Optimization of the Lowry Method of Protein Precipitation from the H. influenzae Type b Conjugate Vaccine Using Deoxycholic Acid and Hydrochloric Acid

  • The Lowry method was used in this study to measure protein in Haemophilus influenzae type b (Hib) conjugate vaccines (polyribosylibitol phosphate-tetanus toxoid; PRP-TT) using deoxycholic acid (DOC) to induce protein precipitation. Trichloroacetic acid (TCA) did not induce precipitation adequately from the Hib conjugate bulk and the freeze-dried Hib conjugate product. Its yield was approximately 50%. The matrix structure of Hib conjugate inhibits precipitation by TCA. Although the Lowry method can be carried out without precipitation in Hib conjugate bulk when no residual impurities (adipic acid dihydrazide [ADH], 1-ethyl-3-(3-dimethylamino-propyl) carbodiimide-HCI [EDAC], phenol and cyanogens bromide [CNBr], etc.) are present, it cannot be used for Hib conjugate products that contain sucrose 8.5%, because 8.5% concentration of sucrose enhanced the protein concentration. DOC- and HCl-induced precipitation is an alternative method for evaluating the protein content of the Hib conjugate bulk and the Hib conjugate product. The precipitation was optimal with a final concentrate of 0.1% for DOC at $4^{\circ}C$ and pH 2. This Lowry method, using DOC/HCI precipitation to induce protein precipitation, was confirmed a consistent, reproducible, and valid test for proteins in Hib conjugate bulk and its freeze-dried product.

The Relationship between Retention of the Maternal Child Health Handbook, Awareness of DPT Additional Immunization and DPT Additional Immunization (모자보건수첩보유, 디피티 추가 예방접종에 대한 인지와 디피티 추가 예방접종 실천간의 관계연구)

  • Purpose: This study was aimed at investigating the relationship between retention of the maternal child health handbook, awareness of DPT additional immunization and DPT additional immunization, and to provide basic information to enhance the DPT additional immunization rate. Method: The study subjects were 312 women whose children were between four and six years old, and residing in six provinces of Gyungsangnam do. Data was collected with a 10 item questionnaire by interviews from July 1st to 30th, 2003, and analyzed with descriptive statistics and X2 test at a significance level of 0.05, by two tailed test. Results: The awareness of DPT additional immunization was significantly higher in the women who retained the maternal child health handbook, than their counterparts. The rate of DPT additional immunization was significantly higher in the women who were aware of the DPT additional immunization or who retained the maternal child health handbook, than their counterpart. Conclusion: The DPT additional immunization rate was related to retention of the maternal child health handbook and awareness of DPT additional immunization. Therefore it is recommended that measures be developed and taken to increase the retention rate of the maternal child health handbook and awareness of DPT additional immunization, to enhance the rate of DPT additional immunization.

Effects of Cicadae Periostracum (CP) in Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice (DNCB로 유발된 생쥐의 알레르기성 접촉피부염에 선태(蟬蛻)가 미치는 영향)

  • Objectives In the theory of Korean Medicine, Cicadae Periostracum (CP) has been used to treat skin diseases such as inflammatory dermatitis, tetanus and pruritus. CP can reduce heat and disperse wind. In prior studies, anti-allergic effect and anti-inflammatory effect of CP were reported. However, there has been no report regarding the correlation of CP and allergic contact dermatitis (ACD). This study was performed to show the effects of CP in ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. Methods In this experiment, the effects of CP on biological changes were measured, such as changes in ear and spleen weight, ear and dorsum skin thickness, clinical aspect on the dorsum skin and histological changes. The effect of proliferation rate of splenocytes was also investigated in vitro and vivo study. Results In results, CP application (CPA) group and CP application and administration (CPAA) group were significantly restrained in ear weight gain and increase in ear and dorsum skin thickness compared to the control group. In addition, CPA and CPAA group showed diminished erythema, desquamation, bloodstain, and marks. Also, the histological assessment showed that CP treatement diminished thickness of epidermis, hyperkeratosis, pigmentation and parakeratosis. Conclusions In conclusion, these data suggest that CP can decrease symptoms of ACD.

  • I have come to conclusion as follows about Bangpungtongsungsan after literatural study. 1. Bangpungtongsungsan is the medical treatment of heat, wind and dryness. 2. Bangpungtongsungsan is used in head-wind, dizziness, both eye disease, tinnitus, both ear deafness, nasal polyp, uriticaria, sajuabi, beard and hair falling, apoplexy, paralysis of hands and feet, dull mentality, tetanus, epidemic disease characterized by swelling and redness of face, carbuncle, daepungchang, pustule, syphilis, tinea capitis and so on. 3. Bangpungtongsungsan is used in the disease of cerebral hemorrhage, hypertension, arteriosclerosis, constipation, rosacea, hemorrhoids, cutaneous disease, empyema, eye disease, diabetes, asthma, obesity, beriberi, erysipelas, baldhead, fatty heart, chronic nephritis and so on. 4. Bangpungtongsungsan fits in Taeumin who has much wetness-heat and those who have much heat or have much heat but don't give off well. And it doesn't fit in the disease of intolerance to cold and fever in the form of weakness headache, asthenia of the spleen and stomach, cold by internal disorder and so on.

An Animal Model to Evaluate the Protective Efficacy of Haemophilus influenzae Type b Conjugate Vaccines

  • An efficacy test of PRP (polyribosylribitol phosphate)-TT (Tetanus toxoid) conjugate vaccines was carried out using BALB/c mice as an animal model by inoculating Haemophilus in­fluenzae type b (Hib) with a virulence enhancement factor (VEF). Three administrations of the conjugate vaccines at 2-week intervals elicited a significantly high level of PRP antibodies (P>0.0001). The protective activity of the PRP immunization was challenged with either Hib with iron dextran (Hib/) or with a combination of mucin and hemoglobin (Hibmh) as a VEF. The me­dium lethal dose $(LD_{50})$ for Hibmh and Hibiwas measured as 10 CFU (Colony Forming Unit) and $2.5{\times}10^{8}$ CFU respectively. Each immunized animal was challenged with five or ten times the $LD_{50}$ level of bacteria with a VEF. A significant difference in mortality between the immunized and control mice (P> 0.01) was observed with the Hibmh challenge inoculation but not with the Hibi challenge inoculation. These results show that a combination of mucin and hemoglobin was able to enhance the virulence of Hib in BALB/c mice to cause a lethal infection, thus suggesting that BALB/c mice introduced to this method can be an effective model animal for testing the protective efficacy of H. influenzae conjugate vaccines.

Primary Structure of the Human VkII Regions Elicited by Haemophilus influenzae Type b Polysaccharide Vaccines; The J Gene Usage Is Restricted in Child Antibodies Using the A2 Gene

  • The Haemophilus influenzae type b (Hib) has been a major cause of bacterial meningitis in children who are less than two years old. The variable (V) region repertoire of adult Caucasian antibodies (Abs) to Hib polysaccharide (PS) has been characterized well. The majority of adult antibodies against Hib uses VL that is derived from the Vk gene A2 and have arginine at the N region. In order to explore the possibility those antibody responses to Hib-PS is variable in various age groups, we examined the VL regions of the antibodies to Hib-PS in Korean adults and children. We immunized Korean adults (n = 8) and children (n = 39) with Hib tetanus conjugated vaccines, isolated RNAs from the peripheral lymphocytes, and amplified the A2-derived VL regions by RT-PCR. The PCR products were subcloned and sequenced. Forty-seven out of 54 independent clones from children used the $J{\kappa}2$, or $J{\kappa}3$ gene in preference. The adults, however, used all of the $J{\kappa}$ genes evenly. With respect to the amino acid sequences of variable regions, adult $A2-J{\kappa}$ recombinants have a germline sequence. But, the 76th codon (AGC) of child $A2-J{\kappa}2$ recombinants was substituted with CGC (arginine) in most cases (88 %) and 77 percent of child clones using the $A2-J{\kappa}3$ genes have isoleucine-109 at the junction of $J{\kappa}-C{\kappa}$ instead of threonine that is found in a germline sequence. These results suggest that the mechanism of antibody production in young children is different from that of adults.

  • To use basic data for health planning and evaluation of Kangwha community health project of Yonsei University, a study on death rates and causes of death were investigated in two townships (Naega and Sunwon Myuns) in Kangwha County from April 1, 1975 to March 31, 1977 All death was identified and reported by family health workers who are living in each village and 2 trained public health nurses confirmed the death. The causes of death were investigated by 2 public physicians. Total number of deaths for 2 years was 230 and the followings are brief summary of the study. 1. Age-adjusted crude death rates of study area were 8.69 per 1,000 population in 1975 and 7.18 per 1,000 population in 1976. Age-adjusted crude death rates for male were 9.18 in 1975 and 6.38 in 1976 and for female were 8.33 and 7.80 per 1,000 population 2. Age specific death rate curves by year and sex showed 'U' shapes. 3. Infant and neonatal death rates were 30.08 and 22.56 per 1,000 live births in 1975, and the rates in 1976 were 18.18 and 13.64. 4. The most common cause of death was cerebrovascular disease and average cause specific death rate for the disease was 215.5 per 100,000 population. 5. Four leading causes of death were non-infectious origin; cerebrovascular disease, malignant neoplasms, senility and suicide. Pulmonary tubeculosis and pneumonia occupied 5th and 9th causes of death. 6. Stomach cancer and hepatoma occupied 61.3% of total death due to malignant neoplasms. 7. Most frequent cause of neonatal death was birth injury. Two deaths due to tetanus were found in 1975, but no death due to this disease was found in 1976. 8. About half of deceased received care from physician before death.

Impacts of Host Immunization on the Translocation of Intestinal Bacteria and Growth Performance in Weanling Piglets

  • Effects of host immunization on bacterial translocation and growth performance in weanling piglets were studied. Twenty four barrows were assigned to one of two immunization treatments: Control group (CON: immunized with placebo) or Immunization group [IMMU: immunized with Antigen cocktail; Keyhole limpet hemocyanin (KLH), Ovalbumin (OA), and Tetanus toxoid (TT)]. On d0, piglets were weaned and intramuscularly immunized with 2 ml of placebo or Antigen cocktail, respectively. Antigen-specific Ig titers were determined by ELISA (Enzyme Linked ImmunoSorbent Assay). Ig titers to E. coli-derived lipopolysaccharides (LPS) were measured as the indicator of bacterial translocation. Ig titers to LPS were higher (p<0.10, 0.05 or 0.01) in CON group before immunization (d0), but the difference disappeared with time and IgA titers to LPS became higher (p<0.05) in IMMU group on d39. In IMMU group, IgG titers to LPS from d28 onwards showed positive correlations (p<0.10, 0.05, 0.01 or 0.001) with IgG titers to KLH from d11 onwards and with IgM titers to KLH from d7 onwards. Generally, growth performance was negatively related to IgG titers to LPS. Average daily gain for d28 to d35 showed negative correlations (p<0.10, 0.05, or 0.01) with IgG titers to LPS on d28 onwards in immunization group. These results reveal some evidences that host immunization might facilitate bacterial translocation and high humoral immune responses to LPS are negatively related with the growth performance.

A Study on Application of Radix Saposhnikoviae Main Blended Prescription From Dongeubogam (동의보감(東醫寶鑑) 중(中) 방풍(防風)이 주약(主藥)으로 배오(配伍)된 방제(方劑)의 활용(活用)에 대한 고찰(考察))

  • This report describes 71 studies related to the use of Radix Saposhnikoviae main blended prescription from Donguibogam. The following conclusion were reached through investigations on the prescriptions that use Radix Saposhnikoviae as a main ingredient. Prescriptions that Radix Saposhnikoviae was taken as a monarch drug are utilized for 20 therapeutic purposes, for example, eye disease, wind disease, head disease and Sang Han syndromes. In particular, 28.1% of prescriptions appear in the chapter of eye, and 25.3% of those appear in the chapter of head. Prescriptions that utilize Radix Saposhnikoviae as the main ingredient are used in the treatment of eye disease. paralysis and tetanus. Radix Saposhnikoviae is used in pathogenic factors such as wind. wind with cold. and used in pathology related to liver system. The dosage of Radix Saposhnikoviae in prescriptions is 7 li(about 0.26 gram) to 2.5 jeon(about 9.38 gram), however 1 jeon(about 3.75 gram) has been taken the most for clinical application. Bangpungtang is the most useful base prescription which use the Radix Saposhnikoviae as the main ingredient.

An analysis of Clinical Studies on Galgeun-tang (갈근탕의 임상연구 논문 분석)

  • Objective : This study's purpose was to review the clinical studies of Galgeun-tang. Method : We searched papers about Galgeun-tang using KISS, RISS, OASIS, PUBMED and J-stage. The key words we used were "Galgeuntang", "Kakkonto", "Ge gen tang", and "Pueraria Decoction". Papers not matched with inclusion criteria were excluded. Results : Until today, there have been 223 studies on Galgeun-tang. Of these, 25 studies were classified as clinical research papers. There were 4 cases of fever, 5 cases of inflammation on respiratory system, 3 cases of head, neck and shoulder disorder, 2 cases of diarrhea, 6 cases of pharmacokinetics and interaction, 5 cases of side effect. Conclusion : It can be seen that Galgeun-tang has established the basis for application to the purpose of treating fever (common cold, influenza), inflammation on respiratory system (nasal obstruction, maxillary sinus retention cyst, mucoid pseudomonas aeruginosa chronic lower respiratory tract infection), head, neck and shoulder disorder (temporomandibular disorders, shoulder stiffness, tetanus), and diarrhea. On the other hand, considering 4 cases of side effect on drug eruption, caution should be exercised when observing the progress of the patient taking Galgeun-tang.
