Effects of Racemic Ketamine on Excitable Membranes of Frog (개구리 세포막에 대한 Racemic Ketamine의 영향)

  • The effect of racemic Ketamine HCl was observed on excitable membranes of sciatic nerve fibres and toe muscles from frog. Ketamine significantly depressed the amplitude of the action potential, maximum rate of rise and that of fall of action potentials of sciatic nerve by dose-dependent and time-course manner, and also it produced the inhibition of $K^+-contracture$ in toe muscle. We used two different ways of sucrose gap method to to obtain the better results from sciatic nerve. We observed and compared the effect of ketamine on sciatic nerve with naloxone, 4-AP (4-aminopyridine) and TEA (Tetraethylammonium). Naloxone significantly but not totally blocked the effect of ketamine both on nerve and on skeletal muscle. 4-AP or TEA by itself had a significant depressant effect on the action potentials on nerve by central perfusion (extracellular perfusion), but both of these drugs did not much affect the action of Ketamine on nerve. The reversibility of effect of Ketamine (10 mM) was observed both on nerve and on skeletal muscles when exposed to drug for short duration. The effects of racemic ketamine described may provide to support that one of the mechanisms of the action of Ketamine on nerve and on muscles of frog might be related to non-specifically effect on receptors within the ion channels $(K^+-channel,\;Na^+-channel\;or\;slow\;Ca^{++}\;channel)$ at higher dose which produces anesthetic effect and also it interacts specifically with one of the opioid receptors or subtype of these receptors which is sensitive to Naloxone at lower dose which produces analgesia.

Silicon Oil-Based 2-Channel Fiber-Optic Temperature Sensor Using a Subtraction Method (감법을 이용한 실리콘 오일 기반의 2채널 광섬유 온도 센서)

  • We developed a 2-channel fiber-optic temperature sensor (FOTS) using a temperature sensing probe, a fiber-optic coupler, transmitting optical fiber, and an optical time domain reflectometer (OTDR). The temperature sensing probe is divided into a sensing probe and a reference probe for accurate thermometry. A sensing probe is composed of a silicon oil, a FC terminator, a brass pipe, and a singlemode optical fiber and the structure of a reference probe is identical with that of the sensing probe excluding a silicon oil. In this study, we measured the modified optical powers of the light signals reflected from the temperature sensing probe placed inside of the water with a thermal variation from 5 to $70^{\circ}C$. Although the optical power of the reference probe was constant regardless of the temperature change, the optical power of the sensing probe decreased linearly as the temperature increased. As experimental results, the FOTS using a subtraction method showed a small difference (i.e., hysteresis) in its response due to heating and cooling. The reversibility and reproducibility of the FOTS were also evaluated.

Preparation of Electrolytic Tungsten Oxide Thin Films as the Anode in Rechargeable Lithium Battery (리튬 이차전지용 텅스텐 산화물 전해 도금 박막 제조)

  • Tungsten oxide films were prepared by an electrochemical deposition method for use as the anode in rechargeable lithium batteries. Continuous potentiostatic deposition of the film led to numerous cracks of the deposits while pulsed deposition significantly suppressed crack generation and film delamination. In particular, a crack-free dense tungsten oxide film with a thickness of ca. 210 nm was successfully created by pulsed deposition. The thickness of tungsten oxide was linearly proportional to deposition time. Compositional and structural analyses revealed that the as-prepared deposit was amorphous tungsten oxide and the heat treatment transformed it into crystalline triclinic tungsten oxide. Both the as-prepared and heat-treated samples reacted reversibly with lithium as the anode for rechargeable lithium batteries. Typical peaks for the conversion processes of tungsten oxides were observed in cyclic voltammograms, and the reversibility of the heat-treated sample exceeded that of the as-prepared one. Consistently, the cycling stability of the heat-treated sample proved to be much better than that of the as-prepared one in a galvanostatic charge/discharge experiment. These results demonstrate the feasibility of using electrolytic tungsten oxide films as the anode in rechargeable lithium batteries. However, further works are still needed to make a dense film with higher thickness and improved cycling stability for its practical use.

Partially Hydrolyzed Crosslinked Alginate-graft-Polymethacrylamide as a Novel Biopolymer-Based Superabsorbent Hydrogel Having pH - Responsive Properties

  • In this study, a series of highly swelling hydrogels based on sodium alginate (NaAlg) and polymethacryl­amide (PMAM) was prepared through free radical polymerization. The graft copolymerization reaction was performed in a homogeneous medium and in the presence of ammonium persulfate (APS) as an initiator and N,N'-methylenebis­acrylamide (MBA) as a crosslinker. The crosslinked graft copolymer, alginate-graft-polymethacrylamide (Alg-g­PMAM), was then partially hydrolyzed by NaOH solution to yield a hydrogel, hydrolyzed alginate-graft-poly­methacrylamide (H-Alg-g-PMAM). During alkaline hydrolysis, the carboxamide groups of Alg-g-PMAM were converted into hydrophilic carboxylate anions. Either the Alg-g-PMAM or the H-Alg-g-PMAM was characterized by FTIR spectroscopy. The effects of the grafting variables (i.e., concentration of MBA, MAM, and APS) and the alkaline hydrolysis conditions (i.e., NaOH concentration, hydrolysis time, and temperature) were optimized systematically to achieve a hydrogel having the maximum swelling capacity. Measurements of the absorbency in various aqueous salt solutions indicated that the swelling capacity decreased upon increasing the ionic strength of the swelling medium. This behavior could be attributed to a charge screening effect for monovalent cations, as well as ionic cross-linking for multivalent cations. Because of the high swelling capacity in salt solutions, however, the hydrogels might be considered as anti-salt superabsorbents. The swelling behavior of the superabsorbing hydrogels was also measured in solutions having values of pH ranging from 1 to 13. Furthermore, the pH reversibility and on/off switching behavior, measured at pH 2.0 and 8.0, suggested that the synthesized hydrogels were excellent candidates for the controlled delivery of bioactive agents. Finally, we performed preliminary investigations of the swelling kinetics of the synthesized hydrogels at various particle sizes.

Can denosumab be a substitute, competitor, or complement to bisphosphonates?

  • Osteoblasts, originating from mesenchymal cells, make the receptor activator of the nuclear factor kappa B ligand (RANKL) and osteoprotegerin (OPG) in order to control differentiation of activated osteoclasts, originating from hematopoietic stem cells. When the RANKL binds to the RANK of the pre-osteoclasts or mature osteoclasts, bone resorption increases. On the contrary, when OPG binds to the RANK, bone resorption decreases. Denosumab (AMG 162), like OPG (a decoy receptor), binds to the RANKL, and reduces binding between the RANK and the RANKL resulting in inhibition of osteoclastogenesis and reduction of bone resorption. Bisphosphonates (BPs), which bind to the bone mineral and occupy the site of resorption performed by activated osteoclasts, are still the drugs of choice to prevent and treat osteoporosis. The merits of denosumab are reversibility targeting the RANKL, lack of adverse gastrointestinal events, improved adherence due to convenient biannual subcutaneous administration, and potential use with impaired renal function. The known adverse reactions are musculoskeletal pain, increased infections with adverse dermatologic reactions, osteonecrosis of the jaw, hypersensitivity reaction, and hypocalcemia. Treatment with 60 mg of denosumab reduces the bone resorption marker, serum type 1 C-telopeptide, by 3 days, with maximum reduction occurring by 1 month. The mean time to maximum denosumab concentration is 10 days with a mean half-life of 25.4 days. In conclusion, the convenient biannual subcutaneous administration of 60 mg of denosumab can be considered as a first-line treatment for osteoporosis in cases of low compliance with BPs due to gastrointestinal trouble and impaired renal function.

Sensitivity of BOD Sensor with Heavy Metal Tolerant Serratia marcescens LSY4 (Serratia marcescens LSY4 중금속 내성주를 이용한 BOD센서의 감응도)

  • A BOD sensor was prepared with S. marcescens LSY4 and was applied for measurement of BOD values of a solution containing the standard organic pollutants. The sensor sensitivity was nearly independent of the culture time in the range of 9-16 hours. It was also affected little by the cell mass in the range of 0.22-0.75 mg $cm^{-2}$. A cyclic change in the solution pH in the range of 4-9 was accompanied by a reversible variation in the sensor sensitivity. However, the reversibility was lost when the solution pH became more acidic or more basic. Heavy metal ions lowered the sensor sensitivity, which took place more precipitously in the presence of $Cu^{2+}$ and $Ag^+$ rather than in the presence of $Zn^{2+}$ and $Cd^{2+}$. The reduction of the sensor sensitivity was significantly attenuated by loading heavy metal ion tolerance induced strain. The $Cu^{2+}$tolerance induced strain was more efficient for the attenuation than $Zn^{2+}$ and $Cd^{2+}$ tolerance induced strain.

Fiber-optic Goniometer to Measure Knee Joint Angle for the Diagnosis of Gait Disturbance (보행장애 진단을 위한 무릎관절 각도 측정용 광섬유 각도센서)

  • In this study, we developed a fiber-optic goniometer for the continuous measurement of knee joint angle which provides important medical information on Alzheimer's disease. The fiber-optic goniometer is composed of a light-emitting diode (LED), a plastic optical fiber (POF), and a voltage output photodiode (PD). As a sensing part of the fiber-optic goniometer, a unclad fiber with regular intervals of 1 mm was fabricated to improve efficiency of bending loss according to the angle variation of knee joint. The emitting light with a center wavelength of 470 nm from a LED is guided by a POF to the PD, the transmitted light is then attenuated by the bending loss inside the bent POF. The intensity variation of the light transmitted from the POF gives rise to a change in output voltage in the fiber-optic goniometer. Therefore, we measured the real-time output voltage of the proposed fiber-optic goniometer using the unclad fiber according to the knee joint angle. Through the repeated experiments, the fiber-optic goniometer shows that it has a reversibility and a wide measurable angle range.

High-Sensitive Fiber-Optic pH Sensor Using Neutral Red Immobilized in Porous Sol-Gel Film (뉴트럴레드가 고정화된 다공성 졸-겔 필름을 이용한 고감도 광섬유 pH 센서의 특성)

  • In this study, a fiber-optic pH sensor based on a pH sol-gel film is fabricated. The sol-gel film is made by co-polymerizing tetramethoxysilane, trimethoxymethylsilane, ethanol and distilled water. As a pH indicator, a neutral red is immobilized in a thin porous film formed by the sol-gel process. The pH change in a sensing probe gives rise to a change in the color of the pH sol-gel film, and the absorbance of reflected light through the pH sol-gel film is also changed. By using a spectrometer, therefore, the spectra of reflected lights in the sensing probe with different pH values are measured. Also, the relationships between the pH values and the absorbance are analyzed on the basis of the color variations of the pH sol-gel films. In repeated experiments, the fiber-optic pH sensor shows that it has reversibility, a high reproducibility and a wide absorbance change in a pH range from pH 5 to 9. Also, we confirmed that the fabricated pH sol-gel film exhibits a fast response time, little or no pH indicator leaching and a dynamic range of 2.04 dB from pH 5 to 9. Based on the results of this study, a fiber-optic pH sensor can be developed for the pH monitoring in the harsh environments.

A Study on the Development of Poly-Urea for Porcelain Restoration (폴리우레아 도자기 복원 재료의 적용)

  • This study synthesized poly-urea and used it as a filler material for the restoration of porcelain. The synthesized poly-urea was manufactured as a resin and hardener mix that does not undergo contraction during curing, and is unfading. Given an adhesion strength of $180kg/cm^2$ and shearing strength of $200kg/cm^2$, the synthesized poly-urea exhibited the same efficacy as the epoxy putty currently sold in the market. Moreover, it also overcame the drawback of foaming encountered by urethane restoratives, which are a structurally similar type. The hardening time and pot life could also be controlled using additives. The poly-urea used for the restoration of modern artifacts presented a pot life of approximately one hour and took 12 h for complete hardening ($T_{90}$). When a $2{\times}2{\times}2cm$-sized test sample was added to xylene, poly-urea started to separate approximately two hours later and completed perfect pulverization within the solution 24 h later, demonstrating its reversibility. When directly applied to contemporary artifacts, it demonstrated the potential for restoration, as well as convenience and colorfulness.

Differentiation of Sorptive Bindings of Some Radionuclides with Sequential Chemical Extractions in Sandstones (순차적화학추출법을 사용한 방사성핵종의 사암에 대한 수착유형 평가)

  • Sorption experiments of $^{60}$ Co, $^{85}$ Sr. and $^{137}$ Cs onto sandstone particles in a batch were carried out to investigate the migration mobility. Sorption kinetics and reversibility as well as sorption mechanisms were examined. Sorption reaction occurred mostly within 10 hours on the outer surface of the sandstone particle but diffusion into the inner surface of the mineral has still occurred after that time. In order to distinguish sorption types of radionuclides, a sequential chemical extraction was introduced. The sorbed radionuclides were then extracted by applying different solutions of synthetic groundwater, CaCl$_2$, KCl and KOX-HA Especially KCl is adopted to extract the ion-exchanged cesium. Sorption types considered are reversible sorption under groundwater condition, ion exchange, association with ferro-manganese oxides or oxyhydroxides, and irreversible fixation. Strontium sorbs onto the sandstone surface mainly by fast and reversible ion exchange reaction. However, cobalt and cesium do not sorb by simple process. The main sorptive binding of cobalt was the association with ferro-manganese oxides and the secondary one was irreversible fixation. Diffusion into the lattice of minerals controlled the sorption rate of cobalt The main sorptin type of cesium was irreversible fixation, while ion exchange reaction was the secondary importance. Hence the oreder of migration mobility for the three radionuclides was Sr$^{2+}$ > Co$^{2+}$ > Cs$^{+}$ in the sandstones.
