  • In the goby Micropercops swinhonis, the follicular layer of full-grown oocytes consists of an outer layer (theca cell) and an inner layer (granulosa cell). As the oocyte grows, columnar cells of inner granulosa layer secrete mucin to their cytoplasm and then surround the oocyte. Such granulosa cells appear to be cuboidal cells in the early vitellogenesis, yolk vesicle stage, to be replaced by columnar cell secreting mucins (adhesive materials) in the middle vitellogenesis, yolk granule stage. The enveloping layer of the oocyte has a muco- follicle layer filled with mucins. The mucins are an amorphous and electron-dense substance. Interestingly, the oocyte enveloping layer becomes thickened towards the animal pole as vitelogenesis proceeds. A zona radiata of about $7.8{\sim}11.5\;{\mu}m$ thick is present below the muco-follicle layer. The zona radiata is composed of an one-layered electron-dense externa and a three to five-layered electron-less interna.

Role of Growth Differentiation Factor 9 and Bone Morphogenetic Protein 15 in Ovarian Function and Their Importance in Mammalian Female Fertility - A Review

  • Growth factors play an important role during early ovarian development and folliculogenesis, since they regulate the migration of germ cells to the gonadal ridge. They also act on follicle recruitment, proliferation/atresia of granulosa cells and theca, steroidogenesis, oocyte maturation, ovulation and luteinization. Among the growth factors, the growth differentiation factor 9 (GDF9) and the bone morphogenetic protein 15 (BMP15), belong to the transforming growth factor beta (TGF-${\beta}$) superfamily, have been implicated as essential for follicular development. The GDF9 and BMP15 participate in the evolution of the primordial follicle to primary follicle and play an important role in the later stages of follicular development and maturation, increasing the steroidogenic acute regulatory protein expression, plasminogen activator and luteinizing hormone receptor (LHR). These factors are also involved in the interconnections between the oocyte and surrounding cumulus cells, where they regulate absorption of amino acids, glycolysis and biosynthesis of cholesterol cumulus cells. Even though the mode of action has not been fully established, in vitro observations indicate that the factors GDF9 and BMP15 stimulate the growth of ovarian follicles and proliferation of cumulus cells through the induction of mitosis in cells and granulosa and theca expression of genes linked to follicular maturation. Thus, seeking greater understanding of the action of these growth factors on the development of oocytes, the role of GDF9 and BMP15 in ovarian function is summarized in this brief review.

Anatomical and Histological Features and Ovarian Hormone Analysis of Ovarian Cysts in Korean Native Cow and Dairy Cow (한우(韓牛) 및 유우(乳牛)의 난소난종(卵巢囊腫)에 관한 해부조직학적(解剖組織學的) 소견(所見) 및 난소(卵巢)호르몬 분석(分析))

  • A total of 1200 Korean native cow and 240 dairy cow genitalia were collected during the slaughtering process in Seoul and Kwang Ju abattoir and were examined from July 1985 to March 1986. Ovarian follicles were classified as cystic if the diameter was greater than 2.5cm or if follicles were multiple. In order to investigate the ovarian cysts, anatomical and histological examinations were performed. In addition progesterone and estrogen level in different types of cystic follicular fluid and serum were measured by radioimmunoassay. The results were summerized as follows: 1. The incidences of ovarian cysts were 2.0% in Korean native cow and 7.9% in dairy cow. 2. In distribution of cysts in the left, right and both ovaries, the most encountered ovary was right one. The frequency was 45.8% in right ovaries, 33.4% in left ovaries and 20.8% in both ovaries in Korean native cow. On the contrary the frequency was 42.1% in right ovaries, 31.8% in both ovaries and 26.3% in left ovaries in dairy cow. 3. Six speciemens (25.0%) of Korean native cow and six specimens (31.6%) of dairy cow were associated with corpora lutes in both ovaries. 4. The luteinization of theca layer was most significant in the group 2Aa (71.4%) and 2Ba (38.5%) which associated with no granulosa cell and corpora lutea in the same cystic ovaries. 5. Correlation of progesterone concentration between cystic fluid and serum was found only in the group 2Aa and 2Ab (r=0.86). Progesterone and estrogen concentrations in cystic fluid were closely related to the degree of degeneration of granulosa cell layer. The cystic follicles that consist of thickened theca and degenerated granulosa cell layers contained a large amount of progesterone, and small amount of estrogen. In conclusion, various types of ovarian cysts with various levels of progesterone and estrogen were observed in Korean native cow.

  • Gangilosides are ubiquitous membrane components in mammalian cells and are suggested to play essential roles in cellular phenomena such as cell-cell interadion, differentiation, and signal transduction. Rat ovary contained GM3 as major gangiloside. Nn order to study GM3 distribution in the atretic follicles and its possible changes during follicular development, frozen sections were stained with spedfic monocional antibodies against eleven gangilo-series gangliosides including GM3. In the atretic follicles, Glf3 was expressed in a spatlo-temporally different manner during foilicular development, but GM1 and other gangliosides were not immunohistochemicaily detected. in atretic follicle from primary follicle stage, GM3 was expressed in all the theca cells and some granulosa cells adjacent to oocyte. In atretic follicle from secondary follicle stage, GM3 were expressed in all theca cells and granulosa cells. in atretic follicles from developing Graaflan follicle stages, GM3 was similarly expressed as in secondary follicle stage.

  • Aquaporin channels (AQPs) are known to play an important role in the development of ovarian follicles through their function in water transport pathways. Compared to other AQPs, research on the role of AQP4 in female reproductive physiology, particularly in cattle, remains limited. In our previous study, gene chip microarray data showed a downregulation of AQP4 in bovine cystic follicles. This study was performed to validate the AQP4 expression level at the protein level in bovine follicles using immunohistochemistry, Western blotting, and immunoprecipitation assays. Immunostaining data showed that AQP4 was expressed in granulosa and theca cells of bovine ovarian follicles. The ovarian follicles were classified according to size as small (< 10 mm) or large (> 25 mm) in diameter. Consistent with earlier microarray data, semi-quantitative PCR data showed a decrease in AQP4 mRNA expression in large follicles. Western blot analysis showed a downregulation of the AQP4 protein in large follicles. In addition, AQP4 was immunoprecipitated and blotted with anti-AQP4 antibody in small and large follicles. Accordingly, AQP4 exhibited a low expression in large follicles. These results show that AQP4 is downregulated in bovine ovarian large follicles, suggesting that the downregulation of AQP4 expression may interfere with follicular water transport, leading to bovine follicular cysts.

  • Sex cord tumors with annular tubules are known to originate from the sex cord of embryonic gonads that synthesize Sertoli cells, Leydig cells, granulosa cells, and theca cells of the ovarian stroma, while ovarian small cell carcinoma of the hypercalcemic type is a type of neuroendocrine tumor. Both these tumors are uncommon, potentially malignant neoplasms in children. We report the case of a sex cord tumor with annular tubules in an 11-year-old girl and a case of small cell carcinoma of the hypercalcemic type in a 10-year-old girl. We also discuss the prognosis and management of these tumors.

  • 60 cases of swine and bovine ovaries were investigated for detecting the homosiderin pigment by means of Perl's prussian blue reaction. The results abtained were as follows; 1. Among the 30 cases of normal adult swine ovaries, 20 cases (66.6%) were presented as positive. 2. The hemosiderin pigment was detected in corpus albicans, connective tissue of corpus luteum, theca externa of follicles and interstitial tissue of ovary in swine. 3. The morphological characteristics of the hemosiderin pigment of swine ovaries were fine and coarse granules or amorphous and homogenous pigment. 4. In 30 cases of bovine ovaries, the hemosiderin pigment was not detected.

Effect of Prostaglandin F2 Alpha on E-cadherin, N-cadherin and Cell Adhesion in Ovarian Luteal Theca Cells (난소의 황체협막세포에서 E-cadherin, N-cadherin과 세포부착에 미치는 Prostaglandin F2 Alpha의 영향)

  • Cadherins are essential transmembrane proteins that promote cell-cell adhesion and maintain the corpus luteum structure in the ovary. This study examined the influence of prostaglandin F2 alpha ($PGF2{\alpha}$) on E-cadherin, N-cadherin, and adhesion in luteal theca cells (LTCs). The luteal cells were isolated from the mid-phase corpus luteum, and the LTCs were cultured separately from the luteal heterogeneous cells according to the morphology of the mesenchymal cells and to determine if steroidogenic and endothelial cells of LTCs, 3beta-hydroxysteroid dehydrogenase ($3{\beta}$-HSD), and vascular endothelial growth factor receptor 2 (VEGFR2) mRNA were used. The LTCs were then incubated in the culture medium supplemented with 0.01, 0.1, and 1.0 mM $PGF2{\alpha}$ for 24 h, and the E-cadherin and N-cadherin proteins in the LTCs were detected by confocal laser scanning microscopy. The results revealed $3{\beta}$-HSD mRNA expression in the LTC but no VEGF2R mRNA expression. The E-cadherin and N-cadherin proteins of the LTCs were damaged in the 0.01, 0.1, and 1.0 mM $PGF2{\alpha}$ treatment groups, and the expression of the N-cadherin protein was reduced significantly in 0.01 mM $PGF2{\alpha}$ compared to the 0 mM $PGF2{\alpha}$ treatment groups (P<0.05). In addition, the number of attached LTCs were significantly lower in the 0.01 mM $PGF2{\alpha}$ treatment group than in the 0 mM $PGF2{\alpha}$ treatment group (P<0.05). In conclusion, $PGF2{\alpha}$ affected the disruption of cadherin proteins and cell adhesion in LTCs. These results may help better understand the cadherin and adhesion mechanism during corpus luteum regression in the ovary.

Expression and Cellular Localization of Gonadotropin-Releasing Hormone (GnRH)-like Messenger Ribonucleic Acid in the Rat Gonad (흰쥐 생식소에서 GnRH-like mRNA의 발현과 세포내 분포)

  • Gonadotropin releasing horrnone (GnRH) is known to be extrahypothalamically localized with a broad range including gonad. It remains, however, unknown whether GnRH is locally synthesized in the gonad. The present srudy aims to identity expression and cellular localization of GnRH-Iike mRNA and immunoreactive GnRH in the rat gonad. GnRH radioimmunoassay and chromatographic extracts on G-50 sephadex column showed that rat gonadal extracts contained a substantial amount of immunoreactive GnRH similar to the hypothalamic and synthetic GnRH. Although a wide distribution of immunostainable GnRH-like molecule with different cell types in the rat ovary was observed, the major cell population hybridized with GnRH probe appears to be granulosa. theca cells and corpus luteum. Immunoreactive GnRH-Iike peptides were distributed m various regions of testis, including spermatogenic cells, Sertoli cells and Leydig cells. In situ hybridization revealed that positive signals of GnRH-Iike mRNA were predominandy present in Sertoli cells within some seminiferous tubules, but absent in the outside of seminiferous tubules in the testis. This study clearly demonstrated that GnRH-Iike molecule present in the rat gonad may be resulted from the local synthetic machinery of GnRH supporting the notion that this peptide may act as autocrine and/or paracrine role in intra-gonadal communication.

Development and Transformation of Oocyte' Follicle Cell for Korean Four Sillurid Fishes of Liobagrus and Pseudobagrus (Pisces: Silluriformes) (한국산 동자개속 및 퉁가리속 4종 어류의 난모세포에서 여포세포의 발달과 변형)

  • A histological study on development and transformation of the oocyte' follicle cell for Korean four sillurid fishes, Liobagrus obesus, L. mediadiposalis, Pseudobagrus koreanus, and P. brevicorpus was performed by light and electron microscopes. The follicular layer surrounding the oocyte consisted of an outer theca cell and an inner follicle cell (granulosa cell). The follicle cells of the oocyte were flatten cells at early oocyte but during vitellogenesis they were transformed it to a single layer of cuboidal cell, then to a single columnar cell layer, and finally to a layer covered with a substance secreted by themselves. Although the development and transformation of the follicle cells was similar to four species, the secreted materials, called an adhesive membrane, were divided into two types in its appearance and nature. Firstly, a jelly coat-like type was found in L. obesus and L. mediadiposalis, which they are presumed to be polysaccharides and mucoproteins in its nature and secondly, a granular type in P. koreanus and P. brevicopus, being mucoprotein. A zona radiata with about $0.6{\sim}3.1{\mu}m$ thin was present below the adhesive material secreted by the transformed-follicle cell's activity. The zona radiata was composed of two layers, a thin externa and a thick interna.
