Objectives: Nail has been a substitute DNA source for genotyping. To investigate the integrity and consistency of nail DNA amplification for biomarker study, nail clippings from 12 subjects were collected at monthly intervals. The possibility of longer amplification and existence of GAPDH RNA/protein, were also investigated with three nail samples. Methods: Three primer sets were designed for quantitative amplification of nuclear and mitochondrial genes and analysis of their consistency. The mean threshold cycles in amplification of the target genes were compared to test the consistency of polymerase chain reaction (PCR) performance among individual factors including age groups, sex, family, the nail source, and by the size of the amplification segments. Results: The amplification of the target genes from nail DNA showed similar integrity and consistency between the nail sources, and among the serial collections. However, nail DNA from those in their forties showed earlier threshold cycles in amplification than those in their teens or seventies. Mitochondrial DNA (mtDNA) showed better DNA integrity and consistency in amplification of all three targets than did nuclear DNA (nucDNA). Over 9 kb of mtDNA was successfully amplified, and nested quantitative PCR showed reliable copy numbers (%) between the two loci. Reverse transcription PCR for mRNA and immunoblotting for GAPDH protein successfully reflected their corresponding amounts. Regarding the existence of RNA and protein in nails, more effective extraction and detection methods need to be set up to validate the feasibility in biomarker study. Conclusions: Nail DNA might be a feasible intra-individual monitoring biomarker. Considering integrity and consistency in target amplification, mtDNA would be a better target for biomarker research than nucDNA.
Interactions such as mergers and flybys play a fundamental role in shaping galaxy morphology. We used the Horizon Run 4 cosmological N-body simulations to study the frequency and the type of halo interactions as a function of the environment, the separation p, the mass ratio q, and the target halo mass. We defined targets as haloes more massive than 10^11 Msun/h, and a target is interacting if it is located within the virial radius of a neighbour halo more massive than 0.4 times the target mass. We find that the interaction rate as a function of time has a universal shape for different halo mass and large-scale density, with an increase and saturation. Larger density yield steeper slopes and larger final interaction rates, while larger masses saturate later. Most interactions happen at large-scale density contrast ${\delta}$ about 10^3, regardless of the redshift. We also report the existence of two modes of interactions in the (p,q) plane, reflecting the nature (satellite or main halo) of the target halo. These two trends strongly evolve with redshift, target mass, and large-scale density. Interacting pairs have similar spins parameters and aligned spins, with radial trajectories, and prograde encounters for non-radial trajectories. The satellite trajectories become less and less radial as time proceed. This effect is stronger for higher-mass target, but independent of the large-scale density.
두 가지 고려사항을 통해 단일 표적 추적 자료결합 기법을 다수 표적 추적 필터로 확장하여 사용하며, 대표적인 알고리듬으로 JIPDAF(Joint Integrated Probabilistic Data Association Filter)와 LM-IPDAF(Linear Multi-target IPDAF)가 있다.
In this paper, we consider a problem of partitioning a search area into smaller rectangular regions, so that multiple platforms can conduct search operations independently without requiring unnecessary coordination among themselves. The search area consists of cells where each cell has some prior information regarding the probability of target existence. The detection probability in particular cell is evaluated by multiplying the observation probability of the platform and the target existence probability in that cell. The total detection probability within the search area is defined as the cumulative detection probability for each cell. However, since this search area partitioning problem is NP-Hard, we decompose the problem into three sequential phases to solve this computationally intractable problem. Additionally, we discuss a special case of this problem, which can provide an optimal analytic solution. We also examine the performance of the proposed approach by comparing our results with the optimal analytic solution.
This article proposes algorithms for the automatic initiation of the tracks of maneuvering targets in cluttered environments. These track initiation algorithms consist of IPDA-AI(Integrated Probabilistic Data Association-Amplitude Information) and MPDA(Most Probable Data Association) in an Interacting Multiple Model(IMM) configuration, and they are referred to as the IMM-IPDAF-AI and IMM-MPDA respectively. The IMM portion consists of several filters based on different dynamical models to handle target maneuvers. Each of the filters utilizes an IPDA-AI(or MPDA) algorithm to deal with the problem of track existence in the presence of clutter. Although the primary purpose of this study is to deal with the track initiation problem, the IMM-IPDAF-AI and IMM-MPDA can also be used for the maintenance of existing tracks and the termination of tracks for targets when they disappear. For illustrative purposes, simulation is used to compare the performance of the algorithms proposed to other track formation algorithms.
In this paper, we propose a new tracking algorithm which uses weighted sum of color bin's centroids to find the main centroid of the target. The weights are determined by the proportion of colors of the target and by the colors of background. That is, A color which has high occupation in forming the target is highly weighted and a color which has low occupation is lowly weighted. Moreover, the proposed algorithm prevent track failure by lowering the weight of the colors which forms the background. Therefore, the proposed algorithm performs stable tracking inspite of occlusion and existence of confusing backgrounds.
Homeodomain (HD) is a highly conserved DNA-binding domain composed of helix-turn-helix motif. Drosophila Vnd (Ventral nervous system defective) containing HD acts as a regulator to either enhance or suppress gene expression upon binding to its target sequence. In this study, kinetic analysis of Vnd binding to DNA was performed. The result demonstrates that DNA-binding affinity of the recombinant protein containing HD and NK2-specific domain (NK2-SD) was higher than that of the full-length Vnd. To access whether phosphorylation sites within HD and NK2-SD affect the interaction of the protein with the target sequence, alanine substitutions were introduced. The result shows that S631A mutation within NK2-SD does not contribute significantly to the DNA-binding affinity. However, S571A and T600A mutations within HD showed lower affinity for DNA binding. In addition, DNA-binding analysis using embryonic nuclear protein also demonstrates that Vnd interacts with other nuclear proteins, suggesting the existence of Vnd as a complex.
Achieving a target operational availability is more economical and efficient than having many quantities of the weapon system, since the cost of weapon system becomes expensive. The intent of this study is twofold; first, we develop the simulation model to determine the optimal inventory level of the maintenance float while achieving a target operational availability of the Korean-made helicopter. The quantity decision model considers following factors such as a reliability. a turn around time(TAT). a protection level for inventory, and so on. Second, we analyze whether the existence of a lateral transshipment among bases and the reduction of TAT relate to an inventory level and the operational availability. The research result shows that both TAT and lateral transshipment have an effect on reducing the inventory level of the maintenance float and improving an operational availability.

The aim of this paper is to present theorems on the exitence of zeros for mappings defined on convex subsets of topological vector spaces with values in a vector space. In addition to natural assumptions of continuity, convexity, and compactness, the mappings are subject to some geometric conditions. In the first theorem, the mapping satisfies a "Darboux-type" property expressed in terms of an auxiliary numerical function. Typically, this functions is, in this case, related to an order structure on the target space. We derive an existence theorem for "obtuse" quasiconvex mappings with values in an ordered vector space. In the second theorem, we prove the existence of a "common zero" for an arbitrary (not necessarily countable) family of mappings satisfying a general "inwardness" condition againg expressed in terms of numerical functions (these numerical functions could be duality pairings (more generally, bilinear forms)). Our inwardness condition encompasses classical inwardness conditions of Leray-Schauder, Altman, or Bergman-Halpern types.
