STAT6 and PARP Family Members in the Development of T Cell-dependent Allergic Inflammation

  • Allergic inflammation requires the orchestration of altered gene expression in the target tissue and in the infiltrating immune cells. The transcription factor STAT6 is critical in activating cytokine gene expression and cytokine signaling both in the immune cells and in target tissue cells including airway epithelia, keratinocytes and esophageal epithelial cells. STAT6 is activated by the cytokines IL-4 and IL-13 to mediate the pathogenesis of allergic disorders such as asthma, atopic dermatitis, food allergy and eosinophilic esophagitis (EoE). In this review, we summarize the role of STAT6 in allergic diseases, its interaction with the co-factor PARP14 and the molecular mechanisms by which STAT6 and PARP14 regulate gene transcription.

생물학적 폭경을 고려한 보철임상 증례 (Clinical Cases on the Restorative Procedure Preserving the Biologic Width)

  • The preservation of a healthy periodontal attachment is the most significant factor in the long-term prognosis of a restored tooth. The 'Biologic Width' is composed of the connective tissue attachment and the epithelial attachment in the dentogingival junction. The violation of the biologic width may result in a progressive inflammatory process and crestal bone loss. So a careful soft tissue management is needed to preserve it for the gingival health and an esthetic restoration. The following clinical cases show the five different situations of the violation of the biologic width and their management.

  • Berger formulation was used to calculate the dose distribution of $^{60}$ Co source in tissue. $^{60}$ Co source was supposed as point source. The effect of the stainless-steel around the source was considered and Taylor Approximation Method was used for calculating exposure build-up factor. Calculated depth dose data was compared with measured data which was measured by the ionization chamber.

일반 Batho방법의 부정형 이질조직에의 적용 (Application of Generalized Batho Method to Arbitrary Shape of Heterogeneous Tissues)

  • The generalized Bathe method, proposed by Webb and Fox, which is a method of calculation of dose correction factor for the purpose of heterogeneous tissue, is complex even for a few kind of tissues. The method was modified for the purpose of getting a simple method that divide the multilayer of heterogeneous tissues into some groups of adjacent-tissue pairs. This new method could reduce the number of exponential terms and the time for calculating the dose correction factors by manual and computer calculation.

심장외막의 지방두께에 따른 대사질환의 위험도 분석 - 무증상의 검진목적으로 내원한 대상자를 위주로 (Risk Analysis of Factors for Metabolic Diseases according to the Epicaridal Adipose Tissue Thickness - which Focused on the Presented Subjects with Asymptomatic Screening Purposes)

  • 심장외막지방(epicardial adipose tissue, EAT)은 여러 호르몬을 분비하는 대사활성 내분비 기관으로 지방의 두께증가는 심혈관질환이나 대사질환의 위험인자이다. 본 연구는 초음파를 이용하여 대상군의 심장외막 지방두께와 복부피하지방두께를 측정한 후 일반적 특성 및 혈액학적 특성과의 상관관계를 분석하고 대사질환의 예측을 위한 지방두께를 제시하고자 하였다. 연구 결과 대상자의 심장외막지방두께의 평균은 각 단면도에서 각각 8.890mm, 4.783mm, 4.777mm, 6.147mm로 측정되었다. 각 단면에서 심장외막지방두께평균과 위험인자와의 상관관계에서는 나이, BMI, 수축기혈압, LDH, LDL, TC가 양의 상관관계(p<0.05)를 나타냈다. 특히 대사질환의 위험인자를 가지고 있는 대상자가 위험인자를 가지고 있지 않은 대상자에 비하여 심장외막지방두께 및 복부피하지방의 두께가 유의(p<0.05)하게 높게 나타났다. EAT1에서 측정한 심장외막지방 두께 8.950 mm에서 대사질환의 위험을 예측할 수 있는 민감도 66.7 %, 특이도 80 %를 보여 가장 신뢰성 있는 cut off value를 나타냈다.

Effects of Soft Tissue Massage of the Posterior Deltoid Muscle on Shoulder Horizontal Adduction

  • Stiffness of the posterior deltoid is as a causative factor in the limited range of glenohumeral horizontal adduction and various other shoulder pathologies including shoulder impingement syndrome, frozen shoulder, and humerus anterior glide syndrome. The purpose of this study was to compare the effects of two techniques (soft tissue massage and cross-body stretch) on increasing the range of horizontal adduction. Thirty-two subjects with a $10^{\circ}$ or greater difference between the right and left sides in horizontal adduction were selected. Sixteen subjects from each group were allocated randomly. Interventions were applied on six occasions for 2 weeks, and the range of horizontal adduction was measured using an inclinometer at pre-and post-intervention. A $2{\times}2$ analysis of variance (intervention${\times}$time) was used to compare the effects of the two techniques. In the soft tissue massage group, the angle of horizontal adduction significantly increased compared with the cross-body stretch group. These findings indicate that the soft tissue massage of the posterior deltoid muscle is a more effective method to increase the flexibility of the glenohumeral horizontal adduction.

Bone Morphogenetic Protein(BMP)이 인체 치주인대 세포의 활성에 미치는 효과 (EFFECTS OF BONE MORPHOGENETIC PROTEIN(BMP) ON HUMAN PERIODONTAL LIGAMENT CELLS IN VITRO)

  • Periodontitis is characterized by gingival inflammation and results in periodontal pocket formation with loss of the supporting alveolar bone and connective tissue around the teeth. Therapeutic modalities should therefore aim not only at eliminating the gingival inflammatory process and preventing the progression of periodontal disease but also at reestablishing and regenerating the periodontal tissue previously lost to the disease. To achieve periodontal regeneration, progenitor cells must migrate to the denuded root surface, attach to it, proliferate and mature into an organized and functional fibrous attachment apparatus. Likewise, progenitor bone cells must also migrate, proliferate, and mature in conjunction with the regenerating periodontal ligament. Significant advances have been made during the last decade in understanding the factors controlling the migration, attachment and proliferation of cells. A group of naturally occuring molecules known as polypeptide growth factors in conjunction with certain matrix proteins are key regulators of these biological events. Of these, the fibroblast growth factor(FGF), platelet-derived growth factor(PDGF) , insulin like growth factor(CIGFs), transforming growth factor(TGFs), epidermal growth factor(EGF) and bone morphogenetic growth factor(BMPs) apper to have an important role in periodontal wound healing. The purpose of this study was to determine the effects of BMP on periodontal ligament cells. Human periodontal ligament cells were cultured from extracted tooth for non-periodontal reason. Cultured periodontal ligament cells were treated with BMP. Cellular activities were determined by MTT(3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay and ALP(alkaline phosphatase) activity. The results were as follows ; Regardless of cultured time, cellular activities were stimulated by BMP. Also, BMP greatly increased alkaline phosphatase(ALP) in periodontal ligament cells. These results suggest that BMP not only have no cytotoxic effect on periodontal ligament cells, but also have osteogenic stimulatory effect on periodontal ligament cells.

진피섬유모세포에서 대복피추출물의 세포외기질 합성 촉진 효과 (Stimulation of the Extracellular Matrix Production in Dermal Fibroblasts by Areca catechu Extract)

  • 교원질을 비롯한 세포외기질의 생합성을 통해 피부장력과 탄력 등 피부 특성을 제공하는 진피섬유모세포는 피부노화와 함께 활성이 감소되어 주름 형성의 이유가 된다. 따라서 젊고 건강한 피부를 유지하기 위해서는 진피섬유 모세포의 활성화가 큰 의미를 지닌다. 본 연구에서는 대복피 에탄올추출물이 진피섬유모세포의 세포외기질 합성에 미치는 영향을 ELISA, Western blot analysis 및 RT-PCR 등의 in vitro 평가법으로 측정하였다. ELISA와 western blot analysis에서 대복피추출물은 제1형 교원질, fibronectin, transforming growth factor-${\beta}1$ (TGF-${\beta}1$)의 생성을 촉진시켰고, RT-PCR에서는 COL1A1, TGF-${\beta}1$, keratinocyte growth factor (KGF), insulin growth factor (IGF)-1의 유전자 발현을 증가시켰다. 이상의 결과로부터 대복피추출물은 진피섬유모세포에서 세포외기질의 생성을 촉진시키는 천연소재인 것으로 판단되었다.
