Investigation of Microbial Contamination in Semisulcospira libertine and Evaluation of Its Reduction Effects by Sediment Removal Treatment (다슬기(Semisulcospira libertine)의 미생물 오염도 평가 및 해감 제거공정에 따른 저감화 효과)

  • In this study, microbial contamivation semisulcospira libertine and effect of sedimentation treatment of major bacterial and fungal pathogens were investigated. The total aerobic bacteria, coliforms, Escherichia coli, Staphylococcus aureus, and yeast and mold present in raw and water-dipped Semisulcospira libertine were enumerated using the standard plate count methods on using the standard plate method on potato dextrose agar (PDA), 3M Petrifilm for coliforms / E. coli, 3M Petrifilm for S. aureus, and plate count agar (PCA), respectively. In analysis of microbial contamination of raw Semisulcospira libertine, the total aerobic bacteria, coliforms, and yeast and mold were monitored as 6.40, 2.70, and $6.79{\log}_{10}CFU/g$, respectively. Both E. coli and S. aureus were not detected (detection limit: 10 CFU/g). However, Semisulcospira libertine dipped in ground water for 3 hours had higher contamination levels of all natural indigenous microorganisms than raw Semisulcospira libertine. Especially, E. coli was detected as $2.46{\log}_{10}CFU/g$ in the ground water-dipped Semisulcospira libertine. The total aerobic bacteria in the ground water-dipped Semisulcospira libertine was not significantly reduced (p>0.05) compared to that in the raw Semisulcospira libertine. Moreover, coliforms were significantly increased (p>0.05) in all water-dipped Semisulcospira libertine. Only fungi were slightly reduced (less than 0.2 log) (p>0.05) in the tap water-dipped Semisulcospira libertine by comparison with the raw Semisulcospira libertine. The results of this study suggest that the use of chemical sterilizing agents and other physical methods in the washing stage will be necessary for the microbial reduction in raw Semisulcospira libertine because the use of sediment removal treatment by ground or tap water did not affect the microbiological safety of the raw Semisulcospira libertine.

  • This study was conducted to investigate the effect of barley tea with roasted grains and barley tea with a tea bag on the reduction of chlorination by-product(CBP) levels in chlorinated drinking water. Since the concentrations of six volatile compounds of eight CBPs were blow their respective detection limits after 10 minute heating, two nonvolatile CBPs dichloroacetic acid and trichloroacetic acid, and total chlorine were compared between tap water and two kinds of barley tea. No significant differences were observed in the relative changes of the amounts of the above three items, and new peaks which were not found in the original water appeared in the chromatograms of gas chromatograph/electron capture detector(GC/ECD). Thirty three organic compounds were identified in the extracts of barley tea with roasted grains, barley tea with a tea bag, corn tea, and Cassia tora seed tea which were prepared with distilled/deionized water, using gas chromatography/mass selective detection(GC/MSD). Exclusive of fatty acids, most of the compounds were aromatic compounds such as phenols, furans, and pyrroles.

Effects of Ozonated Water Treatment on Pesticide Residues and Catechin Content in Green Tea Leaves (녹차의 잔류농약과 카테친 함량에 미치는 오존수 처리 효과)

  • This study examined the effects of treating green tea leaves with ozonated water by evaluating pesticide residue levels and catechin content. The pesticide residue levels of tea leaves treated with carbendazim, captain, diazinon, fenthim, dichlorvos, and chlorpyrifos ranged from 43.2 to 48.2 ppm. For leaves treated by soaking or watering with tap water, or with 0.25 ppm of ozone water for 30 min. Pesticide residue levels were reduced by 24.0-30.2%, 30.3-33.6%, 52.4-70.5%, and 65.5-80.2%, respectively. No major differences in catechin content were observed in the leaves according to the soaking and rinsing treatments using ozonated or tap water.

Health Risk Assessment of Disinfection By-products by Chlorination in Tap Water Ingestion (수도수중 염소 소독부산물로 인한 건강위해성 평가에 관한 연구 - 서울시 수도수중 Trihalomethanes 및 Haloaceticnitriles을 중심으로 -)

  • Public concerns about hazardous health effect from the exposure to organic by-products of the chlorination have been increased. There are numerous studies reporting that chlorination of drinking water produces numerous chlorinated organic by-products including THMs, HAAs, HANs. Some of these products are known to be animal carcinogens. The purpose of this study was to estimate health risk of DBPs by chlorinated drinking water ingestion in Seoul based on methodologies that have been developed for conducting risk assessment of complex-chemical-mixture. The drinking water sample was collected seperately at six water treatment plant in Seoul at March, April, 1996. In tap water of households in Seoul, DBPs were measured wilfh the mean value of 36.6 $\mu$g/L. Risk assessment processes,. which include processes for the estimation of human cancer potency using animal bioassay data and calculation of human exposure, entail uncertainties. In the exposure assessment process, exposure scenarios with various assumptions could affect the exposure amount and excess cancer risk. The reference dose of haloacetonitriles was estimated to be 0.0023 mg/kg/day by applying dibromoacetonitrile NOAEL and uncertainty factor to the mean concentration. In the first case, human excess cancer risk was estimated by the US EPA method used to set the MCL (maximum contaminant level). In the second and third case, the risk was estimated for multi-route exposure with and without adopting Monte-Carlo simulation, respectively. In the second case, exposure input parameters and cancer potencies used probability distributions, and in the third case, those values used point estimates (mean, and maximum or 95% upper-bound value). As a result, while the excess cancer risk estimated by US EPA method considering only direct ingestion tended to be underestimated, the risk which was estimated by considering multi-route exposure without Monte-Carlo simulation and then using the maximum or 95% upper-bound value as input parameters tended to be overestimated. In risk assessment for Trihalomethanes, considering multi-route exposure with adopting Monte-Carlo analysis seems to provide the most reasonable estimations.

Effects of ultrasonic thawing on the physicochemical properties of frozen pork (주파수에 따른 초음파 해동처리가 냉동 돈육의 이화학적 특성에 미치는 효과)

  • This study was carried out to investigate the effects of ultrasonic thawing on physicochemical characteristics of frozen pork sirloin. To determine the velocity of an ultrasonic thawing machine, $-80^{\circ}C$ frozen pork sirloins were thawed using 132, 580, and 1,000 kHz ultrasonic thawing machines, and the physicochemical properties of pork sirloin thawed with $15^{\circ}C$ tap water and those using the ultrasonic thawing machines were compared. As a result, thawing speed by ultrasonic thawing was three times faster than that of tap water, but drip weight loss rate is increased by about 5% during ultrasonic thawing compared to that during thawing in tap water. However, biochemical properties, such as pH, volatile basic nitrogen, thiobarbituric acid, and total aerobic bacteria, were not improved by ultrasonic thawing. Hunter values, such as total color change and redness, improved slightly after 580 kHz ultrasonic thawing. The hardness and chewiness of the pork sirloin decreased significantly after 580 kHz ultrasonic thawing; however, springiness was not changed. These results indicate that ultrasonic thawing is faster than thawing in tap water, but the physicochemical characteristics of frozen pork sirloin was not significantly improve by ultrasonic sound.

LAND FARMING OF WATER PLANT ALUM SLUDGE ON ACID MINERAL SOIL AFFECTED BY ACID WATER

  • An acid forest surface soil as a land farming medium was treated with a water plant alum sludge at 0 to 18%. Indian mustard was grown in the treated soil in a greenhouse for 5 weeks and watered with pH 4 tap water adjusted with a mixed acid (1HNO$_3$: 2H$_2$SO$_4$) during plant growth. Changes in soil property, leachate chemistry, plant growth, and plant uptake of elements by the sludge treatment were determined. The alum sludge treatment increased buffer capacity to acidity, hydraulic conductivity, water holding capacity, and phosphate adsorption of the soil and decreased bulk density and mobility of small particles. The sludge treatment reduced leaching of Al, Mg, K, Na, and root elongation. Plant did uptake less amount of the cations and P but more Ca with the sludge treatment.

Detection of Trace Copper Metal at Carbon Nanotube Based Electrodes Using Squarewave Anodic Stripping Voltammetry

  • We investigate sensitivity and limit of detection (LOD) of trace copper (Cu) metal using pristine carbon nanotube (CNT) and acidified CNT (ACNT) electrodes. Squarewave based anodic stripping voltammetry (SWASV) is used to determine the stripped Cu concentration. Prior to performing the SWASV measurements, its optimal conditions are determined and with that, effects of potential scan rate and $Cu^{2+}$ concentration on stripping current are evaluated. The measurements indicate that (1) ACNT electrode shows better results than CNT electrode and (2) stripping is controlled by surface reaction. In the given $Cu^{2+}$ concentration range of 25-150 ppb, peak stripping current has linearity with $Cu^{2+}$ concentration. Quantitatively, sensitivity and LOD of Cu in ACNT electrode are 9.36 ${\mu}A\;{\mu}M^{-1}$ and 3 ppb, while their values are 3.99 ${\mu}A\;{\mu}M^{-1}$ and 3 ppb with CNT electrode. We evaluate the effect of three different water solutions (deionized water, tap water and river water) on stripping current and the confirm types of water don't affect the sensitivity of Cu. It turns out by optical inspection and cyclic voltammetry that superiority of ACNT electrode to CNT electrode is attributed to exfoliation of CNT bundles and improved interfacial adhesion occurring during oxidation of CNTs.

Influence of Pipe Materials on Corrosion and Bacteria Regrowth in A Model Home Plumbing System (급수관에서 관재에 따른 부식특성과 미생물 재성장 고찰)

  • This study performed in order to evaluate the effects of pipe materials on corrosion and bacteria regrowth using a laboratory scale batch test. Two varieties of feed water with different microbial conditions were selected: tap water, surface river water (Han River water), and five pipe materials; carbon steel, copper, galvanized iron, stainless steel, and PVC, Carbon steel and galvanized iron pipes showed higher corrosion rates than other materials. In terms of attached bacterial growth, pipes with PVC and stainless steel showed higher bacteria concentration compared to other materials. Pseudomonas vesicularis was the predominant bacteria found on biofilm. The behavior of bacterial growth in the pipes was observed using a scanning electron microscope.

  • The purpose of this study was to estimate the microbial quality of some raw vegetables, and suggest a safer method for their sanitization and pre-preparation in foodservice operations. Baechu-geotjeori was monitored from the ingredient to the final product, during different holding temperature. Three sanitization methods were performed during the pre-preparation (tap water, chlorine water and electrolyzed water). The largest reduction in the microbial counts was shown with the electrolyzed water pre-preparation (after treatment; the total plate counts decreased to 3.34-4.06 Log CFU/g, coliform counts decreased to 1.40-1.45 CFU/g). Prior to immersion in the chlorine water, washing was first peformed to see if the was a larger effective reduction in the microbial counts.

  • A series of laboratory experiments carried out to investigate the particle separation efficiency and flotation characteristics using $CO_2$ bubbles. The primary objective of this study was to find out the feasibility of $CO_2$ bubbles as an applicable unit of flotation process in tap-water and wastewater treatment plant. The fundamental measurements were conducted to characterize the $CO_2$ bubble from the physical viewpoint in water including bubble size distribution and rising velocity under various operational conditions. In addition, the removal efficiency of solid was experimented using the lab scale plant applied $CO_2$ bubbles, namely the dissolved carbon dioxide flotation (DCF) process. The DCF process using carbon dioxide bubble, which is an advantage as the decrease and the reuse of Green-House gas, can be a promising technology as an water treatment process. On the other hand, the further research to decrease the bubble size distribution of $CO_2$ is required to enhance the particle separation efficiency.
