The Distribution and Diversity of Freshwater Fishes in Korean Peninsula (한반도 담수어류의 분포와 다양성)

  • A recent topic of interest within ecology is ecosystem services that are importantly connected with biodiversity. Biodiversity is a fundamental component of ecological studies and has been the subject of many studies worldwide. However, studies on the diversity and distribution of freshwater fish were not undertaken in earnest until 2000, and the number of internationally available studies is therefore still limited. In this study, we used data from "The Survey and Evaluation of Aquatic Ecosystem Health (hereafter SEAEH)" within the framework of which whole water systems on the Korean peninsula are annually monitored to investigate and determine the current status of the diversity and distribution of freshwater fish, as well as to elucidate major factors impacting freshwater fish. A total of 130 species from 28 families were identified at around 953 sites in the SEAEH investigation of 2014. The species Zacco platypus (relative abundance: 28.2%) and Z. koreanus (19.3%) were identified as the most abundant species on the Korean peninsula, and besides these 20 endangered species, 51 endemic species and 4 exotic species were also collected. Highest (96 species) and lowest (72 species) diversity were identified in the Han River Watershed and the Yeongsan/Seomjin River Watershed, respectively. The mean endemic rate was 32.7%, which is higher than the previous rate determined in 1995. The exotic species Micropterus salmoides, Lepomis macrochirus and Carassius cuvieri were found in the whole river system except north Yeongdong, whereas Oreochromis niloticus occurred only at three sites due to the cold water temperatures in winter. Totally, 28 species were found to be translocated to different water systems not connected with known original habitats, most translocations occurring in the Nakdong River and the Yeongdong region. Among the translocated species, Opsariichthys uncirostris amurensis, and Erythroculter erythropterus were dominantly distributed in the mid- to lower reaches of Nakdong River where they may be harmful to the native fish assemblages due to their strong predation. The construction of weirs in connection with the "4 River Project" generated changes in fish assemblages before, during and after the project, resulting in altered habitat condition. Especially, a decrease in the number of endemic species and an increase in the abundance of lentic species were seen. Human-induced global warming will allow exotic species requiring high water temperatures to adapt to the environmental conditions of the Korean peninsula, which may enhance the diversity of native fishes and create changes in distribution. For the establishment of realistic and efficient management and protection strategies for Korean freshwater fish, SEAEH, which produces fundamental and quantitative data for Korean streams, is an important and necessary tool.

Characteristics of Phytoplankton Succession Based on the Functional Group in the Enclosed Culture System (대형 배양장치에서 기능그룹에 기초한 식물플랑크톤 천이 특성)

  • The present study was conducted from August to December 2016 in a cylindrical water tank with a diameter of 1 m, a height of 4 m and a capacity of 3,000 L. The field water and sediment from the Nakdong River were also sampled for the experimental culture (field water+sediment) and control culture (field water), respectively. In this study, we aimed to investigate phytoplankton succession pattern using the phytoplankton functional group in the enclosed culture system. A total of 50 species in 27 genera including Chlorophyceae (30 species), Bacillariophyceae (11 species), Cyanophyceae (7 species), and Cryptophyceae (2 species) were identified in the experimental and control culture systems. A total of 19 phytoplankton functional groups (PFGs) were identified, and these groups include B, C, D, F, G, H1, J, K, Lo, M, MP, N, P, S1, $T_B$, $W_0$, X1, X2 and Y. In particular, $W_0$, J and M groups exhibited the marked succession in the experimental culture system with higher biovolumes compared to those of the control culture system, which may be related to the internal cycling of nutrients by sediment in the experimental culture system. The principal component analyses demonstrated that succession patterns in PFG were associated with the main environmental factors such as nutrients(N, P), water temperature and light intensity in two culture systems. In conclusion, the present study showed the potential applicability of the functional group for understanding the adaptation strategies and ecological traits of the phytoplankton succession in the water bodies of Korea.

Assessing removal effects on particulate matters using artificial wetland modules (인공 습지 모형을 활용한 습지의 미세먼지 저감 효과)

  • To assess the wetland systems' capability to reduce fine dust, we used an artificial wetland module of small-sized greenhouse (70cm W × 70cm L × 60cm H) which creates a closed system. Experiment was performed twice using four species in each experiment. Non-plantation, one species, or two species condition was created in each mesocosm. We measured air quality, primarily PM2.5 and PM10 at the initial open mesocosms and 1hr later since mesocosms were closed. The dry weight of vegetation was measured at the 2nd experiment. The decreased amount of PM2.5 and PM10 was 13.7±1.3 and 13.2±1.3 ㎍·m-3hr-1 in wetland condition and 15.0±1.4 and 13.8±1.5 ㎍·m-3hr-1 in dryland condition, respectively. In 2nd experiment, the decreased amount of PM 2.5 and PM 10 in wetland condition was 13.7±1.3 and 9.2±1.5 ㎍·m-3hr-1, 15.0±1.4 and 8.8±1.4 ㎍·m-3hr-1 in dryland condition, respectively. Wetland showed higher removal effect due to its high productivity leading to more effective absorption of particulate matter. Furthermore, the aquatic characteristics of wetland system and high humidity helped purifying the air quality. This can be seen as another value of wetlands, which can be presented as one of the solutions to the problem of fine dust.

ESR-Spin Trapping Detection of Radical Center Formed on the Reaction of Metmyoglobin with Hydrogen Peroxide

  • The radical centers detected in the reaction of metmyoglobin (MetMb) with hydrogen peroxide ($H_2O_2$) have been studied by using a spin trapping technique. A broad 5-line asymmetric electron spin resonance (ESR) spectrum, with $2A_{max}=4.07\;mT$ and $2A_{min}=2.97\;mT$, obtained after incubation of MetMb with $H_2O_2$ in the presence of a spin trap, 5,5-dimethyl pyrroline-N-oxide (DMPO) was gradually weakened with time and disappeared completely by 6 min after addition of guanidine-HCl (14 M). When a higher concentration (6 M) of the agent was added, the signal disappeared within 40 see and the DMPO/OH signal appeared immediately. Then, a new 8-line signal with similar intensities grew gradually and was fixed by 45 min, coexisting with the DMPO/OH signal. This new signal was found to be composite, consisting of two different radical species. One of the 6-line signals, with $a_N$ 1.49 mT and $a_H$ 0.988 mT, was assigned to the DMPO/phenoxyl radical adduct. The second 6-line signal with $a_N$ 1.55 mT and $a_H$ 2.22 mT was assigned to carbon-centered radical adduct. When 3,3,5,5-tetramethylpyrrolin-N-oxide (TMPO), was employed in the place of DMPO, another broad asymmetric 5-line signal was detected with $2A_{max}=3.99\;mT$ and $2A_{min}=3.04\;mT$, which is virtually identical to that obtained from the DMPO system The shape of the spectrum of the TMPO adduct changed drastically, with lapse of time resulting in a broad singlet after 40 min. The broad singlet was assigned to the porphyrin radical adduct. Incubation of globin with Fenton reagent in the presence of DMPO initially gave a DMPO/OH signal. Then, a new 12-line signal began to grow after one minute and fixed after 15 min. coexisting with the DMPO/OH signal, This 12-line signal was assigned to DMPO/phenoxyl with $a_N$ 1.47 mT, $a_{{\beta}H}$ 0.99 mT and $a_{{\gamma}H}$ 0.13 mT. A minor concentration of carbon-centered radical adduct was also detected. This radical composition is identical to that of guanidine HCl treated MetMb/DMPO/$H_2O_2$ system, indicating that the radical producing conditions are somehow common in both systems. Heme iron can be released by excess $H_2O_2$ in the MetMb/$H_2O_2$ system, providing for Fenton reagent. When MetMb was pretreated with tyrosine blocking agent, $KI_3$ the broad 5.line MetMb-derived signal was not detected in the MetMb/DMPO/$H_2O_2$ system, whereas no such effect was detected on such system of Hb in which the radical center was assigned to cysteine residue not tyrosine, indicating that tyrosine residue is a main radical center produced in the MetMb/$H_2O_2$ system Thus, the present data strongly support the previous indication that the apomyoglobin-derived radical center formed in the reaction of MetMb with $H_2O_2$ is a tyrosine residue.

Virus Disease Incidences and Transmission Ecology of Oriental Melons in Seongju Area (성주지역 참외 바이러스병의 발생실태와 전염생태)

  • Throughout the years 2008 to 2010, we analyzed approximately two thousand oriental melon samples collected from Seongju, using electron microscopy and testing by RT-PCR using primers specific for eight cucurbit-infecting viruses. Data from RT-PCR indicated that Cucumber green mottle mosaic virus (CGMMV), Watermelon mosaic virus 2 (WMV2) and Zucchini yellow mosaic virus (ZYMV) were present and the other viruses were not detected. Among them, CGMMV and WMV2 were the most prevalent pathogens. CGMMV was thought to infect oriental melon from the early growing season, and reached nearly 100% in the later of growing period. Otherwise, WMV2 emerged from June, several months later compared to CGMMV. CGMMV was detected from all aerial parts of the oriental melon including seeds, but not from the roots of the grafted pumpkin rootstock. Seed of two out of five commercial varieties were shown to be CGMMV positive. Nine varieties of pumpkins used as rootstocks were not infected with CGMMV. When the seedlings of grafted oriental melon were transplanted into pots mixed with the oriental melon debris infected with CGMMV, they were not infected by CGMMV. Cutting of pruning shear and the contact of tendrils contributed 48% and 30% to the transmission of the virus, respectively.

Beliefs About Gifted Education and Classroom Practices of the Science Teachers at Science Academy in Korea (과학영재학교 과학교사들의 영재교육에 대한 신념과 교수활동 유형)

  • The most important factor in providing education to gifted students as well as to students in general are the teachers themselves. However, at present in Korea, most of the teachers in charge of education for the gifted are educated by in-service training programs only for a short period of time. It is doubtful whether the teachers, who have taught ordinary students in general, can teach gifted students effectively only after completing such a short course. This research investigated the relationship between the teachers' beliefs about educating the gifted and the teachers' classroom practices in a Science Academy through case studies. The guiding questions for this study are as follows: First, what beliefs do the participating teachers have about education for the gifted? Second, how are the participants' beliefs reflected in their classroom practices? Of the five participants, two are physics teachers, two are biology teachers, and one is an earth science teacher. I observed and videotaped four classroom practices for each participant and conducted an in-depth interview with each participant. Further data were collected through e-mails with the participants. All data were carefully transcribed and analyzed. The results are as follows: Beliefs about education for the gifted do not exist independently, and form a belief system connecting with beliefs about teaching and learning, and subject matter. And the belief systems of participants can be divided into "student-centered," "teacher-centered," and "conflict chaos." In the classes of the participants who have "student-centered" belief system, students' questions or opinions played an important role and the participation structure in the classroom was determined by the students. On the contrary, participants who have "teacher-centered" belief system focused on teaching contents as much as possible in their classes. These teachers played a heavy role and formed a participation structure where students depended on their teacher's intellectual authority and therefore participated in their class passively. A participant who have "conflict chaos" belief did not form a firm belief system yet, and traditional beliefs about teaching and learning were reflected a lot in her classes. The research results imply teachers' beliefs play an important role in classroom practices and beliefs about teaching and learning and subject matter as well as beliefs about education for the gifted are important factors for teachers who guide gifted students. Additionally, I make some suggestions for the improvement of teacher education for the gifted.

First Report of the Virus Diseases in Victory Onion (Allium victorialis var. platyphyllum) (산마늘(Allium victorialis var. platyphyllum)에서 바이러스병의 최초보고)

  • In 2005, a survey was conducted to identify virus diseases on victory onion, Allium victorialis var. platyphyllum grown in Ulleung island located in the East Sea. A total of 61 samples were collected from victory onion in the neighborhood of Seonginbong. The identification of viruses from the samples were carried out by electron microscopy and RT-PCR using primers species specific to GCLV, LYSV, SLV, OYDV and genus specific to Allexivirus, respectively. From sixty-one samples, filamentous rod particles (600-900 nm) were detected from four victory onion samples in EM, three samples containing SLV and one sample containing both SLV and Allexivirus in RT-PCR analysis, respectively. Victory onions naturally infected by the viruses were asymptomatic apparently. The viruses detected by RT-PCR were further characterized by the nucleotide sequence analysis of the coat protein region. Three isolates of SLV showed approximately 99% identities in the nucleotide and amino acid sequences, suggesting that they were likely to be the same strain. On the other hand, they showed approximately 75.7~83.7% identities in the nucleotide and 89.2~97.0% in amino acid sequences compared with the previously reported SLV isolates in Allium. The CP gene of the Allexivirus showed approximately 99.2% nucleotide identities and 98.8% amino acid identities with Garlic virus A. However, there was relatively low homology ranging from 60.6% to 81.5% compared with other Allexiviruses (GarV-C, GarV-E, GarV-X, GMbMV, and Shal-X). These data suggested that two viruses, SLV and GarV-A identified from victory onion, are named SLV-Ulleungdo and GarV-A-Ulleungdo, respectively. This is the first report of viruses infecting victory onion.

Comparative Functional Analysis of the Malate Dehydrogenase(Mor2) during in vitro Maturation of the Mouse and Porcine Oocytes (체외성숙 과정 중 생쥐와 돼지 난자의 Malate Dehydrogenase(Mor2)의 기능에 대한 비교 분석)

  • Contrast to mouse where its in vitro maturation rates are high without specific supplements or presence of the cumulus cells, there are some species, such as porcine, where its in vitro oocyte maturation rates are still very low. This comparative study was conducted to investigate the role of malate dehydrogenase(Mor2) during oocyte maturation by RNAi in the mouse and porcine. The Mor2 double-stranded RNA(dsRNA) was prepared speciesspecifically and microinjected into the cytoplasm of denuded germinal vesicle(GV) oocytes. Oocytes were cultured for 48 h(porcine) and 16 h(mouse) in M199 with 10% porcine follicular fluid, pyruvate, p-FSH, EGF, cystein, and estradiol-$17{\beta}$. We measured changes in oocyte morphology, maturation rates and mRNA levels after Mor2 RNAi. We confirmed gene sequence-specific knock down of Mor2 mRNA in both species after Mor2 RNAi. In contrast to our previous finding that mMor2 RNAi resulted in GV arrest in the mouse, we found that pMor2 RNAi resulted in MI arrest in denuded porcine oocytes(58%), but developed to MII(84.4%) in COCs. To determine whether this difference between mouse and porcine RNAi is due to differences in culture media, we cultured mouse oocytes in the M199 media for 16 h after mMor2 RNAi. Mouse oocytes were developed to MII stage(62%) and there was no statistical difference compared to that of non-injected(76.8%) and buffer-injected(73.3%) control groups. Therefore, we concluded that the mouse and porcine oocytes are having different metabolic systems in relation to malate dehydrogenase for oocyte maturation. This could be a basis for differences in maturation rates in vitro in two species. Further scrutinized studies on the metabolic pathways would led us in finding better culture system to improve oocyte maturation rates in vitro, especially in more challenging species like the porcine.

Growth, Photosynthesis and Chlorophyll Fluorescence of Chinese Cabbage in Response to High Temperature (고온 스트레스에 대한 배추의 생장과 광합성 및 엽록소형광 반응)

  • In order to gain insight into the physiological responses of plants to high temperature stress, the effects of temperature on Chinese cabbage (Brassica campestris subsp. napus var. pekinensis cv. Detong) were investigated through analyses of photosynthesis and chlorophyll fluorescence under 3 different temperatures in the temperature gradient tunnel. Growth (leaf length and number of leaves) during the rosette stage was greater at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures than at ambient temperature. Photosynthetic $CO_2$ fixation rates of Chinese cabbage grown under the different temperatures did not differ significantly. However, dark respiration rate was significantly higher in the cabbage that developed under ambient temperature relative to elevated temperature. Furthermore, elevated growth temperature increased transpiration rate and stomatal conductance resulting in an overall decrease of water use efficiency. The chlorophyll a fluorescence transient was also considerably affected by high temperature stress; the fluorescence yield $F_J$, $F_I$, and $F_P$ decreased considerably at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, with induction of $F_K$ and decrease of $F_V/F_O$. The values of RC/CS, ABS/CS, TRo/CS, and ETo/CS decreased considerably, while DIo/CS increased with increased growth temperature. The symptoms of soft-rot disease were observed in the inner part of the cabbage heads after 7, 9, and/or 10 weeks of cultivation at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, but not in the cabbage heads growing at ambient temperature. These results show that Chinese cabbage could be negatively affected by high temperature under a future climate change scenario. Therefore, to maintain the high productivity and quality of Chinese cabbage, it may be necessary to develop new high temperature tolerant cultivars or to markedly improve cropping systems. In addition, it would be possible to use the non-invasive fluorescence parameters $F_O$, $F_V/F_M$, and $F_V/F_O$, as well as $F_K$, $M_O$, $S_M$, RC/CS, ETo/CS, $PI_{abs}$, and $SFI_{abs}$ (which were selected in this study), to quantitatively determine the physiological status of plants in response to high temperature stresses.

Impact of Rising Global Temperatures on Growth, Mineral Composition, and Photosynthesis in Radish in a Winter Cropping System (월동무의 생장, 무기성분 조성과 광합성에 미치는 온도 상승의 영향)

  • We investigated the effects of rising temperatures on the photosynthesis, mineral composition, and growth of radish (Raphanus sativus var. hortensis) in a winter cropping system using a temperature gradient tunnel to predict the impact of rising global temperatures. Vegetative growth, including shoot and root fresh and dry weights, shoot length, and root length and diameter, was high under elevated temperatures (ambient $+4^{\circ}C$ and $+7^{\circ}C$) compared with ambient temperature. At elevated temperatures, the N, P, Ca, Mg, and Fe contents were high in shoots, whereas in roots, the K, Ca, Mg, and Fe contents were high and the Cu content was low. The maximum photosynthetic rates ($22.1{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at ambient temperature $+4^{\circ}C$ and $22.9{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at ambient temperature $+7^{\circ}C$) at elevated temperatures were more than twice that ($9.7{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) at ambient temperature, whereas the water use efficiency was lower at elevated temperatures. These results suggest that rising global temperatures will lead to increased mineral absorption and photosynthesis in radish in winter cropping systems, subsequently favoring plant growth, although the water requirements will be high.
