In Vitro Plantlet Regeneration from Axillary Buds of Tilia amurensis Mature Trees and Clonal Variation in Tissue Culturability (피나무 성숙목(成熟木)의 액아배양(腋芽培養)에 의한 유식물체(幼植物體) 재생(再生)과 조직배양능력(組織培養能力)에 있어서의 클론간(間) 변이(變異))

  • The axillary buds of 15-year-old Tilia amurensis were cultured on Saito and Ide (IS), Murashige and Skoog (MS) media and woody plant medium (WPM) to establish an effective micropropagation method. Five levels of 6-benzylaminopurine (BAP) were tested. On IS medium and WPM addition of 1.0/l BAP enhanced shoot development and shoot elongation, whereas addition of 0.5/l BAP was effective on MS medium. A better results were obtained from WPM with 1.0/l BAP and MS with 0.1/l BAP. Developed shoots were subcultured on each basal media but with 0.2/l BAP, Multiple shoots were almost doubled in a month. Root formation could be enhanced at higher concentration of indole-3-butyric acid (IBA). Better rooting rate (83.3%) was achieved on a half-strength MS medium with 3.0 /l IBA. Regenerated plantlets were successfully transferred to soil. To investigate the clonal variation in shoot development and shoot elongation by axillary bud culturing, seven plus tree clones were tested, Clonal variation in tissue culturability among plus trees was recognized by the Duncan's multiple range test at the 5% level. Kang Won No. 12 showed the best response on WPM with 1.0/l BAP.

  • Rice (Oryza sativa L.) anthers collected from different genotypes were cultured to investigate the priority to increase callus production rate among several factors affecting on callus formation. Rice varieties, F$_1$ hybrids and F$_2$ plants differed greatly in their abilities to produce callus from anthers, and it was confirmed that the culturability of rice anther was a heritable characteristic. Cold shock treatment before plating anthers promoted callus formation rate, but combined effect with genotype having high culturability was more significant. The response to sucrose concentration in culture medium in callus induction rate was different between rice genotypes, but combined effect with genotype was not significant. Single supplement of NAA to R$_2$ medium increased callus production rate remarkably only in rice genotype having high culturability. Conclusively selection of genotypes is most important to increase callus initiation frequency from rice anthers.

  • In an effort to optimize tissue culture responses of tall fescue, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. The MS basal medium was superior to $N_{5}$ 6/ and $B_{medium}$ in enhancing callus growth and plant regeneration. To determine the effect of carbon sources on plant regeneration, the seeds were cultured on medium with 30 g/L sucrose and maltose, respectively. Medium supplemented with 30 g/L sucrose resulted in regeneration of shoots from 50% of the calli. The genotypic difference in plant regenerability was obvious among five cultivars of tall fescue tested. \\`KY31\\` and \\`Hokuryo\\` showed to have higher regenerability with the frequency of 33% and 51%, respectively.

  • In order to optimize the conditions of callus induction and plant regeneration in seed culture of Italian ryegrass, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. MS medium was better than N6 and B5 medium in enhancing callus growth and plant regeneration. Sucrose was superior to maltose in plant regeneration as carbon source in the medium. The genotypic difference in plant regenerability was obvious among four cultivars of Italian ryegrass tested. 'Rio' and 'Jeanne' showed to have higher regenerability with the frequency of 38% and 56%, respectively.

Reciprocal effects were not noticed in anther culturability There was no variants related to rice leaf discoloration in pollen derived lines from parental varieties, regardless of days of cold stress. The regeneration and recombination of rice leaf discoloration in 146 pollen-derived lines, 70 pollen-derived lines from cold stress at $0^{\circ}C$ for 10 days, and 830 F$_2$ plants presented normal distribution curves with skewness in tolerance and no significant difference among 3 populations. Direct in vitro selection for rice cold tolerance through cold stress on primary pollen callus derived from anther culture, therefore, was revealed ineffective as a in vitro technology.

Varietal Difference and Inheritance of Plant Regenerability in Anther Culture of Rice (벼 악배양 효율의 품종간 차이와 식물체 재분화 능력의 유전)

  • This study was carried out to identify varietal difference and the inheritance of the ability of plant regeneration in anther culture of rice. The anthers of 33 Japonica, 11 Indica/Japonica, and 3 Indica rice cultivars were culture on $N_6-Y$_1$$ medium with 1 mg/L NAA and 2 mg/L kinetin. The remarkable variability in plant regeneration were observed among the genotypes, ranging from 0.0% to 19.4% Most of Japonica rices were resulted to have better culturability than that of Indica/Japonica and Indica type cultivars. Newly developed Japonica rices, 'Ilmibyeo' and 'Hangnambyeo' showed to have regenerability with the frequency of 19.4% and 18.1% respectively. The segregation mode for callus formation and plant regenerationin anther culture of F$_2$ population of 'Milyang 23/Chucheongbyeo' showed a continuous variation. The variation of plant regeneration frequency in anther culture of F$_2$ plants ranged from 0% to 33.3% with a mean of 6.3% The broad-sense heritability estimated from the ability of plant regeneration was considerably high (82.7%).
