Studies on the Immunoblot Characterization of Clonorchis sinensis Worm Antigens at Carly Development Stages (Immunoblot 법을 이용한 간흡충항원(肝吸蟲抗原)의 발육단계별(發育段階別) 항원성분석(抗原性分析)에 관한 연구(硏究))

  • Serodiagnosis of Clonorchis sinensis infections will probably be a first choice tool for screening of clonorchiasis in a future because of increasing difficulties in collection and examination of stools. The sensitive test such as ELISA can he used effectively. However there are some limitations in serological diagnosis for the detection of serum antibody. One of the major problems is the non-specificity of the antigens which produce cross reaction with other helminthic infection sera. To solve this problem. many investigators have tried to purify the antigens used. In this study, we determined the antigenic profile of the crude saline extract antigen of C. sinensis at early developmental stage based on SDS-PAGE and immunoblotting techniques for the purpose of understanding the nature of C. sinensis worm antigen The following results were obtained : 1) The SDS-PAGE showed many protein hands ranging from 10Kd to 91Kd relative molecular weight. Among them, 66, 46, 40, 33, 27, 24, 16, 14 and 10Kd bands were observed as a principle bands. The protein components of C. sinensis changed chronologically during their early developmental period. 44Kd band was stained unclearly in antigen of 2 weeks worm, but changed to concentrated state in antigen of 5 weeks worm. 35Kd band was found in antigen of 2 weeks worm, however this band was disappeared in antigen of 5 weeks worm. 22Kd band also lost its staining property gradually. 2) In spite of differences in antigenic profile, there was no differences in the data obtained by microplate ELISA using each antigen preparation. Absorbance value began to rise in between 2 to 3 weeks after infection. 3) By EITB. serum antibody recognized major protein bands with molecular weight of 91, 85, 63, 46, 40, 33, 24, 14 and 10Kd hand respectively. Among them 66, 33, 17 and 14Kd bands were observed as non-specific band because they reacted even in normal control sera. Generally, gradual increase of positive reactions were observed as the infection period of C. sinensis was prolonged. In other hand, the reaction of 10Kd hand did not occurred when 26th week sera was tested. 4) The positive reactions using antigens of 2 weeks worm, especially on 40 and 24Kd bands, were most strong and sharply demarcated compared to those of 3~5 weeks worm antigen.

Preparation and Properties of Autoxidation Drying Type Waterborne Coatings Containing Bentonite (벤토나이트가 포함된 자동산화 건조형 수성코팅제의 제조 및 특성)

  • Four different composition of autoxidation drying type waterborne coatings (WBC-1, WBC-2, WBC-3, WBC-4) were prepared by the compounding of bentonite (BEN) as a water swellable clay and organometallic soaps as a drier with acrylic binder and coating additives. The solution viscosity, solid content, rheological properties and drying rate of WBCs were investigated. Also the thermal stability, the transmittance and the water-resistance of the films casted by WBCs were measured, and the surface topology of WBC films were investigated by the scanning probe microscopy. As WBC-2, WBC-3 and WBC-4 containing BEN showed the thixotropy with the shear rate, the storage stability of WBC was a excellent. When the driers was mixed in the ratio of Mn/Zn/Ba=1/2/3, the dry ability of WBCs showed maximum as 5.0 sec at 60$\^{C}$. The initial decomposition temperature and the transmittance of WBC films containing BEN increased in range of 32.2∼54.7$\^{C}$ and 5.1∼8.6% than the commercial WBC (MC-21W), respectively. The Effect of Passing Aged Years to the Polarization Characteristics of Embedded Steel Bar of Mortar Specimen(W/C:0.4) (몰타르 시험편(W/C:0.4) 내부철근의 분극특성에 미치는 재령년수의 영향)

  • The structures of reinforced concrete has been extensively increased with rapid development of industrial society. Futhermore, these reinforced concretes are easy to expose to severe corrosive environments such as sea water, contaminated water, acid rain and seashore etc.. Thus, corrosion problem of inner steel bar embedded in concrete is very important in terms of safety and economical point of view. In this study, multiple mortar test specimen(W/C:0.4) with six types having different cover thickness each other was prepared and was immerged in seawater solution for five years to evaluate the effect of cover thickness and immersion years to corrosion property of embedded steel bar. And the polarization characteristics of these embedded steel bars was investigated using electrochemical methods such as measuring corrosion potential, cathodic polarization curve, and cyclic voltammogram. At the beginning of immersion, the corrosion potentials exhibited increasingly nobler values with increasing cover thickness. However, after immersed for 5 years, the thicker cover of thickness, the corrosion potentials shifted in the negative direction, and the relationship between corrosion potential and cover thickness was not in good agreement with each other. Therefore, it is considered that the thinner cover of thickness, corrosive products deposited on the surface of the embedded steel bar plays the role as a resistance polarization which is resulted in decreasing the corrosion rate as well as shifting the corrosion potential in the positive direction. As a result, it seemed that the evaluation which corrosion possibility of the reinforced steel would be estimated by only measuring the corrosion potential may not be a completely desirable method. Therefore, it is suggested that we should take into account various parameters, including cover thickness, passed aged years as well as corrosion potential for more accurate assessment of corrosion possibility of reinforced steel which is exposed to partially or fully in marine environment for long years.

  • Bio diesel has advantages to reduce GHG(Greenhouse Gas) compare with the fossil fuel by using oil comes from plant/animal sources and even waste such as used cook oil. The diversity of energy feeds brings the positive effects to secure the national energy mix. In this circumstance, micro-algae is one of the prospective source, though some technical barriers. We analyzed the bio diesel which was derived from Dunaliella tertiolecta LB999 through the BD100 quality specifications designated by the law. From that result, it is revealed that the oxidation stability is one of the properties to be improved. In order to find the reason for low oxidation stability, we analyzed the oxidation tendency of each FAME components through some methods(EN 14111, EN14112, EN16091). In this study, we could find the higher double bond FAME portion, the more oxidative property(C18:1${\ll}C18:3$) in bio diesel and main unsaturated FAME group is acted as the key component deciding the bio diesel's oxidation stability. It is proved experimentally that C18:3 FAME are oxidized easily under the modified accelerated oxidation test. We also figure out low molecular weight hydrocarbon and FAME were founded as a result of thermal degradation. Some alcohol and aldehydes were also made by FAME oxidation. In conclusion, it is necessary to find the way to improve the micro-algal bio diesel's oxidation stability.

A study on the black core formation of artificial lightweight aggregates at various sintering atmospheres (인공경량골재의 소성조건이 블랙코어에 미치는 영향)

  • The lightweight aggregates made of bottom ash (70 wt%) and dredged soil (30 wt%) were prepared to investigate the property differences at various sintering atmospheres. The green aggregates were sintered at $1150^{\circ}C$ and $1200^{\circ}C$ with oxidized, neutralized and reduced atmospheres. The aggregates sintered with oxidized atmosphere showed a clear border between shell and black core area. However, the aggregates sintered with a reduced atmosphere showed only black core area in the entire cross-section of the aggregates. The black core area of the aggregates sintered with a neutralized atmosphere increased with increasing $N_2$ gas flow rates. It was determined that the sintering atmosphere was similar to that of rotary kiln when the CO gas flow was 100 cc/min to make a reduced atmosphere in tube furnace. The water absorption rates of both aggregates from tube furnace with reduced atmosphere and rotary kiln were very similar to each other.

  • Discharge property of plasma jet devices is investigated for the application to the doping processes of crystalline solar cells and others. Current-voltage characteristics are shown as the typical normal-glow discharge in the various gas pressure of plasma jets, such as in the atmospheric plasma jets of Ar-discharge, in the ambient pressure of atmospheric discharge, and in the ambient Ar-pressure of Ar-discharge. The discharge voltage of atmospheric plasma jet is required as low as about 2.5 kV while the operation voltage of low pressure below 200 Torr is low as about 1 kV in the discharge of atmospheric and Ar plasma jets. With a single channel plasma jet, the irradiated plasma current on the doped silicon wafer is obtained high as the range of 10~50 mA. The temperature increasement of wafer is normally about $200^{\circ}C$. In the result of silicon wafers doped by phosphoric acid with irradiating the plasma jets, the doping profiles of phosphorus atoms shows the possibility of plasma jet doping on solar cells.

Quality Characteristics of Aronia (Aronia melanocarpa) Juice added Yogurt Dressing (아로니아즙 첨가량에 따른 요구르트 드레싱의 품질특성)

  • This study sought to determine the physicochemical and sensory qualities of yogurt dressing prepared with different levels Aronia juice(0, 10, 20, 30, and 40%). As the amount of added Aronia juice increased, malic acid, acetic acid, and citric acid contents increased, although tartaric acid and lactic acid contents decreased. The viscosity of the control group(4,296.67 cP) was higher than those of the other groups(1,466.67~3,496.67 cP). The pH of the yogurt dressing containing 10% Aronia juice(3.83) was higher than those of the other groups(3.59~3.74). The sweetness of the yogurt dressing containing 10% Aronia juice($5.17^{\circ}Brix$) was higher than the other groups($2.47{\sim}3.47^{\circ}Brix$). Lightness decreased significantly, whereas a value and b value increased significantly with added levels of Aronia juice. DPPH radical scavenging activity of the control group was 7.42%, whereas those of the yogurt dressing samples with Aronia juice ranged from 26.30-69.86%. The sensory property results showed that the 20% Aronia juice added yogurt dressing group had the highest value in terms of color, thickness, taste, mouth feel, and overall acceptability. Overall, the results of this study indicates that the yogurt dressing containing 20% Aronia juice was the most preferred among the groups.

A study on the improvement of TiN diffusion barrier properties using Cu(Mg) alloy (Cu(Mg) alloy 금속배선에 의한 TiN 확산방지막의 특성개선)

  • The diffusion barrier properties of TiN by using Cu(Mg) alloy film have been investigated. Cu(Mg) alloy film was deposited on air-exposed TiN film. Upon annealing, interfacial MgO of 100 $\AA$ has been formed due to the reaction of Mg with oxygen existed on the surface of TiN. Combined MgO/TiN structure prevented the interdiffusion of Cu and Si up to $800^{\circ}C$. To improve the adhesion of Cu(Mg) alloy film to the TiN, TiN layer was treated by $O_2$ plasma, followed by vacuum annealing at $300^{\circ}C$. It was found that increased oxygen on the surface of TiN film by plasma treatment enhanced segregation of Mg toward the interface, resulting in the formation of dense MgO layer. Improved adhesion characteristics have been formed through this treatment. However, increased power of $O_2$ plasma led to the formation of TiO$_2$ and decreased the Mg content to be segregated to the interface, resulting in the decrease in adhesion property. In addition, the deposition of 50 ${\AA}$ Si on the TiN enhanced the adhesion of Cu(Mg) alloy to TiN without deteriorating the TiN diffusion barrier characteristics.

The Properties of Permeability and Freeze-Thaw Resistance of Water-Permeable Paving Brick Using Wastes (폐기물을 이용한 투수블록의 투수성 및 동결융해저항 특성)

  • Porous ceramics for water-permeable paving brick was prepared by the sintering of mixed materials comprising of sewage sludge ash, waste porcelain fragment, waste glaze and low-grade clay at 1,000$^{\circ}C$ for 2 h, and the physical $.$mechanical properties, the permeability and the freeze-thaw resistance of specimens with preparation parameters were investigated. The physical mechanical properties were increased in specimens while porosity and permeability were decreased with increasing sewage sludge ash content and sintering temperature on the properties of specimens showed the opposite results. The bulk density, porosity, compressive strength and permeability (passed charge) of 30A60F specimens with 30 wt% of sewage sludge ash content, waste porcelain fragment size with 1∼2 mm and sintered at 1,000$^{\circ}C$ for 2 h were 2.17, 46.2%, 221 kgf/$\textrm{cm}^2$ and 3,150 coulombs, respectively. The permeability was increased with increasing waste porcelain fragment size, however compressive strength was decreased. The freeze-thaw resistance of 30A60F specimen with 1∼2 mm of fragment size was superior to that of the other specimens. The 30A60F specimens can be used for the water-permeable paving brick with the high permeability and adequate strength. The heavy metals included in the all specimens showed lower than the standard level.

Quality Characteristics of Yogurt Dressing Added with Mulberry Juice (오디즙을 첨가한 요구르트 드레싱의 품질특성)

  • This study aims to determine the physicochemical and sensory qualities of yogurt dressing prepared with mulberry juice(0, 10, 20, 30 and 40%). As the amount of organic acid added with mulberry juice increased, malic acid, acetic acid and citric acid contents increased, while tartaric acid content decreased. The viscosity of the control group(5033.33 cP) was higher than that of the others(3043.33~736.67 cP). The pH of the control group(3.38) was lower than that of the others(3.38~3.45). The sweetness of the control group(15.80) was lower than that of the others(16.30~16.83). As for the colors, L value decreased significantly, whereas a value and b value increased significantly with more mulberry juice added. DPPH radical scavenging activity of the control group was 8.61%, whereas that of yogurt dressing samples with mulberry juice ranged from 21.98~50.65%. The sensory property results showed that yogurt dressing with 20% mulberry juice added group had the highest value in terms of color, taste, mouth feel and overall acceptability. Overall, the result of this study indicates that the yogurt dressing containing 20% of mulberry juice was most preferred among the groups.
