The Effect of Photomodulation in Human Dermal Fibroblasts (피부 섬유아세포에서 광자극의 효과)

  • Skin is exposed to sunlight or artificial indoor light on a daily. The reached solar light on the earth surface consist of 50% visible light and 45% infrared (IR) except for ultra violet (UV). The negative effects of UV including UVB and UVA have been steadily investigated within the last decades. However, little is known about the effects of visible or IR light. In this study, we irradiated human dermal fibroblasts using light emitting diode (LED) to investigate the optimal parameter for enhancing cell growth and collagen synthesis. We found that red of 630 nm and green of 520 nm enhance the cell proliferation, but irradiation with purple and blue light exerts toxic effects. To examine the response of irradiation time and light intensity on the fibroblasts, cells were exposed to red or green light with intensities from 0.05 to $0.75mW/cm^2$. Procollagen secretion was increased of 1.4 fold by 10 min irradiation, while 30 min treatment decreased the collagen synthesis of dermal fibroblasts. Treatment with red of $0.3mW/cm^2$ and green of 0.15 and $0.3mW/cm^2$ resulted in enhancement of collagen mRNA. Lastly, we investigated the combinatorial effect of red and green light on dermal fibroblasts. The sequential irradiation of red and green light is an efficient way for the purpose of the increase in the number of fibroblasts than single light treatment. On the other hand, the exposure of red light alone was more effective method for enhancing of collagen secretion. Our study showed that specific light parameters accelerated cell proliferation, gene expression and collagen secretion on human dermal fibroblasts. In conclusion, we demonstrate that light exposure with specific parameter has beneficial effects on the function of dermal fibroblasts, and suggests the possibility of its cosmetically and clinical application.

Preliminary Research on the Effect of Cosmetic Containing Ginseng Extract on Quality of Life of Healthy Women Based on Skindex-16 (인삼 추출물 함유 한방화장품이 건강한 성인 여성의 삶의 질에 미치는 영향에 관한 예비 연구; Skindex-16을 중심으로)

  • This study is designed to analyze the effect of skincare using cosmetic containing ginseng extract, on improving quality of life (QOL) of healthy women, with blind testing. QOL is a concept that represents how one's disease or health condition can physically, psychologically, and socially influence his or her daily life. The study was conducted to assess the effect of a ginseng cosmetic preparation on quality of life (QOL) using the Skindex-16 score, stratified by blind versus non-blinded option. 45 healthy women aged between 30 and 49 years with no skin disease were recruited for this study. Volunteers were divided into two groups. Group A (n = 22) received anti-aging cream with ginseng extract in the original packaging, which included the brand name and logo. Group B (n = 23) received the same cream in a plain white jar without any package decoration or logo. Both groups used the cream for 8 weeks. For the skin-related QOL assessment, Skindex-16 was used at baseline, forth, and eighth week. All volunteers except two dropouts in Group A completed the dermatology-specific QOL measure, Skindex-16, at baseline, after 4 weeks, and after 8 weeks of treatment with the provided samples. As a result, the mean score of 43 participants at baseline was $22.70{\pm}4.82$. There was a significant difference between the baseline score and the score after 8 weeks in both groups: The scores changed from $23.30{\pm}5.14$ to $20.20{\pm}4.83$ in Group A, from $22.17{\pm}4.58$ to $20.52{\pm}3.60$ in Group B. The "Symptom" subscale of Skindex-16 improved after 4 weeks and the "Emotion" subscale improved after 8 weeks in Group A. The "Function" subscale did not show improvement in either groups. Both groups showed no interaction effect between follow up time and groups in Skindex-16 and subscale. This research opens up the possibility of skincare using ginseng cream having a positive effect on QOL in healthy women. Moreover, one can predict that skincare ritual itself may have greater impact on the improvement of QOL, compared to the product packaging.

Component Analysis of Suaeda asparagoides Extracts (나문재 추출물의 성분 분석)

  • In the previous study, the anti-oxidant activity of oxtract/fraction of Sueada aspparagoides(SA) and the stability test for the cream containing SA extract were investigated respectively[1,2]. In this study, the components of SA extract were analyzed by TLC, HPLC, and LC/ESI-MS/MS, $^1H$-NMR. TLC chromatogram of ethyl acetate fraction of SA extract revealed 5 bands $(SA1{\sim}SA5)$. HPLC chromatogram of aglycone fractions obtained from deglycoylation reaction of ethyl acetate fraction showed 2 bands (SAA 2 and SAA 1), which were identified as quercetin (composition ratio, 16.88%) and kaempferol (83.12%) in the order of elution time. Among 5 bands of TLC chromatogram, 4 bands $(SA2{\sim}SA5)$ also were Identified as kaempferol-3-O-glucoside (SA 2), quercetin-3-O-glucoside (SA3), kaempferol-3-O-rutinoside (SA 4), quercetin-3-O-rutinoside (SA 5) by LC/ESI-MS/MSMS/MS. respectively. The spectrum generated for SAA 1 by LC/ESI-MS/MS in the negative ion mode also gave the ion corresponding to the deprotonated aglycone $[M-H]^-$ (285m/z), the $^1H$-NMR spectrum contained signals [${\delta}$ 6.19 (1H, d, J=1.8Hz, H-6), ${\delta}$ 6.44 (1H, d, J=1.8Hz, H-8), ${\delta}$ 6.92 (2H, d, J=9.0Hz, H-3', 5'), ${\delta}$ 8.04 (2H, d, J=9.0Hz, H-2', 6', thus SAA 1 was identified as kaempferol. SAA 2 yielded the deprotonated agycone ion $[M-H]^-$ (301m/z), $^1H$-NMR spectrum showed signals [${\delta}$ 6.20 (1H, d, J=2.0Hz, H-6), ${\delta}$ 6.42 (1H, d, J=2.0Hz, H-8), ${\delta}$ 6.90 (1H, d, J=8.6Hz, H-5'), ${\delta}$ 7.55 (1H, dd, J=8.6, 2.2Hz, H-6'), ${\delta}$ 7.69 (1H, d, J=2.2Hz, H-2', thus SAA 2 was Identified as quercetin. In conclusion, with the anti-oxidant activity and the stability test reported previously, component analysis of SA extracts could be applicable to new cosmeceuticals.

  • Objectives: This study was carried out to analyze and understand the trends of research papers published In the Journal of Oriental Medical Surgery?phthalmology & Otolaryngology Society(JOMSOOS). Methods: We studied the 308 research papers that had been published in JOMSOOS from 1988 Vol. 1. No. 1. to 2002 Vol. 15. No. 2. Our study was developed through the four stages in order to analyze the papers; 1) we analyzed all the papers overall to classify them into three categories; original article. review article and case report. 2) we classified the original articles in terms of methodology. 3) we also analyzed the case reports according to the sort of disease each paper dealt with. 4) we had another statistical approach to each paper to figure out the distribution of diagnoses in detail. Results: We have got the following outcomes from our analysis of the papers in terms of the four stages. 1. Overall Analysis. 1) Classification of 308 research papers between 1988 and 2002: 137 original articles(44.48$\%$), 111 review articles(36.04$\%$), 56 case reports(18.18$\%$). 2) Used language: Korean(99.03$\%$). English(0.97$\%$). 3) The Number of Authors: 2 persons(42.86$\%$). 3 persons(29.87$\%$), 1 person(14.61$\%$). 2. Original Article Analysis 1) Classification of 137 original articles in terms of methodology: 90 experimental studies(65.69$\%$)46 descriptive studies(33.58$\%$), 1 analytic study(0.73$\%$). 2) Classification of the original articles according to the use of statistical methods: No statistical methods(36.42$\%$), Descriptive methods only(1.99$\%$), Not defined(23.18$\%$), t-test(24.50$\%$), ANOVA(3.97$\%$), Multiple comparison(2.65$\%$), Non-parametric test(2.65$\%$), Other methods(1.32$\%$). 3) Classification of 46 descriptive articles in terms of diseases: otorhinolaryngology(43.48$\%$), dermatology(23.91$\%$), ophthalmology(13.04$\%$), facial palsy(13.04$\%$). 4) Classification of descriptive articles in terms of the number of patients: the highest was 'more than 26 but less than 50 persons'(19 articles - 41.30$\%$). 5) Classification of descriptive articles in terms of the period for patients observation: the highest was the time 'more than 9 but less than 12 months(34.78$\%$)' Out of the 34.78$\%$, the number of articles with the patients observed for more than 12 months was 13(28.26$\%$). 3. Case Report Analysis 1) Classification of 56 case reports in terms of the sort of disease: dermatology(44.64$\%$), ophthalmology(19.64$\%$), otorhinolaryngology(14.29$\%$), facial palsy(8.93$\%$). 2) Classification in terms of the number of patients: 1 person(50$\%$), 3 persons(16.07$\%$), 2 persons(14.29$\%$). 4. Diagnosis Distribution of Each Disease. 1) Studies regarding ophthalmology : the percentage of 'strabismus' cases was the highest(33.33$\%$). 2) Studies regarding otorhinolaryngology : nasal inflammation(34.48$\%$), tinnitus(20.69$\%$). 3) Studies regarding dermatology: the percentage of 'allergic skin disease' was the highest(33.33$\%$). Conclusions: We analyzed the trends of research papers that have been published in JOMSOOS in detail. We came to understand the trends of the research through this study. However, we acknowledge that we only adopted the quantitative method out of various possible analysis methods. For further studies, we strongly urge to adopt the qualitative methods as well.

Among members ofcitizen movement groups,government workers,and students who are living inInchon were selected as 613 samples using a purposive sampling method. Thermal history of the Jecheon granite pluton in the Ogcheon Fold Belt, South Korea (남한의 옥천습곡대에 분포되어 있는 제천화강암체의 열역사)

  • Whole rock and mineral ages for the Jecheon Granite distributed in the Ogcheon Fold Belt were dated by three radiometric methods, and its thermal history was elucidated as follows, on the basis of isotopic age data. Rb and Sr isotopic compositions of three whole rock and seven mineral concentrates made an isochron of 202.7${\pm}$ 1.9 Ma with a strontium initial ratio of 0.7140. Different age data of twelve mineral concentrates agree closely with the retention temperature of each mineral in K-Ar and Fission Track methods. The Jecheon granitic magma was generated by partial melting of crustal materials (S-type), or by mixins between mantle and crustal materials, intruded into the katazone or mesozone (7∼9 km) of the Ogcheon Fold Belt, at least in the Early Jurassic (about 203 Ma), and then crystallized and cooled down rapidly from about 600$^{\circ}C$ to 300$^{\circ}C$ (more than 20$^{\circ}C$/Ma), owing to thermal differences between the magma and the wall-rock. During the Middle to Late Jurassic (190∼140 Ma), the cooling of the granite was likely to stop and keep thermal equilibrium with the wall-rock. The severe tectonism associated with igneous activities and active weathering on the surface in Early to Late Cretaceous time (140∼70 Ma) might have accelerated the granite pluton to uplift rapidly (40∼60 m/Ma in average) up to 3∼4 km and cooled down from 300$^{\circ}C$ to 200$^{\circ}C$ (1.4 $^{\circ}C$/Ma). The granite pluton was likely to keep different uplifting and cooling rate of about 120 m/Ma and 5$^{\circ}C$/Ma in average from the Late Cretaceous to Early Tertiary (70∼50 Ma), and about 60 m/Ma and 2$^{\circ}C$/Ma in average from about 50 Ma up to the present, respectively.

(U-Th)/He Dating: Principles and Applications ((U-Th)/He 연령측정법의 원리와 응용)

  • The (U-Th)/He dating utilizes the production of alpha particles ($^4He$ atoms) during natural radioactive decays of $^{238}U$, $^{235}U$ and $^{232}Th$. (U-Th)/He age can be determined from the abundances of the parent nuclides $^{238}U$, $^{235}U$ and $^{232}Th$ and the radiogenic $^4He$. Because helium is one of the noble gases (non-reactive) with a relatively small radius, it diffuses rapidly in many geological materials, even at low temperatures. Therefore, ingrowth of $^4He$ during radioactive decay competes with diffusive loss at elevated temperatures during the geologic time scale, determining the amount of $^4He$ existing today in natural samples. For example, He diffusion in apatite is known to be very rapid compared to that in most other minerals, causing a significant diffusive loss at ${\sim}80^{\circ}C$ or higher. At ${\sim}40^{\circ}C$, He diffusion in apatite becomes slow enough to preserve most $^4He$ in the sample. Thus, an apatite's (U-Th)/He age represents the timing when the sample passed through the temperature range of $80-40^{\circ}C$. The crustal depth corresponding to this temperature range is called a "partial retention zone." Normal closure temperatures for a typical grain size and cooling rate are ${\sim}60-70^{\circ}C$ for apatite and ${\sim}200^{\circ}C$ for zircon and titanite. Because the apatite He closure temperature is lower than that of most other thermochronometers, it can provide critical constraints on relatively recent or shallow-crustal exhumation histories.

Internal Structure and Movement History of the Keumwang Fault (금왕단층의 내부구조 및 단층발달사)

  • Detailed mapping along the Keumwang fault reveals a complex history of multiple brittle reactivations following late Jurassic and early Cretaceous ductile shearing. The fault core consists of a 10~50 m thick fault gouge layer bounded by a 30~100 m thick damaged zone. The Pre-cambrian gneiss and Jurassic granite underwent at least six distinct stages of fault movements based on deformation environment, time and mechanism. Each stage characterized by fault kinematics and dynamics at different deformation environment. Stage 1 generated mylonite series along the Keumwang shear zone by sinistral ductile shearing during late Jurassic and early Cretaceous. Stage 2 was a mostly brittle event generating cataclasite series superimposed on the mylonite series of the Keumwang shear zone. The roundness of pophyroclastes and the amount of matrix increase from host rocks to ultracataclasite indicating stronger cataclastic flow toward the fault core. At stage 3, fault gouge layer superimposed on the cataclasite generated during stage 2 and the sedimentary basins (Umsung and Pungam) formed along the fault by sinistral strike-slip movement. Fragments of older cataclasite suspended in the fault gouge suggest extensive reworking of fault rocks at brittle deformation environments. At stage 4, systematic en-echelon folds, joints and faults were formed in the sedimentary basins by sinistral strike-slip reactivation of the Keumwang fault. Most of the shearing is accommodated by slip along foliations and on discrete shear surfaces, while shear deformation tends to be relatively uniformly distributed within the fault damage zone developed in the mudrocks in the sedimentary basins. Fine-grained andesitic rocks intruded during stage 4. Stage 5 dextral strike-slip activity produced shear planes and bands in the andesitic rocks. ESR(Electron Spin Resonance) dates of fault gouge show temporal clustering within active period and migrating along the strike of the Keumwang fault during the stage 6 at the Quaternary period.

The Qualities of Northern and Southern Ecotype Garlic Bulbs at Different Storage Temperature (한지형' 및 '난지형' 마늘의 저장온도에 따른 품질 특성)

  • This study was conducted to investigate the optimal sub-zero temperature for storage of two garlic cultivars, the northern ecotype 'Jaerae' and the southern ecotype 'Daeseo'. These cultivars of garlic bulbs were stored at different temperature (room temperature, $0^{\circ}C$, and $-4^{\circ}C$) conditions after predrying. Southern ecotype garlic showed higher weight loss than northern ecotype garlic. Northern ecotype garlic at $-4^{\circ}C$ exhibited less than 5% of weight loss during 10 month storage. Sprouting rate was higher in southern than in northern ecotype at room temperature storage. The value of injury by disease and insect was similar between southern ecotype and northern ecotype for 5 month storage, and then northern ecotype showed higher value of injury by disease and insect than southern ecotype. Hunter 'b' value of northern ecotype garlic was the lowest at $-4^{\circ}C$ storage. Northern ecotype garlic showed no cold injury at freezing point temperature storage. However, southern ecotype garlic had $0{\sim}5.3%$ cold injury occurrence at $-4^{\circ}C$ storage. Respiration and ethylene production exhibited the highest at room temperature storage, those had the slight increase at $0^{\circ}C$ and $-4^{\circ}C$ storages. Northern ecotype showed higher enzymatic pyruvic acid and fructan contents than those of southern ecotype. Enzymatic pyruvic acid content increased and fructan contents decreased during storage time.

Monitoring on Physicochemical Properties of Liriope platyphylla by the Use of Four Dimensional Response Surface (4차원 반응표면분석을 통한 맥문동의 이화학적 특성 모니터링)

  • Four dimensional response surface methodology was used to monitor the extraction conditions and predict the optimum extraction conditions on physicochemical properties of Liriope platyphylla. Maximum yield of total soluble solid was 66.02% into range of 35.06~65.70%, and maximum extraction conditions were 16.86 mL/g in ratio of solvent to sample, $99.55^{\circ}C$ in extraction temperature and 3.20 hr in extraction time. Maximum extraction conditions of total phenolics were 18.78 mL/g, $97.09^{\circ}C$ and 3.71 hr. Maximum content of crude saponin was 6.51% into range of 2.22~6.21 %, and maximum extraction conditions were 21.33 mL/g, $95.49^{\circ}C$ and 3.00 hr. Maximum content of reducing sugar was 6.75% into range of 2.43~6.51%, and maximum extraction conditions were 22.93 mL/g, $89.64^{\circ}C$ and 3.75 hr. Electron donating ability was maximized in 16.74 mL/g, $99.63^{\circ}C$ and 3.16 hr. The range of optimum conditions gained by the superimposed four dimensional response surfaces on total soluble solid, crude saponin and reducing sugar of Liriope platyphylla was 15~23 mL/g, 92~$100^{\circ}C$ and 2.4~5.0 hr. And total soluble solid, total phenolics, crude saponin, reducing sugar, browning color intensity and electron donating ability at the given conditions(20 mL/g, $100^{\circ}C$, 3 hr) within the range of optimum conditions were 65.75%, 1.30 mg/g, 6.33%, 5.93%, 0.11 and 10.52%, respectively.
