The localization of paclitaxel was investigated in suspension culture cells of Taxus chinensis. Over 93% of the cell-associated paclitaxel were detected throughout the entire culture period. Intracellular localization of paclitaxel over the culture time was analyzed further by cell fractionation for days 21 and 42. Paclitaxel contents in intracellular organelles were decreased at day 42, while the content in the cell wall fraction was increased at day 42 compared to the value for day 21. The localization of paclitaxel in the cell wall was confirmed by using the immunocytochemical method with the aid of a confocal laser scanning microscope.
The growth promotion of a Taxus brevifolia cell suspension culture was investigated using conditioning factors. The conditioning factors produced and secreted from cultured cells usually stimulate cell division and the production of secondary metabolites. Therefore, the effective incubation time for the optimal secretion of conditioning factors was firstly determined for the promotion of cell growth. Conditioned media obtained by cultivating for 2 and 5 days showed the promotion of initial cell growth during the early cell growth period. However, the positive effect of the conditioning factors on the initial cell growth did not continue because of the depletion of the medium nutrients. Accordingly, the addition of a carbon source to the conditioned medium prolonged the positive effect on the cell growth. The addition of sucrose to the conditioned medium resulted in the maximum cell density being reached 4 days earlier compared to the control group and an increased substrate yield.
A large number of chemical constituents have been reported from the Himalayan yew [Taxus baccata (Linn) or T. wallichiana (Zucc)]. These constituents are mainly taxoids and phenolics. Taxol, a lead anticancer agent, is the most important constituent. Other compounds have also been found to possess interesting biological properties. The literature concerning the chemistry and bioactivity of the constituents of the Himalayan yew has been briefly reviewed.
Anticancer agent taxol was synthesized from baccatin III which was extracted from cell cultures of European yew, Taxus baccata Pendula. Callus and suspension cultures of T. baccata Pendula showed the formation baccatin III. The content of baccatin III in cell cultures reached 0.015% (w/w) on dry weight basis. The semisynthetic approaches were made with baccatin III and taxol side chain. To prepare taxol side chain, (-)-N-((S)-2-hydroxy-1-phenylethyl) benzamide was synthesized first from (S)-(+)-phenylglycine. Then (-)-N-((IS,2S)-2-hydroxy-1-phenyl-3-butenyl) benzamide was synthesized with vinyl magnesium bromide. The synthesis of (2R, 3S)-(-)-2-(1-ethoxyethoxy)-3-phenyl-3-(phenylmethanamido) propanoic acid with RuCl3, catalyst was the final step to prepare taxol side chain. The semisynthetic reactions yielded 0.0002% taxol, 0.0005% 7-epi-10-deacetyltaxol, and unidentified taxanes on dry weight basis. It is suggested that the semisynthesis of taxol from baccatin III could be an alternative source of taxol and related taxanes.

This study was conducted by using a Nikon Coolpix S800c camera equipped with a NIR filter to measure the NDVI(Normalized Difference Vegetation Index). It was used for the measurement of the three trees of Pinus densiflora, Taxus cuspidata, Zelkova serrata and Acer palmatum in Kangwon National University. The NDVI value of the surface of the building was compared and analyzed. The average value of NDVI in August and September was high in all species. The NDVI distribution of Taxus cuspidata was higher than the other trees. The NDVI distribution of Pinus densiflora and Taxus cuspidata did not show any significant seasonal differences, but The NDVI distribution of Zelkova serrata and Acer palmatum were relatively low in May and June, which are leafless periods. Previous studies related to NDVI value were generally analyzed using satellite imagery. However, it was scarce related to study the NDVI value of each tree or study the changing process of NDVI by time series. Previous studies have used NDVI values on the ground but this study used NDVI values in the ground section. Future studies will be necessary to measure the NDVI value at different times for various species and also to make efforts to generalize the measurement method. In addition, research related to various fields such as the relationship between NDVI and carbon stocks and the relationship with LAI needs to be conducted.

Objectives : This study was designed to investigate the analgesic effects of Taxus cuspidata pharmacopuncture(TCP) at KI03 on formalin-induced pain in rats and to evaluate the efficiency of different extraction methods of Taxus cuspidata(TC). Methods : 30 rats were divided into 5 groups, each consisting of 6 rats. Each of the groups was treated two times. The first treatment for its right KI03 was as follows: rats were treated with normal saline(NOR), normal saline(CON), hot-water extraction of TC(W), ethanol extraction of TC(E), and ultrasonification extraction of TC(U). The second treatment was given 35 mins after the first one. Rats in NOR were treated with normal saline at their hind-paw. All groups, except NOR, were treated with formalin(5 %, $40{\mu}{\ell}$) at their hind-paw. To evaluate pain behavior, Ultrasonic vocalization(USV) was examined to be around 18~30 kHz. After analysis of USV, blood samples were taken from the rats for analysis of Substance P, aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Results : In the experimental groups, USV was significantly decreased compared with CON and similar to that of NOR in the early phase. But there were no significant differences among the three extraction methods of TC. In addition, Substance P of W was significantly decreased. AST and ALT activation showed no significant differences among the 5 groups. Conclusion : These results show that regardless of extraction methods of TC, TCP at KI03 has analgesic effects in the early phase on formalin-induced pain in rats. In particular, hot-water extraction of TC significantly reduces Substance P activation.
To seek for a stable supply source of taxol as an anticancer agent urgently needed for the treatment of ovarian cancer, the concentrations of taxol and cephalomannine in bark and needles of 37 yew (Taxus cuspidata S. et Z.) trees grown in 6 locations of Cheju Island were analyzed by using HPLC. A considerable variation in the content was observed in plant part and the location. The average taxol content in the needles ($220.3{\;}{\pm}{\;}56{\;}{\mu}g$ per g dry wt) was ca. 4 times hgher than that in the bark, whereas the content of cephalomannine in the needles $(353{\;}{\pm}{\;}141{\;}{\mu}g)$ was ca. 5 times higher than that in the bark. Particularly, even within Cheju Island, needles from trees grown near the coast including Sogwipo area contained much higher taxol contents than needles from trees grown at mountain locations, indicating that taxane level is affected by environmental effects. Taxol in the needles was confirmed by electrospray mass spectrometry. These results suggest that the needles of yew trees growing in Cheju Island may provide a renewable source for taxol.
In this study, an ultrasound-assisted micellar extraction process was developed to efficiently purify the anticancer substance paclitaxel from the plant cell Taxus chinensis. The problem of many extraction steps and long phase separation time in the traditional micellar process could be dramatically improved. The highest paclitaxel yield (~96%, extracted twice) was obtained at 180 W of ultrasonic power and 1.5 h of ultrasonic irradiation time, which was 24.7% higher than that of the traditional method. In addition, the partition coefficient (K) showed a maximum value (24.0) at 180 W of ultrasonic power and 1.5 h of irradiation time. There was no significant difference in the purity of paclitaxel, and the purity of initial paclitaxel (6.81%) increased to 22.0% after purification. Compared to the traditional method, the phase separation time of the back extraction decreased by 40.7-56.2% (ultrasonic power 80 W), 46.3-67.6% (ultrasonic power 180 W), and 51.9-67.6% (ultrasonic power 250 W), respectively. The phase separation time decreased as the ultrasonic power (80-250 W) and irradiation time (0.5-2.5 h) increased.
Antimicrobial and antioxidative activities of heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. Four lignan derivatives and one taxane were isolated from heartwood of Taxus cuspidata which has been selected due to its high antioxidative activity among the tested species. The chemical structures were identified as : taxusin, isolariciresinol (4, 4', 9, 9'-tetrahydroxy-3', 5-dimethoxy-2, 7'-cyclolignan), lariciresinol (4, 4', 9-trihydroxy-3, 3'-dimethoxy-7, 9'-epoxylignan), taxiresinol (3, 4, 4', 9-tetrahydroxy-3'-methoxy-7, 9'-epoxylignan) and isotaxiresinol (3', 4, 4', 9, 9'-pentahydroxy- 5-methoxy-2, 7'-cyclolignan) on the basis of spectroscopic data and their chemical correlations. According to the results of free radical scavenging activity, isolariciresinol, lariciresinol and isotaxiresinol showed higher radical scavenging activity than those of 𝛼-tocopherol and butylated hydroxytoluene (BHT), the strongest natural and synthetic antioxidants. However, taxusin did not show any free radical scavenging activity. In this regard, it could inferred that high antioxidative activity of extractives of T. cuspidata was derived from isolariciresinol, lariciresinol and isotaxiresinol.
The effects of various elicitors, metabolic inhibitors and growth regulators on the production of diterpenoid anticancer agent taxol were investigated in cell suspension cultures of Taxus brevifolia. Cell cultures of T. brevifolia were treated by 5 kinds of biotic elicitors, 5 kinds of abiotic elicitors, 2 kinds of metabolic inhibitors and 8 kinds of growth regulators at the end of exponential growth phase. Among those treatments, chlorocholine chloride-an inhibitor of plant steroid metabolism-increased the taxol production most significantly. From a series of optimization studies, it was found that the addition of 1mM of chlorocholine chloride at the 9th day of culture was the best for taxol production. Taxol yield under this condition was 0.72mg/$\ell$.
