Study of Effects of Crude Extracts of Three Plants Concerned on Optic Development of African Clawed Frog, Xenopus laevis (아프리카발톱개구리의 시각계 발생에 관여하는 식물추출물 3종에 관한 연구)

  • In this study we investigated the embryo toxicity of three kinds of plant extracts during early development of African clawed frog, Xenopus laevis through FETAX assay (Frog Embryo Teratogenesis Aassay with Xenopus). The plants used in this study were the materials of the Korean herbal medicines, Polygala tenuifolia, Lycium chinensis and Comus officinalis. The test embryos exposed to 1, 10 and $100{\mu}g/ml$ of each plant extract and control embryos were incubated for 96h at $24{\pm}0.5^{\circ}C$. The focus of this study is to elucidate the malformation due to toxicity of plant extracts, especially, to elucidate plant inducing optic malformation. As a result, the growth inhibition of embryos, optic malformation, axial distortion, cephalic and abdominal edema, dysplasia of digestive track and hyper-pigmentation were occurred in all of extracts, and these malformations were increased to the increase of extract concentration. The rate of optic malformation was highest in $100{\mu}g/ml$ of Lycium chinensistreated group and 27% of tested 150 individuals showed optic hernia. The histological results showed enlarged ventriculum in brain, dysplasia of vitreous chamber in eye and unclear retinal layers.

Development of a Bioassay Method Using Aluminium Foil Sheet for Screening Ovicidal Activity Against Diamondback Moth Egg, Plutella xylostella L. and Selection of Plant Extracts with High Ovicidal Activity (알루미늄호일 이용 배추좀나방(Plutella xylostella L.) 살란활성 검정법 개발 및 살란활성 식물추출물의 선발)

  • Diamondback moth (DBM, Plutella xylostella L.) is known as the most destructive pest of cruciferous crops worldwide. As most insecticides targeted to mainly larval stage, new insecticides which have hatching-inhibitory or ovicidal activity could be more efficient to control DBM. Therefore, we developed an easy and efficient method for screening ovicidal activity of DBM eggs using aluminum foil. The aluminum foil ($4{\times}12$ cm) coated with filtered juice of chinese cabbage leave (hereinafter called oviposition foil) exposed to 300 newly-emerged adults for 24 hours inside the rearing container. The oviposition foils were replaced every 4 days consecutively after mating, but it was better to discarded over then. Oviposition foil were divided into 6 to 12 pieces depending on egg mass volume. After dipping into test solutions for 10 seconds using faucet, oviposition foil pieces were placed into common petri dish, and then investigated hatchability. The effect of methanol solvent (50%) for 10 seconds dipping on the toxicity against DBM eggs was negligible. In addition, whether covering the petri dish or not should be dependent on nature of active compounds tested. With applying the new bioassay method, methanol extracts from 50 plants were tested the ovicidal activity to DBM eggs. Among them, four plant extracts; Angelica tenuissima root, Lycium chinense root, Cnidium officinale root and Polygala tenuifolia root, showed high ovicidal activity of over 90% control efficacy, against DBM eggs.

Comparison of Growth Rate and Cold Tolerance with Basic Species, Commercial Lines, and Breeding Lines of Zoysiagrass (한국잔디류 기본종, 상업종 및 육종계통들의 생육속도 및 내한성 비교)

  • This study was conducted to compare cold tolerances and growth rates of 5 basic species, commercial lines, and breeding lines of zoysiagrass. Total 41 zoysiagrasses were tested at the northern part (Jeokseong-myeon) and the middle part (Cheonan) of South Korea. Cold tolerance (survival rate during winter), growth rate, green-up speed, and density data were collected visually. Four types of zoysiagrasses (Z. matrella, Z. tenuifolia, USm, and S4M2) with genetic characteristics of Z. matrella died during winter cold, while the others survived. Midium leaf type zoysiagrasses, such as Anyang1, Samdeock1, Anyang2, SJ21, and Pyeongdong, with genetic characteristics of both Z. sinica and Z. macrostachya showed vigorous growth rate one year after planting at the northern part (Jeokseong-myeon). Twenty nine ($70.7\%$) zoysiagrasses showed vigorous growth rate, but the other twelve ($29.3\%$) zoysiagrasses died by cold and drought condition at the middle (Cheonan) part of S. Korea. Anyang2, AJ9-7, Samdeock1, Samdeock2, Samdeock3, and Zenith-C showed vigorous growth rate even though cold and drought environmental condition were artificially provided at these region. But, SJ2-19, NSm, 88Mey, 88Mey-7S, 88Mey-9S, SJ2l-10, ASm, and DBm which did not die at the northern part, showed serious injury at the middle part of S. korea. From this results, we can conclude that drought is as important factor as low temperature for zoysiagrasses during winter.

Antioxidant Activities and Nitric Oxide Production of Medicine Plants in Gyeongsangbukdo (Carthamus tinctorius seed, Cyperus rotundus, Schizonepeta tenuifolia, Polygonatum odoratum var. pluriflorum, Paeonia lactiflora) (경북약용작물(홍화자, 향부자, 형개, 위유, 작약)의 항산화 및 Nitric Oxide 저해활성)

  • This study was carried out to search for natural anti-oxidants and anti-inflammatory compounds from 5 medicinal plants (Carthamus tinctorius seed, Cyperus rotundus, Schizonepeta tenuifolia, Polygonatum odoratum var. pluriflorum, and Paeonia lactiflora). These plants were extracted with 70% ethanol. In order to measure total antioxidant activity of flavonoids, polyphenol content was measured. Radical scavenging activities of extracts were examined using a-a-Diphenyl-${\beta}$-picrylhydrazyl ($DPPH{\cdot}$), 2,2-azino-bis 3-ethylbenzthiazoline-6-sulfonic acid ($ABTS{\cdot}$), ferric reducing antioxidant power (FRAP) and superoxide anion radical assays. C. tinctorius seed extracts showed the highest polyphenol and flavonoid contents as well as strong $DPPH{\cdot}$, $ABTS{\cdot}$, FRAP, and superoxide anion radical scavenging activity. Also, C. tinctorius seed extracts showed the highest nitric oxide (NO) production inhibitory effect. Theses results indicate that the C. tinctorius seed extracts can be used as a functional material due to their effective anti-oxidative and antiinflammatory activities.

Study on the Flora and Vegetation of the High Moor in Mt. Daeam (대암산 고층습원의 식물상과 식생)

  • The flora and vegetaion of the high moor in Mt. Daeam($128^{\circ}07^{\prime}E$, $38^{\circ}13^{\prime}N$) was investigated by the phytosociological method of Braun-Blanquet's from 13 - 14th June to 7 - 8th August, 1998. The flora of plants recorded in high moor was 191 taxa belonging to 53 families, 131 genera, 160 species, 30 varieties, 1 formas. The high moor vegetation of Sanguisorba tenuifolia var. alba community group in this survey trip was classified into 11 communities as follows : Carex dispalata community and Sphagnum palustre community at the interior of moor, Carex biwensis community and Juncus papillosus community at the regenerated area and Malinia japonica community, Spiraea salicifolia community, Geranium eriosteman var. megalanthum - Calamagrostis langsdorffii community, Juncus effusus var. decipiens community, Salix koreensis community, Caltha palustris var. membranacea community and Sparganium stoloniferum community at the margin of moor. As a result of this study, the change of plant communities on high moor was closely influenced by landization in relation to the disturbance of human since 1969.

Volatile Aroma Compounds and Their Characteristics of Labiatae by Solid-Phase Microextraction (SPME) (Solid-Phase Microextraction (SPME)에 의한 꿀풀과 약초의 향기성분과 그 특성)

  • This study was carried out to find the possibility of use of Solid Phase Microextraction (SPME) for extracting the volatile aroma compounds in the five aromatic plants (Agastache rugosa O. Kuntze, Mentha arvensis Linne, Thymus quinquecostatus Celakovsky, Elsholtzia splendens Nakai, Schizonepta tenuifolia Briquet) belongs to the Labiatae. In the result of the analysis, the volatile aroma compounds were mainly composed monoterpene alcohol (linanol, menthol, ${\alpha}-terpineol$, borneol), monoterpene ketone (limonene, menthone) and sesquiterpene (trans-caryophyllene,${\delta}-cadinene)$. The volatile aroma compounds of Agastache rugosa O. Kuntze and Mentha arvensis Linne were extracted by SPME more identified than the SDE. However, Schizonepta tenuifolia Briquet more identified by the SDE and in Elsholtzia splendens Nakai similar to the SDE. Especially, the SPME showed the sesquiterpene contents was more than the SDE. The major volatile aroma compounds were difference but the composition of those between the SPME and the SDE showed no difference. Within the results, the SPME showed the most convenient and a rapid extraction method to analysis of the volatile aroma compounds.

DNA Polymorphism and Assessments of Genetic Relationships in genus Zoysia Based on Simple Sequence Repeat Markers (ISSR에 의한 잔디속 식물의 DNA 다형성과 유전적 관계 평가)

  • The genetic variability of four species of the genus Zoysia collected from South Korea was analyzed using an inter-simple sequence repeat (ISSR) marker system. Polymerase chain reactions (PCR) with eight ISSR primers generated 86 amplicons, 76 (87.1%) of which were polymorphisms. The polymorphism information content (PIC) value of the ISSR marker system was 0.848. The percentage of polymorphic loci (Pp) ranged from 41.2% to 44.7%. Nei’s gene diversity (H) ranged from 0.149 to 0.186, with an average overall value of 0.170. The mean of Shannon’s information index (I) value was 0.250. Total genetic diversity values (HT) varied between 0.356 (ISSR-1) and 0.418 (ISSR-16), for an average overall polymorphic loci of 0.345. Interlocus variation in within-species genetic diversity (HS) was low (0.170). On a per-locus basis, the proportion of total genetic variation due to differences among species (GST) was 0.601. This indicated that about 60.1% of the total variation was among species. Thus, about 39.9 of genetic variation was within species. The estimate of gene flow, based on GST, was very low among species of the genus Zoysia (Nm = 0.332). The phylogenic tree showed three distinct groups: Z. macrostachya and Z. tenuifolia clades and other species were formed the separated clusters. In conclusion, the ISSR assay was useful for detecting genetic variation in the genus Zoysia, and its discriminatory power was comparable to that of other genotyping tools.

The Origins, Changes, and Compositive Principles of Hyungbangjihwang-tang (형방지황탕(荊防地黃湯)의 기원(基源), 변천과정(變遷過程) 및 구성원리(構成原理))

  • Objectives This paper investigates the origins, changes, and the constructive principles of Hyungbangjihwang-tang(荊防地黃湯). Methods Hyungbangjihwang-tang and other related prescriptions were analyzed in terms of their pathological indications, based on previous literature including 『Gogeumuigam(古今醫鑑)』, 『Sanghannon(傷寒論)』, 『Uihagjeongjeon(醫學正傳)』, 『Donguisusebowon·Sasangchobongwon(東醫壽世保元·四象草本卷)』, 『Donguisusebowon·Gabobon(東醫壽世保元·甲午本)』, 『Donguisusebowon·Sinchukbon(東醫壽世保元·辛丑本)』. Results and Conclusions The origins of Hyungbangjihwang-tang can be seen in three sections depending on the classification of medicinal herbs. Schizonepeta tenuifolia Briq.(荊芥), Saposhnikovia divaricata Schischk.(防風), Notopterygium incisum Ting(羌活), Aralia cordata Thunb.(獨活), and Plantago asiatica L.(車前子) originated from Hyungbangpaedok-san(荊防敗毒散) included in 『Gogeumuigam』. Poria cocos Wolf.(茯苓) and Alisma canaliculatum(澤瀉) originated from Ohryoung-san(五苓散) included in 『Sanghannon』. Rehmannia glutinosa for hueichingensis(熟地黃) and Cornus officinalis S. et Z.(⼭茱萸) originated from Yukmijihwang-tang(六味地黃湯) included in 『Uihagjeongjeon』. Hyungbangpaedok-san was transformed into "Paedok-san" in 『Chobongwon(草本卷)』, Hyungbangpaedok-san in the 『Gabobon(甲午本)』 and 『Sinchukbon(辛丑本)』. Yukmijihwang-tang was mentioned as a Soyangin medicine in the 『Chobongwon』 and was later transformed into a Suhwagije-tang(水火旣濟湯) in 『Gabobon』. The meaning of Yukmijihwang-tang inherited in 『Sinchukbon』 as Dokhwaljihwang-tang and Hyungbangjihwang-tang. The medicinal herbs in Hyungbangjihwang-tang can be categorized in three sections by their functions, Schizonepeta tenuifolia Briq., Saposhnikovia divaricata Schischk., Notopterygium incisum Ting, Aralia cordata Thunb., and Plantago asiatica L. scatter the Pyohan(表寒) and make the Pyoeum(表陰) drop. Poria cocos Wolf.(茯苓) and Alisma canaliculatum(澤瀉) make the Pyoeum(表陰) drop through Lee-su(利水). Rehmannia glutinosa for hueichingensis(熟地黃) and Cornus officinalis S. et Z.(⼭茱萸) tonify Shin-won(腎元). The each principles of medicinal herbs corresponds to the stage of the "Cold-related Diarrhea in Soyangin Spleen Cold-based Exterior Cold disease(少陽人 脾受寒表寒病 亡陰病 身寒腹痛亡陰證)".

Antimicrobial Activity and Chemical Composition of Some Essential Oils

  • In this study the composition and antimicrobial properties of essential oils obtained from Origanum onites, Mentha piperita, Juniperus exalsa, Chrysanthemum indicum, Lavandula hybrida, Rosa damascena, Echinophora tenuifolia, Foeniculum vulgare were examined. To evaluate the in vitro antibacterial activities of these eight aromatic extracts; their in vitro antimicrobial activities were determined by disk diffusion testing, according to the NCCLS criteria. Escherichia coli (ATTC 25922), Staphylococcus aureus (ATCC 25923) and Pseudomonas aeruginosa (ATTC 27853 were used as standard test bacterial strains. Origanum onites recorded antimicrobial activity against all test bacteria, and was strongest against Staphylococcus aureus. For Rosa damascena, Mentha piperita and Lavandula hybrida antimicrobial activity was recorded only to Staphylococcus aureus. Juniperus exalsa, and Chrysanthemum indicum exhibited antibacterial activities against both Staphylococcus aureus and Escherichia coli. We also examined the in vitro artimicrobial activities of some components of the essential oils and found some components with antimicrobial activity.

Effects of Some Sedative Oriental Medicines on Neurotransmission and Antioxidative System in vitro (신경안정 생약 추출몰이 in vitro에서 신경전달효소 및 항산화계에 미치는 영향)

  • The extracts of Euphoria longan, Ziryphus jujuba, Thuja orientalis, Polygala tenuifolia, Acorus gramineus, Cyperus rotundus, Poria cocos, Uncaria rhynchophylla, and Albizzia julibrissin, which have been used as sedative drugs in Korean folk medicine, were evaluated for their effects on neurotransmission and antioxidative system in vitro. Among the tested drugs, Acorus gramineus showed most inhibitory activities on monoamine oxidase, xanthine oxidase, aldehyde oxidase, and lipid peroxidation and Uncaria rhynchophylla also inhibited most effectively GABA transaminase and DPPH radical.
