Characteristics and Sampling of Dioxins/Furans from Emission Gas and Fly Ash Produced in Municipal Waste Incinerator (도시 소각로 쓰레기 소각후 생성된 폐가스 및 비산재중에 포함된 다이옥신류의 측정 및 특성고찰)

  • Polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofuransfurans are the archetype of toxic chemicals. So it has absorbed public attention. The majors primary sources of PCDDS and PCDFs are chemical, thermal and photochemical reactions. Municipal solid waste incinerator facilities has been reported as the major contributors of dioxins to the environment. In this paper, Dioxins and furans were examined emission gas and fly ash produced during combustion in municipal solid waste incinerator. More effective method for sampling, extraction was described. The sample was extracted using a soxhlet method and purified using silicagel, alumina and carbon fibre HPLC to remove interfering compound. The extract was then analyzed by HRGC/HRMS. The result of this study showed recovery standard was good and the data resembled those of thermal processes. Total dioxins and furans were $1076.20pg/Nm^3$ and $1452.34pg/Nm^3$ respectively. The amount of highly chlorinated compound was more than that of lowly chlorinated compound. The 2,3,7,8-substituted TCDD was Just 0.34% of the total dioxins/furans amount.

Endocrine Disruptive Potentials in Surface Water Samples from Taihu Lake, Yangtze Delta

  • Taihu Lake is a major water source for part of Yangtze Delta, which is one of the most urbanized and economically prosperous areas in China. In last couple of decades, some parts of the lake were highly polluted due to eutrophication. This study analyzed dioxin-like potential and mutagenic potential in surface water samples from Taihu Lake. The samples were prepared by XAD-2 resin procedure. A batch of biological assays, including dioxin-like potential microassay with the rat hepatocyte cell line H411E, and Ames test was employed in the research. Results showed that jour water samples have high content of dioxin-like biological potential, the highest activity TEQ to 2,3,7,8-TCDD was 48 pg/ι in sample 1. The mutagenic effect with reading-frame shifting mechanism was confirmed in 3 of 4 samples. The effective sewage treatment facilities and reliable monitoring surveillance system are urgently needed for this area.

  • This study analyzed and standardized the harmful substances of organic compounds(dioxins) in disposable diapers for infant's clothing. Commercial samples obtained from Korea, Japan, America and German were determined by HRGC/HRMS. Experimental Methods were consisted of comparision of extracting solvent, analysis of PCDD/Fs, and analysis of PCDD/Fs of extraction in disposable diaper. The results were as follows. No high TEQ values in TCDD(1), PCDD(0.5) and HxCDD(0.1) were found for commercial disposable diapers from country Korea, Japan, U.S.A and German. In extraction for 6hrs experiments, HpCDD and OCDD were detected in disposable diapers of U1, U2 and G2, respectively. In extraction fur 24hrs experiments, HpCDD, OCDD and OCDF were detected in disposable diapers of J3, U1, U2 and U3. So we need to set standards of measurement for harmful materials contained in textile goods. This will make a safe and comfortable clothing environment for all users, regardless of age.

  • The research in this paper centers on a comparative risk assessment for nearby air pollution by carcinogenic metal emission from municipal solid waste incinerators. If a substance is identified as a potential human carcinogen, the carcinogenicity may be related to the chemical form of a substance and the route of exposure. This type of information with regard to carcinogenic uncertainty is incorporated into hazard quantification. In addition to the dioxin emission, the metal emission from municipal solid waste incineration is found to be a major contributor to human cancer risk via the inhalation route. The magnitude of risk by metals is about 5 times greater than that of risk by dioxins. Hexavalent form of chromium and cadmium compounds are major contributors to cancer risk from metal emission. In addition, hexavalent chromium is known to be human carcinogen while 2,3,7,8-TCDD is known to be only probable human carcinogen.

  • To evaluate the effect of Choakwiyeum water extract on the male reproductive functions, experiments were performed on male rats. The results obtained were as follows: 1. Intromission frequency was increased significantly after the administration of Choakwiyeum water extract in male rats. 2. Intromission latency was increased significantly after the administration of Choakwiyeum water extract and ketoconazole in male rats. 3. Mount frequency was increased significantly after the administration of Cboakwiyeum water extract and TCDD in male rats. 4. Serum testosterone concentration was increased significantly after the administration of Choakwiyeum water extract in male rats. 5. Serum testosterone concentration was increased significantly after the administration of Choakwiyeum water extract and ketoconazole in male rats.

  • Objectives : To propose a feasible, valid and appropriate study designs and epidemiologic methods for evaluating the adverse health effects of Agent Orange-chemical defoliants used in Vietnam- in Korea. Methods : A literature study was peformed on Agent Orange, herbicides, pesticides and dioxins. The study subjects, study design, exposure assessment and health outcomes assessment were examined in each study. The potential data sources for the study subjects, study design, exposure assessment and health outcomes assessment in Korea were investigated. Results and Conclusion : In earlier Korean studies, research subjects for studying the effects of Agent Orange were identified from the patients or persons who claimed to have Agent Orange-related diseases due to the difficulties in identifying the entire population of Vietnam veterans in Korea. In this study, an attempt was made to identify the total number of Vietnam veterans in Korea. As a result, the addresses of 20,000 Vietnam veterans were obtained. It is proposed that a retrospective cohort design on a sample of the total number of Vietnam veterans is a feasible and appropriate study design. Self report questionnaires and military records were proposed to assess the exposure level. It is believed that measuring the plasma or tissue TCDD should be used only for a validation study assessing the level of exposure. For the health outcomes assessment, it is possible to obtain the mortality, cancer frequency, physical examination, screening and medical insurance record data.

  • The purpose of this meta-analysis was to investigate the anti-hepatotoxic effect of ginseng in rats induced with CC14 or TCDD, the toxicities that cause liver damages. Primary studies were collected from the ScienceDirect database, the DBpia, and the KISS. The data on the effect factors in plasma and in enzyme are listed as many as possible: The effect factors were alanine transaminase(ALT), aspartate transaminase(AST), liver aminopyrine N-demethylase(AD), liver aniline hydroxylase(AH), liver 3,4-Methylenedioxyamphetamine(liver MDA), cytochrome P450(P450), serum alkaline phosphatase(ALP), serum lactate dehydrogenase(LDH), cytochrome b5(Cyto b5), glutathione reductase (GR), Liver glutathione S-transferase(GST), liver glutamyltransferase (GT), Liver($\gamma$-GCS), serum liver 3,4-Methylenedioxyamphetamine(serum MDA), serum sorbitol dehydrogenase(SDH), serum total protein(TP), and serum $\gamma$-glutamyltransferase($\gamma$-GT). In order to investigate the effect of ginseng, the standard mean difference(HG) between the group of rats induced with toxicity(RH) and the group of rats induced with ginseng(RHG) were combined, and the significance of HGs were tested. The combined HGs checked the biases caused by heterogeneity among studies and the publication biases. Then they were adjusted by using the random effect model and trim and fill method. Although the publication biases were assumed, among all plasma factors the HGs of ALT, AST, serum MDA, SDH, TP, and $\gamma$-GT were significant, and among all enzyme factors the HGs of liver MDA, Cyto b5, GR, GST, and GT were significant. The treatment with ginseng significantly affected the plasma and enzyme levels in rats induced with toxicity.

Analysis of biological functions of rpt-1 in human cells with exposure to environmental pollutants (환경오염물질 폭로에 따른 인체세포에서의 rpt-1 발현 및 역할의 분석)

  • Abel et al. in Germany discovered a new dioxin-responsive gene, which has later been identified as rpt-1 (regulatory protein T-lymphocyte 1). While it is speculated that rpt-1 may play a role in signal transduction and carcinogenesis, its roles and functions remain unknown. The present study attempted to analyze functions of rpt-1 in human epithelial cells following the xenobiotic exposures. While German counterpart analyzed expressionn of rpt-1 in spleen and thymus cells from mouse and rat and characterizes molecular properties of the gene, our work mainly focused on analyzing function of rpt-1 in human skin cells. Expression of rpt-1 in human cells were analyzed by western and northern blot RT-PCR analysis. Expression of rpt-1 as well as Staf-50 in human cells with or without exposure to environmental pollutants were also analyzed by northern blot analysis, since Staf-50 is homologous with rpt-1 and found in human cells. To help study roles of rpt-1 in human cell system, retroviral vector system carrying rpt-1 gene under the CMV promoter were constructed and transfected. Cells overexpressing the gene after the transfection showed an increase of cell density and soft agar colony formations, as compared to the control cells, suggesting that rpt-1 may play a certain role in the transformation processes of human cells. While the expression of rpt-1 in spleen and thymus is known to be strong in the laboratory animals, both the basal and TCDD-induced expression of rpt-1 in the current cellular system remained insignificant. It is speculated that the expression pattern of rpt-1 may be tissue- and species-specific. The present study demonstrated a strong expression of rpt-1 protein in the brain of SD rat model. Since there is no previous report on the expression of rpt-1 in the brain tissue, the result may play a significant role in understanding dioxin-induced neurotoxicities in the future. The present study provides an opportunity to understand a role of rpt-1 in human cell system and suggest a possible lead and basis for the future study of dioxin-induced neurotoxicities.

Effect of Benzo(k)fluoroanthene and Genistein on CYP1A1 Gene Expression in Human Breast Cancer MCF-7 Cells. (사람 유방암 세포 MCF-7에서 Benzo(k)fluoroanthene과 genistein이 CYP1A1 유전자 발현에 미치는 영향)

  • CYP1A1 is known to be inducible by xenobiotic compouds such as polyciclic aromatic hydrocarbons(PAHs) and 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). These chemicals have been identified worldwide and can have a significant impact on the human health and well being of human and wildlife. Given these issues, the detection and quantification of these chemicals in biological, environmental and food samples is important. First, we investigated the effect of on CYP1A1 promoter activity, 7-ethoxyresorufin-O-deethylase(EROD) activity and CYP1A1 mRNA expression induced by benzo(k)fluoranthene(B(k)F) in MCF-7 cells. We found that B(k)F significantly up-regulates the level of CYP1A1 prompter activity, EROD and CYP1A1 mRNA. When cells were treated with genistein, it was not changed that EROD and CYP1A1 mRNA, compared to that of control. However, genistein inhibited the B(k)F-induced CYP1A1 promoter activity and mRNA level at high concentration. Furthermore, in this study, effects of HDAC(histone deacetvlase) inhibitors on human prostate cancer cells proliferation were examined. HC-toxin, SAHA and TSA inhibited cell proliferation in PC3 cells. A novel HDAC inhibitor, IN2001 also suppressed the growth of PC3 cells. And IN2001 and SAHA increased S phase and G2/M phase at 12 hrs treatment but cells were arrested G0/G1 phase at 45 hrs treatment. The HC-toxin treatment for 24 hrs and 48 hrs increased G0/G1 at low concentration ($0.1\mu\textrm{m}$) but increased G2/M at more than concentration of $1\mu\textrm{m}$. TSA increased G2/M phase. These findings height the possbility of developing HDAC inhibitors as potential anticancer therapeutic agents for the treatment of prostate cancer.

Effects of Formononetin on the Aryl Hydrocarbon Receptor and 7,12-Dimethylbenz[a]anthracene-induced Cytochrome P450 1A1 in MCF-7 Human Breast Carcinoma Cells

  • Formononetin is an isoflavonoid phytoestrogen found in certain foodstuffs such as soy and red clover. In this study, we examined the action of formononetin with the carcinogen activation pathway mediated through the aryl hydrocarbon receptor (AhR) in MCF-7 breast carcinoma cells. Treating the cells with formononetin alone caused the accumulation of CYP1A1 mRNA as well as elevation in CYP1A1-specific 7-ethoxyresorufin O-deethylase (EROD) activity in a dose dependent manner. However, a concomitant treatment with 7,12-dimethylbenz[a]anthracene (DMBA) and formononetin markedly reduced both the DMBA-inducible EROD activity and CYP1A1 mRNA level. Under the same conditions, formononetin inhibited the DMBA-induced AhR transactivation, as shown by reporter gene analysis using a xenobiotic responsive element (XRE). Additionally, formononetin inhibited both DMBA-inducible nuclear localization of the aryl hydrocarbon receptor (AhR) and metabolic activation of DMBA, as measured by the formation of the DMBA-DNA adducts. Furthermore, formononetin competed with the prototypical AhR ligand, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), for binding to the AhR in an isolated rat cytosol. These results suggest that formononetin might be considered as a natural ligand to bind on AhR and consequently produces a potent protective effect against DMBA-induced genotoxicity. Therefore, that's the potential to act as a chemopreventive agent that is related to its effect on AhR pathway as antagonist/agonist.
