The Simultaneous Extraction and Determination of Trace Copper and Zinc in Solvent Extraction (용매추출법에 의한 토양중의 미량 구리와 아연원소의 동시추출 및 정량에 관한 연구)

  • A rapid and selective co-extraction systems of copper and zinc-thiocyanate complex into various types of alkylamine for the simultaneous determination of two metal ions by atomic absorption spectrometry and ion chromatograph have been proposed. The quantitative extractions of Cu(II) and Zn(II) at 0.1 M-thiocyanate and 0.1 M-HCI were achieved with Aliquat 336-$CHCl_3$. The detection limits of Cu and Zn were 2 ppb and 0.9 ppb respectively.

  • A tri-thiocyanate molybdenum (Ⅴ) complex containing pyridine has been prepared by one step process; namely, the solvent extraction of molybdenum (Ⅴ)-thiocyanate complex into organic solvents followed by precipitation of the compound by addition of pyridine to the extract. It is concluded that the compound has a definite composition regardless of the various mole ratios of molybdenum to thiocyanate ion employed in the preparation. The use of hydrazine as the reducing agent eliminates the necessity of working under inert atmosphere and of further purification of the product. Molybdenum (Ⅴ)-thiocyanate (1:3) complex can be quantitatively and selectively extracted with slightly polar organic solvents such as ethyl acetate, and the pyridine complex is quantitatively obtained due to the insolubility of the compound in them. The oxidation state of molybdenum in the compound is + 5 and the formula appears to be $MoH_2O_2(NCS)_3Py_2$ from the contents of Mo and NCS in the compound. The solubility of the compound in various has been studied at $25^{\circ}C$.

Conversion of Apricot Cyanogenic Glycosides to Thiocyanate by Liver and Colon Enzymes

  • Some of the edible plants like apricot kernel, flaxseed, and cassava generate hydrogen cyanide (HCN) when cyanogenic glycosides are hydrolyzed. Rhodanese (thiosulfate: cyanide sulfurtransferases of TSTs; EC: 2.8.1.1) is a sulfide-detoxifying enzymes that converts cyanides into thiocyanate and sulfite. This enzyme exists in a liver and kidneys in abundance. The present study is to evaluate the conversion of apricot cyanogenic glycosides into thiocyanate by human hepatic (HepG2) and colonal (HT-29) cells, and the induction of the enzymes in the rat. The effects of short term exposure of amygdalin to rats have also been investigated. Cytosolic, mitochondrial, and microsomal fractions from HepG2 and HT-29 cells and normal male Spraque-Dawley rats were used. When apricot kernel extract was used as substrate, the rhodanese activity in liver cells was higher than the activity in colon cells, both from established human cell line or animal tissue. The cytosolic fractions showed the highest rhodanese activity in all of the cells, exhibiting two to three times that of microsomal fractions. Moreover, the cell homogenates could metabolize apricot extract to thiocyanate suggesting cellular hydrolysis of cyanogenic glycoside to cyanide ion, followed by a sulfur transfer to thiocyanate. After the consumption of amygdalin for 14 days, growth of rats began to decrease relative to that of the control group though a significant change in thyroid has not been observed. The resulting data support the conversion to thiocyanate, which relate to the thyroid dysfunction caused by the chronic dietary intake of cyanide. Because Korean eats a lot of Brassicaceae vegetables such as Chinese cabbage and radish, the results of this study might indicate the involvement of rhodanese in prolonged exposure of cyanogenic glycosides.

  • In this study, the extraction of Co(Ⅱ) and Cu(Ⅱ) into chloroform solution of dithizone, distribution ratios, extractabilities and extraction rate constants of the metal complexes were compared in each case with which thiocyanate ion was or not used as the auxiliary ligand. The use of the thiocyanate ion increased threefold the distribution ratio for Co(Ⅱ) complex in a basic solution and twofold for Cu(Ⅱ) complex in a wide pH range. And the extractability was also augmented from about 90 to 99 for Co(Ⅱ) and from 95 to 99 for Cu(Ⅱ) in a given period. The extraction rate constants were $k_1\;:\;1.2{\times}10^5$, $k_2\;:\;1.34{\times}10^{17}\; mol^{-1}dm^3s^{-1}$ in case of Co(Ⅱ) and$k_1\;:\;1.1{\times}10^8$, $k_2\;:\;2.83{\times}10^{10}\; mol^{-1}dm^3s^{-1}$ in case of Cu(Ⅱ) on the extraction of dithizonate complexes into chloroform solution.

  • K-bentonite was made by ion exchange reaction in $K^{\ast}$ ion saturated aqueous solution. K-bentonite had a slow releasing effect in different soils such as sand, sandy loam and clayey loam, but the effect was the best in sand. The growth of radish and lettuce was better in the plot fertilized with K-bentonite than with KCl in sand culture in field condition. There was no effects on the growth of radish grown in pot in glass house. Vitamin C, nitrate content, thiocyanate ion content and dry weight of radish were not affected by K-bentonite and KCl in both pot and field culture. The commercial production of K-bentonite was discussed.

  • The silver selenide electrode has been prepared and its properties as an indicating electrode for silver ion have been investigated. Epoxy resin was used as a filler of silver selenide electrode. Silver metal plate was directly connected with the membrane of the electrode and the silver paste was used as its binder. The sintered electrode was more sensitive and stable than the pressed electrode, and the silver selenide electrode more sensitive than the silver sulfide electrode to silver ion. The linear relationship between the electrode potential and logarithmic concentration of silver ion has been observed down to 10-6 M for the electrode. Several heavy metal ions except mercuric ion did not interfere this linearity, but halide, cyanide, and thiocyanate ions did intensively interfere owing to the formation of silver compounds and complexes. This electrode has been applied to the potentiometric titration for determining halide ion. It is concluded that interferences from ,$CN^-, SCN^-, S^-, I^-, Br^-, Cl^- and Hg^{2+}$ ions are detrimental to the practical use of the electrodes for measuring pAg.

  • A stepwise gradient elution with two wavelengths detection was performed for the separation and determination of some anions in saline water. The eight anions such as iodate, bromate, nitrite, bromide, nitrate, chromate, iodide and thiocyanate were successfully separated using AS-7 column and sodium chloride/sodium phosphate buffer solution as an eluant within 40 min. The separation behaviors of anions were studied at various sodium chloride concentrations. The peak shapes of anions of bromate, nitrite, bromide and nitrate gradually broadened as the concentration of sodium chloride increased until 1.0 M in the sample solutions. However, no effect was observed in the peak shapes of chromate, iodide and thiocynate. A good linearity was obtained at the range of ppm(mg/L). The detection limit was proved to be $10-720{\mu}g/L$ for the eight anions with $50{\mu}L$ injection volume. This method was applied to the determination of $Br^-$, ${NO_3}^-$ and $I^-$ in sea water.

  • Glucosinolates from radish (Wangkwan, Daejangkun, Taebaek, Daebooryeung and No. 364) and its seed (Wangkwan) were identified, and their degradation product by heat treatment was analyzed. The Wangkwan variety contained much more types of glucosinolates than other radish varieties. Total glucosinolate contents of Wangkwan, Daejangkun, Taebaek, Daebooryeung, No. 354 and Wangkwan seed by GC method were 1.25, 1.10, 0.97, 0.96, 0.90 and 2.14 ${\mu}mole/g$, respectively. The indoleacetonitrile contents after 20 minutes' heat treatment at $100^{\circ}C$ from Wangkwan, Daejangkun, Taebaek, Daebooryeung, No. 364 and Wangkwan seed were 0.28, 0.20, 0.23, 0.21, 0.24 and 0.58 ${\mu}mole/g$, respectively. The heat treatment increased the thiocyanate ion contents in radish and its seed, and the contents were maximum at 30 min.

  • The perchlorate, thiocyanate, and nitrate ion-selective PVC membrance electrode for measuring hazardous anion in waste water were developed by incorporating the quaternary ammonium salts as active material. Ion-selective characteristics in waste water were studied by the useful pH range, the selective coefficients to various interfering anions, and the stability of electrode potential. DBP was best as a plasticizer. The effect of the membrane thickness on the electrode characteristics was improved with decreasing the membrane thickness, but below the optimum membrane thickness the electrode exhibited an inverse trend. The electrode potential of perchlorate, thiocyanate, and nitrate electrode with TDDA, as active material, was stable within the pH range 4-11, 3-12, and 4-10 repectively. And the long-term potential stability of these electrodes were 3.0, 3.5, and 3.5 months respectively. The order of the selectivity coefficients was as shown below ; $ClO_4{^-}$ > $SCN^-$ > $I^-$ > $NO_3{^-}$ > $Br^-$ > $CN^-$ > $F^-$ > $Cl^-$ > $Ac^-$ > $H_2PO_4{^-}$, $SO_4{^-}$.
