Gene therapy with nonviral vectors using the suicide gene/prodrug activating system of herpes simplex virus type-1 thymidine kinase (HSV1-TK)/ganciclovir (GCV) is inefficient in killing malignant tumor cells due to two major factors: (a) an unsatisfactory bystander effect; (b) short-lived expression of the protein. To study the capacity of the protein transduction domain (PTD) of HIV-1 TAT protein to enhance HSV1-TK/GCV cancer gene therapy, we constructed three fusion proteins TAT-TK, TK-TAT and TK. TAT-TK retained as much enzyme activity as TK, whereas that of TK-TAT was much lower. TAT-TK can enter HepG2 cells and much of it is translocated to the nucleus. The transduced HepG2 cells are killed by exogenously added GCV and have bystander effects on untransduced HepG2 cells. Most importantly, the introduced recombinant protein is stable and remains functional for several days at least, probably because nuclear localization protects it from the cytoplasmic degradation machinery and provides access to the nuclear transcription machinery. Our results indicate that TAT fusion proteins traffic intercellularly and have enhanced stability and prodrug cell killing activity. We conclude that TAT has potential for enhancing enzyme prodrug treatment of liver cancers.
연구배경 : HSVtk 유전자를 암세포에 이입하여 GCV에 대해 선택적으로 감수성을 증가시키는 HSVtk/GCV 유전자치료에서 bystander effect는 모든 암세포에 유전자를 이입하지 않고도 치료효과를 얻을 수 있도록 한다. 그러나 현존하는 viral vector는 유전자이입 효율이 낮아 임상적으로 치료효과를 기대하는데는 한계가 있다. 따라서 유전자의 이입효율이 높은 새로운 vector의 개발과 함께 bystander effect의 극대화를 통해 치료효과를 증가시킬 수 있는 방법 등이 요구된다. 최근 gap junction을 통한 세포간의 metablic cooperation이 bystander effect의 주요기전임이 밝혀졌고 retinoids는 gap junction을 통한 세포간의 communication을 증가시킨다고 보고되었다. 저자들은 HSVtk/GCV 유전자치료에서 bystander effect에 미치는 retinoids의 효과를 조사하였다. 방 법 : Adenovus와 retrovirus vector를 이용하여 connexin 43를 발현하는 악성중피종세포와 connexin 43를 발현하지 않는 SKHep-J 세포주에 HSVtk 유전자를 이입한 후 HSVtk 유전자가 이입된 세포와 HSVtk 유전자가 이입되지 않은 세포들을 여러가지 비율로 혼합한 mixing study를 시행하였으며 $10^{-10}M-10^{-6}M$ RA 처리 유무에 따른 bystander effect에 의한 살상효과를 비교하였다. 그리고 gap junction을 통한 세포간의 communication에 대한 retinoids의 영향을 조사하기 위해 retinoids 처리에 따른 세포간 communication을 FACS를 이용하여 double-dye transfer study로 측정하였다. 결 과 : Connexin 43를 발현하지 않는 SKHep-J 세포주에서는 RA 처리유무에 따른 bystander effect에 의한 살상효과의 차이가 없었다. 그러나 connexin 43를 발현하는 악성중피종 세포주에서는 $10^{-8}M-10^{-6}M$ RA처리시 세포간의 communication과 bystander effect가 RA를 처리하지 않은 대조군에 비해 유의하게 증가되었다. 결 론 : RA는 gap junction을 통한 세포간의 communication을 증가시켜 HSVtk/GCV 유전자치료에서 bystander effect에 의한 살상효과를 증가시켰다. 이러한 결과는 HSVtk/GCV 유전자치료의 효과를 증가시킬 수 있는 새로운 방법이 될 수 있을 것으로 생각된다.
연구배경: 유전자치료에 이용되는 adenovirus를 기초로 하는 벡타는 상피세포에 강한 친화력이 있고 증식하지 않는 세포에도 유전자의 이입이 가능하며, in vivo에서 다른 백타에 비해 유전자이입 효율이 높은 장점이 있다. 그러나 adenovirus에 이해 전달된 유전자는 세포의 chromosome에 삽입되지 않기 때문에 세포가 분열하면 딸세포에 형질이 전달되지 않을 뿐 아니라 adenovirus가 숙주의 면역반응을 초래하여 이입된 유전자가 제거됨에 따라 전달된 유전자의 발현기간이 짧은 단점이 있다. 포화지방산인 butyrate는 histone의 acetylation을 증가시켜 전달된 유전자의 발현을 증가시킨다고 한다. 저자들은 유전자치료시 butyrate를 병용투여 할 경우 이입한 유전자의 발현이 증가되고, 따라서 유전자치료의 효과를 증가시킬 수 있을 것이라는 가정하에 butyrate가 herpes simplex virus thymidine kinase 유전자를 이용한 유전자치료에 미치는 영향을 조사하였다. 방 법: 악성 중피종세포인 REN세포에 Ad.CMVtk를 이입한 후 세포들을 3군으로 분류하였다. 1 군은 butyrate를 처리하지 않고 7일간 배양하였으며, 2군은 이입 후 1일째에 그리고 3군은 이입 후 2일째 1.5mM/L butyrate를 함유하는 배지에서 24시간 동안 배양한 후 butyrate가 함유되지 않은 배지로 교환하여 배양하였다. 유전자 이입 후 2일, 3일, 그리고 7일에 Western blotting을 시행하여 유전자의 발현정도를 조사하였으며, butyrate 처리 유무에 따른 bystander effect에 의한 살상효과의 차이를 조사하였다. 결 과: Butyrate를 처리하지 않은 대조군은 유전자이입 후 7일째에 유전자의 발현이 없었으나 butyrate를 처리한 군은 7일째에도 유전자의 발현이 있었다. 그리고 butyrate를 처리한 경우 bystander effect에 의한 살상효과가 유의하게 증가되었다. 결 론: 이상의 결과로 butyrate는 유전자의 발현을 증가시킴에 의해 bystander effect를 증가시킴을 알 수 있었으며, 향 후 adenovirus를 이장한 유전자치료의 효과를 증가시킬 수 있는 새로운 방법이 될 수 있을 것으로 생각된다.
연구배경: HSVtk/GCV를 이용한 유전자치료에서 면역반응은 1) adenovirus 혹은 retrovirus와 같이 벡타로 사용된 virus의 단백질, 2) 치료목적으로 이입된 HSVtk 유전자의 생성물, 3) 암세포에 대해서 일어날 수 있다. 그리고 이러한 면역반응은 cytokines의 생성 혹은 cytotoxic tumor-specific T-cell의 생성을 초래하여 bystander effect에 의한 살상효과를 증가시키거나, anti-tumor immunity를 유도하여 tumor vaccine의 효과를 나타낼 수 있다. 한편 이와는 대조적으로 면역반응용 HSVtk 유전자를 발현하는 세포들을 파괴하여 이입된 HSVtk 유전자의 발현기간을 제한함으로서 유전자치료의 효과를 감소시킬 수도 있다. 본 연구는 retrovirus 벡타로 이입한 HSVtk 유전자치료에서 면역체계가 bystander effect에 의한 살상효과에 미치는 영향을 규명하고 면역체계가 이입한 유전자의 발현에 미치는 영향을 조사하고자 하였다. 방 법: Immunocompetent mice인 Balb/c mouse와 immunodeficient mouse인 Balb/c-nude 및 SCID mouse에서 retrovirus 벡타를 사용하여 HSVtk 유전자를 이입하고 치료효과를 조사하였다. 그리고 Balb/c mouse에 면역억제제인 cyclosporin을 투여하여 면역억제제가 bystander effect 및 유전자치료 효과와 유전자의 발현기간에 미치는 영향을 조사하였다. 결 과: Balb/c mouse에 HSVtk 유전자를 이입하고 GCV를 투여한 군은 GCV를 투여하지 않은 대조군에 비해 종양의 성장이 유의하게 억제되었으나 Balb/c-nude mouse와 SCID mouse의 경우 GCV를 투여한 군과 대조군 사이에 유의한 차이가 없었다. 면역억제제인 cyclosporin을 투여한 군에서 유전자 치료 효과가 cyclosporin을 투여하지 않은 정상 mouse에 비해 치료효과가 유의하게 작았다. Cyclosporin 투여에 따른 유전자의 발현기간에는 유의한 차이가 없었다. 결 론: Retrovirus 벡타를 사용한 HSVtk 유전자치료에는 면역증강이 치료효과를 증가시킬 것으로 생각된다.
목적: 단순 헤르페스 제 1형 티미딘 키나제(herpes simplex virus type 1 thymidine kinase gene: HSV1-tk)는 GCV와 함께 유전자치료의 한 방법으로서 가장 활발하게 연구되어왔으며, HSV1-tk 효소에 대한 다양한 기질들이 연구되어서 이를 보고 기질로 한 비침습적인 HSV1-tk 유전자 영상시스템에서 가장 널리 사용되고 있다. 본 연구에서는 보고기질로서 방사성 요오드가 표지된 5-iodovinyl-2-deoxyuridine (IVDU) and 5 -iodovinyl-2-fluoro-2-deoxyuridine (IVFRU)를 보고 기질로 하여 HSV1-tk 유전자 영상시스템에서의 유용성을 확인하고자 하였다. 대상 및 방법 HSV1-tk 유전자 영상을 위하여 HSV1-tk 유전자를 레트로 바이러스 벡터를 이용하여 Morris hepatoma 세포주(MCA-tk)에 이입한 세포주를 제조하였으며, HSV1-tk 유전자의 발현을 확인하기 위하여 Northern blotting과 Western Blotting을 시행하였다. 대조세포주인 MCA와 제조된 MCA-tk 세포주에 방사표지 IVDU와 IVFRU를 이용하여 480분까지 세포내 섭취율을 평가하였으며, 또한 MCA-tk 세포주의 백분율을 증가시키면서 이에 따른 IVDU와 IVFRU의 섭취율을 평가함으로서 섭취율과 세포수와의 상관관계를 평가하였다. 결과: MCA-tk 세포주에서 HSV1-tk의 mRNA의 발현과 HSV1-TK 단백질의 발현을 확인하였다. 방사성 요오드 표지 IVDU와 IVFRU 모두는 MCA 세포주에서는 아주 낮은 섭취율을 나타내었으며, MCA-tk 세포주에서는 모두 증가된 섭취를 보였다. IVDU가 480분에서 IVFRU보다 4배 높은 섭취를 나타내었다(p<0.01). 방사표지 IVDU와 IVFRU 모두에서 MCA-tk의 백분율의 증가에 따라서 직선적인 상관관계($R^2>0.96$)를 나타내었다. 결론: 방사성 요오드 표지 IVDU와 IVFRU는 HSV1-tk유전자가 이입된 간암세포주에서 모두 특이적인 높은 섭취율을 나타내고 직선적인 상관관계가 나타나서 두 기질 모두 HSV1-tk유전자 영상시스템에서 보고 기질로서 유용하게 사용될 수 있을것으로 기대된다.
목 적: 본 연구를 통해 GnRH 의한 세포 분열의 억제는 integrin, FAK 빛 ERK를 통한 세포 내 신호전달 기전을 통하여 일어남을 규명하고자 하였다. 연구방법: 연구에 사용된 인간자궁내막암 세포주는 DMEM/F12 (10% FBS)의 조건에서 배양 하였다. GnRH-I과 -II는 실험 목적에 따라 100 nM 농도로 0, 5, 10, 15, 20, 30분간 또는 10 nM or 100 nM의 농도로 20분간 처리 하였다. 세포의 분열 정도는 [$^3H$] thymidine incorporation assay를 이용하여 정량적으로 측정 하였으며, Immunoblotting 방법을 이용하여 단백질의 발현을 확인 하였다. 결 과: GnRH-I과 -II 모두 HEC1A 세포의 세보분열을 억제하였으며 integrin ${\beta}3$의 발현을 증가 시켰다. GnRH-I과 -II를 처리 후 FAK 및 ERK의 안산화가 증가됨을 관찰할 수 있었다. 결 론: GnRH에 의한 세포분열의 억제는 integrin의 발현과 FAK 및 ERK의 인산화 과정을 통하여 일어남을 알 수 있었다.
목적: 만성 골수성 백혈병 세포인 K562 세포주는 방사선 및 다양한 항암제에 대한 apoptosis에 저항성을 가진다. 지난 연구에서 K562 세포는 방사선에 대하여 내성반응을 보이며, 세포내 PTK의 작용을 억제하고자 방사선 조사와 함께 투여한 herbimycin A (HMA)에 의하여 방사선에 대한 apoptosis와 같은 감수성반응이 유도되는 반면, genistein에 의하여 방사선에 대한 apoptosis 반응이 저해됨을 확인하였다. 본 연구에서는 타이로신 인산화효소 억제에 의한 K562 세포의 방사선 반응변화를 조절하는 신호전달경로를 조사하였다. 대상 및 방법: K562 세포를 지수증식기의 세포들만 선택하여 실험에 이용하였다. 방사선조사는 6 MeV 선형가속기(Clinac 1800C, Varian)를 이 용하여 $200\~300$ cGy/min 선량률로 $0.5\~12 $ Gy를 균일하게 조사하였다. HMA와 genistein은 각각 $0.25/muM,\;25\muM$을 방사선 조사 후 즉시 투여하였다. 실험에서 신호전달 경로로 abl kinase, MAPK family, NF-kB, c-fos, c-myc, thymidine kinase1 (TK1) 등에서의 단백질 또는 유전자 발현 및 활성을 조사하였다. 또한 약제 투여에 따른 유전자 발현차이(differential gene expression)를 조사하였다. 결과: Abl kinase의 발현 및 활성 변화를 조사하였으나 PTK 저해제에 의한 방사선 유도 세포사의 변화와의 연관성을 찾을 수 없었다. 세포 생존 및 사멸의 신호전달체계에서 주요 조절과정인 MAPK family의 관여 여부 확인에서 방사선으로 인한 SAPK/JNK의 활성화의 유도가 관찰되었으나, PTK 저해제에 따른 변화는 없었으며, 또한 MAPK/ERK와 p38 MAPK 활성은 모든 조건에서 변함 없이 일정하였다. 전사인자 활성화에 대한 조사에서 방사선 조사와 함께 genistein을 투여한 경우에 NF-kB활성이 증가하였다. 유전자 발현 차이의 조사에서 genistein 투여에 의한 TK 1 유전자 발현 및 단백질 활성이 증가하였다. 결론: PTK 억제에 의한 K562 세포의 방사선에 대한 반응 변화는 bcrabl kinase 활성과는 무관하게 진행되며, MAPK family 경로 외의 다른 경로를 통한 전사인자 활성화 과정이 연관되어 있음을 확인하였다.
Objective : To characterize the antitumorigenic potential of Apamin, one of the major components of bee venom, its effects on cell proliferation and the mitogen-activated protein kinase (MAPK) signal transduction pathway were characterized using the human melanoma cell line SK-MEL-2. Methods & Results : Cell counting analysis for cell death demonstrated that consistent with a previous results, SK-MEL-2 cells treated with $0.5-2.0{\mu}g/ml$ of Apamin showed no recognizable cytotoxic effect whereas detectable induction of cell death was identified at concentrations over $5.0{\mu}g/ml$. [3H]thymidine incorporation assay for cell proliferation demonstrated that DNA replication of SK-MEL-2 cells is inhibited by Apamin in a dose- and time-dependent manner. To explore whether Apamin-induced growth suppression is associated with the MAPK signaling pathway, phosphorylation of Erk, a function mediator of MAPK growth-stimulating signal, was examined Western blot assay using a phospho-specific Erkl/2 antibody. A significant increase of Erkl/2 phosphorylation level was observed in Apamin-treated cells compared with untreated control cells. Qantitative RT-PCR analysis revealed that Apamin inhibit expression of MAPK downstream genes such as c-Jun, c-Fos, and cyclin D1 but not expression of MAPK pathway component genes including Ha-Ras, c-Raf-1, MEK1, and Erk. Conclusion : It is strongly suggested that the antitumorigenic activity of Apamin might result in part from its inhibitory effect on the MAPK signaling pathway in human melanoma cells SK-MEL-2.
Objectives: Hepatoma is a very serious disease in Korea and vvorldwiclc. Hepatitis B vims (HBV) has proved the most significant cause of hepatoma. We canied out this study to investigate the effect of Acanthopanax sessilifloms on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell line and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT/3T3-HBx ceIl. Methods: To investigate the anti-cancer effect of Acanthopanax sessiliflorus, we did the CellTiter 96 Aqueous Non-radioactive Cell Proliferation assay (Promega); MTS/PMS assay, [$^3H$]-thymicline incorporation assay, and we measured the gene expression through westem blotting. Results: Acanthopanax sessiliflorus showed an inhibiting effect on the increase of HepG2.2.15 in the NTS/PMS assay. It also showed an inhibiting effect on DNA synthesis of HepG2.2.15 in the [$^3H$]-thymidine incorporation assay. Acanthopanax sessiliflorus showed an inhibiting effect of phosphorilation of MAP kinase in HBV - X genes. too. Conclusions: The results suggested that this herb had an anti cancer effect. We may discover an effective anti-cancer herb medicine through further studies on this herb medicine.
Objective : Triptolide (TP) has been reported to suppress the expression of mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1), of which main function is to inactivate the extracellular signal-regulated kinase-1/2 (ERK-1/2), the p38 MAPK and the c-Jun N-terminal kinase-1/2 (JNK-1/2), and to exert antiproliferative and pro-apoptotic activities. However, the mechanisms underlying antiproliferative and pro-apoptotic activities of TP are not fully understood. The purpose of this study was to examine whether the down-regulation of MKP-1 expression by TP would account for antiproliferative activity of TP in immortalized HT22 hippocampal cells. Methods : MKP-1 expression and MAPK phosphorylation were analyzed by Western blot. Cell proliferation was assessed by $^3H$-thymidine incorporation. Small interfering RNA (siRNA) against MKP-1, vanadate (a phosphatase inhibitor), U0126 (a specific inhibitor for ERK-1/2), SB203580 (a specific inhibitor for p38 MAPK), and SP600125 (a specific inhibitor for JNK-1/2) were employed to evaluate a possible mechanism of antiproliferative action of TP. Results : At its non-cytotoxic dose, TP suppressed MKP-1 expression, reduced cell growth, and induced persistent ERK-1/2 activation. Similar growth inhibition and ERK-1/2 activation were observed when MKP-1 expression was blocked by MKP-1 siRNA and its activity was inhibited by vanadate. The antiproliferative effects of TP, MKP-1 siRNA, and vanadate were significantly abolished by U0126, but not by SB203580 or SP600125. Conclusion : Our findings suggest that TP inhibits the growth of immortalized HT22 hippocampal cells via persistent ERK-1/2 activation by suppressing MKP-1 expression. Additionally, this study provides evidence supporting that MKP-1 may play an important role in regulation of neuronal cell growth.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
