adverse effects were nausea, dizziness, and somnolence, in order of frequency. Conclusions: An intravenous continuous infusion of NFP reduces spontaneous and evoked neuropathic pain with tolerable adverse effects during the titration of oral medications in inpatients with PHN.

  • Caustic Soda Treatment Systems need a concentration measurement device in order to monitor the weight reduction amount of polyester fabrics. Since the reduction process requires several concentration measurements, we have to do the unit titration fast and exactly. Therefore, a proposition of estimation algorithm for finding the equivalent point in the titration process is needed. In this paper, we used the cardinal spline algorithm, to estimate the proper curve with the measured pH-values after each injection of titration sdution, to predict the equivalent point. While the processing time is elapsed, several equivalent points are estimated and drawed the line graph of fabrics weight reduction automatically. Finally, we designed the hardware and the software of an automatic titration system that can generate the reduction ending signal of Caustic Soda Treatment System.

  • Control of pH neutralization process plays a very important role in some chemical process. Because of their high nonlinearity, frequent disturbance, and time-varying characteristics, it is difficult to control and estimate pH processes. For the adaptive control of pH processes, a lot of researchers have made an efforts in the modeling and control of pH processes. It is very difficult to obtain information of influent stream such as concentrations and dissociation constants and the titration curve equation is very complex. Therefore, several simple models, which hate small number of unknown parameters and estimate the titration curve, have been available, These models were considered here and were transformed into forms that can applied the linear least square method.

  • Kinetic and thermodynamic studies have been made on the effect of the inosine product on the activity of adenosine deaminase in a 50 mM sodium phosphate buffer, pH 7.5, at $27^{\circ}C$ using UV spectrophotometry and isothermal titration calorimetry (ITC). A competitive inhibition was observed for inosine as a product of the enzymatic reaction. A graphical-fitting method was used for determination of the binding constant and enthalpy of inhibitor binding by using isothermal titration microcalorimetry data. The dissociation-binding constant is equal to $140\;{\mu}M$ by the microcalorimetry method, which agrees well with the value of $143\;{\mu}M$ for the inhibition constant that was obtained from the spectroscopy method.
