The localization of paclitaxel was investigated in suspension culture cells of Taxus chinensis. Over 93% of the cell-associated paclitaxel were detected throughout the entire culture period. Intracellular localization of paclitaxel over the culture time was analyzed further by cell fractionation for days 21 and 42. Paclitaxel contents in intracellular organelles were decreased at day 42, while the content in the cell wall fraction was increased at day 42 compared to the value for day 21. The localization of paclitaxel in the cell wall was confirmed by using the immunocytochemical method with the aid of a confocal laser scanning microscope.
The growth promotion of a Taxus brevifolia cell suspension culture was investigated using conditioning factors. The conditioning factors produced and secreted from cultured cells usually stimulate cell division and the production of secondary metabolites. Therefore, the effective incubation time for the optimal secretion of conditioning factors was firstly determined for the promotion of cell growth. Conditioned media obtained by cultivating for 2 and 5 days showed the promotion of initial cell growth during the early cell growth period. However, the positive effect of the conditioning factors on the initial cell growth did not continue because of the depletion of the medium nutrients. Accordingly, the addition of a carbon source to the conditioned medium prolonged the positive effect on the cell growth. The addition of sucrose to the conditioned medium resulted in the maximum cell density being reached 4 days earlier compared to the control group and an increased substrate yield.
A large number of chemical constituents have been reported from the Himalayan yew [Taxus baccata (Linn) or T. wallichiana (Zucc)]. These constituents are mainly taxoids and phenolics. Taxol, a lead anticancer agent, is the most important constituent. Other compounds have also been found to possess interesting biological properties. The literature concerning the chemistry and bioactivity of the constituents of the Himalayan yew has been briefly reviewed.
유럽 주목(Taxus baccata Pendula) 의 세포배양에 서 추출한 taxol 전구체 baccatin ill를 이용하여 항암제 taxol을 반합성할 수 있었다. 유럽주목의 캘러 스배양과 현탁배양 모두에셔 baccatin ill의 생성을 확인할 수 있었으며 함량은 건조중량 기준으로 평균 0.015%(w/w)에 달하였다. Taxol의 반합성은 세포 배양에서 생성된 baccatin ill에 taxol side chain을 합성시킴으로서 가능하였다. Taxol side chain을 합 성하기 위하여 (S)-(+)-phenylglycine로부터 (-) - -N-(S)-2-hydroxy-l-phenyl ethyl)benzamide를 합성하였고 다음 단계로 (-)-N-(lS,2S)-2-hyd droxy-l-phenyl-3-butenyl) benzamide를 합성하 였고 최종적으로 산화과정을 거쳐 taxol side chain 인 (2R,3S) - (-) -2-(1-ethoxyethoxy)-3-phenyl-3 (phenylmethanamido) propanoic acid를 얻을 수 있었다. 이러한 반합성 과정을 통하여 0.0002 % 수준의 taxol 및 0.0005 %의 관련 taxane을 합성하였다. 이와 같이 주목 세포배양을 이용한 taxol 반합성 방법은 기존의 주목 잎에서 추출한 baccatin ill를 이용하는 방법에 비하여 충분한 경제성이 있을 수 있으며 이에 따라 taxo!의 새로운 공급원의 역할 및 기타 여러 종류의 taxol 유사체의 효율적인 이용 등 앞으로 기대 효과가 크다고 본다.
This study was conducted by using a Nikon Coolpix S800c camera equipped with a NIR filter to measure the NDVI(Normalized Difference Vegetation Index). It was used for the measurement of the three trees of Pinus densiflora, Taxus cuspidata, Zelkova serrata and Acer palmatum in Kangwon National University. The NDVI value of the surface of the building was compared and analyzed. The average value of NDVI in August and September was high in all species. The NDVI distribution of Taxus cuspidata was higher than the other trees. The NDVI distribution of Pinus densiflora and Taxus cuspidata did not show any significant seasonal differences, but The NDVI distribution of Zelkova serrata and Acer palmatum were relatively low in May and June, which are leafless periods. Previous studies related to NDVI value were generally analyzed using satellite imagery. However, it was scarce related to study the NDVI value of each tree or study the changing process of NDVI by time series. Previous studies have used NDVI values on the ground but this study used NDVI values in the ground section. Future studies will be necessary to measure the NDVI value at different times for various species and also to make efforts to generalize the measurement method. In addition, research related to various fields such as the relationship between NDVI and carbon stocks and the relationship with LAI needs to be conducted.
Objectives : This study was designed to investigate the analgesic effects of Taxus cuspidata pharmacopuncture(TCP) at KI03 on formalin-induced pain in rats and to evaluate the efficiency of different extraction methods of Taxus cuspidata(TC). Methods : 30 rats were divided into 5 groups, each consisting of 6 rats. Each of the groups was treated two times. The first treatment for its right KI03 was as follows: rats were treated with normal saline(NOR), normal saline(CON), hot-water extraction of TC(W), ethanol extraction of TC(E), and ultrasonification extraction of TC(U). The second treatment was given 35 mins after the first one. Rats in NOR were treated with normal saline at their hind-paw. All groups, except NOR, were treated with formalin(5 %, $40{\mu}{\ell}$) at their hind-paw. To evaluate pain behavior, Ultrasonic vocalization(USV) was examined to be around 18~30 kHz. After analysis of USV, blood samples were taken from the rats for analysis of Substance P, aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Results : In the experimental groups, USV was significantly decreased compared with CON and similar to that of NOR in the early phase. But there were no significant differences among the three extraction methods of TC. In addition, Substance P of W was significantly decreased. AST and ALT activation showed no significant differences among the 5 groups. Conclusion : These results show that regardless of extraction methods of TC, TCP at KI03 has analgesic effects in the early phase on formalin-induced pain in rats. In particular, hot-water extraction of TC significantly reduces Substance P activation.
To seek for a stable supply source of taxol as an anticancer agent urgently needed for the treatment of ovarian cancer, the concentrations of taxol and cephalomannine in bark and needles of 37 yew (Taxus cuspidata S. et Z.) trees grown in 6 locations of Cheju Island were analyzed by using HPLC. A considerable variation in the content was observed in plant part and the location. The average taxol content in the needles ($220.3{\;}{\pm}{\;}56{\;}{\mu}g$ per g dry wt) was ca. 4 times hgher than that in the bark, whereas the content of cephalomannine in the needles $(353{\;}{\pm}{\;}141{\;}{\mu}g)$ was ca. 5 times higher than that in the bark. Particularly, even within Cheju Island, needles from trees grown near the coast including Sogwipo area contained much higher taxol contents than needles from trees grown at mountain locations, indicating that taxane level is affected by environmental effects. Taxol in the needles was confirmed by electrospray mass spectrometry. These results suggest that the needles of yew trees growing in Cheju Island may provide a renewable source for taxol.
본 연구에서는 식물세포 Taxus chinensis로부터 항암물질 파클리탁셀을 효율적으로 정제하기 위하여, 초음파 기반 마이셀 추출 공정을 개발하였다. 전통적 마이셀 공정(대조군)에서의 많은 추출 단계 및 긴 상 분리 시간 문제를 획기적으로 개선하였다. 초음파 파워 180 W, 초음파 조사 1.5 시간에서 가장 높은 파클리탁셀 수율(~96%, 2회 추출)을 얻었으며, 이는 대조군의 수율에 비해 24.7% 증가하였다. 또한 분배 계수(K)는 초음파 파워 180 W, 초음파 조사 1.5 시간에서 최대치(24.0)를 보였다. 파클리탁셀 순도에는 큰 차이가 없었으며, 초기 파클리탁셀의 순도(6.81%)가 정제 후 22%까지 증가하였다. 역 추출(back extraction)의 상 분리 시간은 대조군 대비 각각 40.7-56.2%(초음파 파워 80 W), 46.3-67.6%( 초음파 파워 180 W), 51.9-67.6%(초음파 파워 250 W) 감소하였다. 초음파 파워(80-250 W)와 초음파 조사 시간(0.5-2.5 시간)이 증가할수록 상 분리 시간이 감소하였다.
항산화활성이 우수한 주목 심재 에탄올 추출물로부터 Sephadex LH-20, centrifugal partition chromatography (CPC), silica gel column chromatography 등을 이용하여 5종의 화합물을 단리하였으며, NMR, Mass spectrometry 등의 기기분석 결과 taxane 화합물인 taxusin를 비롯하여 lignan인 isolariciresinol (4, 4', 9, 9'-tetrahydroxy-3', 5-dimethoxy-2, 7'-cyclolignan), lariciresinol (4, 4', 9-trihydroxy-3, 3'-dimethoxy-7, 9'-epoxylignan), taxiresinol (3, 4, 4', 9-tetrahydroxy-3'-methoxy-7, 9'-epoxylignan) 및 isotaxiresinol (3', 4, 4', 9, 9'-pentahydroxy-5-methoxy-2, 7'-cyclolignan)을 각각 동정하였다. 이들 단리된 화합물의 프리라디칼 소거능을 측정하여 항산화활성을 검정한 결과 taxusin을 제외한 3종의 lignan 화합물인 isolariciresinol, lariciresinol, isotaxiresinol 모두 천연항산화제 및 합성항산화제보다 높은 라디칼 소거능을 나타내어 항산화활성이 우수한 것으로 판명되었다. 이상의 결과 주목의 심재 에탄올 조추출물의 높은 항산화활성은 이들 성분에 기인하는 것으로 사료되었다.
Taxus brevifolia 현탁세포배양에 서 항암제 taxol의 생산을 향상시키기 위해 5종류의 biotic elicitor 와 5종류의 abiotic elicitor, 2종류의 대사억제 및 8종류의 생장조절제를 세포배양 중에 첨가하여 효과 가 았는 물질을 선별하였다. T. brevifo/ia 현탁세포 배양의 대수증식기 말기인 10일째에 각각의 물질을 첨가하여 배양액 내의 taxol의 함량을 측정한 결과 steroid계 억제제 인 chlorocholine chloride를 처리 하였을 때 taxol의 생성이 현저히 증가되었다. Chloro choline chloride의 처리시기 및 처리농도를 다르게 하여 최적조건을 찾은 결과 9일째에 1mM을 첨가했 을 경우 taxol 생성이 가장 좋았다.
