Antimutagenic Compounds Indentified from Chloroform Fraction of Persimmon Leaves

  • Methanol extract of dried persimmon leaves was refractionated using hexane, chloroform, ethylacetate, n-butanol aqueous fractions. Among these chloroform fraction showed the highest inhibition rate on the mutagenicities of aflatoxin B₁(AFB₁) and 3,2' -dimethyl-4-amino-bipheny1(DMAB) in Salmonella typhimurium TA 100. Chloroform fraction was further fractionated into eight fractions by silica gel column chromatography and thin layer chromatography(TLC). The fraction 5 on TLC exhibited the highest antimutagenic activities in AFB₁and DMAB. 2,4-Decadienal, dihydro-4-methyl-2(3H)-furanone, hexanoic acid 1,4-bis(1-methy1 ethy1)benzene, heptanoic acid, phenol, octanoic acid, nonanoic acid and benzoic acid were tentatively identified from this antimutagenic fraction by GC-MS.

Antioxidative Substances in Mulberry Leaves (뽕잎(桑葉)에 함유된 항산화성 물질)

  • Antioxodative substances in Mulberry leaves were examined. Antioxidative substances in Mulberry leaves were extracted by 80% methanol agueous solution. Antioxidative activity of extract was determined by examining hydrogen donating ability on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the inhibitory effect on the formation of the peroxide from Linoleic acid in the test tube at $50^{\circ}C$. Antioxidative substance were, then, separated and indentified by thin layer chromatography(TLC), UV-Vis spectrum and High performance liquid chromatography(HPLC) methods. Hydrogen donating ability on DPPH and antioxidative ability on linoleic acid of the extracted antioxidative substance were higher than those of 100ppm butylated hydroxy toluene(BHT). The extracted antioxidative substances were separated by TLC using ethylacetate : chloroform : formic acid : water(8 : 1 : 1 : 1 v/v) as a solvent, and a spot at Rf=0.35 was detected. The spot was scraped from the plate, and extrated by methanol. The extract was analyzed by UV-Vis spetra and HPLC, and chlorogenic acid was identified as a antioxidative substance.

Antimicrobial Activity of The Volatile Components from Fruit Peel of Chopi(Zanthoxylum piperitum DC) (초피(Zanthoxylum piperitum DC) 과피의 휘발 성분의 항균작용)

  • The volatile components (essential oil) showing antimicrobial activity were extracted from the fruit peel of Zanthoxylum piperitum DC by distillation and separated by thin layer chromatography (TLC). The crude volatile components exhibited antimicrobial activity only at very high concentration. The active fraction obtained by TLC inhibited noticeably the growth of bacteria. The minimum inhibitory concentration (MIC) of the fraction were 150ppm, 300ppm, and 300ppm against Escherichia coli, Staphylococcus aureus, and Salmonella enteritidis, respectively. The components in the active fraction were identified by gas chromatography/mass spectrometry to be geranlyl acetate (60.23%), citronellal(36.01%), citronellol(3.77%), geraniol(0.46%), and cumin ldehyde(0.43%).

Study on the molecular weights of radioprotective ginseng proteins by HPLC method

  • Partialy purified ginseng proteins were either treated with sodium dodecyl sulfate (SDS) and .betha.-mercaptoethanol to denature the proteins or not, and subjected to Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC) to compare the components of each fraction. Standard proteins of known molecular weights (MW) were also either treated with SDS and .betha.-mercaptoethanol or not, and subjected to HPLC to obtain regression lines for MW determination. From the retention times obtained from samples in eiether case by HPLC, the MW were estimated as following in SDS treated condition, Gl fraction showed three peaks each with MW of above 100, 000, 51, 000 and 19, 000. Gll showed one original peak with MW of 21, 000 and Gll, two peaks each with MW of 19, 000 and 14, 000. On the other hand, in non-SDS treated condition, GI fraction showed two peaks each with MW of above 200, 000 NA 52, 000. Gll showed one original peak with MW of 41, 000 and Gill, three peaks each with MW of 28, 000, 19, 000 and 14, 000.

  • An investigation of certain tar food colorants required a relatively inexpensive TLC adsorbent to be used in preparative thin-layer chromatography procedure. As will be shown activated clay and Kieselguhr has excellent physical characteristics for T LC applications, is easily applied, and is good for analytical work as well as showing good separator capabilities with selected groups of compounds. In addition to its usefulness its economical price suggested this article.

  • One of the most important characteristics in the seventh educational course is an enforcement of differentiated level curriculum. Therefore, development of various different level educational subjects is needed. A new chromatographic experimental kit which is helpful for middle school students to understand chromatographic principles is needed to correct some mistakes of chromatographic part within the 6th educational course's middle school textbook. In this research, a new thin layer chromatography(TLC) experimental kit for middle school students are developed by using acid-base indicator as samples and ethyl alcohol, ethylacetate, hexane as eluents. The kit is applied to two kinds of middle school student groups(general group & genius group) and two kinds of teacher groups(science teacher & non-science teacher) and an efficacy of the new chromatographic kits are evaluated through making up some questions.

  • This study was carried out to utilize non-toxic solvents instead of harmful solvents for the TLC(Thin Layer Chromatography) identification test of herbal medicines. It is recommended not to use harmful solvent such as chloroform at a viewpoint of clean analysis. In this study, we revised the identification test of 10 items in the Korea Pharmacopoeia(KP) and the Korean Herbal Pharmacopoeia(KHP) such as Cornus officinalis S. containing the harmful solvents on the developing solvent and established identification test that is utilized non-toxic solvents by HPTLC(High Performance Thin Layer Chromatography).

Screening of Deoxynivalenol Producing Fungi from Greenhouse Horticulture by Enzyme Amplification System Immunoassay (Enzyme Amplification System Immunoassay에 의한 시설원예산물의 Deoxynivalenol 생성곰팡이의 검색)

  • In order to evaluate the safety of greenhouse horticulture products in Korea, we carried out this work by screening of Fusarium species, which produce deoxynivalenol (DON) from greenhouse horticulture in Western Gyeongnam and Northern Gyeongbuk, Korea. For this study, high sensitive enzyme-linked immunosorbent assay, ALP/NADP method, was applied to detection of DON by enzyme amplification system. From 192 samples of greenhouse horticulture soil and its products, 103 isolates of Fusarium species were obtained. The isolates were cultured at 28C for 15 days and the cultured mediums were extracted by ethyl acetate. The production of DON was verified by thin layer chromatography (TLC). As the results of TLC, 8 strains were identified as DON producing strain. We screened potential producers of DON by ALP/NADP. The levels of DON production were shown from 0.007 to 1.21 g/ml of YES medium. The maximum DON producing strain No. 32-D-3 was isolated from soil in Namhae, Korea. In conclusion, the above results indicate that DON producing fungi contaminated greenhouse horticulture products in Korea. Therefore, further studies are required to accumulate more detailed data about the contamination of DON in various cereals.

A Study on the Metabolism of Carbon Disulfide by Isolated Rat Liver Perfusion (적출 흰쥐 간 관류법에 의한 이황화탄소 대사에 관한 연구)

  • The purposes of this study are the identification and determination of metabolites in the isolated rat liver perfusate of carbon disulfide by two-dimentional thin-layer chromatography and high performance liquid chromatography for understanding the metabolism of carbon disulfide. 2-Thio-thiazolidine-4-carboxylic acid(TTCA) was synthesized by the reaction of carbon disulfide and cysteine, and confirmed by two-dimentional thin-layer chromatography, high performance liquid chromatography, UV spectroscopy, and IR spectroscopy. The absorbance of UV detector for the simultaneous determination of TTCA and thiocarbamide was 254 nm although their maximum spectra were 273 nm and 237 nm, respectively. Two kinds of the developing solvent in the two-dimentional thin-layer chromatography were 2-butanol : 80% HCOOH : $H_2O$ (7 : 2 : 1) as the first developing solvent and 2-propanol : $H_2O$ (4 : 1) as the second developing solvent. After perfusion of carbon disulfide ($8274.23{\mu}mol$), the amount of TTCA and thiocabamide of the perfusate(100 ul) were $12.02-16.4{\mu}mol$ and $5.25-8.15{\mu}g$, respectively. The mean amount of them were $14.08{\mu}mol$ and $6.41{\mu}mol$ respectively, and the former was 2.20 times greater than the latter. For conforming the mechanism of formations of TTCA and thiocarbamide in vivo, we have to clarify whether the reactions between carbon disulfide and ammonia, ammonium salts, amides, cysteine, cystine, or proteins will be formed in vitro.

Screening of Deoxynivalenol Producing Strains from Agricultural Products by Immunoanalytical Method (면역분석기법에 의한 농산물에서의 Deoxynivalenol생성균 검색)

  • In order to evaluate the safety of agricultural products in Korea, we carried out work by screening of Fusarium species. which can produce deoxinivalenol(DON) from agricultural products in Western Gyeongnam, Korea. From 215 samples of soil agricultural products, 129 strains of Fusarium species were obtained. The production of DON was verified by thin layer chromatography(TLC) As the results of TLC, 25 strains were identified as DON producing strain. But only 10 strains were identified as DON producing strains by enzyme-linked immunosorbent assay (ELISA). The maximum DON producing strain No.41 was isolated from corn. In conclusion. the above results indicate that DON producing fungi contaminated agricultural products in Korea. Therefore further studies are required to accumulate more detailed data about the contamination of DON in various agricultural products.
