Effects of Reticuloendothelial Hyperfunction on Preservation of Lung (망내계기능 항진이 폐장보존에 미치는 영향)

  • The effect of reticuloendothelial hyperfunction on hypothermic preservation of lung was studied in dogs. In order to evaluate the viability after hemodynamic_ load in preserved isolated lung, observations were made on the rate of increase in weight, degree of edema,compliance and surface activity of lung. The results obtained as follows: l. In the group of activating of the reticuloendothelial system by injection of sodium thiosulfate intravenously before pneumonectomy and infusion of naphthionine through the pulmonary artery before hypothermic preservation of isolated lung the limit of preservation was eight hours whereas four hours in non-treated control group. 2.Therefore the method of activating of the reticuloendothelial system before and after pulmonary resection seems effective in preserving for prolonging the period of preservation of lung by means of inhibition of pulmonary edema. 3. Pulmonary surface activity is expected to be valuable as a method in evaluation of the viability of preserved lung along with compliance and rate of increase in weight of lung.

Isolation and Characterization of a Dibenzothiophene Degrading Sulfate-Reducing Soil Bacterium

  • Sulfate-reducing bacteria have been isolated from soil and their abilities to degrade dibenzothiophene (DBT) were compared with those of type cultures. Among the strains tested a soil isolate M6 showed the highest ability to degrade DBT. Isolate M6 was characterized as a mesophilic obligatory anaerobe. The morphology of the bacterium was vibrioid with the size of $0.4-0.7{\;}\mu\textrm{m}{\;}by{\;}1.0-1.5{\;}\mu\textrm{m}$. Gram reaction was negative and nonsporulating. Desulfoviridin is present. Lactate, pyruvate, ethanol and malate supported growth of the bacterium in the presence of sulfate. Sulfate, sulfite, thiosulfate and sulfur served as electron acceptors for growth. Hydrogenase was present. The mol% of guanine and cytosine of DNA was determined as 56%. The bacterium produced viscous material. From these results, the isolate M6 was identified as Desulfovibrio desulfuricans.

  • This study was carried out to simultaneous determination of penicillin G and chloramphenicol in livestock products by HPLC. The results obstained were as follows; 1. Penicillin G and chloramphenicol were analyzed by HPLC on symmetry $C_{18}$ column with acetonitrile-0.1 M phosphate buffer containing 0.0157 M thiosulfate (25 : 75) as mobile phase at UV 325nm and 280nm, respectively. 2. Samples were applied to a SeP-Pak $C_{18}$ cartridge, from which eluted penicillin derivatized with 2 M 1,2,4-triazole containing 0.001 M mercuric chloride. 3. The average recovery rates of penicillin G and chloramphenicol were 81.8% and 80.3%, respectively, and the detection limits were 5 ppb (5$\mu\textrm{g}$/kg: 7.9IU/kg) for penicillin G and chloramphenicol in porcine and bovine muscle.

Development and culture optimization of mutants of Thiobacillus sp. IW for elimination of hydrogen sulfide

  • UIW-10 mutant obtained by UV treatment using sulfur-oxidizing bacteria, Thiobacillus sp. IW was studied. The colony size of UIW-10 was found 2 $^{\sim}$ 3 times bigger in diameter than the parent colony on TAM medium. UIW-10 mutant growth was two times higher than parent strain at 6 h culture in liquid medium containing sulfides such as sulfur and sodium thiosulfate. Initial pH and temperature for the optimum growth of UIW-10 were 6.0 and $35-40^{\circ}C$, respectively. It was found that addition of 0.5% yeast extract and 0.5 to 2.0% tryptone as nitrogen sources and the constant agitation at 150 to 200 rpm had a positive effect and the growth of UIW-10 was increased.

Impact of Dissolved Wastewater Constituents on Laccase-Catalyzed Treatment of Bisphenol A

  • The impact of dissolved wastewater constituents on the treatment of synthetic bisphenol A (BPA) solutions was investigated under a variety of reaction conditions. The laccase enzyme from Trametes vesicolor was used for the BPA treatment. The constituents studied included various inorganic salts, organic compounds and heavy metal ions. BPA degradation was inhibited by sulfate, thiosulfate, sulfide, nitrite, and cyanide ions at 25 mg/$\ell$, 100mg/$\ell$, 25 mg/$\ell$ 150 mg/$\ell$, and 2.5 mg/$\ell$, respectively. However, the inhibitive effects of sulfide and sulfite on BPA degradation were diminished by additional aeration to oxidize them. Formaldehyde significantly reduced the rate of BPA degradation at 1.0% among organic compounds studied. Among heavy metal ions tested, Fe(II) substantially suppressed BPA removal at 1 mM. MgCl$_2$ and CaCl$_2$ exhibited great inhibition of BPA degradation at 25mM.

Growth and Physiology of Thiobacillus novellus under Autotrophic and Heterotrophic Conditions (자가영양과 타가영양 조건하에서 Thiobacillus novellus의 생리 및 성장)

  • The growth of T. novellus in auto trophic and geterotrophic media was studied to determine the time required for cells to enter stationary phase and relative percentage of ribosomal proteins. When T. novellus was grown autotrophically, growth proceeded at a slow rate characteristic of autotrophs and did not enter log phase until the end of the first day. Logarithmic growth proceeded for 3-4 days at which time the cells entered the stationary phase. In particular, logarithmic growth was accompanied by decreasing pH of culture media and in the stationary phase the pH levelled off at 6.0, a decrease of 1.6 pH value compared to original pH of media. The pH decrease was greatest during log phase when cells oxidized thiosulfate to $H_{2}$$SO_{4}$. The doubling time was about 26h. In heterotrophic media growth proceeded at a much faster rate and cells entered stationary phase 20-22h after inoculation. The doubling time was 3h. The protein content of the ribosomes in T. novellus grown heterotrophically was 4.2% greater than those from the organism grown autotrophically.

A Study on Detecting Dross in Coating Layer on Hot-dip Galvanizing Steels (용융아연 도금강판의 도금층에 잔류한 드로스 검출에 관한 연구)

  • To develop a method of detecting dross in coating layer on hot-dip galvanizing steel, chemical etching behavior of the artificial coating layers with top and bottom dross were investigated. After chemical etching with the mixture of picric acid and sodium thiosulfate, each of the top and bottom dross take its distinct color, and alloy layer in coating is also observed. Defects in the coating layers of HGI(hot rolled galvanized iron), CGI(continuous galvanized steel sheet) and GA(galvannealed steel) were analysed, and methods of dross detection which can be applied to inspection process in manufacture were suggested.

Effect of a Combined Treatment with Uniconazole, Silver Thiosulfate on Reduction of Ozone Injury in Tomato Plant (Uniconazole 과 Silver Thiosulfate 의 복합처리가 토마토의 오존피해경감에 미치는 효과)

  • Studies were conducted to determine the combined effect of uniconazole [(E) -1-(4-chlorophenyl)-4, 4-demethyl 2-(1,2,4 triazol-1-yl)-1-penten-3-ol] and silver thiosulfate $[Ag {(S_2O_3)}^3\;_2-]$ (STS) on reduction of ozone injury in tomato plants(Lycopersicon esculentum Mill. 'Pink Glory'). Plants were given a 50ml soil drench of uniconazole at concentrations of 0, 0.001, 0.01 and 0.1 mg/pot at the stage of emerging 4th leaf. Two days prior to ozone fumigation, STS solution contained 0.05% Tween-20 was also sprayed at concentrations of 0, 0.3 and 0.6 mM. Uniconazole at 0.01 mg/pot and STS at 0.6 mM were effective in providing protection against ozone exposure(20h at 0.2ppm) without severe retardation of plant height and chemical phytotoxicity, respectively. Combined treatment with uniconazole, STS significantly reduced ozone injury at the lower concentration than a single treatment with uniconazole or STS. Uniconazole treatment reduced plant height, stem elongation and transpiration rate on a whole plant level and increased chlorophyll concentration. STS did not give any effect on plant growth and chlorophyll content but increased transpiration rate in non-ozone-fumigated plants. Ethylene production in the leaves of ozone-fumigated plants was decreased by uniconazole and STS pretreatment, but there was no protective effect on epinasty of leaves in uniconazole-treated plants. STS increased ethylene production in non-ozone-fumigated plants, but it significantly reduced the degree of epinasty and defoliation of cotyledons when plants were exposed to ozone. Uniconazole slightly increased superoxide dismutase and peroxidase activities. But STS showed little or no effects on such free radical scavengers. Day of flowering after seeding was shortened and percentages of fruit set were increased by uniconazole treatment. STS was highly effective on protecting reduction of fruit set resulting from ozone fumigation. These results suggest that combined use of uniconazole and STS should provide miximum protection against ozone injury without growth retardation resulting in yield loss.

FACTORS AFFECTING THE PRODUCTION OF SULFUR COMPOUNDS BY FUSOBACTERIUM NUCLEATUM (Fusobacterium nucleatum의 유황화합물 생성에 영향을 미치는 인자)

  • Fusobacterium nucleatum, one of the bacteria causing halitosis, produces the volatile sulfur compounds (VSC) such as $H_2S$ in the media containing sulfur components, and forms FeS by binding with iron component. The various factors of oral cavity affect the concentration of sulfur compounds produced by Fusobacterium nucleatum. In this study, the effect of nutrients and pH on the production of sulfur compounds by Fusobacterium nucleatum was studied with the following results. 1. The optical density of broth was increased to $0.817{\pm}0.032$ and $1.297{\pm}0.024$ by adding 1.0% sodium thiosulfate and 0.05% L-cysteine hydrochloride in the media, respectively. 2. Though the optical density of broth was $0.799{\pm}0.032$ by adding volatile sulfur compounds (VSC) only in the media, it was increased to $1.775{\pm}0.003$ and $1.648{\pm}0.022$ by adding xylitol combined with glucose and fructose, respectively. 3. The concentration of VSC was above 20,000 ppb in the media above pH 5.5. The optical density of broth was still high in the media with L-cysteine hydrochloride of higher concentration, being low in the media of lower pH. 4. The concentration of VSC was high when there was distilled water or saline solution on the media, and their amount was small. These results suggest that the production of sulfur compounds by Fusobacterium nucleatum was inhibited by xylitol and acid.
