This study was conducted to determine the inheritance of duvatrienediol(DVTs) and cis-abienol concentration from flesh tobacco leaves. NC744, which has a very low levels of DVTs and no cis-abienol was used as a parent in cross to TI 1068 producing a normal amount of DVTs and cis-abienol. Presence of DVTs and cis-abienol on the leaf surface was determined using thin layer chromatography(TLC). Segregation pattern from F$_2$ and BC$_2$[ (TI 1068 $\times$ NC744) $\times$ NC744J generations revealed that TI 1068 have a single dominant gene controlling DVTs and cis-abienol production. And DVTs production was inherited independently of the ability to produce cis-abienol.
This study was conducted to obtain basic information for varietal difference of leaf breakdown in field of flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1987. The experiment was designed in randomized block with 3 replications, data ware analysed as Split split-plot design. Main plots were varieties, sub-plots were leaf positions, 4, 5 and 6th from bottom, and each sub-plot was divided into 3 parts with distance of midrib, 7 10 and 13em from stalk. Four varieties, NC 95, NC 2326, NC 82 and BY 4 were transplanted in 15 April, and measured the weight of leaf breakdown by artificially weighted in 5 June. The results obtained are as follows : 1. Weights of leaf breakdown according to leaf position, NC 95 were lower as 358-5799 than those of other varieties as 555-597g, were not significantly different regardless of varieties. 2. Weights of leaf breakdown in relation to distance of midrib from stack, NC 95 were lower as 309-419g than those of other varieties as 472-710g. 3. Weights of leaf breakdown were significantly different according to distance of midrib from stalk, and not significantly different according to leaf position under the same distance of midrib from stalk regardless of varieties.
Dihaploid lines were derived from the anthers of the F, of single cross, "Va 115" x"SC 72" (Nicotiana tabacum L.). The path-analysis, correlation coefficient, and variation of characteristics among dihaploid families were evaluated. Dihaploid lines displayed much greater variation than their parents. The genetic correlation coefficient between yield and total alkaloids of dihaploids was significantly lower than that of conventional varieties and lines. In path-analysis which relates yield components to yield, number of leaves harvested and leaf width had a direct effect on yield. a direct effect on yield.
There was no significant difference in length and width of leaf and number of leaves per plant between tissue-cultured plants and conventionally propagated ones but chlorophyll content increased in tissue-cultured ones. Percent of sprouting from planted root segments significantly increased in tissue-cultured plants, resulting in yield increase of more than 200% per 10a. Root thickness of tissue-cultured plants at the time of planting influenced percent of sprouting and yield. Plants with root diameter ranging from 3 to 6mm gave good yield. When virus infection was monitored with N. tabacum and C. amaranticolor as indicator plants, 100% infection occurred in vegetatively propagated plants and introduced plants from China. whereas plants obtained from apical meristem showed 0% and 40% to 45% infection in vitro plantlets and 1 year old plants in vivo, respectively. Tobamovirus and unidentified virus particles were detected in electron microscopy.
Narrow and broad sense heritabilities for the characters ranged from 0.1618 to 0.6914 and from 0.7494 to 0.9357, respectively, in F$_1$ hybrids of $5{\times}5$ full diallel crosses. Plant height and days to flowering had the highest heritabilities and yield and quality the lowest. Partial dominance was exhibited by plant height, number of leaves, leaf area per plant and days to flowering. Over dorminance was detected by leaf width and yield of cured leaf. Leaf length and quality would be considered being controlled by a complete dominance effect.
To enhance the growth of cryopreserved cells of transgenic Nicotiana tabacum, Pluronic F-68 was supplemented in a recovery medium during post-thaw period. As cryoprotective agents, 1 M sucrose, 0.5 M glycerol and 0.5 M dimethyl sulfoxide (DMSO) were added before freezing steps. The post-thaw growth of the cells was improved with Pluronic F-68, ranged from 0.1 to 10 g/L. The interactions of Pluronic F-68 with the cells were confirmed by the changes of hydrophobicity or permeability of the cells. Pluronic F-68 did not show any effect on the activity of $\beta$-glucuronidase (GUS) in all treatments. Therefore, the addition of Pluronic F-68 in a recovery medium was found to be beneficial to enhance the post-thaw growth of cryopreserved transgenic tobacco cells without affecting the production of recombinant protein.
To investigate the function of chloroplast small HSP, transgenic tobacco plants (Nicotiana tabacum L. cv. Samsun) that constitutively overexpress the chloroplast small HSP (NtHSP21) from N. tabacum cv. Petit Havana SR1 were generated. Five homozygous lines of transformants showing different constitutive expression levels of the NtHSP21 were selected. To determine whether constitutive overexpression of NtHSP21 protein affects thermotolerance, wild-type and transformants were grown in Petri dishes, heat-stressed at 52$^{\circ}C$ for 45 min, and then incubated in normal growth condition. When heat-stressed wild-type plantlets were incubated at $25^{\circ}C$, leaf color gradually became white and all trio plantlets finally died within a week. As for the transformants, however, more than 70% of them remained green and survived under the conditions in which all the wild-type plants were dying. It was also found that the levels of NtHSP21 were correlated with the degree of thermotolerance. These results suggest that the NtHSP21 protein in transformants is responsible for the increase in thermotolerance.

The leaf blade-segments of albino tobacco (Nicotiana tabacum L.) were cultured on MS media containing different concentrations of BAP (0, 0.4, 2.2, 4.4, 22.2 ${\mu}{\textrm}{m}$) with or without NAA (0, 0.5, 2.7 ${\mu}{\textrm}{m}$). Multiple shoots were induced on the media containing 0.4 to 2.2 ${\mu}{\textrm}{m}$ BAP. The best condition for multiple shoot induction with root formation was MS media containing 4.4 ${\mu}{\textrm}{m}$ BAP and 0.5 ${\mu}{\textrm}{m}$ NAA. The regenerated albino plants showed a significant reduction in accumulation of chlorophylls and carotenoids. The drastic reduction of the pigments content was associated with the distinct alterations in gene expression in the albino plants. firstly, the expression of plastid genes, such as rbcL, psbA, 165 rDNA and 235 rDNA, was reduced at the level of transcripts in the regenerated albino plants. Secondly, the alteration of structure of the plastid genes was not detected in the albino plants. However, the copy number of the plastid genes whose transcription level was reduced greatly was increased approximately two-fold, although the transcriptions of nuclear gene (255 rDNA) showed the wild-type level.

To enhance the production of heavy chain immunoglobulin G (HC IgG) in the suspension cultures of transgenic tobacco cells (Nicotiana tabacum), the effects of adding various cytokinins (i.e., growth regulators) and organic nitrogen sources to culture media were investigated. Four different cytokinins including kinetin, isopentenyladenine (IPA), 6-benzylaminepurine (BA), and zeatin were tested with or without dichlorophenoxyacetic acid (2,4-D), which is a typical growth regulator supplemented in the standard Murashige and Skoog (MS) medium. The productivity of intracellular HC IgG was increased by 36 and $42\%$, compared to the control, especially when IPA (2 mg/l) or BA (0.2 mg/l) was added to the media in the presence of 2,4-D, respectively. In the study of organic nitrogen sources, addition of each casein hydrolysate and tryptone to the culture media at a final concentration of 0.01 and 1 g/l, respectively. increased the productivity or he IgG as much as 68 and $67\%$, respectively, in comparison with the control, which was is MS medium without supplementation of any organic nitrogen sources. This study shows that the optimization of media composition could offer significant improvements in the production of foreign proteins in the suspension cultures of transgenic plants.
Effects of the heavy-ion ($^{14}$ N or $^{20}$ Ne) beam irradiation on the response of anthers, growth of calli, germination of seeds, and the early growth after the germination of tobacco (Nicotiana tabacum L. cv. BY-4) were investigated. Anthers precultured for 10 days before the irradiation became brown without callus or shoot induction over 20 Gy of $^{14}$ N and $^{20}$ Ne ion beam irradiation. Relative growth rate of the cultured calli was reduced by the irradiation and became brown significantly 2 weeks after the $^{14}$ N and $^{20}$ Ne ion beam irradiation over 50 Gy. The increased intensity of the heavy-ion ($^{14}$ N, $^{20}$ Ne) beam irradiation resulted in the delay of seed germination and the inhibition of the early growth both in water-treated and non-treated seeds before the irradiation. In addition, the heavy-ion beam irradiation to the imbibed seeds inhibited seed germination more than that to the non-imbibed seeds. The screening approach of non-imbibed seeds with heavy-ion beam irradiation using in vitro culture system was more useful than the filter-paper germination method in investigating the characteristics of heavy-ion beam-irradiated seed population and the screening of morphological variants at the early stage of the plant growth.
