Molecular Cloning of Adipose Tissue-specific Genes by cDNA Microarray

  • In an attempt to isolate novel molecules that may play a regulatory role in adipocyte differentiation, we devised an experimental strategy to identify adipose tissue-specific genes by modifying cDNA microarray technique. We used genefilter membranes containing approximately 15,000 rat non-redundant EST clones of which 4,000 EST were representative clones of known genes and 11,000 ESTs were uncharacterized clones. A series of hybridization of genefilter membranes with cDNA probes prepared from various rat tissues and nucleic acids sequence analysis allowed us to identify two adipose-tissue specific genes, adipocyte-specific secretory factor (ADSF) and H-rev107. Verification of tissue-specific expression patterns of these two genes by Northern blot analysis showed that ADSF mRNA is exclusive expressed in adipose tissue and the H-rev107 mRNA is predominantly expressed in adipose tissue. Further analysis of gene expression of ADSF and H-rev107 during 3T3-L1 adipocyte differentiation revealed that the ADSF and H-rev107 gene expression patterns are closely associated with the adipocyte differentiation program, indicating their possible role in the regulation of adipose tissue development. Overall, we demonstrated an application of modified cDNA microarray technique in molecular cloning, resulting in identification of two novel adipose tissue-specific genes. This technique will also be used as a useful tool in identifying novel genes expressed in a tissue-specific manner.

  • Tissue microarry is one of the high throughput technologies in the post-genomic era. Using tissue microarray, the researchers are able to investigate large amount of gene expressions at the level of DNA, RNA, and protein The important aspect of tissue microarry is its ability to assess a lot of biomarkers which have been used in clinical practice. To manipulate the categorical data of tissue microarray, we applied Bayesian network classifier algorithm. We identified that Bayesian network classifier algorithm could analyze tissue microarray data and integrating prior knowledge about gastric cancer could achieve better performance result. The results showed that relevant integration of prior knowledge promote the prediction accuracy of survival status of the immunohistochemical tissue microarray data of 18 tumor suppressor genes. In conclusion, the application of Bayesian network classifier seemed appropriate for the analysis of the tissue microarray data with clinical information.

Assessment of The Accuracy of The MR Abdominal Adipose Tissue Volumetry using 3D Gradient Dual Echo 2-Point DIXON Technique using CT as Reference

  • In this study, in order to determine the validity and accuracy of MR imaging of 3D gradient dual echo 2-point DIXON technique for measuring abdominal adipose tissue volume and distribution, the measurements obtained by CT were set as a reference for comparison and their correlations were evaluated. CT and MRI scans were performed on each subject (17 healthy male volunteers who were fully informed about this study) to measure abdominal adipose tissue volume. Two skilled investigators individually observed the images acquired by CT and MRI in an independent environment, and directly separated the total volume using region-based thresholding segmentation method, and based on this, the total adipose tissue volume, subcutaneous adipose tissue volume and visceral adipose tissue volume were respectively measured. The correlation of the adipose tissue volume measurements with respect to the observer was examined using the Spearman test and the inter-observer agreement was evaluated using the intra-class correlation test. The correlation of the adipose tissue volume measurements by CT and MRI imaging methods was examined by simple regression analysis. In addition, using the Bland-Altman plot, the degree of agreement between the two imaging methods was evaluated. All of the statistical analysis results showed highly statistically significant correlation (p<0.05) respectively from the results of each adipose tissue volume measurements. In conclusion, MR abdominal adipose volumetry using the technique of 3D gradient dual echo 2-point DIXON showed a very high level of concordance even when compared with the adipose tissue measuring method using CT as reference.

  • Objectives : The purpose of this study is to report the relationship between iridological constitution and interleukin 1 beta (IL-1 $\beta$) gene polymorphism. Methods : Iris constitution were diagnosed by automatic Iris analysis system, Bexel Irina(Korea). The blood was stored at - $20^{\circ}$... until it was ready to be extracted. The genomic DNA was extracted by inorganic procedure. The concentration of DNA was estimated by absorbance at 260 nm. The interleukin-1 beta (IL-1 $\beta$) gene polymorphism was detected by PCR amplification. Results & Conclusions : The author classified 166 individuals according to Iris constitution, and determined IL-1 $\beta$ genotype. The frequencies of Iris constitutions as follows : neurogenic type, 41 (24.7%); abdominal connective tissue weakness type, 53(31.9%); cardio-renal connective tissue weakness type, 50 (30.1%); the others type, 22 (13.3%). Especially, the frequency of abdominal connective tissue weakness type was significantly higher in err genotype than in the remaining constitutions. As a result, The author demonstrated the association among IL-1 $\beta$ genotype, IBD and Iris constitution.

  • Objectives : We performed this study to analyze correlation among hair tissue minaral ratio, autonomic function and obesity. Methods : Subjects were gathered from January 2005 to March 2007. This study was carried out on 263 subjects who had visited Garosero oriental clinic and had no previous cardiovascular disease and thyroid disease. Heart rate variability (HRV) parameters, tissue mineral ratio and obesity degree were statistically compared with correlation and T-test analysis. Results : The results of this study are summarized as follows : 1. Normal group were predominant over obesity group in HRV parameters(SDNN, RMSSD, VLF, LF, HF). 2. Ca/P, Ca/K, Na/K and Fe/Cu, Na/Mg ratio in hair tissue mineral ratio have correlation with BMR, BMI and waist circumference. 3. Ca/P ratio has correlation with LF norm in HRV, and Na/K with HF, Na/Mg with LF, equally. Conclusions : Taken together these results may suggest that there are significant relationships between hair tissue mineral analysis and HRV.

Numerical Analysis of the Wavelength Dependence in Low Level Laser Therapy (LLLT) Using a Finite Element Method

  • Purpose: The aim of this study was to do numerical analysis of the wavelength dependence in low level laser therapy (LLLT) using a finite element method (FEM). Methods: Numerical analysis of heat transfer based on a Pennes' bioheat equation was performed to assess the wavelength dependence of effects of LLLT in a single layer and in multilayered tissue that consists of skin, fat and muscle. The three different wavelengths selected, 660 nm, 830 nm and 980 nm, were ones that are frequently used in clinic settings for the therapy of musculoskeletal disorders. Laser parameters were set to the power density of 35.7 W/$cm^2$, a spot diameter of 0.06 cm, and a laser exposure time of 50 seconds for all wavelengths. Results: Temperature changes in tissue based on a heat transfer equation using a finite element method were simulated and were dominantly dependent upon the absorption coefficient of each tissue layer. In the analysis of a single tissue layer, heat generation by fixed laser exposure at each wavelength had a similar pattern for increasing temperature in both skin and fat (980 nm > 660 nm > 830 nm), but in the muscle layer 660nm generated the most heat (660 nm ${\gg}$ 980 nm > 830 nm). The heat generation in multilayered tissue versus penetration depth was shown that the temperature of 660 nm wavelength was higher than those of 830 nm and 980 nm Conclusion: Numerical analysis of heat transfer versus penetration depth using a finite element method showed that the greatest amount of heat generation is seen in multilayered tissue at = 660 nm. Numerical analysis of heat transfer may help lend insight into thermal events occurring inside tissue layers during low level laser therapy.

  • A digital spectrum analysis technique was used to estimate the tissue characteristic parameters (transmission velocity and attenuation coefficient) in the phantom study and the human liver's ultrasound scanning. The soft tissue equivalent phantom was made with the combination materials of agar, water, powdered graphite, and n-propyl alcohol. In the human study, twenty five normal subjects and three patients with liver diseases were studied using the ultrasonic reflection signals and the spectrum analysis method The following results were obtained; 1. The soft tissue-equivalent materical could be produced with various acoustic parameters by changing the composition amount of the powdered graphite and n-propyl alcohol. 2. Attenuation coefficients of normal human liver tissue were estimated to be 0. 36 dB/cm MHz$\pm$0.11. In patients with liver disese, tile attenuation coefficients were shown to be different from the above normal values.

Proteomic Analysis of Colonic Mucosal Tissue from Tuberculous and Ulcerative Colitis Patients

  • Changes in the expression profiles of specific proteins leads to serious human diseases, including colitis. The proteomic changes related to colitis and the differential expression between tuberculous (TC) and ulcerative colitis (UC) in colon tissue from colitis patients has not been defined. We therefore performed a proteomic analysis of human TC and UC mucosal tissue. Total protein was obtained from the colon mucosal tissue of normal, TC, and UC patients, and resolved by 2-dimensional electrophoresis (2-DE). The results were analyzed with PDQuest using silver staining. We used matrix-assisted laser desorption ionization time-of-flight/time-of-flight spectrometry (MALDI TOF/TOF) to identify proteins differentially expressed in TC and UC. Of the over 1,000 proteins isolated, three in TC tissue and two in UC tissue displayed altered expression when compared to normal tissue. Moreover, two proteins were differentially expressed in a comparative analysis between TC and UC. These were identified as mutant ${\beta}$-actin, ${\alpha}$-enolase and Charcot-Leyden crystal protein. In particular, the expression of ${\alpha}$-enolase was significantly greater in TC compared with normal tissue, but decreased in comparison to UC, implying that ${\alpha}$-enolase may represent a biomarker for differential diagnosis of TC and UC. This study therefore provides a valuable resource for the molecular and diagnostic analysis of human colitis.

온침 시술 시 생체 조직 내 열분포 분석에 관한 연구 (Thermal Distribution in Living Tissue during Warm Needling Therapy)

  • Objectives This study aims to analyze a thermal distribution in biological living tissue during warm needling therapy by using a finite element method. The analysis provides an understanding of warm needling's efficacy and safety. Methods A model which consisted of four-layered tissue and stainless steel needle was adopted to analyze the thermal distribution in living tissue with a bioheat transfer analysis. The governing equation for the analysis was a Pennes' bioheat equation. A heat source characteristic of warm needling therapy was obtained by previous experimental measurements. The first analysis of the time-dependent temperature distribution was conducted through points on a boundary between the needle and the tissue. The second analysis was conducted to visualize the horizontal temperature distribution. Results When heat source's peak temperatures was above $500^{\circ}C$ and temperature rising rates were relatively slow, the peak temperature at skin surface exceeded a threshold of pain and tissue damage ($45^{\circ}C$), whereas when the peak temperature was around $400^{\circ}C$, the peak temperature at the skin surface was within a safe limit. In addition, the conduction of combustion energy from the moxa was limited to the skin layer around the needle. Conclusions The results suggest that the skin layer around the needle can be heated effectively by warm needling therapy, but it appears to have little effect at the deeper tissue. These findings enhance our understanding of the efficacy and the safety of the warm needling therapy.

Elevated Aurora Kinase A Protein Expression in Diabetic Skin Tissue

  • Background Aurora kinase A (Aurora-A) plays an important role in the regulation of mitosis and cytokinesis. Dysregulated Aurora-A leads to mitotic faults and results in pathological conditions. No studies on Aurora-A expression in human diabetic skin tissue have been reported. In light of this, we explored the expression of Aurora-A in human diabetic skin tissue. Methods Aurora-A protein was evaluated by western blotting in 6 human diabetic skin tissue and 6 normal skin specimens. Results Increased expression of Aurora-A protein was detected in all diabetic skin tissue samples in both western blot analysis and immunohistochemical staining. However, in the case of the normal skin tissue, no bands of Aurora-A protein were detected in either the western blotting analysis or the immunohistochemical staining. Conclusions Thus far, there have been no studies on the expression of Aurora-A in diabetic skin tissue. However, we believe that oxidative DNA damage related to the expression of Aurora-A protein and Aurora-A could be involved inhuman diabetic skin tissue.
