A Comparison between Diploid and Tetraploid Cultivars of Lolium multiflorum Lam, italicum (이탈리안 라이그라스의 2배체와 4배체 품종간 비교)

  • The influence of ploidy level on agronomic characteristics in italian ryegrass, Lolium multiflorum Lam, italicum, was studied using diploids (2x) and tetraploids (4x) on the upland of Livestock Experiment Station, Suweon. The results are summarized as follow: 1. Tetraploid cultivars are in general dark green in leaf color and strong glittered. 2. Diploid cultivars are more resistant to cold than tetraploids but degree of resistance to cold depends on cultivars. The di-and tetraploids did not differ in summer survival and coldand heat resistance of di-Itetraploid cultivars were not related. 3. Tetraploid cultivars have larger leaf-blades, more rapid growth and higher leaf ratio to stem by heading than diploids. 4. Tetraploids possess significantly lower dry matter content than diploids. 5 . Fresh- and dry weight of tetraploids are similar with that of diploids but tetraploids are more leafy than diploids. 6. Seasonal yield depends more on the type of the cultivar than on its tetraploid nature.

Morphological and Cytogenetic Analysis of Colchicine-induced Tetraploids of Fallopia multiflolra Haraldson (Colchicine 처리에 의해 유기된 4배체 하수오의 형태 및 세포유전학적 특성)

  • Background: For stable induction of tetraploidy in Fallopia multiflora Haraldson, colchicine was treated to establish the condition of induction and investigated the morphological and cytogenetic traits of the tetraploid plants obtained compared to those of diploid ones. Methods and Results: For the induction of tetraploidy, F. multiflora plants were soaked in aqueous solutions of colchicine at various concentration (0.1, 0.5, and 1.0%). After this, 2% dimethyl sulfoxide (DMSO) was added at room temperature on a shaker set at 150 rpm for periods of 12, 24, and 48 h. The induction rate of tetraploids appeared to be the highest in plants treated with 0.5% colchicine for 24 h. As the colchicine concentration and soaking time increased above these levels, the growing tip of the roots did not develop and they began to rot. When compared to diploid plants, tetraploids differed greatly in various characteristics, including the sizes and shapes of the leaves, fruits, flowers and roots. The induced tetraploid F. multiflora had larger guard cells, and chloroplasts, increased number of chloroplast in the guard cells and decreased stomatal densities. Conclusions: When colchicine induced plants for tetraploid, it can be distinguished from diploids, in various characteristics such as morphological changes as stomatal size, number of chloroplasts per guard cell, number of chromosomes and flow cytometry. Therefore, it proved that these methods are suitable, quick and easy methods for the identification of the ploidy level of F. multiflora.

Cytotype distribution and ecology of Allium thunbergii (= A. sacculiferum) with a special reference to South Korean populations

  • Polyploidization plays an important role in generating the current high diversity of plants. Studies of the distributional patterns of diploid and derivative polyploid races have provided important insights into the evolutionary process and cryptic speciation by polyploidization within and between closely related taxa defined on the basis of their morphology. Allium thunbergii and A. sacculiferum, occurring throughout eastern Russia, eastern China, Korea, and Japan, are examples of closely related species with unsolved taxonomic relationships. A total of 97 and 65 individuals from 26 and 13 populations of A. thunbergii (including var. thunbergii, var. deltoids, and var. teretifolium) and A. sacculiferum, respectively, were studied to determine their ploidy. The geographic structure and habitat differentiation of the cytotypes were also analyzed. The main cytotype of A. thunbergii was diploid (92.3% in total; the rest were tetraploids). In contrast, the majority of A. sacculiferum plants were tetraploids (69.2% of the total; the rest were diploids). No populations of the studied taxa harbored both cytotypes. Allium thunbergii was more often found at higher elevations than A. sacculiferum, and it tended to occur more frequently on rocky slopes and below forests in mountainous areas. On the other hand, A. sacculiferum occurred at forest margins and in lowland pastures. The cytotypes differed with respect to the elevation; diploids were found more frequently at higher elevations than tetraploids. The results of this study and additional biosystematics data indicate that the morphological characteristics of A. thunbergii and A. sacculiferum may be influenced by polyploidization and by their adaptation to various habitat conditions and that A. thunbergii and A. sacculiferum do not clearly fulfill the requirements of any species concept. Consequently, we propose that A. sacculiferum be considered as an additional synonym of A. thunbergii. Additionally, Allium thunbergii var. deltoides is unified into A. thunbergii var. thunbergii.

An efficient strategy for blocking the 1st mitotic cleavage of fish zygote using combined thermal treatment, exemplified by mud loach (Misgurnus mizolepis)

  • Blocking the first mitotic cleavage of the zygote is a key tool for chromosome-set manipulations in fish. We developed an improved method for inducing tetraploidy by blocking the mitosis with a combination of heat shock at 40.5$^{\circ}C$ for 1, 2 or 3 min followed by cold shock at $1.5^{\circ}C$ for 30, 45 or 60 min. When applied during the first cleavage metaphase of mud loach (Misgurnus mizolepis) zygotes, the optimal combination was heat for 2 min followed by cold for 45 min. At 1 month, the frequency of 4N survivors and the yield from total eggs fertilized was 55.7% and 14.4%, respectively, compared to heat shock alone with 20.0% efficiency and 3.6% yield. The effectiveness of the procedure was confirmed by diploid mitotic gynogenesis using transgenic markers. The overall yield of homozygous diploids, 34.0%, was better than that for single heat shock, 17.3%. The tetraploids and homozygous diploids had higher early mortality than normal diploid controls. However at 1 month, the viability of the tetraploids was the same as normal diploids. For gynogenetic diploids, the survival was similar to normal diploids after 3 months. The high efficiency of this new protocol extends the opportunity to study polyploidy in basic and applied research.

Difference of Susceptibility on Bentgrass Cultivars to Pink Snow Mold Caused by Microdochium nivale (벤트그라스 품종이 Microdochium nivale에 의한 자주색설부병에 대한 감수성 차이)

  • The susceptibility of cultivars of three bentgrass species (creeping, colonial, and velvet bentgrass) was evaluated on detached leaves assays with pink snow mold 9 isolates caused by Microdochum nivale in Petri dishes and whole plants under controlled conditions. The pink snow mold isolates obtained from infected turfgrasses on golf courses in Wisconsin were tested on response of fungicides and temperature. Detached leaf assay and susceptibility of bentgrass cultivars were evaluated with potted adult seeding during 80 days. Nine isolates were susceptible to two fungicides and were significantly different among isolates. Mycelial growth was varied in response of temperatures among isolates. There were significant differences in development and colonization of the fungus on detached leaf assay among bentgrass species include culvitars. There were significant differences on whole plants in disease severities among the three bentgrass species, particularly between tetraploids (creeping and colonial) and diploid (velvet) species, and among cultivars within each species, indicating that there are varying levels of susceptibility in species and cultivars to M. nivale. This study could be applied to evaluate the susceptibility of bentgrass to pink snow mold and also to predict a prospective evaluation of bentgrass cultivars to pink snow mold in fields in a breeding program.

The effect of hydropriming and explants on shoot initiation and tetraploids in small watermelons

  • Hydropriming had positive effects on the time for germination to reach 50%, the germination index, the time to final germination percentage, and the number of uniform seedlings with enlarged cotyledons in in vitro germination of small watermelon. In addition, the highest shoot initiation was obtained from hydroprimed cotyledonary nodes ($95{\pm}6%$), followed by non-primed cotyledonary nodes ($78{\pm}6%$), hydroprimed cotyledons ($72{\pm}4%$), and non-primed cotyledons ($48{\pm}4%$). Meanwhile, no shoots were initiated from hypocotyls. The total number of shoots that initiated from cotyledonary nodes and cotyledon explants was insignificant, indicating that both cotyledons and cotyledonary node were good sources for the in vitro culture. Choosing explant sources that favor tetraploidy should be the key for producing higher polyploidy plants; a total of 10.5% of tetraploid regenerants were entirely identified from cotyledon explants. Cotyledons with highly differentiated cells might show higher variations than cotyledonary nodes with more preexisting meristematic cells. Cells of cotyledon tissue might undergo changes in ploidy level during differentiation of the culture, or it might be that some of the variations were already present in the tissues of the donor plant. Morphological changes in fruit length of tetraploid regenerants are genotype-dependent.

Effect of Colchicine on the Induction of Prunella vulgaris for. albiflora Nakai

  • This study was conducted to find out the effective induction method of tetraploid plants to obtain potential data for cultivating superior varieties by colchicine treatment. The seed germination were decreased by the higher concentration of colchicine treatment and longer soaking time. A total of 907 individuals were germinated in 16 treated plots except control (untreated plot) and 28 tetraploids were induced which was about 3.1% of the number of seed germinated. The plant regeneration rate by colchicine treatment on explant of Prunella vulgaris for. albiflora Nakai under in vitro culture was decreased with the higher concentration of colchicine. While a total of 312 individuals were regenerated in all treatments, the explant was soaked in more than 0.05% for over 1 hour, tetraploid could be obtained. In particular, for the soaking treatment in 0.05% for 6 hours and 12 hours, 37 tetraploids were induced, which was about 57.8% of the number of plant regenerated. In accordance with the observation on doubling of DNA contents in leaf in order to identify polyploid, the peak DNA content of G1 phase was 101.3 for diploid and 197.2 for tetraploid. The result confirmed the doubling of DNA content. Furthermore, the number of chloroplasts per guard cell depending on polyploid was around 10 in diploid and 19.3 in tetraploid, which was around 1.9 times as much as diploid.

AFLP analysis to assess genomic stability in Solanum regenerants derived from wild and cultivated species

  • The cultivated potato as well as its tuber-bearing relatives are considered model plants for cell and tissue culture, and therefore for exploiting the genetic variation induced by in vitro culture. The association between molecular stability and tissue culture in different genetic backgrounds and ploidy levels has already been explored. However, it still remains to be ascertained whether somaclonal variation differs between callus-derived chromosome-doubled and undoubled regenerants. Our research aimed at investigating, through amplified fragment length polymorphism (AFLP) markers, the genetic changes in marker-banding patterns of diploid and tetraploid regenerants obtained from one clone each of Solanum bulbocastanum Dunal and S. cardiophyllum Lindl (both 2n = 2x = 24) and tetraploids from cultivated S. tuberosum L. (2n = 4x = 48). Pairwise comparisons between the banding patterns of regenerants and parents allowed detecting considerable changes associated to in vitro culture both at diploid and tetraploid level. The percentages of polymorphic bands between diploid and tetraploid regenerants were, respectively, 57 and 69% in S. bulbocastanum and 58 and 63% in S. cardiophyllum. On average, the frequencies of lost parental fragments in regenerants were significantly higher than novel bands both in S. bulbocastanum (48 vs. 22%) and S. tuberosum (36 vs. 18%) regenerants. By contrast, in S. cardiophyllum, a similar incidence of the two events was detected (32 vs. 29%). Our results revealed that structural changes after tissue culture process strongly affected the genome of the species studied, but diploid and tetraploids regenerated plants responded equally.

Effect of Antimitotic Agent Colchicine on In Vitro Regeneration of Watermelon

  • In vitro cultures of watermelon were treated with antimitotic agent colchicine to induce ploidy alterations, particularly the induction of tetraploids. Explants cotyledon, embryonic end of seed, transverse sections of epicotyl and hypocotyl were cultured on MS media supplemented with BA ($1{\mu}M$) and colchicine ($0.01\%,\;0.05\%\;and\;0.1\%$). Explants were subcultured on colchicine free media after 4 and 7 days. Colchicine had negative effect on in vitro regeneration but this exhibited explants related response. However, hypocotyl section of seedlings induced maximum callus on $0.01\%$ colchicine. Shoot proliferation was more in cotyledon explants cultured on colchicine ($0.01\%$) for four days. Maximum root induction and root number were recorded in embryonic end explants. Overall, cotyledon and embryonic end explants, and low colchicine concentration ($0.01\%$) was found optimal in watermelon regeneration.

Cytogenetic Studies of Some Tetraploids in Allium (Allium속내 수종의 사배체에 대한 세포유전학적 연구)

  • The present paper was carried out to clear up the polyploidal constitution and the banding pattern of three species in tetraploidal Allium(X=8) through the studies of meiosis, karyotype and G-bands. From the meiotic data and G-banding patterns obtained under this study, it is confirmed that A. tuberosum and A. chinense are autotetraploids, and A. senescens is allotetraploid. Some aneuploids out of the employed species were found; A. senescens is 2n=34, and A. chinense is 2n=33. The chromosome types of these species are meta-and submetacentrics except the sat-chromosomes and the f-chromosomes. G-bands of these species are generally located in the end of each arm. A. senescens is similar in the quantity of heterochromatin with A. chinense, but A. tuberosum has a little than the other species. The quantity of heterochromatin is higher in small groups of chromosome than large ones, and higher in short arms than long arms.
