Study of Antioxidative Substances from Some Indonesian Plants

  • Various natural compounds act as antioxidants in protection against lipid peroxidation. Lipid peroxidation yields a variety of decomposition products which have been implicated in not only decreasing the nutritional value of food, but also in developing an off-flavor and toxic substances. As a source of safer and more effective natural antioxidants of natural origin have been widely investigated. Some Indonesian plants have evaluated for their antioxidative activity, and suggested the possible existence of various antioxygenic compounds in them. We attempted to study such antioxygenic compounds with simple method evaluation. As we are interested in the natural product compounds, we examined of several sample such as edible sea-weeds, and some edible fruits. Sea-weed, Eisenia bicyclis, one of the edible brown algae, exhibited the activity. As a traditional food additive consumed by Indonesian, Garcinia parvifolia is used as taste supplement in region West Sumatra, have been studied. Our current studies on the semi-polar fractions shows the activity by the thiocyanate method test. Another sample, Garcinia mangostana, a famous fruit with sweet taste, the part kernel have also evaluated. The acidic fraction of the extract showed antioxidative activity. Some other active components were found in the neutral and BuOH fractions.

천연 강황 추출물의 약리, 화학적 특성 및 분석 (A Study on the Pharmaceutical & Chemical Characteristics and Analysis of Natural Curcumin Extract)

  • Natural Curcumin belongs to Zingiber Officinale Roscoe was known to possess natural odor, natural taste, natural color, and other pharmaceutical & chemical characteristics. Natural Curcumin extract was made to use ethanol as a solvent was to show a yellow color having state of solid powder and an active component. Natural Curcumin extract tested pharmaceutical & chemical experiment to dilute in curcumin 1%-water solution. Curcumin extract tested antimicrobial experiment using microbe, and tested dye experiment using fiber. Some conclusions in the result of characteristics experiment was obtained as follow. The result of antimicrobial experiment showed that the growth of staphylococcus aureus (ATCC-001) and aspergillus niger (ATCC-002) as microbes decreased according to passage of time. This phenomenon could know that Curcumin compoment showed influence to antimicrobial effect. Also, the result of dye experiment showed that cotton and sick with fiber dyeing dyed in direction of dark yellow color. This phenomenon could know that Curcumin extract showed influence to dyeing effect in observation of optical electron microscope(OEM.) The result of instrument analysis ascertained inorganic components of K(53.300ppm), Na(1.150ppm), Ca(0.711ppm), Ti(0.351ppm), Li(0.256ppm), Cu(0.233ppm) etcs from Curcumin component with ICP/OES, and ascertained organic components of propanoic acid(1.859), benzene(10.814), phenol(14.194) etcs from Curcumin component with GC/MSD.

복합 처방 품질 관리를 위한 시스템 차원의 연구 동향 (Trends in System-level Research on Quality Control of Complex Herbal Formulation)

  • The quality control is a fundamental procedure for the standardization of herbal medicine to guarantee the consistency of efficacy and safety. For a long time, the quality analysis of herbal medicine has been largely dependent on the routine sensory evaluation, such as taste, smell, color, and shape. However, with the recent development of analytical instruments, various scientific approaches have been introduced in this field. On the basis of the theory that the biological activities of herbal medicine are mainly contributed by its chemical compositions, several types of chemical markers have been suggested for the quality evaluation. In addition to the analytical methods for the specific marker compound(s), including analytical marker and active marker, recently, chemical fingerprinting, a method comparing the chromatographic pattern of the whole chemical components, has been developed and widely accepted as a reliable approach for the quality control of herbal medicine. Moreover, in order to exactly understand the relationship between complex compounds and their holistic biological activities in herbal medicine, unique strategies, such as "BECCs (bioactive equivalent combinatorial components)" and "PhytomicsQC" have been established. In this article, we give an overview of the several categories of chemical markers and the recent research trends for the quality evaluation of herbal medicine.

  • 원료배합(原料配合)을 다리한 간장용(用) 고일(一)지의 제국중(製麴中)에 있어서와 건조(乾燥)고일(一)지의 각종효소(各種酵素) activity 및 성분(成分)의 변화(變化)를 측정(測定)하여 다음결과(結果)를 얻었다. 1. 건조(乾燥)고일(一)지는 중성부근(中性附近)(pH 6.0)에 활성(活性)을 갖는 Protease가 주체(主體)를 이루며 산성(酸性) 및 alkali성측(性側)의 Protease activity는 이보다 미약(微弱)하고 밀을 많이 배합(配合)한 구(區)일수록 산성(酸性) 및 중성측(中性側) Protease activity가 높아지는 반면(反面)에 alkali성측(性側)은 이와 반대(反對)되는 경향(傾向)을 보였다. 2. 제국중(製麴中) 각종효소(各種酵素) activity는 모두 증가(增加)되었고 밀이 많은 배합구(配合區)일수록 ${\alpha}-Amylase$는 그 activity가 높아졌다. 3. 제국중(製麴中) 전당(全糖)은 감소(減少)되었으나 직당(直糖) 및 아미노 태질소(態窒素)와 전질소(全窒素)는 증가(增加)되었다. 4. 건조(乾燥)고일(一)지에 있어서 밀이 많은 배합구(配合區)일수록 직당(直糖) 및 전당(全糖)의 함량(含量)이 많어졌으나 전질소(全窒素)는 적어졌고 아미노(態窒素)는 차이(差異)가 없었다.

자연산 홍합(Mytilus coruscus)과 양식산 진주담치(Mytilus edulis)의 성분 특성 (Food Component Characteristics of Wild Hard-shelled Mussel Mytilus coruscus and Cultured Sea Mussel Mytilus edulis in Korea)

  • To characterize the food components of two mussels (wild hard-shelled mussel (HM) Mytilus coruscus, and cultured sea mussel (SM) Mytilus edulis) in Korea, we examined the proximate composition, fatty composition, amino acid/mineral content, texture, collagen content and chemical and taste compounds. Regarding the proximate composition, HM had lower moisture levels and higher crude protein and carbohydrate contents than SM. The amino nitrogen, volatile basic nitrogen and total amino acid contents of HM and SM were 250.6 and 227.3 mg/100 g, 11.2 and 12.0 mg/100 g, and 17,451.1 and 15,334.8 mg/100 g, respectively. The major amino acids were glutamic acid, aspartic acid, glycine, alanine, lysine and arginine. The major fatty acids of HM and SM were 14:0, 16:0, 16:1n-7, 20:5n-3, and 22:6n-3, which did not differ significantly between the two mussels. HM had a higher n-3 polyene ratio, and a lower saturate and monoene ratio than SM. Regarding the taste-active compounds, the free amino acid contents of HM and SM were 1,116.5 and 961.8 mg/100 g, respectively, and the major free amino acids were taurine, glutamic acid, glutamine, glycine, citrulline, lysine and arginine. The primary minerals in both HM and SM were Na, Cl, K and P which did not differ significantly between the two mussels. The soluble and insolube collagen contents of HM and SM were 265.8 and 228.4 mg/100 g, and 119.5 and 121.8 mg/100 g, respectively.

굴 스파게티 소스의 개발 (Development of Spaghetti Sauce with Oyster)

  • 굴을 보다 효율적으로 이용하고자 신세대 기호에 맞는 굴 스파게티 소스의 제조를 시도하였다. 유기산, 색조, 점도 및 관능검사의 결과로 미루어 보아 스파게티 소스의 굴 최적첨가량은 11%로 판단되었고, 저장성 부여를 위한 최적 $F_0$ value는 4분이라고 판단되었다. 최적조건에서 제조한 굴 스파게티 소스의 수분, 단백질, 지방 및 회분은 각각 71.2%, 2.8%, 6.9% 및 3.2%이었다. 굴 스파게티 소스의 관능검사 결과 향과 조직감의 경우 시판 스파게티 소스보다 좋았으며, 색의 경우 차이가 없었다 굴 스파게티 소스의 총 아미노산 함량은 2,532.2 mg/100 g이었고 주요 아미노산으로는 glutamic acid, aspartic acid 및 phenylalanine 등이었다. 굴 스파게티 소스는 칼슘/인의 비율이 1.88로 칼슘이 흡수되기 좋은 비율로 구성되었다. 유리아미노산 함량은 시판 스파게티소스(1,165.5 mg/100 g)가 굴 스파게티 소스(1,040.2 mg/100 g)에 비하여 높았고, taste value 또한 시판 스파게티 소스 (189.35)가 굴 스파게티 소스(151.26)보다 높았다. Taste value의 결과로 미루어 보아 맛에 관여하는 주 아미노산은 aspartic acid와 glutamic acid로 판단되었다.

죽방 마른멸치의 성분조성 및 품질 (Food Components and Quality Characteristics of Boiled-Dried Anchovies Caught by Pound Net)

  • 우리나라 전통수산식품의 성분조성 및 품질특성에 관한 연구의 일환으로, 우리나라 사람들이 즐겨 먹고 있는 일반 마른 멸치와 지역특산품인 죽방멸치의 이화학적 식품성분 조성을 분석하여 이들의 품질 특성을 비교, 검토하였다. 시판 마른 멸치의 수분 함량은 일반 마른 멸치가, 조단백질 및 조지방 함량은 죽방멸치가 다소 많았으며, 휘발성 염기질소 함량은 일반 마른멸치가 다소 많았다. 한편, 지질의 산패는 TBA값으로 보아 죽방멸치가 약간 더 진행되어 있는 것으로 나타났다. 일반 마른 멸치 및 죽방멸치의 구성아미노산함량은 각각 41.g∼46.5 g/100 g 및 42.0∼49.2 g/100 g으로서 glutamic acid, aspartic acid, histidine, leucine, alanine 및 arginine 등이 주요 구성 아미노산이었으며, 양 시료 모두 중대멸이 소멸에 비해 구성아미노산들의 함량이 많았다. 일반 마른멸치와 죽방멸치의 지방질 조성에서 중성 및 극성지방질의 조성은 양자간에 큰 차이가 없었으며, 대체로 극성지방질의 조성비가 상당히 높았다. 각 지방질의 주요 구성 지방산은 14:0, 16:0, 16:1n-7, 18:1n-9, 20:5n-3 및 22:6n-3 등이었으며, n-3계열의 고도불포화지방산의 조성비는 일반 마른멸치가 높았다. 일반 마른멸치와 죽방멸치의 유리아미노산의 총량은 각각 953.7∼1,044.8 mg/100g 및 968.8∼ 1,306.4 mg/100 g이었으며, taurine, histidine, alanine, 1-methyl histidine, Lysine, glutamic acid및 glycine의 함량이 많았다. IMP 및 TMAO, creatinine등의 염기류는 죽방멸치 쪽의 함량이 약간 많았으나 큰 차이는 없었다. 주요 정미 발현성분의 하나인 무기성분 함량은 소멸쪽이 중대멸에 비해 대체로 많았으며, 일반 마른멸치와 죽방멸치 간에는 별 차이를 보이지 않았다. 관능검사 결과, 중대멸은 조직감을 제외한 맛, 외관 및 냄새에서 죽방멸치 쪽이 우수하였으나, 소멸의 경우에서는 양자간에 관능적인 차이가 거의 인지되지 않았다. 시판 마른멸치류의 품질은 이화학적 구성 성분의 함량 차이보다는 마른멸치의 외관, 맛, 냄새 등 관능적 면에서 결정되었다.

진주담치(Mytilus edulis) 추출물의 제조 및 품질특성 (Evaluating Cultured Sea Mussels Mytilus edulis Extractions Methods and Extract Quality Characteristics)

  • Extraction methods for cultured sea mussels Mytilus edulis and the quality characteristics of resulting extracts were investigated. The crude protein, carbohydrate and volatile basic nitrogen content of raw sea mussels was 15.2%, 1.9%, and 11.2 mg/100 g, respectively. Extracts were prepared using three different methods: hot-water extract (WE), scrap enzymatic hydrolysate extraction (SE), and complex extraction (CE). The respective extracts contained 5.5%, 8.6%, and 6.6% crude protein; 281.7, 366.0, and 343.0 mg/100 g amino nitrogen,: and 2.0%, 1.1% and 1.8% salinity. Their extraction yields were 689, 323, and 1,012 mL/kg. The CE method was superior to the traditional WE method in terms of extraction yield, amino-nitrogen content, and organoleptic qualities, but not odor. Active taste components were evaluated and the total free amino acid content of the WE and CE methods was 5,667.0 and 7,006.3 mg/100 g, respectively. The concentrations of major components (for WE and CE methods, respectively) were as follows: glutamic acid (1,244.0 and 955.4 mg/100 g), taurine (987.9 and 746.8 mg/100 g), glycine (721.2 and 847.0 mg/100 g), alanine (341.9 and 423.8 mg/100 g), arginine (265.5 and 376.5 mg/100 g), lysine (199.8 and 270.4 mg/100 g), and proline (253.9 and 220.3 mg/100 g). In conclusion, these results demonstrate that there is potential for using the CE method to expand the commercial utilization of sea mussels as a flavoring substance resource.

Comparison of Tastes-Related Components and Eating Quality between Hanwoo Steer and Cow Longissimus thoracis Muscles

  • The objective of this study was to compare tastes-related components and eating quality properties between Hanwoo steer and cow meats. Longissimus thoracis (LT) muscles with same quality grade (1+ grade) collected from left sides of the steer and cow carcasses were used for analyses of technological quality traits, free amino acids (FAAs), metabolites, nucleotides, fatty acids and sensory attributes. there were no differences occurring in the chemical composition (fat, protein, moisture and collagen) and technological quality traits (cooking loss, water holding capacity, shear force and color) between the two beef types (p>0.05). The cow meat exhibited significantly higher amounts of some FAAs associated with umami (e.g., glutamic acid and lysine), sweetness (e.g., proline and glutamic acid) and saltiness (e.g., histidine and glutamic acid) compared to the steer meat (p<0.05). Regarding the nucleotides, no differences occurred in all the identified nucleotides between the two beef types (p>0.05). A total of 27 metabolites were identified, however, only some compounds (e.g., acetate, creatine, creatinine, glucose and inosine, etc.) showed their significantly higher amounts in the cow meat compared with those in the steer meat (p<0.05). In terms of sensory aspects, the panelists found no differences in scores of all the eating quality traits between the two sex types of beef (p>0.05). Overall, excepts some variations in tastes-active compounds, there were no differences in the quality characteristics in general and eating quality in particular between the cow and steer meats when they were in the same quality grade.

콩 Saponin의 생리활성 기능과 함량변이 (Biological Activities of Soyasaponins and Their Genetic and Environmental Variations in Soybean)

  • There is much evidence suggesting that compounds present in soybean can prevent cancer in many different organ systems. Especially, soybean is one of the most important source of dietary saponins, which have been considered as possible anticarcinogens to inhibit tumor development and major active components contributing to the cholesterol-towering effect. Also they were reported to inhibit of the infectivity of the AIDS virus (HIV) and the Epstein-Barr virus. The biological activity of saponins depend on their specific chemical structures. Various types of triterpenoid saponins are present in soy-bean seeds. Among them, group B soyasaponis were found as the primary soyasaponins present in soybean, and th e 2, 3-dihydro-2, 5-dihydroxy-6- methyl-4H-pyran-4-one(DDMP)-conjugated soyasaponin $\alpha\textrm{g}$, $\beta\textrm{g}$, and $\beta$ a were the genuine group B saponins, which have health benefits. On the other hand, group A saponins are responsible for the undesirable bitter and astringent taste in soybean. The variation of saponin composition in soybean seeds is explained by different combinations of 9 alleles of 4 gene loci that control the utilization of soyasapogenol glycosides as substrates. The mode of inheritance of saponin types is explained by a combination of co-dominant, dominant and recessive acting genes. The funtion of theses genes is variety-specific and organ specific. Therefore distribution of various saponins types was different according to seed tissues. Soyasaponin $\beta\textrm{g}$ was detected in both parts whereas $\alpha\textrm{g}$ and $\beta$ a was detected only in hypocotyls and cotyledons, respectively. Soyasaponins ${\gamma}$g and $\gamma\textrm{g}$ were minor saponin constituents in soybean. In case group A saponins were mostly detected in hypocotyls. Also, the total soyasaponin contents varied among different soy-bean varieties and concentrations in the cultivated soy-beans were 2-fold lower than in the wild soybeans. But the contents of soyasaponin were not so influenced by environmental effects. The composition and concentration of soyasaponins were different among the soy products (soybean flour, soycurd, tempeh, soymilk, etc.) depending on the processing conditions.
