Investigation of Soil and Groundwater Contaminated by Gasoline and Lubricants Around a Railroad Station in S City, Korea

  • Objective: This research was performed to evaluate the state of oil pollution in an area surrounding a railway station that has over 100 years of business history as a railway station in S City, Korea. The amount of polluted soil was estimated, and the target area for remediation was assessed in this study to restore the oil-polluted area. Methods: To accomplish this aim, five observation wells were installed for the sampling of groundwater, and soil was sampled at 33 points. Electric resistance studies and a trench investigation were undertaken to understand the geological conditions of the site, and the groundwater movement in this area was simulated by MODFLOW. Physiochemical analyses were conducted to determine the quality of the groundwater and the current state of oil pollution influenced by that of the soil. Results: The mean level of total petroleum hydrocarbons (TPHs) in this area was 1,059 mg/kg, and the area for remediation was determined to be 7,610 mg/kg. Levels of benzene, toluene, ethylbenzene, and xylene (BTEX) were determined to be under the legal standard. Conclusion: In terms of depth, the biggest area polluted by TPH found was between 0 and 1 m from ground level, and the affected area was 5,900 $m^3$. TPHs were not detected in groundwater. Diesel and lubricating oil were the main causes of TPH pollution at this railway station.

A Study on Strengthening Exercise for Stroke Patients through Comparison of Literature between Domestic and Foreign (국내·외 문헌적 비교를 통한 뇌졸중 환자에 대한 근력강화 운동의 고찰연구)

  • Purpose : The purpose of this study is to investigate the effect of strengthening program for improving the muscle strength and body function in rehabilitation of patients with stroke Methods : We found the 15 precedent studies at online portal site of "Korea education and Research information service" and found 5 precedent studies at "Pubmed". We analyzed and describe the total 19 studies involving national and international research. Results : The strengthening exercise was adapted to lower extremities and trunk muscle of participants and the isokinetic (concentric and eccentric) exercise was most of exercise type and then manual isometric exercise, functional activities, progressive task oriented resistance exercise, PNF pattern exercise was following. The studies reported that the strengthening program increased target muscle strength and improved balance capacity and walking function for the participants positively. Conclusion : Conclusionally the several strengthening program can be the intervention to increase the muscle strength without increasing spasticity for patients with stroke. We think that the strengthening of lower extremities an improve balance capacity and walking ability and it can use the intervention to change the quality of life in patients with stroke. More than 3weeks strength program might be effectiveness, in case of acute patients with stroke, the improving of muscle strength is available but need to study for improving balance and walking capacity more in the future.

Traffic-induced vibrations at the wet joint during the widening of concrete bridges and non-interruption traffic control strategies

  • The rapid development of road transport has increased the number of bridges that require widening. A critical issue in the construction of bridge widening is the influence of vibrations of the old bridge on the casting of wet joint concrete between the old and new bridges owing to the running traffic. Typically, the bridge is closed to traffic during the pouring of wet joint concrete, which negatively affects the existing transportation network. In this study, a newly developed microscopic traffic load modeling approach and the vehicle-bridge interaction theory are incorporated to develop a refined numerical framework for the analysis of random traffic-bridge coupled dynamics. This framework was used to investigate traffic-induced vibrations at the wet joint of a widened bridge. Based on an experimental study on the vibration resistance of wet joint concrete, traffic control strategies were proposed to ensure the construction performance of cast-in-site wet joint concrete under random traffic without interruption. The results show that the vibration displacement and frequency of the old bridge, estimated by the proposed framework, were comparable with those obtained from field measurements. Based on the target peak particle velocity and vibration amplitude of the wet joint concrete, it was found that traffic control measures, such as limiting vehicle gross weight and limiting traffic volume by closing an additional traffic lane, could ensure the construction performance of the wet joint concrete.

Application of In-Situ Mixing Hydration Accelerator on Polymer Modified Concrete for Bonded Concrete Overlay (접착식 콘크리트 덧씌우기를 위한 초속경화 첨가재 현장 혼합 폴리머 개질 콘크리트의 적용성 연구)

  • PURPOSES : Recently, bonded concrete overlay has been used as an alternative solution in concrete pavement rehabilitation since its material properties are similar to those of the existing concrete pavements. Deteriorated concrete pavements need rapid rehabilitation in order to prevent traffic jams on Korean expressways. Moreover, speedy and effective repair methods are required. Therefore, the use of bonded concrete overlay with ultra-rapid hardening cement has increased in an effort to reopen promptly the expressways in Korea. However, mobile mixer is required for ultra-rapid hardening cement concrete mixing in the construction site. The use of mobile mixer causes various disadvantages aforementioned such as limitation of the construction supply, open-air storage of mixing materials, increase in construction cost, and etc. In this study, therefore, hydration accelerator in-situ mixing on polymer modified concrete produced in concrete plant is attempted in order to avoid the disadvantages of existing bonded concrete overlay method using ultra-rapid hardening cement. METHODS : Bonded concrete overlay materials using ultra-rapid hardening cement should be meet all the requirements including structural characteristics, compatibility, durability for field application. Therefore, This study aimed to evaluate the application of hydration accelerator in-situ mixing on polymer modified concrete by evaluating structural characteristics, compatibility, durability and economic efficiency for bonded concrete overlay. RESULTS : Test results of structural characteristics showed that the compressive, flexural strength and bond strength were exceed 21MPa, 3.15MPa and 1.4MPa, respectively, which are the target strengths of four hours age for the purpose of prompt traffic reopening. In addition, tests of compatibility, such as drying shrinkage, coefficient of thermal expansion and modulus of elasticity, and durability (chloride ions penetration resistance, freezing-thawing resistance, scaling resistance, abrasion resistance and crack resistance), showed that the hydration accelerator in-situ mixing on polymer modified concrete were satisfied the required criteria. CONCLUSIONS : It was known that the hydration accelerator in-situ mixing on polymer modified concrete overlay method was applicable for bonded concrete overlay and was a good alternative method to substitute the existing bonded concrete overlay method since structural characteristics, compatibility, durability were satisfied the criteria and its economic efficiency was excellent compare to the existing bonded concrete overlay methods.

Levels of Resistance and Fitness in Glufosinate-ammonium-Resistant Transgenic Rice Plants (Glufosinate-ammonium 저항성 형질전환벼의 저항성 수준과 적응성에 관한 연구)

  • The objectives of this research were to quantify resistance levels of transgenic rice expressing the bar gene to glutamine synthetase (GS)-inhibiting, and methionine sulfoximine and photosynthesis-inhibiting herbicide, paraquat, and compare the ammonium accumulation, chilling injury, and yield between transgenic and non-transgenic rice. The transgenic rice lines were 45-96-fold more resistant to glufosinate-ammonium than non-transgenic rice. The transgenic rice lines were also 18-fold more resistant to methionine sulfoximine, but was not resistant to paraquat, which has different target site. Glufosinate-ammonium increased the ammonium accumulation in leaves of non-transgenic rice plants, but had minimal or no effect on leaves of transgenic lines. The transgenic lines except for 258, 411, 607 and 608 were more susceptible during chilling and recovery than non-transgenic rice plants. The yield of transgenic lines 142, 144, 258 and 608 was similar or higher than that of non-transgenic rice in pot conditions.

The Attenuation Mechanism and Live Vaccine Potential of a Low-Virulence Edwardsiella ictaluri Strain Obtained by Rifampicin Passaging Culture

  • The rifampicin-resistant strain E9-302 of Edwardsiella ictaluri strain 669 (WT) was generated by continuous passage on BHI agar plates containing increasing concentrations of rifampicin. E9-302 was attenuated significantly by 119 times to zebrafish Danio rerio compared to WT in terms of the 50% lethal dose (LD50). Zebrafish vaccinated with E9-302 via intraperitoneal (IP) injection at a dose of 1 × 103 CFU/fish had relative percentage survival (RPS) rates of 85.7% when challenged with wild-type E. ictaluri via IP 14 days post-vaccination (dpv). After 14 days of primary vaccination with E9-302 via immersion (IM) at a dose of 4 × 107 CFU/ml, a booster IM vaccination with E9-302 at a dose of 2 × 107 CFU/ml exhibited 65.2% RPS against challenge with wild-type E. ictaluri via IP 7 days later. These results indicated that the rifampicin-resistant attenuated strain E9-302 had potential as a live vaccine against E. ictaluri infection. A previously unreported amino acid site change at position 142 of the RNA polymerase (RNAP) β subunit encoded by the gene rpoB associated with rifampicin resistance was identified. Analysis of the whole-genome sequencing results revealed multiple missense mutations in the virulence-related genes esrB and sspH2 in E9-302 compared with WT, and a 189 bp mismatch in one gene, whose coding product was highly homologous to glycosyltransferase family 39 protein. This study preliminarily explored the molecular mechanism underlying the virulence attenuation of rifampicin-resistant strain E9-302 and provided a new target for the subsequent study of the pathogenic mechanism of E. ictaluri.

Evaluation of Field Applicability of Slope of Improved Soil for Ground Stabilizer (지반안정재 개량토의 토사 비탈면 현장 적용성 평가)

  • This research studies the stabilization method for improved soil sloped through the on-site application of Paper Flyash ground stabilizers. The target strength required for improved soil is 500 kPa, and the compressive strength for the slope surface needs to be less than 1,000 kPa after the improvement in order to plant vegetation. To meet this condition, we mixed soil from the site and the ground stabilization material, which is the main material for surface improvement material, performed mixing design and conducted various tests including strength test, permeability test and plantation test. After analyzing the results of the compression test on improved soil slope, we proposed soil constants for the improved soil. In order to evaluate the applicability of the improved soil on the slope, the site construction was carried out on the collapsed slope and the reinforcement evaluation of the surface of the improvement soil was conducted. The stability was not secured before the reinforcement, but the test shows after the reinforcement with improved soil, the safety rate is secured up to 48 hours during the raining period. In addition, the compressive strength of the improved soil at the site was secured at more than 200 kPa adhesion as planned, and the soil hardness test result was also found to be within the specified value of 18-23 mm, which increased the resistance to rainfall and ability to grow plant on the surface for improved soil.

Construction of a Pure Cryparin-null Mutant for the Promoter Analysis of Cryparin Gene (Cryparin 유전자의 promoter 분석을 위한 cryparin 유전자 치환체의 순수 제조)

  • The cryparin of Cryphonectria parasitica belongs to a cell wall associated fungal hydrophobin. The cryparin, though it is encoded by a single copy gene, is known for the high expression during the liquid culture of C. parasitica, and it turns out that 22% of total mRNA was transcribed for cryparin at 48hr after the liquid culture. In addition, it is also known as one of down-regulated fungal proteins by the presence of double stranded RNA virus, Cryphonectria hypovirus 1. In previous studies (Kim et al., 1999), we have constructed a cryparin-null mutant by replacing the cryparin gene with hygromycin B resistance gene due to site directed homologous recombination. In order for the promoter analysis of cryparin which seems to be very strong as well as mycoviral specific, it is preferable to have a strain with only a target promoter replaced and a discernable target site for incoming vectors. However, the cryparin-null mutant revealed the presence of an additional copy of transforming vector except the one which replaced the cryparin gene. In addition, the cryparin-null mutant did not contain any markers for targeted integration of incoming vectors. This prompts us to design an experiment to obtain a strain for promoter analysis of cryparin gene. A different mating type strain EP6(Mata, $met^-$) was mated with the cryparin-null mutant ${\triangle}$Crp194-7(MatA, Crp${\triangle}$::hph) to make the progenies with only a single replacement vector and $met^-$ characteristic remained. Nutritional assay as well as Southern blot analysis revealed that the progeny, ${\triangle}$Crp194-a6, was the methionine auxotroph with a single replacing vector in genome. Northern blot analysis and PAGE showed that there was no cryparin produced in this bred strain either.

Co-expression of MDRI and HLA-B7 Genes in a Mammalian Cell Using a Retrovirus

  • Using a retrovirus, foreign genes can be introduced into mammalian cells. The purpose of this study is to produce a retrovirus that can make the infected cells express two genes; the human multidrug resistance gene (MDR1) and the HLA-B7 gene, which is one of the major human histocompatibility complex (MHC) class I genes. For the expression of these genes, the internal ribosome entry site (IRES) was used, which was derived from the encephalomyocarditis (EMC) virus. In order to produce retroviruses, a retroviral vector was transfected into a packaging cell line and the transfected cells were treated with vincristine, which is an anti-cancer drug and a substrate for the MDRI gene product. This study revealed that two genes were incorporated into chromosomes of selected cells and expressed in the same cells. The production of the retrovirus was confirmed by the reverse transcription (RT)-PCR of the viral RNA. The retrovirus that was produced infected mouse fibroblast cells as well as the human U937. This study showed that packaging cells produced the retroviruses, which can infect the target cells. Once the conditions for the high infectivity of retrovirus into human cells are optimized, thus virus will be used to infect hematopoietic stem cells to co-express MDRl and HLA-B7 genes, and develop the lymphocytes that can be used for the immnogene therapy.

Enhancement of antimicrobial peptide genes expression in Cactus mutated Bombyx mori cells by CRISPR/Cas9

  • CRISPR/Cas9 gene editing system is an efficient method to mutation in a sequence specific manner. Here we report the direct transfection of the Cas9 nuclease and gene specific guide RNA can be used in BM-N cell line derived from Bombyx mori ovarian tissue to enfeeble function of endogenous gene in vitro. We have used gene editing system to negative regulation components of major signaling cascade, the Toll pathway, which controls B. mori resistance to microbe infections, such as fungi and gram positive bacteria. We demonstrate that the $I{\kappa}B-like$ protein Cactus may controls the activation of transcription factors such as Rel A and Rel B. The direct transfection of Cas9 nuclease and Cactus-specific guide-RNA complex may be used in BM-N cells to disrupt the function of endogenous genes in vitro. A mutation frequency of 30-40% was observed in the transfected cells, and various mutations caused the target region. Moreover, RT-PCR analysis revealed that Cactus gene was down regulated after these mutations. More importantly, mutation of BmCactus stimulated expression of lysozyme, moricin, and lebocin genes. These results suggest that the CRISPR/Cas9 systems are expected to efficiently induce site-specific mutations and it was possible to produce antimicrobial peptide through the gene editing.
